Reading and language disorders: the importance of both quantity and quality.

ABSTRACT: Reading and language disorders are common childhood conditions that often co-occur with each other and with other neurodevelopmental impairments. There is strong evidence that disorders, such as dyslexia and Specific Language Impairment (SLI), have a genetic basis, but we expect the contributing genetic factors to be complex in nature. To date, only a few genes have been implicated in these traits. Their functional characterization has provided novel insight into the biology of neurodevelopmental disorders. However, the lack of biological markers and clear diagnostic criteria have prevented the collection of the large sample sizes required for well-powered genome-wide screens. One of the main challenges of the field will be to combine careful clinical assessment with high throughput genetic technologies within multidisciplinary collaborations.

Quantity and/or Quality? The Importance of Publishing Many Papers.

Project description:Do highly productive researchers have significantly higher probability to produce top cited papers? Or do high productive researchers mainly produce a sea of irrelevant papers-in other words do we find a diminishing marginal result from productivity? The answer on these questions is important, as it may help to answer the question of whether the increased competition and increased use of indicators for research evaluation and accountability focus has perverse effects or not. We use a Swedish author disambiguated dataset consisting of 48.000 researchers and their WoS-publications during the period of 2008-2011 with citations until 2014 to investigate the relation between productivity and production of highly cited papers. As the analysis shows, quantity does make a difference.

Importance of Surfactant Quantity and Quality on Growth Regime of Iron Oxide Nanoparticles.

Project description:This study shows the influence of selected nonstandard surfactants on the growth and properties of magnetite nanoparticles. Particles were obtained using thermally decomposed iron (III) acetylacetonate in an organic environment. For synthesis, three different concentrations (4, 8, and 16 mmol) of tested surfactants were used. Five types of each long-chain carboxylic acid and amines were selected for stabilization of nanoparticles. Nanoparticles were characterized by X-ray diffraction, transmission electron microscopy, and infrared spectroscopy. Magnetic properties of the nanoparticles were tested by conventional room temperature Mössbauer spectroscopy with and without external magnetic field. TEM images clearly showed that application of tertiary amines causes the nanoparticles to form nanoflowers, in contrast to other compounds, which do not show such growth. Influence of surfactant amount on growth regime depends on the nature of the substances. Mössbauer spectroscopy confirms differences in magnetic core composition as a result of the surfactant amount present in synthetic procedure.

Dyslexia and Developmental Language Disorder: comorbid disorders with distinct effects on reading comprehension.

Project description:BackgroundReading comprehension draws on both decoding and linguistic comprehension, and poor reading comprehension can be the consequence of a deficit in either of these skills.MethodsUsing outcome data from the longitudinal Wellcome Language and Reading Project, we identified three groups of children at age 8 years: children with dyslexia (N = 21) who had deficits in decoding but not oral language, children with Developmental Language Disorder (DLD; N = 38) whose decoding skills were in the normal range, and children who met criteria for both dyslexia and DLD (N = 29).ResultsAll three groups had reading comprehension difficulties at the ages of 8 and 9 years relative to TD controls though those of the children with dyslexia were mild (relative to TD controls, d = 0.51 at age 8, d = 0.60 at age 8); while the most severe problems were found in the comorbid dyslexia + DLD group (d = 1.79 at age 8, d = 2.06 at age 9) those with DLD also had significant difficulties (d = 1.56 at age 8, d = 1.56 at age 9).ConclusionsThese findings confirm that children with dyslexia or DLD are at-risk for reading comprehension difficulties but for different reasons, because of weak decoding in the case of dyslexia or weak oral language skills in the case of DLD. Different forms of intervention are required for these groups of children, targeted to their particular area(s) of weakness.

The regulatory element READ1 epistatically influences reading and language, with both deleterious and protective alleles.

Project description:BACKGROUND:Reading disability (RD) and language impairment (LI) are heritable learning disabilities that obstruct acquisition and use of written and spoken language, respectively. We previously reported that two risk haplotypes, each in strong linkage disequilibrium (LD) with an allele of READ1, a polymorphic compound short tandem repeat within intron 2 of risk gene DCDC2, are associated with RD and LI. Additionally, we showed a non-additive genetic interaction between READ1 and KIAHap, a previously reported risk haplotype in risk gene KIAA0319, and that READ1 binds the transcriptional regulator ETV6. OBJECTIVE:To examine the hypothesis that READ1 is a transcriptional regulator of KIAA0319. METHODS:We characterised associations between READ1 alleles and RD and LI in a large European cohort, and also assessed interactions between READ1 and KIAHap and their effect on performance on measures of reading, language and IQ. We also used family-based data to characterise the genetic interaction, and chromatin conformation capture (3C) to investigate the possibility of a physical interaction between READ1 and KIAHap. RESULTS AND CONCLUSIONS:READ1 and KIAHap show interdependence--READ1 risk alleles synergise with KIAHap, whereas READ1 protective alleles act epistatically to negate the effects of KIAHap. The family data suggest that these variants interact in trans genetically, while the 3C results show that a region of DCDC2 containing READ1 interacts physically with the region upstream of KIAA0319. These data support a model in which READ1 regulates KIAA0319 expression through KIAHap and in which the additive effects of READ1 and KIAHap alleles are responsible for the trans genetic interaction.

Quality and readability of English-language Internet information for vestibular disorders.

Project description:BackgroundThe Internet has become a powerful, accessible resource for many patients to use for their own medical management and knowledge. Vestibular disorders are prevalent, especially in the elderly. As the Internet is increasingly a major source of health-related information to the general public, it is often used to search for information regarding dizziness and vertigo. Ensuring that the information is accessible, unbiased, and appropriate can aid informed decision-making.ObjectiveTo evaluate the quality and readability of English-language Internet information related to vestibular disorders.MethodsA cross-sectional website search using three keywords (nausea, dizziness, and vertigo) in five country-specific versions of the most commonly used Internet search engine was conducted in March 2018. The language was limited to English for all websites. Quality was assessed by presence of Health on the Net (HON) certification and DISCERN scores. Readability was assessed using the Flesch Reading Ease (FRE) score, Flesch-Kincaid Grade Level Formula (F-KGL), and Simple Measure of Gobbledygook (SMOG).ResultsIn total, 112 websites were included and analyzed. The majority were commercial (61%) websites. A total of 42% had obtained HON certification. No association was found between the presence of HON certification and the resource of the website. The DISCERN scores had a mean of 2.52 (SD 1.1). Readability measures indicated that an average of 14-18 years of education was required to read and understand the Internet information provided regarding vestibular disorders.ConclusionsTo ensure the accessible to the general population, it is necessary to improve the quality and readability of Internet-based information regarding vestibular disorders.

Reading Akkadian cuneiform using natural language processing.

Project description:In this paper we present a new method for automatic transliteration and segmentation of Unicode cuneiform glyphs using Natural Language Processing (NLP) techniques. Cuneiform is one of the earliest known writing system in the world, which documents millennia of human civilizations in the ancient Near East. Hundreds of thousands of cuneiform texts were found in the nineteenth and twentieth centuries CE, most of which are written in Akkadian. However, there are still tens of thousands of texts to be published. We use models based on machine learning algorithms such as recurrent neural networks (RNN) with an accuracy reaching up to 97% for automatically transliterating and segmenting standard Unicode cuneiform glyphs into words. Therefore, our method and results form a major step towards creating a human-machine interface for creating digitized editions. Our code, Akkademia, is made publicly available for use via a web application, a python package, and a github repository.

Quality and quantity of leaf litter: Both are important for feeding preferences and growth of an aquatic shredder.

Project description:The study of leaf litter as a resource for shredders has emerged as a key topic in trophic links in ecology. However, thus far, most studies have emphasized the leaf quality as one of the main determinants of shredder behaviour and growth without simultaneously considering the leaf quantity availability. Nevertheless, the combined effects of leaf quantity and quality on shredder behaviour and growth is particularly crucial to further understand how ecosystem functioning may respond to the increasing flow intermittency due to climate change. In this study, we explore how changes in the leaf litter quality and quantity influence the feeding preferences and growth of an invertebrate shredder (Potamophylax latipennis). To do so, we used black poplar leaves conditioned in two streams with different flow regimens as a food resource. Afterwards, using a microcosm approach, we offered leaf discs that varied in terms of leaf quantity and quality to P. latipennis. Our results showed that flow intermittency had a negative effect on the quality of the food resource, and a lower quality had a negative effect on the consumption and growth rates of P. latipennis. Furthermore, we found that P. latipennis fed selectively on higher quality leaves even though the availability (quantity) of this resource was lower. In the context of climate change, with higher aridity/drier conditions/scenarios, our findings suggest that a decrease in the availability (quantity) of high-quality resources could potentially threaten links in global fluvial food webs and thus threaten ecosystem functioning.

The human lexinome: genes of language and reading.

Project description:UNLABELLED:Within the human genome, genetic mapping studies have identified 10 regions of different chromosomes, known as DYX loci, in genetic linkage with dyslexia, and two, known as SLI loci, in genetic linkage with Specific Language Impairment (SLI). Further genetic studies have identified four dyslexia genes within the DYX loci: DYX1C1 on 15q, KIAA0319 and DCDC2 on 6p22, and ROBO1 on 13q. FOXP2 on 7q has been implicated in the development of Speech-Language Disorder. No genes for Specific Language Impairment have yet been identified within the two SLI loci. Functional studies have shown that all four dyslexia genes play roles in brain development, and ongoing molecular studies are attempting to elucidate how these genes exert their effects at a subcellular level. Taken together, these genes and loci likely represent only a fraction of the human lexinome, a term we introduce here to refer to the collection of all the genetic and protein elements involved in the development of human language, expression, and reading. LEARNING OUTCOMES:The reader will become familiar with (i) methods for identifying genes for complex diseases, (ii) the application of these methods in the elucidation of genes underlying disorders of language and reading, and (iii) the cellular pathways through which polymorphisms in these genes may contribute to the development of the disorders.

Small transcriptome analysis indicates that the enzyme RppH influences both the quality and quantity of sRNAs in Neisseria gonorrhoeae.

Project description:Prokaryotic mRNA turnover can be initiated by the removal of pyrophosphate from the 5' end of a transcript using the RNA pyrophosphohydrolase enzyme RppH. Following the initial dephosphorylation step, RNaseE then degrades the message into small oligonucleotide segments. This study assessed the small RNA transcriptome of Neisseria gonorrhoeae strain MS11 in two genetic backgrounds; using wild type cells as well as cells carrying a rppH insertional mutation. It was found that the presence of the RppH enzyme affected both the quantity and length of small RNAs (sRNAs) in various chromosomal locations and involved sense transcripts (seRNAs), transcripts originating from the opposite strand (asRNAs) as well as inter-genic-derived RNAs (IGRs). In comparing the two transcriptomes, we found that not all small RNAs were expressed in both genetic backgrounds, suggesting that RppH apparently targets only a subset of transcripts. Overall, this study shows that small RNAs can be detected from the majority of genes within the chromosome, as well as from inter-genic regions, and that more sRNA transcripts are detected in the absence of the RppH enzyme.

The impact of second language learning on semantic and nonsemantic first language reading.

Project description:The relationship between orthography (spelling) and phonology (speech sounds) varies across alphabetic languages. Consequently, learning to read a second alphabetic language, that uses the same letters as the first, increases the phonological associations that can be linked to the same orthographic units. In subjects with English as their first language, previous functional imaging studies have reported increased left ventral prefrontal activation for reading words with spellings that are inconsistent with their orthographic neighbors (e.g., PINT) compared with words that are consistent with their orthographic neighbors (e.g., SHIP). Here, using functional magnetic resonance imaging (fMRI) in 17 Italian-English and 13 English-Italian bilinguals, we demonstrate that left ventral prefrontal activation for first language reading increases with second language vocabulary knowledge. This suggests that learning a second alphabetic language changes the way that words are read in the first alphabetic language. Specifically, first language reading is more reliant on both lexical/semantic and nonlexical processing when new orthographic to phonological mappings are introduced by second language learning. Our observations were in a context that required participants to switch between languages. They motivate future fMRI studies to test whether first language reading is also altered in contexts when the second language is not in use.
