Complete genome sequences of a cytopathic/noncytopathic pair of border disease viruses.

ABSTRACT: The complete genome sequences of a cytopathic/noncytopathic pair of border disease viruses (BDV) that were isolated from a sheep with mucosal disease-like lesions were determined. The cytopathic virus possessed an insert of 288 nucleotides that was derived from the Jiv cellular transcript and had 37 nucleotide differences from the noncytopathic virus.

Cytopathic and noncytopathic interferon responses in cells expressing hepatitis C virus subgenomic replicons.

Project description:Hepatitis C virus (HCV) is the only known positive-stranded RNA virus that causes persistent lifelong infections in humans. Accumulation of HCV RNA can be inhibited with alpha interferon (IFN-alpha) in vivo and in culture cells. We used cell-based assay systems to investigate the mechanisms responsible for the cytokine-induced inhibition of HCV replication. The results showed that IFN-alpha could suppress the accumulation of viral RNA by a noncytopathic pathway and could also induce apoptosis of virally infected cells in a concentration- and cell line-dependent fashion. Whereas the noncytopathic IFN-alpha response depended on a functional Jak-STAT signal transduction pathway, it did not appear to require double-stranded RNA-dependent pathways. Moreover, we found that functional proteasomes were required for establishment of the IFN-alpha response against HCV. Based on the results described in this study we propose a model for the mechanism by which IFN-alpha therapy suppresses HCV replication in chronic infections by both cytopathic and noncytopathic means.

First Complete Genome Sequences of Two Keystone Viruses from Florida.

Project description:We report here the first complete sequences of two Keystone virus (KEYV) genomes isolated from Florida in 2005, which include the first two publicly available complete large (L) gene sequences. The sequences of the KEYV L segments show 75.99 to 83.86% nucleotide similarity with those of other viruses in the California (CAL) serogroup of bunyaviruses.

Project description:Here, we isolated two chikungunya virus strains (SZ1050 and SZ1239) from patients infected with chikungunya virus who were returning to China from India and Indonesia, respectively. Strain SZ1050 was classed in the Indian Ocean lineage, and strain SZ1239 belonged to the Asian lineage. No A226V mutation in the envelope protein E1 was found in either strain.

Seven of the eight segments were most closely related to H16 and H13 isolates from eastern North America and Iceland. Of note is a C-terminal truncation of the nonstructural 1 (NS1) protein in one of the isolates that is usually found in swine H1N1 virus.
