Arthrobacter siccitolerans sp. nov., a highly desiccation-tolerant, xeroprotectant-producing strain isolated from dry soil.

ABSTRACT: A novel desiccation-tolerant, xeroprotectant-producing bacterium, designated strain 4J27(T), was isolated from a Nerium oleander rhizosphere subjected to seasonal drought in Granada, Spain. Phylogenetic analysis based on 16S rRNA gene sequencing placed the isolate within the genus Arthrobacter, its closest relative being Arthrobacter phenanthrenivorans Shep3 DSM 18606(T), with which it showed 99.23?% 16S rRNA gene sequence similarity. DNA-DNA hybridization measurements showed less than 25?% relatedness between strain 4J27(T) and Arthrobacter phenanthrenivorans DSM 18606(T). The DNA base composition of strain 4J27(T) was 65.3 mol%. The main fatty acids were anteiso C15?:?0, anteiso C17?:?0, C16?:?0 and iso C16?:?0 and the major menaquinone was MK-9 (H2). The peptidoglycan type was A3? with an l-Lys-l-Ser-l-Thr-l-Ala interpeptide bridge. The bacterium tested positive for catalase activity and negative for oxidase activity. Phylogenetic, chemotaxonomic and phenotypic analyses indicated that the desiccation-tolerant strain 4J27(T) represents a novel species within the genus Arthrobacter, for which the name Arthrobacter siccitolerans is proposed. The type strain is 4J27(T) (?=?CECT 8257(T)?=?LMG 27359(T)).

Project description:A novel yellow colony-forming bacterium, strain P3B162T was isolated from the pokkali rice rhizosphere from Kerala, India, as part of a project study aimed at isolating plant growth beneficial rhizobacteria from saline tolerant pokkali rice and functionally evaluate their abilities to promote plant growth under saline conditions. The novel strain P3B162T possesses plant growth beneficial traits such as positive growth on 1-aminocyclopropane-1-carboxylic acid (ACC), production of indole acetic acid (IAA) and siderophore. In addition, it also showed important phenotypic characters such as ability to form biofilm and utilization of various components of plant root exudates (sugars, amino acids and organic acids), clearly indicating its lifestyle as a plant rhizosphere associated bacterium. Taxonomically, the novel strain P3B162T was affiliated to the genus Arthrobacter based on the collective results of phenotypic, genotypic and chemotaxonomic analyses. Moreover, molecular analysis using 16S rRNA gene showed Arthrobacter globiformis NBRC 12137T, Arthrobacter pascens DSM 20545T and Arthrobacter liuii DSXY973T as the closely related phylogenetic neighbours, showing more than 98% 16S rRNA similarity values, whereas the recA gene analysis displayed Arthrobacter liuii JCM 19864T as the nearest neighbour with 94.7% sequence similarity and only 91.7% to Arthrobacter globiformis LMG 3813T and 88.7% to Arthrobacter pascens LMG 16255T. However, the DNA-DNA hybridization values between strain P3B162T, Arthrobacter globiformis LMG 3813T, Arthrobacter pascens LMG 16255T and Arthrobacter liuii JCM 19864T was below 50%. In addition, the novel strain P3B162T can be distinguished from its closely related type strains by several phenotypic characters such as colony pigment, tolerance to NaCl, motility, reduction of nitrate, hydrolysis of DNA, acid from sucrose, cell wall sugars and cell wall peptidoglycan structure. In conclusion, the combined results of this study support the classification of strain P3B162T as a novel Arthrobacter species and we propose Arthrobacter pokkalii sp.nov.as its name. The type strain is P3B162T (= KCTC 29498T = MTCC 12358T).

Project description:Heavy-metal-tolerant bacteria, GIMN1.004(T), was isolated from mine soils of Dabaoshan in South China, which were acidic (pH 2-4) and polluted with heavy metals. The isolation was Gram-negative, aerobic, non-spore-forming, and rod-shaped bacteria having a cellular width of 0.5-0.6 µm and a length of 1.3-1.8 µm. They showed a normal growth pattern at pH 4.0-9.0 in a temperature ranging from 5 °C to 40 °C.The organism contained ubiquinone Q-8 as the predominant isoprenoid quinine, and C(16:0), summed feature 8 (C(18:1)?7c and C(18:1)?6c), C(18:0), summed feature 3 (C(16:1)?7c or iso-C(15:0) 2-OH), C(17:0) cyclo, C(18:1)?9c, C(19:0) cyclo ?8c, C(14:0) as major fatty acid. These profiles were similar to those reported for Burkholderia species. The DNA G+C % of this strain was 61.6%. Based on the similarity to 16S rRNA gene sequence, GIMN1.004(T) was considered to be in the genus Burkholderia. The similarities of 16S rRNA gene sequence between strain GIMN1.004(T) and members of the genus Burkholderia were 96-99.4%, indicating that this novel strain was phylogenetically related to members of that genus. The novel strain showed the highest sequence similarities to Burkholderia soli DSM 18235(T) (99.4%); Levels of DNA-DNA hybridization with DSM 18235(T) was 25%. Physiological and biochemical tests including cell wall composition analysis, differentiated phenotype of this strain from that closely related Burkholderia species. The isolation had great tolerance to cadmium with MIC of 22 mmol/L, and adsorbability of 144.94 mg/g cadmium,and it was found to exhibit antibiotic resistance characteristics. The adsorptive mechanism of GIMN1.004(T) for cadmium depended on the action of the amide,carboxy and phosphate of cell surface and producing low-molecular-weight (LMW) organic acids to complex or chelated Cd(2+).Therefore, the strain GIMN1.004(T) represented a new cadmium resistance species, which was tentatively named as Burkholderia dabaoshanensis sp. nov. The strain type is GIMN1.004(T) (= CCTCC M 209109(T) = NRRL B-59553(T)).
