Project description:In the past decade, the fusion between diffusion magnetic resonance imaging (dMRI) and functional magnetic resonance imaging (fMRI) has opened the way for exploring structure-function relationships in vivo. As it stands, the common approach usually consists of analysing fMRI and dMRI datasets separately or using one to inform the other, such as using fMRI activation sites to reconstruct dMRI streamlines that interconnect them. Moreover, given the large inter-individual variability of the healthy human brain, it is possible that valuable information is lost when a fixed set of dMRI/fMRI analysis parameters such as threshold values are assumed constant across subjects. By allowing one to modify such parameters while viewing the results in real-time, one can begin to fully explore the sensitivity of structure-function relations and how they differ across brain areas and individuals. This is especially important when interpreting how structure-function relationships are altered in patients with neurological disorders, such as the presence of a tumor. In this study, we present and validate a novel approach to achieve this: First, we present an interactive method to generate and visualize tractography-driven resting-state functional connectivity, which reduces the bias introduced by seed size, shape and position. Next, we demonstrate that structural and functional reconstruction parameters explain a significant portion of intra- and inter-subject variability. Finally, we demonstrate how our proposed approach can be used in a neurosurgical planning context. We believe this approach will promote the exploration of structure-function relationships in a subject-specific aspect and will open new opportunities for connectomics.

Project description:BACKGROUND:Drifts between wakefulness and sleep are common during resting state functional MRI (rsfMRI). Among healthy adults, within-scanner sleep can impact functional connectivity of default mode (DMN), task-positive (TPN), and thalamo-cortical networks. Because dysfunctional arousal states (i.e., sleepiness, sleep disturbance) are common in affective disorders, individuals with affective psychopathology may be more prone to unstable wakefulness during rsfMRI, hampering the estimation of clinically meaningful functional connectivity biomarkers. METHODS:A transdiagnostic sample of 150 young adults (68 psychologically distressed; 82 psychiatrically healthy) completed rsfMRI and reported whether they experienced within-scanner sleep. Symptom scales were reduced into depression/anxiety and mania proneness dimensions using principal component analysis. We evaluated associations between within-scanner sleep, clinical status, and functional connectivity of the DMN, TPN, and thalamus. RESULTS:Within-scanner sleep during rsfMRI was reported by 44% of participants (n?=?66) but was unrelated to psychiatric diagnoses or mood symptom severity (p-values > 0.05). Across all participants, self-reported within-scanner sleep was associated with connectivity signatures akin to objectively-assessed sleep, including lower within-DMN connectivity, lower DMN-TPN anti-correlation, and altered thalamo-cortical connectivity (p < 0.05, corrected). Among participants reporting sustained wakefulness (n?=?84), depression/anxiety severity positively associated with averaged DMN-TPN connectivity and mania proneness negatively associated with averaged thalamus-DMN connectivity (p-values < 0.05). Both relationships were attenuated and became non-significant when participants reporting within-scanner sleep were included (p-values > 0.05). LIMITATIONS:Subjective report of within-scanner sleep. CONCLUSIONS:Findings implicate within-scanner sleep as a source of variance in network connectivity; careful monitoring and correction for within-scanner sleep may enhance our ability to characterize network signatures underlying affective psychopathology.

Project description:Confirming the presence (or absence) of dynamic functional connectivity (dFC) states during rest is an important open question in the field of cognitive neuroscience. The prevailing dFC framework aims to identify dynamics directly from connectivity estimates with a sliding window approach, however this method suffers from several drawbacks including sensitivity to window size and poor test-retest reliability. We hypothesize that time-varying changes in functional connectivity are mirrored by significant temporal changes in functional activation, and that this coupling can be leveraged to study dFC without the need for a predefined sliding window. Here, we introduce a data-driven dFC framework, which involves informed segmentation of fMRI time series at candidate FC state transition points estimated from changes in whole-brain functional activation, rather than a fixed-length sliding window. We show our approach reliably identifies true cognitive state change points when applied on block-design working memory task data and outperforms the standard sliding window approach in both accuracy and computational efficiency in this context. When applied to data from four resting state fMRI scanning sessions, our method consistently recovers five reliable FC states, and subject-specific features derived from these states show significant correlation with behavioral phenotypes of interest (cognitive ability, personality). Overall, these results suggest abrupt whole-brain changes in activation can be used as a marker for changes in connectivity states and provides new evidence for the existence of time-varying FC in rest.

Project description:Delirium is characterized by inattention and other cognitive deficits, symptoms that have been associated with disturbed interactions between remote brain regions. Recent EEG studies confirm that disturbed global network topology may underlie the syndrome, but lack an anatomical basis. The aim of this study was to increase our understanding of the global organization of functional connectivity during delirium and to localize possible alterations. Resting-state fMRI data from 44 subjects were analyzed, and motion-free data were available in nine delirious patients, seven post delirium patients and thirteen non-delirious clinical controls. We focused on the functional network backbones using the minimum spanning tree, which allows unbiased network comparisons. During delirium a longer diameter (mean (M)?=?0.30, standard deviation (SD)?=?0.05, P?=?.024) and a lower leaf fraction (M?=?0.32, SD?=?0.03, P?=?.027) was found compared to the control group (M?=?0.28, SD?=?0.04 respectively M?=?0.35, SD?=?0.03), suggesting reduced functional network integration and efficiency. Delirium duration was strongly related to loss of network hierarchy (rho?=?-0.92, P?=?.001). Connectivity strength was decreased in the post delirium group (M?=?0.16, SD?=?0.01) compared to the delirium group (M?=?0.17, SD?=?0.03, P?=?.024) and the control group (M?=?0.19, SD?=?0.02, P?=?.001). Permutation tests revealed a decreased degree of the right posterior cingulate cortex during delirium and complex regional alterations after delirium. These findings indicate that delirium reflects disintegration of functional interactions between remote brain areas and suggest long-term impact after the syndrome resolves.

Project description:Socio-affective touch communication conveys a vast amount of information about emotions and intentions in social contexts. In spite of the complexity of the socio-affective touch expressions we use daily, previous studies addressed only a few aspects of social touch mainly focusing on hedonics, such as stroking, leaving a wide range of social touch behaviour unexplored. To overcome this limit, we present the Socio-Affective Touch Expression Database (SATED), which includes a large range of dynamic interpersonal socio-affective touch events varying in valence and arousal. The original database contained 26 different social touch expressions each performed by three actor pairs. To validate each touch expression, we conducted two behavioural experiments investigating perceived naturalness and affective values. Based on the rated naturalness and valence, 13 socio-affective touch expressions along with 12 corresponding non-social touch events were selected as a complete set, achieving 75 video clips in total. Moreover, we quantified motion energy for each touch expression to investigate its intrinsic correlations with perceived affective values and its similarity among actor- and action-pairs. As a result, the touch expression database is not only systematically defined and well-controlled, but also spontaneous and natural, while eliciting clear affective responses. This database will allow a fine-grained investigation of complex interpersonal socio-affective touch in the realm of social psychology and neuroscience along with potential application areas in affective computing and neighbouring fields.

Project description:BACKGROUND:Previous studies have demonstrated that patients with treatment-resistant depression (TRD) and treatment-sensitive depression (TSD) differed at neural level. However, it remains unclear if these two subtypes of depression differ in the interhemispheric coordination. This study was undertaken for two purposes: (1) to explore the differences in interhemispheric coordination between these two subtypes by using the voxel-mirrored homotopic connectivity (VMHC) method; and (2) to determine if the difference of interhemispheric coordination can be used as a biomarker(s) to differentiate TRD from both TSD and healthy subjects (HS). METHODS:Twenty-three patients with TRD, 22 with TSD, and 19 HS participated in the study. Data of these participants were analyzed with the VMHC and seed-based functional connectivity (FC) approaches. RESULTS:Compared to the TSD group, the TRD group showed significantly lower VMHC values in the calcarine cortex, fusiform gyrus, hippocampus, superior temporal gyrus, middle cingulum, and precentral gyrus. Lower VMHC values were also observed in the TRD group in the calcarine cortex relative to the HS group. However, the TSD group had no significant change in VMHC value in any brain region compared to the HS group. Receiver operating characteristic curves (ROC) analysis revealed that the VMHC values in the calcarine cortex had discriminatory function distinguishing patients with TRD from patients with TSD as well as those participants in the HS group. CONCLUSIONS:Lower VMHC values of patients with TRD relative to those with TSD and those in the HS group in the calcarine cortex appeared to be a unique feature for patients with TRD and it may be used as an imaging biomarker to separate patients with TRD from those with TSD or HS.

Project description:Novel methods for measuring large-scale dynamic brain organisation are needed to provide new biomarkers of schizophrenia. Using a method for modelling dynamic modular organisation (Mucha et al., 2010), evidence suggests higher 'flexibility' (switching between multilayer network communities) to be a feature of schizophrenia (Braun et al., 2016). The current study compared flexibility between 55 patients with schizophrenia and 72 controls (the COBRE Dataset). In addition, novel methods of 'between resting state network synchronisation' (BRSNS) and the probability of transition from one community to another were used to further describe group differences in dynamic community structure. There was significantly higher schizophrenia group flexibility scores in cerebellar (F (1124) = 9.33, p (FDR) = 0.017), subcortical (F (1124) = 13.14, p (FDR) = 0.005), and fronto-parietal task control (F (1124) = 7.19, p (FDR) = 0.033) resting state networks (RSNs), as well as in the left thalamus (MNI XYZ: -2, -13, 12; F(1, 124) = 17.1, p (FDR) < 0.001) and the right crus I (MNI XYZ: 35, -67, -34; F (1, 124) = 19.65, p (FDR) < 0.001). Flexibility in the left thalamus reflected transitions between communities covering default mode and sensory-somatomotor RSNs. BRSNS scores suggested altered dynamic inter-RSN modular configuration in schizophrenia. This study suggests less stable community structure in a schizophrenia group at an RSN and node level and provides novel methods of exploring dynamic community structure. Mediation of group differences by mean time window correlation did however suggest flexibility to be no better as a schizophrenia biomarker than simpler measures and a range of methodological choices affected results.

Project description:In graph theory, "multilayer networks" represent systems involving several interconnected topological levels. A neuroscience example is the hierarchy of connections between different cortical depths or "lamina". This hierarchy is becoming non-invasively accessible in humans using ultra-high-resolution functional MRI (fMRI). Here, we applied multilayer graph theory to examine functional connectivity across different cortical depths in humans, using 7T fMRI (1-mm3 voxels; 30 participants). Blood oxygenation level dependent (BOLD) signals were derived from five depths between the white matter and pial surface. We then compared networks where the inter-regional connections were limited to a single cortical depth only ("layer-by-layer matrices") to those considering all possible connections between regions and cortical depths ("multilayer matrix"). We utilized global and local graph theory features that quantitatively characterize network attributes such as network composition, nodal centrality, path-based measures, and hub segregation. Detecting functional differences between cortical depths was improved using multilayer connectomics compared to the layer-by-layer versions. Superficial aspects of the cortex dominated information transfer and deeper aspects clustering. These differences were largest in frontotemporal and limbic brain regions. fMRI functional connectivity across different cortical depths may contain neurophysiologically relevant information. Multilayer connectomics could provide a methodological framework for studies on how information flows across this hierarchy.

Project description:Concurrent EEG and fMRI acquisitions in resting state showed a correlation between EEG power in various bands and spontaneous BOLD fluctuations. However, there is a lack of data on how changes in the complexity of brain dynamics derived from EEG reflect variations in the BOLD signal. The purpose of our study was to correlate both spectral patterns, as linear features of EEG rhythms, and nonlinear EEG dynamic complexity with neuronal activity obtained by fMRI. We examined the relationships between EEG patterns and brain activation obtained by simultaneous EEG-fMRI during the resting state condition in 25 healthy right-handed adult volunteers. Using EEG-derived regressors, we demonstrated a substantial correlation of BOLD signal changes with linear and nonlinear features of EEG. We found the most significant positive correlation of fMRI signal with delta spectral power. Beta and alpha spectral features had no reliable effect on BOLD fluctuation. However, dynamic changes of alpha peak frequency exhibited a significant association with BOLD signal increase in right-hemisphere areas. Additionally, EEG dynamic complexity as measured by the HFD of the 2-20 Hz EEG frequency range significantly correlated with the activation of cortical and subcortical limbic system areas. Our results indicate that both spectral features of EEG frequency bands and nonlinear dynamic properties of spontaneous EEG are strongly associated with fluctuations of the BOLD signal during the resting state condition.

Project description:The physiological adaptive regulation of healthy population with a high fitness level is associated with enhanced cognitive control in brain. This study further investigated the effects of different levels of sporting experience on intrinsic brain networks involved in central autonomic processing using resting-state functional magnetic resonance imaging. We explored functional connectivity of four core regions within central autonomic network (CAN), namely posterior midcingulate cortex (pMCC), left amygdala (AMYG), and right anterior (aINS) and left posterior insular cortices, in advanced and intermediate baseball players, and compared their strength of connectivity with individuals without baseball-playing experience. Functional connectivity maps across three groups confirmed a close relationship between CAN and large-scale brain networks in sensory, motor and cognitive domains. Crucially, both advanced and intermediate batters demonstrated enhanced connectivity between pMCC and sensorimotor network, between right aINS and dorsal anterior cingulate cortex, and between left AMYG and right putamen, than controls. These results reflected a stronger interregional coupling in sensorimotor and cognitive control, and in motor skill consolidation. In conclusion, we provided evidence that different levels of sporting experience could reorganize/enhance intrinsic functional connectivity for central autonomic processing.
