The Juquitiba virus, an agent of Hantavirus Cardiopulmonary Syndrome, is one of the most widely distributed hantavirus found in South America. It has been detected in Oligoryzomys nigripes, Akodon montensis, Oxymycterus judex, Akodon paranaensis in Brazil and in O. nigripes, Oryzomys sp. and Oligoryzomys fornesi rodents in Argentine, Paraguay and Uruguay. Project description:Nutrients are known to limit productivity of plant communities around the world. In the Brazilian Cerrado, indirect evidences point to phosphorus as the main limiting nutrient, but some fertilization experiments suggest that one or more micronutrients might play this role. Boron is one of the essential micronutrients for plants. Agronomically, it received some attention, but it has mostly been neglected in ecological studies assessing the effects of nutrients on plant growth. Through field fertilization and mesocosm experiments in a degraded area in the Cerrado, we show that boron addition increased biomass production of herbaceous vegetation. This could be related to a lower aluminum uptake in the boron fertilized plants. Even considering that plant growth was promoted by boron addition due to aluminum toxicity alleviation, this is the first study reporting boron limitation in natural, noncultivated plant communities and also the first report of this kind in vegetative grasses. These results contribute to disentangling patterns of nutrient limitation among plant species of the species-rich, aluminum-rich, and nutrient-poor Cerrado biome and highlight the potential role of micronutrients, such as boron, for growth of noncrop plants. Understanding how nutrient limitation differs among functional groups in the highly biodiverse areas founded on ancient tropical soils may help managing these plant communities in a changing world.

Project description:Cytogenetic data can be useful for taxonomic and phylogenetic studies, as well as to provide information about chromosome evolution. Therefore, it may help design conservation priorities for some threatened species, such as anurans. Herein, we describe the karyotypes of Scinax constrictus and Ololygon centralis, native endemic species from the Brazilian Cerrado. Chromosome preparations for both species were stained with Giemsa for morphological analyses and then impregnated by the Ag-NOR method for localization of the nucleolar organizer region (NOR). Both species had 24 chromosomes, as confirmed by meiotic analyses, which showed 12 bivalents. Chromosome morphologies presented the same pattern for Scinax and Ololygon compared to species already karyotyped in both genera. The NOR was interstitially located in the long arm of pair 7 in S. constrictus, whereas in O. centralis it was found near the centromere in the long arm of pair 1, thus diverging from what is commonly found for other Ololygon species. Therefore, we provide the first description of the karyotype of O. centralis and the first report of the localization of the NOR for the karyotype of both species. Our study increases the cytogenetic knowledge in species of the genera Scinax and Ololygon, and provide support for further studies on the taxonomy, ecology, and evolution of hylid anurans.

Project description:We used 16S rRNA sequencing to assess the archaeal communities across a gradient of Cerrado. The archaeal communities differed across the gradient. Crenarcheota was the most abundant phyla, with Nitrosphaerales and NRPJ as the predominant classes. Euryachaeota was also found across the Cerrado gradient, including the classes Metanocellales and Methanomassiliicoccaceae.

Project description:Pesticides aerial application may results in the drift to neighboring areas if some application technology is not well executed. This phenomenon should be minimized to reduce environmental risks and agricultural production costs. This work aimed to investigate the interaction of environmental conditions, surrounding distance, and application conditions influencing spray drift in aerial applications. Sampling data from aerial sprays were collected during three agricultural years (from 2012 to 2014) in fields cultivated with sorghum, millet, soybean, corn, and cotton. The following variables were evaluated: application swath width, application rate, distance from the applied field, wind speed, relative humidity, and temperature. The estimated Pearson's correlations and path analysis identified that application rate and distance from the applied field and application were the variables that most influenced drift. Equations relating spray drift in function of distance from the applied field and application rate were adjusted in function of the variable, and a response surface model was constructed to estimate drift. The major pesticide class sprayed with aircraft in the Brazilian Cerrado was insecticide, followed by fungicide. This scenario shows the potential hazard risk of spray drift over the environment. The concentration of the drift deposits decreased as surrounding distance and application rate were increased. A mathematical equation of drift prediction was established, where the variables that contributed most to drift deposits were surrounding distance and wind speed. Thus, it is very important to monitor and respect the wind speed limits during the aerial spraying, mainly when there is any risk potential associated with pesticide exposure over the downwind direction.

Project description:The Araguaia River basin runs through the states of Goiás, Mato Grosso, Tocantins, and Pará, covering 373,000 Km(2), mostly within the Brazilian Cerrado. The region has a wide variety of wetlands. The climate is characterized by high temperatures and strongly seasonal precipitation. There are two well defined seasons: the dry season (winter-spring) and the rainy season (summer- fall). The Araguaia River basin is dominated by plinthosoils that are found in low flat areas, poorly drained and prone to flooding, yielding wetland habitats of high plant diversity. Since the 1970s, human activities have led to reduction in both the diversity and area of wetlands. The construction of the Belém-Brasília highway and hydroelectric dams, as well as the expansion of agricultural and mining activities, have had major impacts on the region.The flora diversity data of the Araguaia River basin was developed through field work, herbarium research, and use of a database (Species Link). The resulting checklist of 162 aquatic and marshy monocotyledons from the Araguaia River basin represents 20 families and 50 genera. Cyperaceae (51 spp.), Poaceae (39 spp.), and Eriocaulaceae (16 spp.) are the most representative families. Life form analysis indicates that helophytes predominate (98 spp.; 60.5%). One hundred one species are native to tropical and/or subtropical America and twenty one are endemic to Brazil. Ninety-three species are new occurrences for the Araguaia River basin. Among them, three species are reported in the Brazilian Cerrado for the first time. This work contributes to the understanding of aquatic plant diversity in the Cerrado and other savanna-like vegetation physiognomies; environments and habitats poorly understood taxonomically and undercollected generally.

Project description:Prior to the discovery of the mimivirus in 2003, viruses were thought to be physically small and genetically simple. Mimivirus, with its ~750-nm particle size and its ~1.2-Mbp genome, shattered these notions and changed what it meant to be a virus. Since this discovery, the isolation and characterization of giant viruses has exploded. One of the more recently discovered giant viruses, Samba virus, is a Mimivirus that was isolated from the Rio Negro in the Brazilian Amazon. Initial characterization of Samba has revealed some structural information, although the preparation techniques used are prone to the generation of structural artifacts. To generate more native-like structural information for Samba, we analyzed the virus through cryo-electron microscopy, cryo-electron tomography, scanning electron microscopy, and fluorescence microscopy. These microscopy techniques demonstrated that Samba particles have a capsid diameter of ~527 nm and a fiber length of ~155 nm, making Samba the largest Mimivirus yet characterized. We also compared Samba to a fiberless mimivirus variant. Samba particles, unlike those of mimivirus, do not appear to be rigid, and quasi-icosahedral, although the two viruses share many common features, including a multi-layered capsid and an asymmetric nucleocapsid, which may be common amongst the Mimiviruses.

Project description:Conserving animals beyond protected areas is critical because even the largest reserves may be too small to maintain viable populations for many wide-ranging species. Identification of landscape features that will promote persistence of a diverse array of species is a high priority, particularly, for protected areas that reside in regions of otherwise extensive habitat loss. This is the case for Emas National Park, a small but important protected area located in the Brazilian Cerrado, the world's most biologically diverse savanna. Emas Park is a large-mammal global conservation priority area but is too small to protect wide-ranging mammals for the long-term and conserving these populations will depend on the landscape surrounding the park. We employed novel, noninvasive methods to determine the relative importance of resources found within the park, as well as identify landscape features that promote persistence of wide-ranging mammals outside reserve borders. We used scat detection dogs to survey for five large mammals of conservation concern: giant armadillo (Priodontes maximus), giant anteater (Myrmecophaga tridactyla), maned wolf (Chrysocyon brachyurus), jaguar (Panthera onca), and puma (Puma concolor). We estimated resource selection probability functions for each species from 1,572 scat locations and 434 giant armadillo burrow locations. Results indicate that giant armadillos and jaguars are highly selective of natural habitats, which makes both species sensitive to landscape change from agricultural development. Due to the high amount of such development outside of the Emas Park boundary, the park provides rare resource conditions that are particularly important for these two species. We also reveal that both woodland and forest vegetation remnants enable use of the agricultural landscape as a whole for maned wolves, pumas, and giant anteaters. We identify those features and their landscape compositions that should be prioritized for conservation, arguing that a multi-faceted approach is required to protect these species.

Project description:The Cerrado biome corresponds to an extensive area of Brazil and is considered a biodiversity hotspot. Frequent fires are a natural feature in this biome and have influences on vegetation structure and composition. However, continuous anthropogenic actions are promoting changes in fire frequency and seasonality. Despite the high biodiversity of the Cerrado, little is known about its microbiome, with few publications describing some aspects of the bacterial and fungal communities found on this biome and almost no references about archaea. In this study, we describe the archaeal diversity in Cerrado sensu stricto soils, comparing the archaeal communities from soils of an area long protected from fires to one exposed to biennial fires, using both 16S rRNA and amoA genes as molecular markers. Almost all 16S rRNA sequences from both studied areas were affiliated with I.1b and 1.1c Thaumarchaeota, groups commonly detected in terrestrial environments. A higher relative abundance of I.1b thaumarchaeal subgroup was detected in the frequently burned area even though no statistically significant differences were observed in archaeal 16S rRNA richness and diversity between the investigated areas. Many ammonia-oxidizing archaea (AOA) are affiliated with this group, which is consistent with the higher amoA diversity and OTU numbers detected in the area periodically burned. Taken together, our results suggest that, although total archaeal community richness and diversity do not seem to greatly differ between the investigated conditions, alterations in wood cover and vegetation structure caused by frequent fires likely cause long-term effects in AOA diversity in Cerrado soils.

Project description:Colorectal cancer (CRC) is the third most common type of cancer among men and the second among women in Brazil. Despite the high incidence and significance of CRC in Brazil, very little is known about its prevalence among the asymptomatic population. Recently, a CRC screening program was implemented at the Cancer Hospital of Barretos. Characterization of the clinical findings detected in the screening population and the prevalence of basal CRC might contribute to better organization of the program and define the best strategy for a future national screening program. We hypothesize that recruitment and the early outcomes of our screening program based on the fecal immunochemical test (FIT) will differ from the outcomes corresponding to other populations due to sociodemographic differences. Aims: i. To implement a data collection and storage system for follow-up of the screening program participants and to measure early outcomes (adenoma, advanced adenoma and cancer) and associate them with sociodemographic risk factors; ii. to quantify the risk of CRC in the Brazilian population and to develop algorithms for risk stratification of CRC screening; and iii. to compare the risk stratification to other countries with low, medium and high incomes. Methods: Individuals aged 50 to 65 years will be included in the HCB screening program from November 2017 to December 2018. The following data will be collected from all participants: sociodemographic and ethnic (skin color) characteristics; risk factors for CRC, such as smoking and drinking; comorbidities, including diabetes mellitus and arterial hypertension; and FIT, colonoscopy and histopathology examination results. Data collection will be performed using the REDCap data collection/database system. The risk score will be formulated using the Chi-square test (or Fisher’s exact test) and simple logistic regression, and the regression coefficients will be calculated. Then, the model identified for the training sample will be replicated with a validation sample. The resulting score will be used to calculate the sensitivity, specificity, positive predictive value, negative predictive value, accuracy, area under the receiver operating characteristic (ROC) curve and Kolmogorov D statistic.

Project description:The threatened caesalpinioid legume Dimorphandra wilsonii, which is native to the Cerrado biome in Brazil, was examined for its nodulation and N(2)-fixing ability, and was compared with another, less-threatened species, D. jorgei. Nodulation and potential N(2) fixation was shown on seedlings that had been inoculated singly with five bradyrhizobial isolates from mature D. wilsonii nodules. The infection of D. wilsonii by two of these strains (Dw10.1, Dw12.5) was followed in detail using light and transmission electron microscopy, and was compared with that of D. jorgei by Bradyrhizobium strain SEMIA6099. The roots of D. wilsonii were infected via small transient root hairs at 42 d after inoculation (dai), and nodules were sufficiently mature at 63 dai to express nitrogenase protein. Similar infection and nodule developmental processes were observed in D. jorgei. The bacteroids in mature Dimorphandra nodules were enclosed in plant cell wall material containing a homogalacturonan (pectic) epitope that was recognized by the monoclonal antibody JIM5. Analysis of sequences of their rrs (16S rRNA) genes and their ITS regions showed that the five D. wilsonii strains, although related to SEMIA6099, may constitute five undescribed species of genus Bradyrhizobium, whilst their nodD and nifH gene sequences showed that they formed clearly separated branches from other rhizobial strains. This is the first study to describe in full the N(2)-fixing symbiotic interaction between defined rhizobial strains and legumes in the sub-family Caesalpinioideae. This information will hopefully assist in the conservation of the threatened species D. wilsonii.
