Response of seed tocopherols in oilseed rape to nitrogen fertilizer sources and application rates.

ABSTRACT: Tocopherols (Tocs) are vital scavengers of reactive oxygen species (ROS) and important seed oil quality indicators. Nitrogen (N) is one of the most important fertilizers in promoting biomass and grain yield in crop production. However, the effect of different sources and application rates of N on seed Toc contents in oilseed rape is poorly understood. In this study, pot trials were conducted to evaluate the effect of two sources of N fertilizer (urea and ammonium nitrate). Each source was applied to five oilseed rape genotypes (Zheshuang 72, Jiu-Er-1358, Zheshuang 758, Shiralee, and Pakola) at three different application rates (0.41 g/pot (N1), 0.81 g/pot (N2), and 1.20 g/pot (N3)). Results indicated that urea increased ?-, ?-, and total Toc (T-Toc) more than did ammonium nitrate. N3 was proven as the most efficient application rate, which yielded high contents of ?-Toc and T-Toc. Highly significant correlations were observed between Toc isomers, T-Toc, and ?-/?-Toc ratio. These results clearly demonstrate that N sources and application rates significantly affect seed Toc contents in oilseed rape.

Project description:In agricultural plant production, nitrate, ammonium, and urea are the major fertilized nitrogen forms, which differ in root uptake and downstream signaling processes in plants. Nitrate is known to stimulate cytokinin synthesis in roots, while for urea no hormonal effect has been described yet. Elevated cytokinin levels can delay plant senescence favoring prolonged nitrogen uptake. As the cultivation of winter oilseed rape provokes high nitrogen-balance surpluses, we tested the hypotheses whether nitrogen use efficiency increases under ammonium nitrate- relative to urea-based nutrition and whether this is subject to genotypic variation. In a 2-year field study, 15 oilseed rape lines were fertilized either with ammonium nitrate or with urease inhibitor-stabilized urea and analyzed for seed yield and nitrogen-related yield parameters. Despite a significant environmental impact on the performance of the individual lines, which did not allow revealing consistent impact of the genotype, ammonium nitrate-based nutrition tended to increase seed yield in average over all lines. To resolve whether the fertilizer N forms act on grain yield via phytohormones, we collected xylem exudates at three developmental stages and determined the translocation rates of cytokinins and N forms. Relative to urea, ammonium nitrate-based nutrition enhanced the translocation of nitrate or total nitrogen together with cytokinins, whereas in the urea treatment translocation rates were lower as long as urea remained stable in the soil solution. At later developmental stages, i.e., when urea became hydrolyzed, nitrogen and cytokinin translocation increased. In consequence, urea tended to increase nitrogen partitioning in the shoot toward generative organs. However, differences in overall nitrogen accumulation in shoots were not present at the end of the vegetation period, and neither nitrogen uptake nor utilization efficiency was consistently different between the two applied nitrogen forms.

Project description:Although the impact of sulfur (S) availability on the seed yield and nutritional quality of seeds has been demonstrated, its impact coupled with nitrogen (N) availability remains poorly studied in oilseed rape. A deeper knowledge of S and N interactions on seed yield components and seed quality could improve S and N fertilization management in a sustainable manner. To address this question, our goals were to determine the effects of nine different S fertilization management strategies (i) in interaction with different levels of N fertilization and (ii) according to the timing of application (by delaying and fractionating the S inputs) on agronomic performances and components of seed yield. The impact of these various managements of S and N fertilizations was also investigated on the seed quality with a focus on the composition of SSPs (mainly represented by napins and cruciferins). Our results highlighted synergetic effects on S and N use efficiencies at optimum rates of S and N inputs and antagonistic effects at excessive rates of one of the two elements. The data indicated that adjustment of S and N fertilization may lead to high seed yield and seed protein quality in a sustainable manner, especially in the context of reductions in N inputs. Delaying S inputs improved the seed protein quality by significantly increasing the relative abundance of napin (a SSP rich in S-containing amino acids) and decreasing the level of a cruciferin at 30 kDa (a SSP with low content of S-amino acids). These observations suggest that fractionated or delayed S fertilizer inputs could provide additional insights into the development of N and S management strategies to maintain or improve seed yield and protein quality. Our results also demonstrated that the S% in seeds and the napin:30 kDa-cruciferin ratio are highly dependent on S/N fertilization in relation to S supply. In addition, we observed a strong relationship between S% in seeds and the abundance of napin as well as the napin:30 kDa-cruciferin ratio, suggesting that S% may be used as a relevant index for the determination of protein quality in seeds in terms of S-containing amino acids.

Project description:BACKGROUND:Acetolactate synthase (ALS)-inhibiting herbicides from the chemical families of sulfonylureas and imidazolinones are used worldwide. However, drift or sprayer contamination from some sulfonylurea herbicides causes a high level of male sterility in cruciferous species, especially oilseed rape (OSR). In this paper, we evaluated the gametocidal effects of 27 ALS-inhibiting herbicides that were sprayed on OSR plants at the bolting stage. RESULTS:OSR anther development was very sensitive to sublethal exposure to most ALS-inhibiting herbicides. The application of 18 out of the 20 tested sulfonylureas (except ethametsulfuron and ethoxysulfuron), two imidazolinones (imazethapyr and imazamox), and one sulfonylamino-carbonyltriazolinone (flucarbazone-sodium) at suitable rates could induce male sterility. Eight of the herbicides, including chlorsulfuron (at application rates of 60-120 mg/ha), halosulfuron-methyl (300-600 mg/ha), sulfosulfuron (400-600 mg/ha), triflusulfuron-methyl (500-750 mg/ha), pyrazosulfuron-ethyl (150-225 mg/ha), nicosulfuron (200-300 mg/ha), imazethapyr (750-1125 mg/ha), and imazamox (400-800 mg/ha), could induce over 90% male sterility and over 60% relative outcrossed seed set in six cultivars with different origins. These eight chemicals could be used as new gametocides for hybrid seed production. This study also examined the possibility of external application of these gametocides on several unstable Polima cytoplasmic male sterile and thermosensitive genic male sterile lines. Although the outcrossed seed set of the treated lines was slightly reduced, the gametocide application significantly increased the seed purity of the resulting hybrid. CONCLUSION:The finding of the gametocidal effects of most sulfonylureas and imidazolinones are of great importance for developing new functions for ALS-inhibiting herbicides. The application of gametocides will also greatly promote the safe utilization of environment-sensitive male sterility in hybrid seed production. Unexpectedly, the application of three triazolopyrimidines (florasulam, flumetsulam, and penoxsulam) and one pyrimidinylthiobenzoate (bispyribac-sodium) did not cause male sterility, although these herbicides obviously inhibited the activity of ALS and plant growth. This result suggests that inhibition of ALS activity does not always lead to male sterility in plants, and these gametocides may also inhibit other biological functions vital for microspore development.

Project description:Nitrogen is essential for plant growth, and N fertilization allows farmers to obtain high yields and produce sufficient agricultural commodities. On the other hand, nitrogen losses potentially cause adverse effects to ecosystems and to human health. Increasing nitrogen use efficiency (NUE) is vital to solve the conflict between productivity, to secure the demand of a growing world population, and the protection of the environment. To ensure this, genetic improvement is considered to be a paramount aspect toward ecofriendly crop production. Winter oilseed rape (Brassica napus L.) is the second most important oilseed crop in the world and is cultivated in many regions across the temperate zones. To our knowledge, this study reports the most comprehensive field-based data generated to date for an empirical evaluation of genetic improvement in winter oilseed rape varieties under two divergent nitrogen fertilization levels (NFLs). A collection of 30 elite varieties registered between 1989 and 2014, including hybrids and open pollinated varieties, was tested in a 2-year experiment in 10 environments across Germany for changes in seed yield and seed quality traits. Furthermore, NUE was calculated. We observed a highly significant genetics-driven increase in seed yield per-se and, thus, increased NUE at both NFLs. On average, seed yield from modern open-pollinated varieties and modern hybrids was higher than from old open-pollinated varieties and old hybrids. The annual yield progress across all tested varieties was ~35 kg ha-1 year-1 at low nitrogen and 45 kg ha-1 year-1 under high nitrogen fertilization. Furthermore, in modern varieties an increased oil concentration and decreased protein concentration was observed. Despite, the significant effects of nitrogen fertilization, a surprisingly low average seed yield gap of 180 kg N ha-1 was noted between high and low nitrogen fertilization. Due to contrary effects of N fertilization on seed yield per-se and seed oil concentration an oil yield of 2.04 t ha-1 was measured at both N levels. Collectively, the data reveal that genetic improvement through modern breeding techniques in conjunction with reduced N fertilizer inputs has a tremendous potential to increase NUE of oilseed rape.
