Project description:The asymmetric unit of the title compound, 2C(8)H(9)N(2) (+)·2ClO(4) (-)·H(2)O, consists of two (4-cyano-benz-yl)>ammonium cations, two disordered ClO(4) (-) anions and one water mol-ecule. The differences in the two cations are reflected in the N-C-C-C torsion angles [-94.7?(3) and -115.9?(3)°]. In addition, the cations show different hydrogen-bonding patterns as one N atom bonds to two O atoms of ClO(4) (-) ions, while the other N atom is involved in hydrogen bonding with the O atoms of the ClO(4) (-) ions and water mol-ecules. In the crystal, N-H?O and O-H?O hydrogen bonds result in a three-dimensional network. An O atom in each of the anions is disordered over two positions of equal occupancy.

Project description:The asymmetric unit of the title salt, C7H7N2 (+)·ClO4 (-), comprises two independent formula units. The solid-state structure comprises corrugated layers of cations and of anions, approximately parallel to (010). The supra-molecular layers are stabilized and connected by C-H?O hydrogen bonding to consolidate a three-dimensional architecture. A close pyridin-ium-perchlorate N?O contact [2.867?(5)?Å] is noted. The crystal was refined as an inversion twin.

Project description:Four complexes containing the [UO(2)(oda)(2)](2-) anion (oda is oxydiacetate) are reported, namely dipyridinium dioxidobis(oxydiacetato)uranate(VI), (C(5)H(6)N)(2)[U(C(4)H(4)O(5))(2)O(2)], (I), bis(2-methylpyridinium) dioxidobis(oxydiacetato)uranate(VI), (C(8)H(8)N)(2)[U(C(4)H(4)O(5))(2)O(2)], (II), bis(3-methylpyridinium) dioxidobis(oxydiacetato)uranate(VI), (C(8)H(8)N)(2)[U(C(4)H(4)O(5))(2)O(2)], (III), and bis(4-methylpyridinium) dioxidobis(oxydiacetato)uranate(VI), (C(8)H(8)N)(2)[U(C(4)H(4)O(5))(2)O(2)], (IV). The anions are achiral and are located on a mirror plane in (I) and on inversion centres in (II)-(IV). The four complexes are assembled into three-dimensional structures via N-H...O and C-H...O interactions. Compounds (III) and (IV) are isomorphous; the [UO(2)(oda)(2)](2-) anions form a porous matrix which is nearly identical in the two structures, and the cations are located in channels formed in this matrix. Compounds (I) and (II) are very different from (III) and (IV): (I) forms a layered structure, while (II) forms ribbons.

Inter-molecular non-classical C-H⋯N hydrogen bonding inter-links the [Cu(C(3)H(5)N)(3)(C(12)H(18)N(6))](2+) cations into a two-dimensional supra-molecular sheet extending along the ab plane. The crystal packing also exhibits weak C-H⋯O inter-actions.

Site-specific protein transamination using N-methylpyridinium-4-carboxaldehyde.

Project description:The controlled attachment of synthetic groups to proteins is important for a number of fields, including therapeutics, where antibody-drug conjugates are an emerging area of biologic medicines. We have previously reported a site-specific protein modification method using a transamination reaction that chemoselectively oxidizes the N-terminal amine of a polypeptide chain to a ketone or an aldehyde group. The newly introduced carbonyl can be used for conjugation to a synthetic group in one location through the formation of an oxime or a hydrazone linkage. To expand the scope of this reaction, we have used a combinatorial peptide library screening platform as a method to explore new transamination reagents while simultaneously identifying their optimal N-terminal sequences. N-Methylpyridinium-4-carboxaldehyde benzenesulfonate salt (Rapoport's salt, RS) was identified as a highly effective transamination reagent when paired with glutamate-terminal peptides and proteins. This finding establishes RS as a transamination reagent that is particularly well suited for antibody modification. Using a known therapeutic antibody, herceptin, it was demonstrated that RS can be used to modify the heavy chains of the wild-type antibody or to modify both the heavy and the light chains after N-terminal sequence mutation to add additional glutamate residues.
