Project description:Four new limonoids, dysohainanins A–D (1–4), and two new triterpenoids, dysohainanins E and F (5 and 6), together with seven known ones were isolated from the twigs and leaves of Dysoxylum hainanense Merr. The structures of the new compounds were determined by a variety of spectroscopic methods. The cytotoxic activities of these compounds were evaluated, and the known compound ent-19-nor-4,16,18-trihydroxy-8(14)-pomaren-15-one (13) showed in vitro cytotoxicity against HL-60, A-549, MCF-7, and SW480 cells, with IC50 values of 24.3, 28.1, 30.7, and 22.5 µM, respectively. Compounds 2 and 3 were tested their insecticidal activities using brine shrimp and both of them were inactive. Project description:Phytochemical investigation of the twigs of Podocarpus nagi (Podocarpaceae) led to the isolation of two new abietane-type diterpenoids, named 1?,16-dihydroxylambertic acid (1) and 3?,16-dihydroxylambertic acid (2), along with two new ent-pimarane-type diterpenoids, named ent-2?,15,16,18-tetrahydroxypimar-8(14)-ene (3) and ent-15-oxo-2?,16,18-trihydroxypimar-8(14)-ene (4). Their respective structures were elucidated on the basis of spectroscopic analyses, including 1D- and 2D-NMR, IR, CD, and HR-ESI-MS. This is the first time ent-pimarane-type diterpenoids from the genus Podocarpus has been reported. All four new compounds were tested for cytotoxic activity. The MTT assay results showed that compounds 3 and 4 significantly inhibited the proliferation of human cervical cancer Hela cells, human lung cancer A549 cells, and human breast cancer MCF-7 cells at a concentration of 10 ?M. Furthermore, using the lipopolysaccharide (LPS)-stimulated RAW264.7 cells, compounds 2 and 4 were found to significantly inhibit nitrogen oxide (NO) production with IC50 values of 26.5 ± 6.1 and 17.1 ± 1.5 ?M, respectively.

Project description:Cytotoxic flavonoids of Murraya tetramera were investigated in this study. A novel flavonoid and twelve known flavonoids, including seven flavones (1-7), three flavanones (8-10), and three chalcones (11-13) were isolated from the leaves and twigs of Murraya tetramera. Chemical structures were elucidated by NMR combined with MS spectral analysis, and the new compound (6) was confirmed as 3',5'-dihydroxy-5,6,7,4'-tetramethoxyflavone. Furthermore, all the isolated flavonoids were evaluated for their cytotoxicities against murine melanoma cells (B16), and human breast cancer cells (MDA-MB-231) by CCK-8 assay. Among them, compounds 7, 13, and 5 exhibited potent cytotoxic activities against B16 cell lines (IC50 = 3.87, 7.00 and 8.66 μg/mL, respectively). Compounds 5, 13, and 12 displayed potent cytotoxicities against MDA-MB-231 cell lines (IC50 = 3.80, 5.95 and 7.89 μg/mL, respectively). According to the correlation of the structure and activity analysis, 5-hydroxyl and 8-methoxyl substituents of the flavone, 8-methoxyl substituent of the flavanone, and 3',5'-methoxyl substituents of the chalcone could be critical factors of the high cytotoxicity. The results indicated that the active flavonoids have potential to be developed as leading compounds for treating cancers.

Project description:Guayule (Parthenium argentatum) is one of the most promising alternatives to produce natural rubber. As the guayule leaves represent a waste byproduct in the rubber extraction process, their exploitation might contribute to the valorization of the whole plant biomass. The specific aim of our study was to determine the antioxidant capacity and the content of phenolic compounds in leaves and twigs of different guayule lines cultivated in a Mediterranean environment. The antioxidant capacity and the contents of phenolic were affected significantly by guayule accession and harvest time. Overall means in twigs represented about 60% of the corresponding leaf values for antioxidant capacity as DPPH (1,1-diphenyl-2-picrylhydrazyl) and not tannic phenolic and about 55% as ABTS (2,2'-azinobis (3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt), total phenolic, tannic phenolic and total flavonoid contents. Five individual phenolic compounds were identified in methanolic extracts of leaves. Neochlorogenic acid (62.5-174.8 mg g-1 DW) and chlorogenic acid (28.6-41.1 mg g-1 DW) were the most abundant phenolic acids. In addition to the compounds quantified in leaves, cynarin was identified only in twigs and for the first time in guayule biomass. Our results indicate that guayule leaves and twigs represent a rich source of antioxidants for potential applications in fodder, nutraceutical and pharmacological sectors.

Project description:Two new seco-labdane diterpenoids, nudiflopene N (1) and nudiflopene O (2), and four known compounds were isolated from the leaves of Callicarpa nudiflora. The structures of the new compounds were established by 1D-, 2D-NMR, and HR-ESI-MS spectral analyses. Compounds 1-3 showed inhibitory activities on lipopolysaccharide-induced nitric oxide (NO) production in RAW264.7 cells, and new compounds 1-2 exhibited more potent inhibitory activity than compound 3. The cytotoxicity of compounds 1-3 against human hepatocellular carcinoma HepG2 cells and human gastric carcinoma SGC-7901 cells were evaluated, while all of them exhibited no cytotoxicity.

Project description:A new non-cytotoxic [(+)-17?-hydroxystrebloside (1)] and two known cytotoxic [(+)-3'-de-O-methylkamaloside (2) and (+)-strebloside (3)] cardiac glycosides were isolated and identified from the combined flowers, leaves, and twigs of Streblus asper collected in Vietnam, with the absolute configuration of 1 established from analysis of its ECD and NMR spectroscopic data and confirmed by computational ECD calculations. A new 14,21-epoxycardanolide (3a) was synthesized from 3 that was treated with base. A preliminary structure-activity relationship study indicated that the C-14 hydroxy group and the C-17 lactone unit and the established conformation are important for the mediation of the cytotoxicity of 3. Molecular docking profiles showed that the cytotoxic 3 and its non-cytotoxic analogue 1 bind differentially to Na+/K+-ATPase. Compound 3 docks deeply in the Na+/K+-ATPase pocket with a sole pose, and its C-10 formyl and C-5, C-14, and C-4' hydroxy groups may form hydrogen bonds with the side-chains of Glu111, Glu117, Thr797, and Arg880 of Na+/K+-ATPase, respectively. However, 1 fits the cation binding sites with at least three different poses, which all depotentiate the binding between 1 and Na+/K+-ATPase. Thus, 3 was found to inhibit Na+/K+-ATPase, but 1 did not. In addition, the cytotoxic and Na+/K+-ATPase inhibitory 3 did not affect glucose uptake in human lung cancer cells, against which it showed potent activity, indicating that this cardiac glycoside mediates its cytotoxicity by targeting Na+/K+-ATPase but not by interacting with glucose transporters.

Project description:Tyrosine kinase (TK), vascular endothelial growth factor (VEGF), and matrix metalloproteinases (MMP) are important cancer therapeutic target proteins. Based on reported anti-cancer and cytotoxic activities of Caesalpinia bonduc, this study isolated phytochemicals from young twigs and leaves of C bonduc and identified the interaction between them and cancer target proteins (TK, VEGF, and MMP) in silico. AutoDock Vina, iGEMDOCK, and analysis of pharmacokinetic and pharmacodynamic properties of the isolated bioactives as therapeutic molecules were performed. Seven phytochemicals (7-hydroxy-4'-methoxy-3,11-dehydrohomoisoflavanone, 4,4'-dihydroxy-2'-methoxy-chalcone, 7,4'-dihydroxy-3,11-dehydrohomoisoflavanone, luteolin, quercetin-3-methyl, kaempferol-3-O-?-d-xylopyranoside and kaempferol-3-O-?-l-rhamnopyranosyl-(1???2)-?-D-xylopyranoside) were isolated. Molecular docking analysis showed that the phytochemicals displayed strong interactions with the proteins compared with their respective drug inhibitors. Pharmacokinetic and pharmacodynamic properties of the compounds were promising suggesting that they can be developed as putative lead compounds for developing new anti-cancer drugs.

Project description:One new neolignan, racelactone A (1), together with seven known compounds (2-8) were isolated from the methanolic extract of the leaves and twigs of Lumnitzera racemosa. The structure of racelactone A (1) was determined on the basis of the mass and NMR spectroscopic data interpretation. With respect to bioactivity, compound 1 displayed an anti-angiogenic effect by suppressing tube formation. Furthermore, compounds 1, 4, and 5 showed significant anti-inflammatory effects with IC50 values of 4.95 ± 0.89, 1.95 ± 0.40, and 2.57 ± 0.23 ?M, respectively. The plausible biosynthesis pathway of racelactone A (1) was proposed.
