Dopamine is required for learning and forgetting in Drosophila.

ABSTRACT: Psychological studies in humans and behavioral studies of model organisms suggest that forgetting is a common and biologically regulated process, but the molecular, cellular, and circuit mechanisms underlying forgetting are poorly understood. Here we show that the bidirectional modulation of a small subset of dopamine neurons (DANs) after olfactory learning regulates the rate of forgetting of both punishing (aversive) and rewarding (appetitive) memories. Two of these DANs, MP1 and MV1, exhibit synchronized ongoing activity in the mushroom body neuropil in alive and awake flies before and after learning, as revealed by functional cellular imaging. Furthermore, while the mushroom-body-expressed dDA1 dopamine receptor is essential for the acquisition of memory, we show that the dopamine receptor DAMB, also highly expressed in mushroom body neurons, is required for forgetting. We propose a dual role for dopamine: memory acquisition through dDA1 signaling and forgetting through DAMB signaling in the mushroom body neurons.

Dopamine Receptor DAMB Signals via Gq to Mediate Forgetting in Drosophila.

Project description:Prior studies have shown that aversive olfactory memory is acquired by dopamine acting on a specific receptor, dDA1, expressed by mushroom body neurons. Active forgetting is mediated by dopamine acting on another receptor, Damb, expressed by the same neurons. Surprisingly, prior studies have shown that both receptors stimulate cyclic AMP (cAMP) accumulation, presenting an enigma of how mushroom body neurons distinguish between acquisition and forgetting signals. Here, we surveyed the spectrum of G protein coupling of dDA1 and Damb, and we confirmed that both receptors can couple to Gs to stimulate cAMP synthesis. However, the Damb receptor uniquely activates Gq to mobilize Ca2+ signaling with greater efficiency and dopamine sensitivity. The knockdown of G?q with RNAi in the mushroom bodies inhibits forgetting but has no effect on acquisition. Our findings identify a Damb/Gq-signaling pathway that stimulates forgetting and resolves the opposing effects of dopamine on acquisition and forgetting.

Dopamine Neurons Mediate Learning and Forgetting through Bidirectional Modulation of a Memory Trace.

Project description:It remains unclear how memory engrams are altered by experience, such as new learning, to cause forgetting. Here, we report that short-term aversive memory in Drosophila is encoded by and retrieved from the mushroom body output neuron MBOn-γ2α'1. Pairing an odor with aversive electric shock creates a robust depression in the calcium response of MBOn-γ2α'1 and increases avoidance to the paired odor. Electric shock after learning, which activates the cognate dopamine neuron DAn-γ2α'1, restores the response properties of MBOn-γ2α'1 and causes behavioral forgetting. Conditioning with a second odor restores the responses of MBOn-γ2α'1 to a previously learned odor while depressing responses to the newly learned odor, showing that learning and forgetting can occur simultaneously. Moreover, optogenetic activation of DAn-γ2α'1 is sufficient for the bidirectional modulation of MBOn-γ2α'1 response properties. Thus, a single DAn can drive both learning and forgetting by bidirectionally modulating a cellular memory trace.

Active forgetting requires Sickie function in a dedicated dopamine circuit in Drosophila

Project description:Forgetting is an essential component of the brain memory management system, providing a balance to memory formation processes by removing unused or unwanted memories, or by suppressing their expression. However, the molecular, cellular and circuit mechanisms underlying forgetting are poorly understood. Here we show that the memory suppressor gene, sickie, functions in a single dopamine neuron (DAn) by supporting the process of active forgetting in Drosophila. RNAi knockdown (KD) of sickie impairs forgetting by reducing the Ca2+ influx and DA release from the DAn that promotes forgetting. Co-immunoprecipitation/mass spectrometry analyses identified cytoskeletal and presynaptic active zone (AZ) proteins as candidates that physically interact with Sickie, and a focused RNAi screen of the candidates showed that Bruchpilot (Brp), a presynaptic AZ protein that regulates calcium channel clustering and neurotransmitter release, impairs active forgetting like sickie KD. In addition, overexpression of brp rescued the impaired forgetting of sickie KD, providing evidence that they function in the same process. Moreover, we show that sickie KD in the DAn reduces the abundance and size of AZ markers but increases their number, suggesting that Sickie controls DAn activity for forgetting by modulating the presynaptic AZ structure. Our results identify a new molecular and circuit mechanism for normal levels of active forgetting and reveal a surprising role of Sickie in maintaining presynaptic AZ structure for neurotransmitter release.

Sleep Facilitates Memory by Blocking Dopamine Neuron-Mediated Forgetting.

Project description:Early studies from psychology suggest that sleep facilitates memory retention by stopping ongoing retroactive interference caused by mental activity or external sensory stimuli. Neuroscience research with animal models, on the other hand, suggests that sleep facilitates retention by enhancing memory consolidation. Recently, in Drosophila, the ongoing activity of specific dopamine neurons was shown to regulate the forgetting of olfactory memories. Here, we show this ongoing dopaminergic activity is modulated with behavioral state, increasing robustly with locomotor activity and decreasing with rest. Increasing sleep-drive, with either the sleep-promoting agent Gaboxadol or by genetic stimulation of the neural circuit for sleep, decreases ongoing dopaminergic activity, while enhancing memory retention. Conversely, increasing arousal stimulates ongoing dopaminergic activity and accelerates dopaminergic-based forgetting. Therefore, forgetting is regulated by the behavioral state modulation of dopaminergic-based plasticity. Our findings integrate psychological and neuroscience research on sleep and forgetting.

Goldilocks Forgetting in Cross-Situational Learning.

Project description:Given that there is referential uncertainty (noise) when learning words, to what extent can forgetting filter some of that noise out, and be an aid to learning? Using a Cross Situational Learning model we find a U-shaped function of errors indicative of a "Goldilocks" zone of forgetting: an optimum store-loss ratio that is neither too aggressive nor too weak, but just the right amount to produce better learning outcomes. Forgetting acts as a high-pass filter that actively deletes (part of) the referential ambiguity noise, retains intended referents, and effectively amplifies the signal. The model achieves this performance without incorporating any specific cognitive biases of the type proposed in the constraints and principles account, and without any prescribed developmental changes in the underlying learning mechanism. Instead we interpret the model performance as more of a by-product of exposure to input, where the associative strengths in the lexicon grow as a function of linguistic experience in combination with memory limitations. The result adds a mechanistic explanation for the experimental evidence on spaced learning and, more generally, advocates integrating domain-general aspects of cognition, such as memory, into the language acquisition process.

Dopamine and Serotonin Are Both Required for Mate-Copying in Drosophila melanogaster.

Project description:Mate-copying is a form of social learning in which the mate-choice decision of an individual (often a female) is influenced by the mate-choice of conspecifics. Drosophila melanogaster females are known to perform such social learning, and in particular, to mate-copy after a single observation of one conspecific female mating with a male of one phenotype, while the other male phenotype is rejected. Here, we show that this form of social learning is dependent on serotonin and dopamine. Using a pharmacological approach, we reduced dopamine or serotonin synthesis in adult virgin females with 3-iodotyrosine (3-IY) and DL-para-chlorophenylalanine (PCPA), respectively, and then tested their mate-copying performance. We found that, while control females without drug treatment copied the choice of the demonstrator, drug-treated females with reduced dopamine or serotonin chose randomly. To ensure the specificity of the drugs, the direct precursors of the neurotransmitters, either the dopamine precursor L-3,4-dihydroxyphenylalanine (L-DOPA) or the serotonin precursor 5-L-hydroxytryptophan (5-HTP) were given together with the drug, (respectively 3-IY and PCPA) resulting in a full rescue of the mate-copying defects. This indicates that dopamine and serotonin are both required for mate-copying. These results give a first insight into the mechanistic pathway underlying this form of social learning in D. melanogaster.

Dopamine D2L Receptor Is Required for Visual Discrimination and Reversal Learning.

Project description:The corticostriatothalamic circuit regulates learning behaviors via dopamine neurotransmission. D2 long (D2L) receptors are an isoform of dopamine D2 receptors (D2Rs) and may act mainly at postsynaptic sites. It is well known that D2Rs influence high brain functions, but the roles of individual D2R isoforms are still unclear. To assess the influence of D2L receptors in visual discrimination learning, we performed visual discrimination and reversal tasks with D2L knockout mice using a touchscreen operant system. There were no significant differences in an operant conditioning task between genotypes. However, D2L knockout mice were impaired in both visual discrimination and reversal learning tasks. D2L knockout mice were also significantly slower than wild-type mice in collecting the reward in the visual discrimination task. These results indicate that D2L receptors play an important role in visual discrimination and reversal learning.

When forgetting fosters learning: A neural network model for statistical learning.

Project description:Learning often requires splitting continuous signals into recurring units, such as the discrete words constituting fluent speech; these units then need to be encoded in memory. A prominent candidate mechanism involves statistical learning of co-occurrence statistics like transitional probabilities (TPs), reflecting the idea that items from the same unit (e.g., syllables within a word) predict each other better than items from different units. TP computations are surprisingly flexible and sophisticated. Humans are sensitive to forward and backward TPs, compute TPs between adjacent items and longer-distance items, and even recognize TPs in novel units. We explain these hallmarks of statistical learning with a simple model with tunable, Hebbian excitatory connections and inhibitory interactions controlling the overall activation. With weak forgetting, activations are long-lasting, yielding associations among all items; with strong forgetting, no associations ensue as activations do not outlast stimuli; with intermediate forgetting, the network reproduces the hallmarks above. Forgetting thus is a key determinant of these sophisticated learning abilities. Further, in line with earlier dissociations between statistical learning and memory encoding, our model reproduces the hallmarks of statistical learning in the absence of a memory store in which items could be placed.

Learning and forgetting using reinforced Bayesian change detection.

Project description:Agents living in volatile environments must be able to detect changes in contingencies while refraining to adapt to unexpected events that are caused by noise. In Reinforcement Learning (RL) frameworks, this requires learning rates that adapt to past reliability of the model. The observation that behavioural flexibility in animals tends to decrease following prolonged training in stable environment provides experimental evidence for such adaptive learning rates. However, in classical RL models, learning rate is either fixed or scheduled and can thus not adapt dynamically to environmental changes. Here, we propose a new Bayesian learning model, using variational inference, that achieves adaptive change detection by the use of Stabilized Forgetting, updating its current belief based on a mixture of fixed, initial priors and previous posterior beliefs. The weight given to these two sources is optimized alongside the other parameters, allowing the model to adapt dynamically to changes in environmental volatility and to unexpected observations. This approach is used to implement the "critic" of an actor-critic RL model, while the actor samples the resulting value distributions to choose which action to undertake. We show that our model can emulate different adaptation strategies to contingency changes, depending on its prior assumptions of environmental stability, and that model parameters can be fit to real data with high accuracy. The model also exhibits trade-offs between flexibility and computational costs that mirror those observed in real data. Overall, the proposed method provides a general framework to study learning flexibility and decision making in RL contexts.

Dissecting neural pathways for forgetting in Drosophila olfactory aversive memory.

Project description:Recent studies have identified molecular pathways driving forgetting and supported the notion that forgetting is a biologically active process. The circuit mechanisms of forgetting, however, remain largely unknown. Here we report two sets of Drosophila neurons that account for the rapid forgetting of early olfactory aversive memory. We show that inactivating these neurons inhibits memory decay without altering learning, whereas activating them promotes forgetting. These neurons, including a cluster of dopaminergic neurons (PAM-β'1) and a pair of glutamatergic neurons (MBON-γ4>γ1γ2), terminate in distinct subdomains in the mushroom body and represent parallel neural pathways for regulating forgetting. Interestingly, although activity of these neurons is required for memory decay over time, they are not required for acute forgetting during reversal learning. Our results thus not only establish the presence of multiple neural pathways for forgetting in Drosophila but also suggest the existence of diverse circuit mechanisms of forgetting in different contexts.
