New observations and insights into the morphology and growth kinetics of hydrate films.

ABSTRACT: The kinetics of film growth of hydrates of methane, ethane, and methane-ethane mixtures were studied by exposing a single gas bubble to water. The morphologies, lateral growth rates, and thicknesses of the hydrate films were measured for various gas compositions and degrees of subcooling. A variety of hydrate film textures was revealed. The kinetics of two-dimensional film growth was inferred from the lateral growth rate and initial thickness of the hydrate film. A clear relationship between the morphology and film growth kinetics was observed. The shape of the hydrate crystals was found to favour heat or mass transfer and favour further growth of the hydrate film. The quantitative results on the kinetics of film growth showed that for a given degree of subcooling, the initial film thicknesses of the double hydrates were larger than that of pure methane or ethane hydrate, whereas the thickest hydrate film and the lowest lateral growth rate occurred when the methane mole fraction was approximately 0.6.

Project description:Background: Research in chlamydial genetics is challenging because of its obligate intracellular developmental cycle. In vivo systems exist that allow studies of different aspects of basic biology of chlamydiae, the murine Chlamydia muridarum model is one of great importance and thus an essential research tool. C. muridarum carries a plasmid that has a role in virulence. Our aim was to compare and contrast the C. muridarum plasmid-free phenotype with that of a chromosomally isogenic plasmid-bearing strain, through the inclusion phase of the developmental cycle. Methods: We measured infectivity for plasmid bearing and plasmid-cured C. muridarum by inclusion forming assays in McCoy cells and in parallel bacterial chromosome replication by quantitative PCR, throughout the developmental cycle. In addition to these studies, we have carefully monitored chlamydial inclusion formation by confocal microscopy and transmission electron microscopy. A new E.coli/chlamydial shuttle vector (pNigg::GFP) was constructed using standard cloning technology and used to transform C. muridarum for further phenotypic studies. Results: We have advanced the definition of the chlamydial phenotype away from the simple static observation of mature inclusions and redefined the C. muridarum plasmid-based phenotype on growth profile and inclusion morphology. Our observations on the growth properties of plasmid-cured C. muridarum challenge the established interpretations, especially with regard to inclusion growth kinetics. Introduction of the shuttle plasmid pNigg::GFP into plasmid-cured C. muridarum restored the wild-type plasmid-bearing phenotype and confirmed that loss of the plasmid was the sole cause for the changes in growth and chromosomal replication. Conclusions: Accurate growth curves and sampling at multiple time points throughout the developmental cycle is necessary to define plasmid phenotypes. There are subtle but important (previously unnoticed) differences in the overall growth profile of plasmid-bearing and plasmid-free C. muridarum. We have proven that the differences described are solely due to the plasmid pNigg.

Project description:Natural methane hydrates are estimated to be the largest source of unexploited hydrocarbon fuel. The ideal conditions for methane hydrate formation are low temperatures and high pressures. On the other hand, recent experimental studies suggest that porous materials, thanks to their confinement effects, can enable methane hydrate formation at milder conditions, although there has not been a consensus on this. A number of studies have investigated methane hydrate growth in confinement by employing molecular simulations; however, these were carried out at either very high pressures or very low temperatures. Therefore, the effects of confinement on methane hydrate growth at milder conditions have not yet been elaborated by molecular simulations. In order to address this, we carried out a systematic study by performing molecular dynamics (MD) simulations of methane water systems. Using a direct phase coexistence approach, microsecond-scale MD simulations in the isobaric-isothermal (NPT) ensemble were performed in order to study the behavior of methane hydrates in the bulk and in confined nanospaces of hydroxylated silica pores at external pressures ranging from 1 to 100 bar and a simulation temperature corresponding to a 2 °C experimental temperature. We validated the combination of the TIP4P/ice water and TraPPE-UA methane models in order to correctly predict the behavior of methane hydrates in accordance to their phase equilibria. We also demonstrated that the dispersion corrections applied to short-range interactions lead to artificially induced hydrate growth. We observed that in the confinement of a hydroxylated silica pore, a convex-shaped methane nanobuble forms, and methane hydrate growth primarily takes place in the center of the pore rather than the surfaces where a thin water layer exists. Most importantly, our study showed that in the nanopores methane hydrate growth can indeed take place at pressures which would be too low for the growth of methane hydrates in the bulk.
