Patterning of nanocrystalline cellulose gel phase by electrodissolution of a metallic electrode.

ABSTRACT: At high concentration or in the presence of electrolytes and organic solvents, solutions of cellulose nanocrystals (CNCs) can form gels exhibiting optical properties similar to the ones of liquid crystal phases. In an attempt to pattern such a gel phase, we have studied the electrodissolution of a metallic electrode in a water suspension of carboxylated CNCs (cCNCs). Depending on the metal used, the electrodissolution process was observed at a different positive potential. In the case of copper the minimum potential at which we could observe optically the growth of the gel phase was 200 mV. The growth rate was current limited indicating that the process was controlled by the electrodissolution of the copper electrode. This hypothesis was confirmed by using circular and square copper patterns as positive electrodes. In both cases, the consumption of the electrode material was observed optically and correlated with the growth of the gel phase.

Theoretical Assessment of Thermodynamic Stability in Nanocrystalline Metallic Alloys.

Project description:Thermal stability in nanocrystalline alloys has been extensively explored while using both experimental and theoretical approaches. From the theoretical point of view, the vast majority of the models proposed in the literature have been implicitly limited to immiscible or dilute systems and thus lack the necessary generality to make predictions for different alloying interactions and in the case of intermetallic compounds formation. In this work, a general theoretical description for the case of binary W-based alloys is presented. It is shown that a critical value ? ? of the interaction energy in the grain boundary ? ( g b ) exists, such that the condition ? ( g b ) < ? ? can be regarded as a criterion for thermodynamic stability assessment. A procedure for calculating the value of ? ? for each specific alloy is illustrated. A preliminary qualitative comparison between the model predictions and properly selected experimental findings taken from the literature and related to the W-Cr system is also provided.

Phase patterning of metallic glasses through superfast quenching of ion irradiation-induced thermal spikes.

Project description:Amorphous metallic glasses (MGs) convert to crystalline solids upon annealing at a high temperature. Such a phase change, however, does not occur with the local melting caused by damage cascades introduced by ion irradiation, although the resulting thermal spikes can reach temperatures > 1000 K. This is because the quenching rate of the local melting zone is several orders of magnitude higher than the critical cooling rate for MG formation. Thus the amorphous structure is sustained. This mechanism increases the highest temperature at which irradiated MG sustains amorphous phase. More interestingly, if an irradiated MG is pre-annealed to form a polycrystalline structure, ion irradiation can locally convert this crystalline phase to an amorphous phase if the grains are nanometers in size and comparable to the damage cascade volume size. Combining pre-annealing and site selective ion irradiation, patterned crystalline-amorphous heterogeneous structures have been fabricated. This finding opens new doors for various applications.

Sol-gel Autocombustion Synthesis of Nanocrystalline High-entropy Alloys.

Project description:A reduction in the particle size is expected to improve the properties and increase the application potential of high-entropy alloys. Therefore, in this study, a novel sol-gel autocombustion technique was first used to synthesize high-entropy alloys. The average grain size of the prepared nanocrystalline CoCrCuNiAl high-entropy alloys showed was 14 nm with an excellent and uniform dispersion, exhibiting a distinct magnetic behavior similar to the superparamagnetic behavior. We show that the metal nitrates first form (Co,Cu,Mg,Ni,Zn)O high-entropy oxides, and then in situ reduce to CoCrCuNiAl high-entropy alloys by the reducing gases, and the chelation between citric acid and the metal ions and the in situ chemical reactions are the dominant reaction mechanisms. We demonstrate that the sol-gel autocombustion process is an efficient way to synthesize solid solution alloys eluding the restriction of a high mixing entropy.

Nanocrystalline Cellulose as a Versatile Engineering Material for Extrusion-Based Bioprinting.

Project description:Naturally derived polysaccharide-based hydrogels, such as alginate, are frequently used in the design of bioinks for 3D bioprinting. Traditionally, the formulation of such bioinks requires the use of pre-reticulated materials with low viscosities, which favour cell viability but can negatively influence the resolution and shape fidelity of the printed constructs. In this work, we propose the use of cellulose nanocrystals (CNCs) as a rheological modifier to improve the printability of alginate-based bioinks whilst ensuring a high viability of encapsulated cells. Through rheological analysis, we demonstrate that the addition of CNCs (1% and 2% (w/v)) to alginate hydrogels (1% (w/v)) improves shear-thinning behaviour and mechanical stability, resulting in the high-fidelity printing of constructs with superior resolution. Importantly, LIVE/DEAD results confirm that the presence of CNCs does not seem to affect the health of immortalised chondrocytes (TC28a2) that remain viable over a period of seven days post-encapsulation. Taken together, our results indicate a favourable effect of the CNCs on the rheological and biocompatibility properties of alginate hydrogels, opening up new perspectives for the application of CNCs in the formulation of bioinks for extrusion-based bioprinting.

Chemical Modification of Nanocrystalline Cellulose for Manufacturing of Osteoconductive Composite Materials.

Project description:Cellulose is one of the main renewable polymers whose properties are very attractive in many fields, including biomedical applications. The modification of nanocrystalline cellulose (NCC) opens up the possibility of creating nanomaterials with properties of interest as well as combining them with other biomedical polymers. In this work, we proposed the covalent modification of NCC with amphiphilic polyanions such as modified heparin (Hep) and poly(αL-glutamic acid) (PGlu). The modification of NCC should overcome two drawbacks in the production of composite materials based on poly(ε-caprolactone) (PCL), namely, (1) to improve the distribution of modified NCC in the PCL matrix, and (2) to provide the composite material with osteoconductive properties. The obtained specimens of modified NCC were characterized by Fourier-transform infrared spectroscopy and solid-state 13C nuclear magnetic resonance spectroscopy, dynamic and electrophoretic light scattering, as well as thermogravimetric analysis. The morphology of PCL-based composites containing neat or modified NCC as filler was studied by optical and scanning electron microscopy. The mechanical properties of the obtained composites were examined in tensile tests. The homogeneity of filler distribution as well as the mechanical properties of the composites depended on the method of NCC modification and the amount of attached polyanion. In vitro biological evaluation showed improved adhesion of human fetal mesenchymal stem cells (FetMSCs) and human osteoblast-like cells (MG-63 osteosarcoma cell line) to PCL-based composites filled with NCC bearing Hep or PGlu derivatives compared to pure PCL. Furthermore, these composites demonstrated the osteoconductive properties in the experiment on the osteogenic differentiation of FetMSCs.

Protein array patterning by diffusive gel stamping.

Project description:Proteins and small molecules are the effectors of physiological action in biological systems and comprehensive methods are needed to analyze their modifications, expression levels and interactions. Systems-scale characterization of the proteome requires thousands of components in high-complexity samples to be isolated and simultaneously probed. While protein microarrays offer a promising approach to probe systems-scale changes in a high-throughput format, they are limited by the need to individually synthesize tens of thousands of proteins. We present an alternative technique, which we call diffusive gel (DiG) stamping, for patterning a microarray using a cellular lysate enabling rapid visualization of dynamic changes in the proteome as well protein interactions. A major advantage of the method described is that it requires no specialized equipment or in-vitro protein synthesis, making it widely accessible to researchers. The method can be integrated with mass spectrometry, allowing for the discovery of novel protein interactions. Here, we describe and characterize the sensitivity and physical features of DiG-Stamping. We demonstrate the biologic utility of DiG-Stamping by (1) identifying the binding partners of a target protein within a cellular lysate and by (2) visualizing the dynamics of proteins with multiple post-translational modifications.

Incorporating nanocrystalline cellulose into a multifunctional hydrogel for heart valve tissue engineering applications.

Project description:Functional tissue engineered heart valves (TEHV) have been an elusive goal for nearly 30 years. Among the persistent challenges are the requirements for engineered valve leaflets that possess nonlinear elastic tissue biomechanical properties, support quiescent fibroblast phenotype, and resist osteogenic differentiation. Nanocellulose is an attractive tunable biological material that has not been employed to this application. In this study, we fabricated a series of photocrosslinkable composite hydrogels mNCC-MeGel (mNG) by conjugating TEMPO-modified nanocrystalline cellulose (mNCC) onto the backbone of methacrylated gelatin (MeGel). Their structures were characterized by FTIR, 1 HNMR and uniaxial compression testing. Human adipose-derived mesenchymal stem cells (HADMSC) were encapsulated within the material and evaluated for valve interstitial cell phenotypes over 14 days culture in both normal and osteogenic media. Compared to the MeGel control group, the HADMSC encapsulated within mNG showed decreased alpha smooth muscle actin (αSMA) expression and increased vimentin and aggrecan expression, suggesting the material supports a quiescent fibroblastic phenotype. Under osteogenic media conditions, HADMSC within mNG hydrogels showed lower expression of osteogenic genes, including Runx2 and osteocalcin, indicating resistance toward calcification. As a proof of principle, the mNG hydrogel, combined with a viscosity enhancing agent, was used to 3D bioprint a tall, self-standing tubular structure that sustained cell viability. Together, these results identify mNG as an attractive biomaterial for TEHV applications.

Preparation and Characterization of 3D-Printed Biobased Composites Containing Micro- or Nanocrystalline Cellulose.

Project description:Stereolithography (SLA), one of the seven different 3D printing technologies, uses photosensitive resins to create high-resolution parts. Although SLA offers many advantages for medical applications, the lack of biocompatible and biobased resins limits its utilization. Thus, the development of new materials is essential. This work aims at designing, developing, and fully characterizing a bio-resin system (made of poly(ethylene glycol) diacrylate (PEGDA) and acrylated epoxidized soybean oil (AESO)), filled with micro- or nanocellulose crystals (MCC and CNC), suitable for 3D printing. The unfilled resin system containing 80 wt.% AESO was identified as the best resin mixture, having a biobased content of 68.8%, while ensuring viscosity values suitable for the 3D printing process (>1.5 Pa s). The printed samples showed a 93% swelling decrease in water, as well as increased tensile strength (4.4 ± 0.2 MPa) and elongation at break (25% ± 2.3%). Furthermore, the incorporation of MCC and CNC remarkably increased the tensile strength and Young's modulus of the cured network, thus indicating a strong reinforcing effect exerted by the fillers. Lastly, the presence of the fillers did not affect the UV-light penetration, and the printed parts showed a high quality, thus proving their potential for precise applications.

Bleaching-free, lignin-tolerant, high-yield production of nanocrystalline cellulose from lignocellulosic biomass.

Project description:Nanocrystalline cellulose (NCC) preparation in an integrated fractionation manner is expected to solve the problems of low yield and environmental impact in the traditional process. An integrated fractionation strategy for NCC production from wood was developed through catalytic biomass fractionation, the partial dissolution of cellulose-rich materials (CRMs) in aqueous tetrabutylphosphonium hydroxide, and short-term ultrasonication. The presented process could tolerate a high CRM lignin content of 21.2 wt % and provide a high NCC yield of 76.6 wt % (34.3 wt % of the original biomass). The increase in the CRM lignin content decreased the NCC yield, facilitated the crystal transition of NCC from cellulose I to cellulose II, and showed no apparent effects on the NCC morphology. A partial/selective dissolution mechanism is proposed for the presented strategy. This study provided a promising efficient fractionation-based method toward comprehensive and high-value utilization of lignocellulosic biomass through effective delignification and high-yield NCC production.

Nanocrystalline Cellulose-Supported Iron Oxide Composite Materials for High-Performance Lithium-Ion Batteries.

Project description:Nanocrystalline cellulose (NCC) can be converted into carbon materials for the fabrication of lithium-ion batteries (LIBs) as well as serve as a substrate for the incorporation of transition metal oxides (TMOs) to restrain the volume expansion, one of the most significant challenges of TMO-based LIBs. To improve the electrochemical performance and enhance the longer cycling stability of LIBs, a nanocrystalline cellulose-supported iron oxide (Fe2O3) composite (denoted as NCC-Fe2O3) is synthesized and utilized as electrodes in LIBs. The obtained NCC-Fe2O3 electrode exhibited stable cycling performance, better capacity, and high-rate capacity, and delivered a specific discharge capacity of 576.70 mAh g-1 at 100 mA g-1 after 1000 cycles. Moreover, the NCC-Fe2O3 electrode was restored and showed an upward trend of capacity after working at high current densities, indicating the fabricated composite is a promising approach to designing next-generation high-energy density lithium-ion batteries.
