On Pulchritia new genus, with a reappraisal of the genera of Trichotriidae (Rotifera, Monogononta).

ABSTRACT: During the study of rotifers collected in Eastern DR Congo, we rediscovered specimens that correspond to Monostyla dorsicornuta Van Oye, 1926. This species, which we redescribe, had not been seen since it's summary description, and lacked type material. Our analysis reveals that the animal belongs to Trichotriidae rather than to Lecane (presently considered to include Monostyla) or Lecanidae, but is nevertheless characterised by a foot structure that is remarkably convergent to that of Lecanidae, and different from all other genera of Trichotriidae. We conclude that the species and the closely related South American Macrochaetus kostei (José de Paggi, Branco & Kozlowsky-Suzuki, 2000) belong to a new genus of Trichotriidae; the two offer a rare example of African-South American vicariance in rotifers.We further provide emended diagnoses of the remaining genera of Trichotriidae, to conform these to the new information and to address some inconsistencies in these.

Project description:A new monogonont rotifer, Cephalodellabinoculatasp. nov., was described from a soil sample collected in Korea. The new species is morphologically similar to C.carina but is distinguished by having two frontal eyespots, a vitellarium with eight nuclei, and the shape of its fulcrum. We also described four other cephalodellid species collected in Korea; Cephalodellaauriculata, C.catellina, C.gracilis, and C.tinca. Of these four species, C.gracilis and C.tinca were newly recorded in Korea. We provided the morphological characteristics of the five Cephalodella species along with photographs of trophi observed with a scanning electron microscope. Furthermore, we provided the mitochondrial cytochrome c oxidase subunit I gene sequences of the five species.

A weighted taxonomic matrix key for species of the rotifer genus Synchaeta (Rotifera, Monogononta, Synchaetidae).

Project description:A new, weighted matrix identification key for 34 largely undisputed species of Synchaeta was created with the aim of providing comparable, detailed and diagnostic character sets for each species that can be applied to live and/or preserved specimens. As part of this process, 14 species of Synchaeta were intensively re-investigated with respect to their habitus and trophi morphology using binocular, light, and scanning electron microscopy, which, together with behavioural observations, revealed several new discriminating characters. Whenever possible, missing information for any character was added for the remaining species from the literature, with the two recently described species Synchaeta arcifera and Synchaeta squamadigitata being considered for the first time in an identification key. Beyond its completeness, our key has two distinct advantages. First, the characters are supported by detailed illustrations of their respective character states whenever possible to both simplify identification and minimize any uncertainty in the descriptions themselves. Second, the new approach of weighting the characters according to their reliability, robustness and/or ease of determination was employed. This latter approach is especially advantageous for soft-bodied rotifers such as species of Synchaeta, where, for example, several external characters can be influenced by preservation and are therefore less diagnostic or reliable. Although the key is as comprehensive as possible, information for many species remains missing for many characters, thereby highlighting the need for additional comprehensive and detailed species (re-)investigations within Synchaeta.

Phylotaxogenomics for the Reappraisal of the Genus Roseomonas With the Creation of Six New Genera.

Project description:The genus Roseomonas is a significant group of bacteria which is invariably of great clinical and ecological importance. Previous studies have shown that the genus Roseomonas is polyphyletic in nature. Our present study focused on generating a lucid understanding of the phylogenetic framework for the re-evaluation and reclassification of the genus Roseomonas. Phylogenetic studies based on the 16S rRNA gene and 92 concatenated genes suggested that the genus is heterogeneous, forming seven major groups. Existing Roseomonas species were subjected to an array of genomic, phenotypic, and chemotaxonomic analyses in order to resolve the heterogeneity. Genomic similarity indices (dDDH and ANI) indicated that the members were well-defined at the species level. The Percentage of Conserved Proteins (POCP) and the average Amino Acid Identity (AAI) values between the groups of the genus Roseomonas and other interspersing members of the family Acetobacteraceae were below 65 and 70%, respectively. The pan-genome evaluation depicted that the pan-genome was an open type and the members shared 958 core genes. This claim of reclassification was equally supported by the phenotypic and chemotaxonomic differences between the groups. Thus, in this study, we propose to re-evaluate and reclassify the genus Roseomonas and propose six novel genera as Pararoseomonas gen. nov., Falsiroseomonas gen. nov., Paeniroseomonas gen. nov., Plastoroseomonas gen. nov., Neoroseomonas gen. nov., and Pseudoroseomonas gen. nov.

Do genome size differences within Brachionus asplanchnoidis (Rotifera, Monogononta) cause reproductive barriers among geographic populations?

Project description:Genome size in the rotifer Brachionus asplanchnoidis, which belongs to the B. plicatilis species complex, is greatly enlarged and extremely variable (205–407 Mbp). Such variation raises the question whether large genome size differences among individuals might cause reproductive barriers, which could trigger speciation within this group by restricting gene flow across populations. To test this hypothesis, we used B. asplanchnoidis clones from three geographic populations and conducted assays to quantify reproductive isolation among clones differing in genome size, and we examined the population structure of all three populations using amplified fragment length polymorphisms (AFLPs). AFLPs indicated that these populations were genetically separated, but we also found hints of natural gene flow. Clones from different populations with genome size differences of up to 1.7-fold could interbred successfully in the laboratory and give rise to viable, fertile ‘hybrid’ offspring. Genome sizes of these ‘hybrids’ were intermediate between those of their parents, and fitness in terms of male production, population growth, and egg development time was not negatively affected. Thus, we found no evidence for reproductive isolation or nascent speciation within B. asplanchnoidis. Instead, our results suggest that gene flow within this species can occur despite a remarkably large range of genome sizes.

Brachionus paranguensis sp. nov. (Rotifera, Monogononta), a member of the L group of the Brachionus plicatilis complex.

Project description:The Brachionus plicatilis complex represents the most studied group of rotifers, although the systematics of the species complex has not been completely clarified. Many studies have been conducted trying to explore the diversity within the complex, leading to the recognition of three major morphotypes: large (L), small-medium (SM), and small (SS). Currently six species have been described and classified under these types and another nine taxa have been identified but not formally described. Within the L group, three species have been officially described [B. plicatilis s.s. (L1), B. manjavacas (L2), and B. asplanchnoidis (L3)], while a formal description of L4, unofficially known as B. 'Nevada', is still lacking. In the present study, a new species, Brachionus paranguensis sp. nov., is formally described and presented as a representative of the L4 clade. The species has been named after a high altitude saline crater lake from Central Mexico, where the specimens were collected. An integrated approach using DNA taxonomy through COI and ITS1 markers, morphology, and ecology was used to confirm the identity of the new species.

Two new species of genus Limnias from Thailand, with keys to congeners (Rotifera, Gnesiotrocha).

Project description:Two new species and two morphological variant taxa of sessile rotifers found during a survey of Thai rotifers are reported upon. Living specimens were collected and identified from twelve sampling sites. The specimens were photographed, and prepared for SEMs of dorsal plates and trophi. Two new species of genus Limnias are recognized. Limniaslenis sp. n. possesses a corona with a relatively shallow ventral sinus, and a dorsal plate without any projection, whereas Limniasnovemceras sp. n. carries a corona with a deep and wide ventral sinus, and a dorsal plate with nine projections. Two morphological variants of Limniasmelicerta are discussed, which are designated as L.melicerta SH and L.melicerta LH on the basis of the length of the horns on their dorsal plates. Moreover, both a dichotomous key and a formula key are provide including all Limnias species known to date.

The genus Cletocamptus (Harpacticoida, Canthocamptidae): a reappraisal, with proposal of a new subfamily, a new genus, and a new species.

Project description:A new species of Cletocamptus closely related to C.helobius was found in sediment samples taken from a polluted estuarine system in north-western Mexico. The genus Cletocamptus was relegated to species incertae sedis in 1986, and this finding prompted us to evaluate the current taxonomic position of the genus within the Canthocamptidae. The latter has been subdivided in several, seemingly unnatural subfamilies in the past to better understand the relationships between its constituent taxa. In this study we propose a new subfamily, the Cletocamptinaesubfam. nov. for Amphibiperita, Cletocamptus, and Cletocamptoides gen. nov., defined by the synapomorphic subdistal ventral spinules on the rostrum. The genus Cletocamptoides gen. nov. is proposed for C.helobius, C.merbokensis, and C.biushelo sp. nov., and is supported by the 'cletodid' shape of the body and the reduced one-segmented endopod of the fourth swimming leg. Cletocamptus includes all the other species with long slender spinules on the posterior margin of prosomites and with the sexually modified inner spine on the second endopodal segment of the second swimming leg in the males. Amphibiperita retained the primitive female fifth leg with exopod and baseoendopod separated, and the primitive prehensile endopod of the first leg, but is defined by the loss of the antennary exopod. Other (syn)apomorphies are given, and the evolution of the mandibular palp is briefly discussed. Additionally, a diagnosis for the new subfamily, Cletocamptinae subfam. nov., an amended narrower diagnosis for Cletocamptus, the diagnosis for Cletocamptoides gen. nov., and a phylogenetic analysis supporting the proposal of these new taxa, are given.

Reappraisal of the genus Alternariaster (Dothideomycetes).

Project description:Alternariaster was erected in 2007 to accommodate Alternaria helianthi, a fungal species known to cause leaf spots on Helianthus annuus (sunflower). It was segregated from Alternaria based on conidial morphology. Recently an unknown alternaria-like dematiaceous fungus was found associated with leaf spots on Bidens sulphurea (yellow cosmos) in Brazil. Based on a multi-gene phylogeny of parts of the ITS and LSU genes, this fungus was placed within the Leptosphaeriaceae with Alternariaster helianthi as its closest neighbour. Additional genes sequenced, RPB2 and GAPDH, confirmed this close relationship. The fungus on B. sulphurea has smaller conidia, 50-97.5 × 12.5-20 μm, compared to Al. helianthi, 80-160 × 18-30 μm, and lacks oblique or transverse septa which can be present in Al. helianthi. Pathogenicity studies on 18 plant species belonging to the Compositae showed that the B. sulphurea fungus only infected B. sulphurea, whereas Al. helianthi infected H. annuus and Galinsoga quadriradiata, a yet unreported host of Al. helianthi. The fungus causing disease on B. sulphurea is hence closely related but phylogenetically, morphologically and pathologically distinct from Al. helianthi, and therefore newly described as Alternariaster bidentis. The collection of a second species in the genus Alternariaster and the multigene phylogenetic analysis of these two species, confirmed Alternariaster to be a well-delimited genus in the Leptosphaeriaceae rather than the Pleosporaceae, to which Alternaria belongs.

Sexual Reproductive Biology of a Colonial Rotifer Sinantherina socialis (Rotifera: Monogononta): Do mating strategies vary between colonial and solitary rotifer species?

Project description:In many aquatic invertebrates including monogonont rotifers, sex provides genetic variation and dormant stages that allows dispersal in time and space. While the reproductive biology of some solitary monogonont rotifer species is known, little is known concerning mating behaviors in colonial rotifers. Coloniality poses unique challenges to the typical mating behavior of solitary rotifers. For instance, most species engage in circling behavior, where the male swims in close proximity to the female. In colonial forms, access to a particular female may be hindered by nearby colony mates. Here we provide descriptions of (1) male morphology, (2) mating behavior, and (3) types of eggs of the widespread colonial rotifer Sinantherina socialis, and discuss modifications in mating strategies as a consequence of coloniality. Two important differences from mating patterns documented in solitary rotifers were found in S. socialis. First, duration of circling phase of mating is protracted for males encountering small colonies of females as compared to solitary females. Males encountering single females removed from their colonies behave similarly to those of solitary species. Second, duration of copulation in S. socialis is the shortest reported for any rotifer species. Endogamy might occur in this species as sons copulate with their sisters and mothers, at least under laboratory conditions. Examples of behaviour in linked video clips.

A taxonomic revision of the genus Conidiobolus (Ancylistaceae, Entomophthorales): four clades including three new genera.

Project description:The genus Conidiobolus is an important group in entomophthoroid fungi and is considered to be polyphyletic in recent molecular phylogenies. To re-evaluate and delimit this genus, multi-locus phylogenetic analyses were performed using the large and small subunits of nuclear ribosomal DNA (nucLSU and nucSSU), the small subunit of the mitochondrial ribosomal DNA (mtSSU) and the translation elongation factor 1-alpha (EF-1α). The results indicated that the Conidiobolus is not monophyletic, being grouped into a paraphyletic grade with four clades. Consequently, the well-known Conidiobolus is revised and three new genera Capillidium, Microconidiobolus and Neoconidiobolus are proposed along with one new record and 22 new combinations. In addition, the genus Basidiobolus is found to be basal to the other entomophthoroid taxa and the genus Batkoa locates in the Entomophthoraceae clade.
