ABSTRACT: It is hypothesised here that some mealybug (Pseudococcidae) and root mealybug (Rhizoecidae) prepupae and pupae are mobile. The prepupa and pupa of the mealybug Promyrmococcus dilli Williams and the prepupa of the root mealybug Ripersiella malschae (Williams) are described and illustrated and their probable mobility is discussed. It is also suggested that the prepupae and pupae of the mealybug Macrocepicoccus loranthi Morrison can move rapidly on the leaves when disturbed.

Preparation of Mealybugs (Hemiptera: Pseudococcidae) for Genetic Characterization and Morphological Examination.

Project description:Mealybugs (Hemiptera: Pseudococcidae) are economically significant agricultural pests on many different crops. Because of their small size and lack of easily visible characters for identification, determination of their taxonomic status is difficult and requires technical competency to prepare a slide-mounted specimen. The standard mounting technique does not allow for analysis of the genome of the specimen. Conversely, preparatory techniques for genetic analysis of mealybugs cause either loss of the entire individual or physical damage that can make morphology-based identification difficult. This study describes a simple protocol that does not impact physical integrity of the specimen for fixation and microscopic examination yet enables simultaneous DNA extraction for DNA-based identification of four mealybug species. All species prepared yielded high quality slide mounts, identified as Planococcus citri Risso, Pseudococcus viburni Signoret, Rhizoecus kondonis Kuwana, or Rhizoecus californicus Ferris. DNA extracted in this manner had higher purity and yield in the final eluate than in samples extracted using standard methods. DNA extracted was successfully amplified by polymerase chain reaction using primers for the cytochrome oxidase I gene and subsequently sequenced for all specimens. This protocol is likely to be applicable to other Hemiptera taxa that are preserved by slide mounting, allowing for both the preparation of a high-quality voucher specimen for morphological identification and simultaneous analysis of DNA for the same specimen. The methods used are technically less challenging than current standard procedures.

Phylogenetic analyses and characteristics of the microbiomes from five mealybugs (Hemiptera: Pseudococcidae).

Project description:Associations between Sternorrhyncha insects and intracellular bacteria are common in nature. Mealybugs are destructive pests that seriously threaten the production of agriculture and forestry. Mealybugs have evolved intimate endosymbiotic relationships with bacteria, which provide them with essential amino acids, vitamins, and other nutrients. In this study, the divergence of five mealybugs was analyzed based up the sequences of the mitochondrial cytochrome oxidase I (mtCOI). Meanwhile, the distinct regions of the 16S rRNA gene of primary symbionts in the mealybugs were sequenced. Finally, high-throughput sequencing (HTS) techniques were used to study the microbial abundance and diversity in mealybugs. Molecular phylogenetic analyses revealed that these five mealybugs were subdivided into two different clusters. One cluster of mealybugs (Dysmicoccus neobrevipes, Pseudococcus comstocki, and Planococcus minor) harbored the primary endosymbiont "Candidatus Tremblaya princeps," and another cluster (Phenacoccus solenopsis and Phenacoccus solani) harbored "Ca. Tremblaya phenacola." The mtCOI sequence divergence between the two clusters was similar to the 16S rRNA sequence divergence between T. princeps and T. phenacola. Thus, we concluded that the symbiont phylogeny was largely concordant with the host phylogeny. The HTS showed that the microbial abundance and diversity within P. solani and P. solenopsis were highly similar, and there was lower overall species richness compared to the other mealybugs. Among the five mealybugs, we also found significant differences in Shannon diversity and observed species. These results provide a theoretical basis for further research on the coevolution of mealybugs and their symbiotic microorganisms. These findings are also useful for research on the effect of symbiont diversity on the pest status of mealybugs in agricultural systems.

DNA barcoding identification of Pseudococcidae (Hemiptera: Coccoidea) using the mitochondrial COI gene.

Project description:DNA barcoding is a recently developed technique for species-level identification that involves the use of short, standard DNA sequences as species labels. It is an effective complement to traditional taxonomic classification based on morphology. At present, research and applications involving the DNA barcoding of the Pseudococcidae are focused primarily on the cytochrome coxidase subunit I (COI) gene, but there is not yet a consensus on the preferred gene region for barcoding. The purpose of this study was to explore the effectiveness of identification of Pseudococcidae beetles using DNA barcoding technology. The COI gene sequences of 97 samples from 21 species of Asemini were analysed, followed by evaluation of the ability to identify species using a tree-building method and distance evaluation. The COI sequences (500 bp) exhibited distinct distributions of intra-specific and inter-specific variation and a significant barcoding gap. The success rate of identification was 97.84%. These results demonstrate the feasibility of using this segment of COI to identify most species of Pseudococcidae.

Mealybugs (Hemiptera, Coccomorpha, Pseudococcidae) on parasitic plants (Loranthaceae) in Indonesia with description of a new species and a new country record.

Project description:Parasitic plants have been known to be attacked by insect pests since ancient times. However, little is known about the mealybug (Hemiptera, Coccomorpha, Pseudococcidae) fauna associated with them. A series of surveys of mealybugs found on Loranthaceae, a semi-parasitic plant family, was conducted in several places in Bengkulu Province, southern Sumatra, Indonesia. In the study, 55 mealybug specimens were collected, consisting of eight species belonging to five genera, namely Chorizococcus McKenzie (1 species), Dysmicoccus Ferris (2 species), Ferrisia Fullaway (1 species), Planococcus Ferris (3 species) and Pseudococcus Westwood (1 species). Chorizococcusozeri Zarkani & Kaydan, sp. nov. is new to science, whilst Planococcusbagmaticus Williams represents the first record in Indonesia. In addition, the mealybugs Dysmicoccuslepelleyi (Betrem), Dysmicoccuszeynepae Zarkani & Kaydan, Ferrisiadasylirii (Cockerell), Planococcuslilacinus (Cockerell) and Pseudococcusjackbeardsleyi Gimpel & Miller are newly recorded from plants of the family Loranthaceae. Figures and illustrations of mealybug species with a taxonomic key to Asian Chorizococcus and a new country record based on morphological characters are also updated.

Cytogenetic and taxonomic studies of some legless mealybugs (Homoptera, Coccinea, Pseudococcidae).

Project description:A new monotypic genus and species, Komodesia circuliplurimagen. et sp. n., from Flores Is. (Indonesia) and the new species, Antonina diversiglandulosasp. n., from Southern Thailand are described and illustrated. Chromosomes of these species and also the ones of Antonina purpurea Signoret, 1872 and Antonina thaiensis Takahashi, 1942 are studied for the first time: 2n = 30, 20, 12 and 22+Bs correspondingly; the male embryos of all four species demonstrate Lecanoid paternal heterochromatinization of one haploid set of chromosomes. The karyotypes of three widely distributed species, Antonina pretiosa Ferris, 1953, Antonina graminis (Maskell, 1897) and Chaetococcus bambusae (Maskell, 1893), are studied based on material from other regions in comparison with previously published data. Photographs of the karyotypes are provided for the first time for all seven species. The terminological problems connected with the identification and naming of the three scale insect genetic systems, Lecanoid, Comstockioid and Diaspidoid, are discussed.

Evidence for gene flow between two sympatric mealybug species (Insecta; Coccoidea; Pseudococcidae).

Project description:Occurrence of inter-species hybrids in natural populations might be evidence of gene flow between species. In the present study we found evidence of gene flow between two sympatric, genetically related scale insect species--the citrus mealybug Planococcus citri (Risso) and the vine mealybug Planococcus ficus (Signoret). These species can be distinguished by morphological, behavioral, and molecular traits. We employed the sex pheromones of the two respective species to study their different patterns of male attraction. We also used nuclear ITS2 (internal transcribed spacer 2) and mitochondrial COI (Cytochrome c oxidase sub unit 1) DNA sequences to characterize populations of the two species, in order to demonstrate the outcome of a possible gene flow between feral populations of the two species. Our results showed attraction to P. ficus pheromones of all tested populations of P. citri males but not vice versa. Furthermore, ITS2 sequences revealed the presence of 'hybrid females' among P. citri populations but not among those of P. ficus. 'hybrid females' from P. citri populations identified as P. citri females according to COI sequences. We offer two hypotheses for these results. 1) The occurrence of phenotypic and genotypic traits of P. ficus in P. citri populations may be attributed to both ancient and contemporary gene flow between their populations; and 2) we cannot rule out that an ancient sympatric speciation by which P. ficus emerged from P. citri might have led to the present situation of shared traits between these species. In light of these findings we also discuss the origin of the studied species and the importance of the pherotype phenomenon as a tool with which to study genetic relationships between congener scale insects.

Protein Profile Analysis of Ericerus pela (Hemiptera: Coccoidea) Egg

Project description:Abstract The transformation from embryo to first instar nymph is an essential process in the insect life cycle. In order to characterize protein expression in the Ericerus pela Chavannes (Hemiptera: Coccoidea) egg, high-throughput proteomics and bioinformatics methods were used. A total of 678 peptides were identified and assigned to 358 protein groups. The proteins exhibited a wide range of molecular weight (3.50–495.12 kDa) and isoelectric points (3.50–13.1). Gene Ontology annotation showed that the majority of proteins were associated with cellular processes, metabolic processes, and response to stimulus processes. The predominant molecular functions of E. pela egg proteins included binding, catalytic activity, transporter activity, and structural molecule activity. Kyoto Encyclopedia of Genes and Genomes annotations identified 137 pathways, and most proteins were assigned to metabolism events, including many enzymes participating in energy metabolism, protein folding, sorting, and degradation. The processes and functions of the identified proteins were closely related to the physiological status of egg and embryo development. We conclude that some identified proteins are related to important egg biological characteristics, and regard them as the target proteins for future study.

The first mitochondrial genome of scale insects (Hemiptera: Coccoidea).

Project description:Here, we report the first mitochondrial genome of scale insects sequenced from Ceroplastes japonicus (Hemiptera: Coccidae). The genome has a circular genome of 14,979 bp in length, with a high A + T content of 85.15%. Twelve protein-coding genes (excluding atp8), 13 tRNA, and 2 rRNA genes were detected and annotated using the MITOS web server. The absence of atp8 and some tRNAs might indicate possible novel structures or loss of genes.

Discovery pattern and species number of scale insects (Hemiptera: Coccoidea).

Project description:Few investigations have been made of the species description trend of scale insects. The present study reports the discovery pattern and taxonomic efforts for this group based on global species and a literature dataset. In addition, three asymptotic models (Logistic, Gompertz, and Extreme Value) based on a discovery curve were used to predict the species number of scale insects. Our results showed that the species description rate has been changing over time, with certain peaks and valleys in the past 250 years. The mean number of species described per year was 30, with the highest number of 195 described species in 1985. The increasing number of authors and the almost constant proportion of species described by 10% most prolific authors since the 1900s suggested that taxonomic effort has been increasing over time. The Gompertz model with lowest AIC value suggested that there are about 10,450 species of scale insects on Earth, nearly 30% of which remain to be described. Our study offers insights into the discovery pattern of scale insect diversity.
