Meta-Analysis for Sociology - A Measure-Driven Approach.

ABSTRACT: Meta-analytic methods are becoming increasingly important in sociological research. In this article we present an approach for meta-analysis which is especially helpful for sociologists. Conventional approaches to meta-analysis often prioritize "concept-driven" literature searches. However, in disciplines with high theoretical diversity, such as sociology, this search approach might constrain the researcher's ability to fully exploit the entire body of relevant work. We explicate a "measure-driven" approach, in which iterative searches and new computerized search techniques are used to increase the range of publications found (and thus the range of possible analyses) and to traverse time and disciplinary boundaries. We demonstrate this measure-driven search approach with two meta-analytic projects, examining the effects of various social variables on all-cause mortality.

Integrative OMICS Data-Driven Procedure Using a Derivatized Meta-Analysis Approach.

Project description:The wealth of high-throughput data has opened up new opportunities to analyze and describe biological processes at higher resolution, ultimately leading to a significant acceleration of scientific output using high-throughput data from the different omics layers and the generation of databases to store and report raw datasets. The great variability among the techniques and the heterogeneous methodologies used to produce this data have placed meta-analysis methods as one of the approaches of choice to correlate the resultant large-scale datasets from different research groups. Through multi-study meta-analyses, it is possible to generate results with greater statistical power compared to individual analyses. Gene signatures, biomarkers and pathways that provide new insights of a phenotype of interest have been identified by the analysis of large-scale datasets in several fields of science. However, despite all the efforts, a standardized regulation to report large-scale data and to identify the molecular targets and signaling networks is still lacking. Integrative analyses have also been introduced as complementation and augmentation for meta-analysis methodologies to generate novel hypotheses. Currently, there is no universal method established and the different methods available follow different purposes. Herein we describe a new unifying, scalable and straightforward methodology to meta-analyze different omics outputs, but also to integrate the significant outcomes into novel pathways describing biological processes of interest. The significance of using proper molecular identifiers is highlighted as well as the potential to further correlate molecules from different regulatory levels. To show the methodology's potential, a set of transcriptomic datasets are meta-analyzed as an example.

Sociology of Chronic Pain.

Project description:Chronic pain is a common, costly, and consequential health problem. However, despite some important analytic contributions, sociological research on pain has not yet coalesced into a unified subfield. We present three interrelated bodies of evidence and illustrative new empirical findings using 2010 to 2018 National Health Interview Survey data to argue that pain should have a central role in sociological investigations of health. Specifically, we contend that (1) pain is a sensitive barometer of population health and well-being, (2) pain is emblematic of many contested and/or chronic conditions, and (3) pain and pain treatment reflect and have wide-ranging implications for public policy. Overall, whether pain is analyzed quantitatively or qualitatively-focusing on its distribution in the population, its social causes and consequences, or its subjective meanings for individuals-pain reflects social conditions, sociopolitical context, and health-related beliefs of a society. Pain is thus an important frontier for future sociological research.

Multisectoral Approach to Address Chikungunya Outbreaks Driven by Human Mobility: A Systematic Review and Meta-Analysis.

Project description:BackgroundThe role of human mobility in the epidemiology of emerging Aedes-transmitted viral diseases is recognized but not fully understood. The objective of this systematic review and meta-analysis was to examine how human mobility patterns are driving chikungunya outbreaks.MethodsLiterature was systematically reviewed for studies on chikungunya prevalence in countries/territories with high-level evidence of human mobility-driven outbreaks, based on: (1) emergence of chikungunya outbreaks with epidemic chikungunya virus genotypes among displaced/migrant populations and their hosting communities; and (2) identification of imported index case(s) with epidemic genotypes phylogenetically related to the genotypes circulating during emerging or subsequent outbreaks.ResultsThe meta-analysis of extracted prevalence data revealed that a large proportion of the population in countries/territories afflicted by outbreaks is still at risk of infection during future outbreaks. On the other hand, approximately one-half of suspected chikungunya cases could be infected with other co-circulating acute febrile illnesses.ConclusionsWe discussed in this paper how human mobility-driven chikungunya outbreaks can be addressed, and how the involvement of several sectors in addition to the health sector in multisectoral approaches (MSAs) is important for prevention and control of chikungunya and other Aedes-transmitted arboviral outbreaks.

Data-driven programmatic approach to analysis of basophil activation tests.

Project description:BACKGROUND:Conventional data analysis of flow cytometry-based basophil activation testing requires repetitive, labor-intensive analysis that hampers efforts to standardize testing for clinical applications. Using an open-source platform, we developed and implemented a programmatic approach to the analysis of the basophil activation test (BAT) by flow cytometry. METHODS:Using the BÜHLMANN FlowCAST® assay, peripheral blood from peanut allergic patients undergoing oral immunotherapy was incubated with peanut allergens (Arah1, Arah2, Arah6, whole peanut extract) and stained with fluorescent antibodies to CCR3 and CD63 for the development of a data-driven programmatic analysis using Bioconductor and R. Basophil identification using clustering and classification was validated using manually gated comparisons in an experimental subset. Reproducibility of CD63 upregulation set on unstimulated or anti-FcERI stimulated basophils was compared. RESULTS:BAT analysis of 294 experiments was successful in 91.5% using the above approach, with a total of 7,166 individual basophil activation tests from 269 experiments. We estimate this represents a net saving of 1340 min of labor by a skilled operator. Medium-based gating correlated to respective manual gating more closely than anti-FcERI based gating (R = 0.96 vs. R = 0.84, P < 0.001). Only 2% of the basophil activation results were significantly different from manual gating. Quality measures of the experiments and other measures of basophil activation were also provided by the analysis. CONCLUSIONS:We present a novel data-driven flow cytometric platform for the analysis of clinical basophil activation testing, providing a high throughput objective approach to basophil activation analysis. © 2017 International Clinical Cytometry Society.

Data-driven analysis of simultaneous EEG/fMRI using an ICA approach.

Project description:Due to its millisecond-scale temporal resolution, EEG allows to assess neural correlates with precisely defined temporal relationship relative to a given event. This knowledge is generally lacking in data from functional magnetic resonance imaging (fMRI) which has a temporal resolution on the scale of seconds so that possibilities to combine the two modalities are sought. Previous applications combining event-related potentials (ERPs) with simultaneous fMRI BOLD generally aimed at measuring known ERP components in single trials and correlate the resulting time series with the fMRI BOLD signal. While it is a valuable first step, this procedure cannot guarantee that variability of the chosen ERP component is specific for the targeted neurophysiological process on the group and single subject level. Here we introduce a newly developed data-driven analysis procedure that automatically selects task-specific electrophysiological independent components (ICs). We used single-trial simultaneous EEG/fMRI analysis of a visual Go/Nogo task to assess inhibition-related EEG components, their trial-to-trial amplitude variability, and the relationship between this variability and the fMRI. Single-trial EEG/fMRI analysis within a subgroup of 22 participants revealed positive correlations of fMRI BOLD signal with EEG-derived regressors in fronto-striatal regions which were more pronounced in an early compared to a late phase of task execution. In sum, selecting Nogo-related ICs in an automated, single subject procedure reveals fMRI-BOLD responses correlated to different phases of task execution. Furthermore, to illustrate utility and generalizability of the method beyond detecting the presence or absence of reliable inhibitory components in the EEG, we show that the IC selection can be extended to other events in the same dataset, e.g., the visual responses.

Boolean logic algebra driven similarity measure for text based applications.

Project description:In Information Retrieval (IR), Data Mining (DM), and Machine Learning (ML), similarity measures have been widely used for text clustering and classification. The similarity measure is the cornerstone upon which the performance of most DM and ML algorithms is completely dependent. Thus, till now, the endeavor in literature for an effective and efficient similarity measure is still immature. Some recently-proposed similarity measures were effective, but have a complex design and suffer from inefficiencies. This work, therefore, develops an effective and efficient similarity measure of a simplistic design for text-based applications. The measure developed in this work is driven by Boolean logic algebra basics (BLAB-SM), which aims at effectively reaching the desired accuracy at the fastest run time as compared to the recently developed state-of-the-art measures. Using the term frequency-inverse document frequency (TF-IDF) schema, the K-nearest neighbor (KNN), and the K-means clustering algorithm, a comprehensive evaluation is presented. The evaluation has been experimentally performed for BLAB-SM against seven similarity measures on two most-popular datasets, Reuters-21 and Web-KB. The experimental results illustrate that BLAB-SM is not only more efficient but also significantly more effective than state-of-the-art similarity measures on both classification and clustering tasks.

Implicit Task-Driven Probability Discrepancy Measure for Unsupervised Domain Adaptation.

Project description:Probability discrepancy measure is a fundamental construct for numerous machine learning models such as weakly supervised learning and generative modeling. However, most measures overlook the fact that the distributions are not the end-product of learning, but are the input of a downstream predictor. Therefore, it is important to warp the probability discrepancy measure towards the end tasks, and towards this goal, we propose a new bi-level optimization based approach so that the two distributions are compared not uniformly against the entire hypothesis space, but only with respect to the optimal predictor for the downstream end task. When applied to margin disparity discrepancy and contrastive domain discrepancy, our method significantly improves the performance in unsupervised domain adaptation, and enjoys a much more principled training process.

Managing ethical uncertainty: implicit normativity and the sociology of ethics.

Project description:This article illustrates and discusses the idea of 'implicit normativity', and specifically its relevance to the management of ethical uncertainty. In particular I consider (i) the role implicit normativity plays in masking and containing potential ethical uncertainty and (ii) the contrast and boundary between implicit normativity and 'overt ethics' where ethical contestation - as well as particular processes and agents - are highlighted as salient. Using examples I show how the idea of implicit normativity can be applied not only to specific practices but also to whole fields of practice. The notion of 'moral settlements' - along with the explanatory role of the threat of 'chaos' - is introduced and elucidated to develop these points. I argue that attention to the management of ethical uncertainty shows the critically important contribution that an ambitious sociology of ethics can make to clinical ethics, including by helping to formulate and drive home questions about the 'ethics of ethics'. The account presented here has resonances with work that seeks to use sociological lenses to move beyond conventional bioethics, including Petersen's (2013) call for a 'normative sociology'.

Can Functionalised Play Make Children Happy? A Critical Sociology Perspective

Project description:The functionalisation of play basically stems from the diagnosis of a global childhood inactivity crisis, the so-called “children's obesity pandemic.” Hence, in the context of the activity-related guidelines, children's play appears no longer to be unproductive and purpose-free. It is rather considered an anti-obesity tool that will help children to meet physical activity recommendations. It is questionable whether such a functionalised tool can also provide what has been called the salience of the pleasures of free-play for children. Furthermore, a “normalization” of functionalised practices of play, in turn, could stigmatize children who do not or cannot adhere to these practices. Based upon this background, this paper will take a critical sociology perspective to analyse the functionalisation and medicalisation of children's play in an individualized, mediatized, and pedagogised society. In this sense, the paper aims to pay attention to how the primary goal of play in the sense of “simply make children happy” has given way to the goal of making them healthy and functional.
