Microsatellite markers for the invasive species Bidens alba (Asteraceae).

ABSTRACT: UNLABELLED: PREMISE OF THE STUDY:Microsatellite markers were developed in the invasive species Bidens alba (Asteraceae) to assess its population structure and to facilitate tracking its expansion in China. • METHODS AND RESULTS:Using 454 pyrosequencing, 20 microsatellite primer sets were developed for B. alba. The markers were tested on one population of B. alba (30 individuals) and one population of the closely related B. pilosa (30 individuals) in China. For B. alba, all of the markers were polymorphic, and the number of alleles per locus ranged from three to 32. The expected heterozygosity values were from 0.3787 to 0.9284, and the Shannon-Wiener index was from 0.6796 to 2.8401. • CONCLUSIONS:These markers will be useful for investigating the genetic structure, genetic diversity, and invasion dynamics of B. alba and will also be useful in studies of B. pilosa.

Development of microsatellite markers for Crepis mollis (Asteraceae).

Project description:Polymorphic microsatellite markers were developed for the threatened species Crepis mollis (Asteraceae) to investigate population and conservation genetics. Illumina sequencing was conducted on pooled genomic DNA from 10 individuals of two populations. Ten polymorphic and 10 monomorphic microsatellite loci with di-, tri-, tetra-, penta-, and hexanucleotide repeat motifs were developed and characterized in C. mollis. In the polymorphic markers, up to 17 alleles per locus were detected with an observed and expected heterozygosity ranging from 0.120 to 0.780 and 0.102 to 0.834, respectively. Furthermore, the polymorphic markers were tested for cross-amplification in three congeneric species (C. biennis, C. foetida, and C. sancta) and amplified in up to three loci. The markers developed in this study are the first microsatellites tested on C. mollis and will be useful for performing population and conservation genetic studies in this threatened species.

Characterization of microsatellite markers for Baccharis dracunculifolia (Asteraceae).

Project description:Baccharis dracunculifolia (Asteraceae) is a native plant of the Atlantic Forest that is used for the production of essential oil. Microsatellite markers were developed for this species to investigate the genetic diversity of three natural populations. Seventeen out of 27 microsatellite loci identified in a genomic library used for the characterization of 315 individuals derived from three natural populations of B. dracunculifolia resulted in successful amplifications. Eleven polymorphic loci, ranging from two to seven alleles per locus, were obtained with expected and observed heterozygosity values ranging between 0.068 and 0.775 and 0.046 and 0.667, respectively. The microsatellite loci described in this study are tools that can be used for further studies of population genetics of B. dracunculifolia with a focus on deforested areas and conservation of natural populations.

Development of 17 microsatellite markers in the federally endangered species <i>Liatris helleri</i> (Asteraceae).

Project description:PremiseMicrosatellite markers were developed in the federally endangered species Liatris helleri (Asteraceae) to evaluate species boundaries with closely related congeners within the genus.Methods and resultsUsing Illumina data, 17 primer pairs were developed in populations of L. helleri. The primers amplified motifs from tri- to hexanucleotide repeats with one to 17 alleles per locus. Primers were also tested for cross-amplification in L. aspera, L. microcephala, and L. pycnostachya.ConclusionsThe developed primers for L. helleri serve as a novel genetic tool for future investigations in this genus, allowing for more explicit species delineation as well as population genetic analyses.

Microsatellite markers for study of the invasive species Rumex alpinus (Polygonaceae).

Project description:Premise of the Study:Polymorphic microsatellite markers were developed to study genetic diversity, population structure, and dispersal strategies of the highly invasive species Rumex alpinus (Polygonaceae). Methods and Results:Fifteen polymorphic microsatellite loci were developed using a 454 sequencing approach and used to genotype 72 individuals from six populations in Austria and France. All markers were polymorphic in at least one investigated population, and the number of alleles ranged from one to four alleles per locus. Observed and expected heterozygosity ranged from 0.06 to 1.0 and from 0.18 to 0.72, respectively. Conclusions:These 15 markers will be useful for characterizing dispersal strategies and gene flow assessment between R. alpinus populations and other, often weedy Rumex species.

Microsatellite markers for <i>Saussurea polylepis</i> (Asteraceae), a vulnerable continental island species endemic to Korea.

Project description:PremiseNuclear microsatellite markers were developed for Saussurea polylepis (Asteraceae), a vulnerable species with very limited distribution in a few southwestern continental islands of the Korean peninsula, in order to facilitate future population genetic studies.Methods and resultsBased on the Illumina sequence data, a total of 21 microsatellite primer pairs were designed and tested for their suitability. Nineteen of these primers, with two to 11 alleles per locus, were polymorphic in three natural populations of S. polylepis. The levels of expected and observed heterozygosity ranged from 0.000 to 0.842 and 0.000 to 0.933, respectively. Sixteen of these simple sequence repeat (SSR) markers were successfully cross-amplified in five congeneric species, namely S. gracilis, S. grandifolia, and S. tanakae for all 21 loci, and S. maximowiczii and S. pulchella for 18 loci.ConclusionsThe SSR markers developed here will be useful for future population genetic studies on S. polylepis and related species.

Fourteen polymorphic microsatellite markers for the threatened Arnica montana (Asteraceae).

Project description:Microsatellite markers were developed to investigate population genetic structure in the threatened species Arnica montana. • Fourteen microsatellite markers with di-, tetra-, and hexanucleotide repeat motifs were developed for A. montana using 454 pyrosequencing without and with library-enrichment methods, resulting in 56,545 sequence reads and 14,467 sequence reads, respectively. All loci showed a high level of polymorphism, with allele numbers ranging from four to 11 in five individuals from five populations (25 samples) and an expected heterozygosity ranging from 0.192 to 0.648 across the loci. • This set of microsatellite markers is the first one described for A. montana and will facilitate conservation genetic applications as well as the understanding of phylogeographic patterns in this species.

Revision of endemic Marquesas Islands Bidens (Asteraceae, Coreopsideae).

Project description:During the preparation of the Vascular Flora of the Marquesas Islands four new species of Bidens (Coreopsideae, Asteraceae) have come to light and are described herein: Bidens woodii W.L. Wagner, J.R. Clark & Lorence, sp. nov. from Ua Pou, and Bidens microcephala W.L. Wagner, J.R. Clark & Lorence sp. nov., Bidens evapelliana W.L. Wagner, J.R. Clark & Lorence, sp. nov., and Bidens wichmanii W.L. Wagner, J.R. Clark & Lorence, sp. nov. from the undercollected island of Fatu Hiva. In addition to these new species, we recognize the following six species of Bidens previously described from the Marquesas Islands: Bidens beckiana (F. Br.) Sherff (Eiao and Hatutaa), Bidens bipontina Sherff and Bidens cordifolia Sch. Bip. (both in Nuku Hiva), Bidens henryi Sherff (Hiva Oa, Tahuata), Bidens uapensis (F. Br.) Sherff (Ua Pou), and Bidens polycephala Sch. Bip. (Nuku Hiva, Ua Huku, Hiva Oa, Tahuata, and Mohotani). Two names are reduced to synonymy under Bidens polycephala: Bidens collina Degener & Sherff, syn. nov. and Bidens jardinii Sch. Bip., syn. nov. Bidens polycephala has the widest distribution of the Marquesan species on five of the islands and exhibits considerable variation.

Characterization of microsatellite markers for the vulnerable grassland forb Senecio macrocarpus (Asteraceae).

Project description:Development of microsatellite markers for the vulnerable forb Senecio macrocarpus was performed to begin an assessment of its population structure and breeding method to aid in the conservation of the species in Victoria, Australia. • Fifteen microsatellite markers were developed for S. macrocarpus from 454 pyrosequencing. The markers were tested on 104 individuals from four populations. The markers produced between two and seven alleles per locus while the expected heterozygosity ranged from 0.20 to 0.67 and the observed heterozygosity ranged from 0.00 to 1.00. The observed heterozygosity is suggestive that the populations may be apomictic. • The microsatellite markers developed for S. macrocarpus are intended to be used on future studies that aim to assess the population genetics and local breeding dynamics of the species with an emphasis on conservation.

Isolation and characterization of microsatellite loci in the invasive herb Solidago altissima (Asteraceae).

Project description:Microsatellite markers were developed in the perennial herb Solidago altissima from populations within its introduced range in Japan to assess its population structure and to facilitate tracking of invasion expansion. • Using 454 pyrosequencing, 16 microsatellite primer sets were developed for S. altissima. The primer sets were tested on 70 individuals sampled from three populations in Japan. The primers amplified di- and trinucleotide repeats with five to 25 alleles per locus, and the expected heterozygosity ranged from 0.46 to 0.92. • These results indicate the utility of primers in S. altissima for future research on a wide range of applications, including tracking of invasion dynamics and investigating population genetics of the species.
