Effects of CSN1S2 Genotypes on Economic Traits in Chinese Dairy Goats.

ABSTRACT: The aim of this study was to investigate allele frequencies at the CSN1S2 locus in two Chinese dairy goat breeds and the effects of its variation on dairy goat economic traits. Seven hundred and eight goats from Xinong Saanen (XS, n = 268) and Guanzhong (GZ, N = 440) breeds were selected. The milk samples of 268 XS goats were collected during the middle of lactation, body size parameters (708 goats) and daily milk yield (202 goats) were registered. The RFLP (restriction fragment length polymorphism) and SSCP (single strand conformation polymorphism) were used to detect the polymorphisms in CSN1S2. The Hardy-Weinberg (HW) equilibrium and the associations between body size, milk yield and composition and the genotypes were calculated. The results revealed that only A and F CSN1S2 alleles were found in the two Chinese dairy goat breeds. Allelic frequencies of A and F were 0.795, 0.205 and 0.739, 0.261 in Xinong Saanen and Guanzhong population respectively. Xinong Saanen breed was in Hardy-Weinberg equilibrium, while Guanzhong breed deviated from Hardy-Weinberg equilibrium (p<0.05). The association of polymorphism with economic traits indicated that the goats with FF genotype have higher milk fat and total solid concentration than those with AA and AF genotypes (p<0.05).

Project description:Dairy goats experience metabolic stress during the peripartal period, and their ability to navigate this stage of lactation is related to the occurrence and development of metabolic diseases. Unlike dairy cows, there is a lack of comprehensive analysis of changes in the plasma profiles of peripartal dairy goats, particularly using high-throughput techniques. A subset of 9 clinically-healthy dairy goats were used from a cohort of 96 primiparous Guanzhong dairy goats (BCS, 2.75 ± 0.15). Blood samples were collected at seven time points around parturition (d 21, 14, 7 before parturition, the day of kidding, and d 7, 14, 21 postpartum), were analyzed using untargeted metabolomics and targeted lipidomics. The orthogonal partial least squares discriminant analysis model revealed a total of 31 differential metabolites including p-cresol sulfate, pyruvic acid, cholic acid, and oxoglutaric acid. The pathway enrichment analysis identified phenylalanine metabolism, aminoacyl-tRNA biosynthesis, and citrate cycle as the top three significantly-altered pathways. The Limma package identified a total of 123 differentially expressed lipids. Phosphatidylserine (PS), free fatty acids (FFA), and acylcarnitines (ACs) were significantly increased on the day of kidding, while diacylglycerols (DAG) and triacylglycerols (TAG) decreased. Ceramides (Cer) and lyso-phosphatidylinositols (LPI) were significantly increased during postpartum period, while PS, FFA, and ACs decreased postpartum and gradually returned to antepartum levels. Individual species of FFA and phosphatidylcholines (PC) were segregated based on the differences in the saturation and length of the carbon chain. Overall, this work generated the largest repository of the plasma lipidome and metabolome in dairy goats across the peripartal period, which contributed to our understanding of the multifaceted adaptations of transition dairy goats. Project description:In ruminants, feeding behaviour variables are parameters involved in feed efficiency that show variation among individuals. This study aimed to evaluate during the first two production cycles in ruminants the repeatability of feed intake pattern, which is an important aspect of feeding behaviour. Thirty-five dairy goats from Alpine or Saanen breeds were housed in individual pens at four periods (end of first gestation, middle of first and second lactations and middle of second gestation which is also the end of first lactation) and fed a total mixed ration (TMR) ad libitum. Individual cumulative dry matter intake (DMI) was automatically measured every 2 min during the last 4 days of each period. Feed intake pattern was characterized by several measures related to the quantity of feed eaten or to the rate of intake during the 15 h following the afternoon feed delivery. Two main methods were used: modelling cumulative DMI evolution by an exponential model or by a segmentation-clustering method. The goat ability to sort against dietary fibre was also evaluated. There was a very good repeatability of the aggregate measures between days within a period for a given goat estimated by the day effect within breed and goat, tested on the residual variance (P > 0.95). The correlations between periods were the highest between the second and either the third or fourth periods. With increasing age, goats sorted more against the fibrous part of the TMR and increased their initial rate of intake. Alpine goats ate more slowly than Saanen goats but ate during a longer duration. Principal component analysis (PCA) was performed on all the aggregate measures of feed intake patterns. The factor score plots generated by the PCA highlighted the opposition between the different measures of feed intake patterns and the sorting behaviour. The projection of the animals on the scoring plots showed a breed effect and that there was a continuum for the feed intake pattern of goats. In conclusion, this study showed that the feed intake pattern was highly repeatable for an animal in a given period and between periods. This means that phenotyping goats in a younger age might be of interest, either to select them on feeding behaviour and choose preferentially the slow eaters or to adapt the quantity offered and restrict feed delivery to the fast eaters in order to increase feed efficiency and welfare by limiting the occurrence of acidosis, for example.
