Self-organized patterning through the dynamic segregation of DNA and silica nanoparticles.

ABSTRACT: Exotic pattern formation as a result of drying of an aqueous solution containing DNA and silica nanoparticles is reported. The pattern due to segregation was found to critically depend on the relative ratio of nanoparticles and DNA, as revealed by polarization microscopy, scanning electron microscopy, and fluorescence microscopy. The blurred radial pattern that is usually observed in the drying of a colloidal solution was shown to be vividly sharpened in the presence of DNA. Uniquely curved, crescent-shaped micrometer-scale domains are generated in regions that are rich in nanoparticles. The characteristic segregated patterns observed in the present study are interpreted in terms of a large aspect ratio between the persistence length (∼ 50 nm) and the diameter (∼ 2 nm) of double-stranded DNA, and the relatively small silica nanoparticles (radius: 5 nm).

Frequency-Driven Self-Organized Helical Superstructures Loaded with Mesogen-Grafted Silica Nanoparticles.

Project description:Adding colloidal nanoparticles into liquid-crystal media has become a promising pathway either to enhance or to introduce novel properties for improved device performance. Here we designed and synthesized new colloidal hybrid silica nanoparticles passivated with a mesogenic monolayer on the surface to facilitate their organo-solubility and compatibility in a liquid-crystal host. The resulting nanoparticles were identified by 1 H NMR spectroscopy, TEM, TGA, and UV/Vis techniques, and the hybrid nanoparticles were doped into a dual-frequency cholesteric liquid-crystal host to appraise both their compatibility with the host and the effect of the doping concentration on their electro-optical properties. Interestingly, the silica-nanoparticle-doped liquid-crystalline nanocomposites were found to be able to dynamically self-organize into a helical configuration and exhibit multi-stability, that is, homeotropic (transparent), focal conic (opaque), and planar states (partially transparent), depending on the frequency applied at sustained low voltage. Significantly, a higher contrast ratio between the transparent state and scattering state was accomplished in the nanoparticle-embedded liquid-crystal systems.

Organized Silica Films Generated by Evaporation-Induced Self-Assembly as Hosts for Iron Oxide Nanoparticles.

Project description:In this work, we prepared oriented mesoporous thin films of silica on various solid substrates using the pluronic block copolymer P123 as a template. We attempted to insert guest iron oxide (FexOy) nanoparticles into these films by two different methods: (a) by co-precipitation-where iron precursors are introduced in the synthesis sol before deposition of the silica film-and subsequent oxide production during the film calcination step; (b) by preparing and calcining the silica films first then impregnating them with the iron precursor, obtaining the iron oxide nanoparticles by a second calcination step. We have examined the structural effects of the guest nanoparticles on the silica film structures using grazing incidence X-ray scattering (GISAXS), high-resolution transmission electron spectroscopy (HRTEM), spectroscopic ellipsometry, X-ray photoelectron spectroscopy (XPS), and Raman microscopy. Formation of nanoparticles by co-precipitation may induce substantial changes in the film structure leading, in our adopted process, to the appearance of lamellar ordering in the calcination stage. On the contrary, impregnation-based approaches perturb the film structures much more weakly, but are also less efficient in filling the pores with nanoparticles.

Self-organized developmental patterning and differentiation in cerebral organoids.

Project description:Cerebral organoids recapitulate human brain development at a considerable level of detail, even in the absence of externally added signaling factors. The patterning events driving this self-organization are currently unknown. Here, we examine the developmental and differentiative capacity of cerebral organoids. Focusing on forebrain regions, we demonstrate the presence of a variety of discrete ventral and dorsal regions. Clearing and subsequent 3D reconstruction of entire organoids reveal that many of these regions are interconnected, suggesting that the entire range of dorso-ventral identities can be generated within continuous neuroepithelia. Consistent with this, we demonstrate the presence of forebrain organizing centers that express secreted growth factors, which may be involved in dorso-ventral patterning within organoids. Furthermore, we demonstrate the timed generation of neurons with mature morphologies, as well as the subsequent generation of astrocytes and oligodendrocytes. Our work provides the methodology and quality criteria for phenotypic analysis of brain organoids and shows that the spatial and temporal patterning events governing human brain development can be recapitulated in vitro.

Self-organized patterning of cell morphology via mechanosensitive feedback.

Project description:Tissue organization is often characterized by specific patterns of cell morphology. How such patterns emerge in developing tissues is a fundamental open question. Here, we investigate the emergence of tissue-scale patterns of cell shape and mechanical tissue stress in the Drosophila wing imaginal disc during larval development. Using quantitative analysis of the cellular dynamics, we reveal a pattern of radially oriented cell rearrangements that is coupled to the buildup of tangential cell elongation. Developing a laser ablation method, we map tissue stresses and extract key parameters of tissue mechanics. We present a continuum theory showing that this pattern of cell morphology and tissue stress can arise via self-organization of a mechanical feedback that couples cell polarity to active cell rearrangements. The predictions of this model are supported by knockdown of MyoVI, a component of mechanosensitive feedback. Our work reveals a mechanism for the emergence of cellular patterns in morphogenesis.

Self-organized podosomes are dynamic mechanosensors.

Project description:Podosomes are self-organized, dynamic, actin-containing structures that adhere to the extracellular matrix via integrins [1-5]. Yet, it is not clear what regulates podosome dynamics and whether podosomes can function as direct mechanosensors, like focal adhesions [6-9]. We show here that myosin-II proteins form circular structures outside and at the podosome actin ring to regulate podosome dynamics. Inhibiting myosin-II-dependent tension dissipated podosome actin rings before dissipating the myosin-ring structure. As podosome rings changed size or shape, tractions underneath the podosomes were exerted onto the substrate and were abolished when myosin-light-chain activity was inhibited. The magnitudes of tractions were comparable to those generated underneath focal adhesions, and they increased with substrate stiffness. The dynamics of podosomes and of focal adhesions were different. Torsional tractions underneath the podosome rings were generated with rotations of podosome rings in a nonmotile, nonrotating cell, suggesting a unique feature of these circular structures. Stresses applied via integrins at the apical surface directly displaced podosomes near the basal surface. Stress-induced podosome displacements increased nonlinearly with applied stresses. Our results suggest that podosomes are dynamic mechanosensors in which interactions of myosin tension and actin dynamics are crucial for regulating these self-organized structures in living cells.

Somitic positional information guides self-organized patterning of snake scales.

Project description:Two influential concepts in tissue patterning are Wolpert's positional information and Turing's self-organized reaction-diffusion (RD). The latter establishes the patterning of hair and feathers. Here, our morphological, genetic, and functional-by CRISPR-Cas9-mediated gene disruption-characterization of wild-type versus "scaleless" snakes reveals that the near-perfect hexagonal pattern of snake scales is established through interactions between RD in the skin and somitic positional information. First, we show that ventral scale development is guided by hypaxial somites and, second, that ventral scales and epaxial somites guide the sequential RD patterning of the dorsolateral scales. The RD intrinsic length scale evolved to match somite periodicity, ensuring the alignment of ribs and scales, both of which play a critical role in snake locomotion.

YAP1 regulates the self-organized fate patterning of hESC-derived gastruloids.

Project description:The gastrulation process relies on complex interactions between developmental signaling pathways that are not completely understood. Here, we interrogated the contribution of the Hippo signaling effector YAP1 to the formation of the three germ layers by analyzing human embryonic stem cell (hESC)-derived 2D-micropatterned gastruloids. YAP1 knockout gastruloids display a reduced ectoderm layer and enlarged mesoderm and endoderm layers compared with wild type. Furthermore, our epigenome and transcriptome analysis revealed that YAP1 attenuates Nodal signaling by directly repressing the chromatin accessibility and transcription of key genes in the Nodal pathway, including the NODAL and FOXH1 genes. Hence, in the absence of YAP1, hyperactive Nodal signaling retains SMAD2/3 in the nuclei, impeding ectoderm differentiation of hESCs. Thus, our work revealed that YAP1 is a master regulator of Nodal signaling, essential for instructing germ layer fate patterning in human gastruloids.

Self-organized cell patterning via mechanical feedback in hindbrain neuropore morphogenesis.

Project description:Cell patterning is essential for organized tissue development, enabling precise geometric arrangement of cells, body axis establishment and developmental timing. Here we investigate the role of physical forces and mechanical cues in organizing and maintaining cell morphological patterns during hindbrain neuropore closure, a critical morphogenetic event in vertebrate development. Through live-imaging in mouse embryos and cell-based biophysical modeling, we demonstrate that active cell crawling and actomyosin purse-string contraction at the neuropore border are insufficient to account for the observed cellular arrangements in space and time. Instead, mechanosensitive feedback between cellular stress, shape, and nematic alignment is required to establish and maintain cell morphological patterns and their spatial order. This feedback-driven model generates persistent shape memory in cells, stalls cell rearrangements, and promotes local tissue solidification to preserve the spatial organization during the closure process. We validate this model experimentally, establishing the critical role of mechanical feedback in guiding tissue-level morphogenesis through active, force-driven patterning.

Controlled mud-crack patterning and self-organized cracking of polydimethylsiloxane elastomer surfaces.

Project description:Exploiting pattern formation - such as that observed in nature - in the context of micro/nanotechnology could have great benefits if coupled with the traditional top-down lithographic approach. Here, we demonstrate an original and simple method to produce unique, localized and controllable self-organised patterns on elastomeric films. A thin, brittle silica-like crust is formed on the surface of polydimethylsiloxane (PDMS) using oxygen plasma. This crust is subsequently cracked via the deposition of a thin metal film - having residual tensile stress. The density of the mud-crack patterns depends on the plasma dose and on the metal thickness. The mud-crack patterning can be controlled depending on the thickness and shape of the metallization - ultimately leading to regularly spaced cracks and/or metal mesa structures. Such patterning of the cracks indicates a level of self-organization in the structuring and layout of the features - arrived at simply by imposing metallization boundaries in proximity to each other, separated by a distance of the order of the critical dimension of the pattern size apparent in the large surface mud-crack patterns.

Dynamic self-assembly and self-organized transport of magnetic micro-swimmers.

Project description:We demonstrate experimentally and in computer simulations that magnetic microfloaters can self-organize into various functional structures while energized by an external alternating (ac) magnetic field. The structures exhibit self-propelled motion and an ability to carry a cargo along a pre-defined path. The morphology of the self-assembled swimmers is controlled by the frequency and amplitude of the magnetic field.
