Project description:The relationships between grain yields and whole-plant accumulation of micronutrients such as zinc (Zn), iron (Fe), manganese (Mn) and copper (Cu) in maize (Zea mays L.) were investigated by studying their reciprocal internal efficiencies (RIEs, g of micronutrient requirement in plant dry matter per Mg of grain). Field experiments were conducted from 2008 to 2011 in North China to evaluate RIEs and shoot micronutrient accumulation dynamics during different growth stages under different yield and nitrogen (N) levels. Fe, Mn and Cu RIEs (average 64.4, 18.1 and 5.3 g, respectively) were less affected by the yield and N levels. ZnRIE increased by 15% with an increased N supply but decreased from 36.3 to 18.0 g with increasing yield. The effect of cultivars on ZnRIE was similar to that of yield ranges. The substantial decrease in ZnRIE may be attributed to an increased Zn harvest index (from 41% to 60%) and decreased Zn concentrations in straw (a 56% decrease) and grain (decreased from 16.9 to 12.2 mg kg-1) rather than greater shoot Zn accumulation. Shoot Fe, Mn and Cu accumulation at maturity tended to increase but the proportions of pre-silking shoot Fe, Cu and Zn accumulation consistently decreased (from 95% to 59%, 90% to 71% and 91% to 66%, respectively). The decrease indicated the high reproductive-stage demands for Fe, Zn and Cu with the increasing yields. Optimized N supply achieved the highest yield and tended to increase grain concentrations of micronutrients compared to no or lower N supply. Excessive N supply did not result in any increases in yield or micronutrient nutrition for shoot or grain. These results indicate that optimized N management may be an economical method of improving micronutrient concentrations in maize grain with higher grain yield.

Project description:Zinc deficiency in agricultural soils is a current global agroecosystems challenge. Maize exhibits elevated susceptibility to Zn deficiency and low response to zinc fertilization. As a result, there are contradicting literature reports on the crop response to zinc fertilization. This meta-analysis synthesized the current evidence on maize response to zinc fertilization from different studies and highlighted the potential innovations to improve the crop response to zinc application. Systematic literature searches were conducted on the Web of Science and Google Scholar for peer-reviewed publications. From the selected publications, data extracted were maize grain yield and maize grain zinc concentration. The meta-analysis was conducted in R statistical environment using the metafor package. The ratio of means was the chosen effect size measure used. The assessment of effect size heterogeneity showed that the study effect sizes were significantly heterogeneous and also publication bias was evident. The analysis showed 17% and 25% maize grain yield and grain zinc concentration response to zinc fertilization. As a result, zinc fertilization was associated with yield increments of up to 1 t ha-1 and 7.19 mg kg-1 grain zinc concentration over the control (no zinc application). Despite the observed maize grain response to zinc application, the median concentration of grain Zn was below the 38 mg kg-1 recommended maize grain zinc concentration to combat human zinc deficiency (hidden hunger). As a result, potential innovations likely to achieve sufficient maize grain zinc content were highlighted including the use of nano-particulate zinc oxide, foliar zinc application, timing of zinc application, precision fertilization and zinc micro-dosing. Due to scanty literature on the progress of these innovations in maize, follow-up studies are recommended to evaluate their potential success in the agronomic bio-fortification of maize with zinc.

Project description:The Zea Mays BIG GRAIN 1 HOMOLOG 1 (ZM-BG1H1) was ectopically expressed in maize. Elite commercial hybrid germplasm was yield tested in diverse field environment locations representing commercial models. Yield was measured in 101 tests across all 4 events, 26 locations over 2 years, for an average yield gain of 355 kg/ha (5.65 bu/ac) above control, with 83% tests broadly showing yield gains (range +2272 kg/ha to -1240 kg/ha), with seven tests gaining more than one metric ton per hectare. Plant and ear height were slightly elevated, and ear and tassel flowering time were delayed one day, but ASI was unchanged, and these traits did not correlate to yield gain. ZM-BG1H1 overexpression is associated with increased ear kernel row number and total ear kernel number and mass, but individual kernels trended slightly smaller and less dense. The ZM-BG1H1 protein is detected in the plasma membrane like rice OS-BG1. Five predominant native ZM-BG1H1 alleles exhibit little structural and expression variation compared to the large increased expression conferred by these ectopic alleles.

Project description:Nitrogen (N) supply could improve the grain yield of maize, which is of great importance to provide calories and nutrients in the diets of both humans and animals. Field experiments were conducted in 2009 and 2010 to investigate dynamic zinc (Zn) accumulation and the pre-silking and post-silking Zn uptake and their contributions to grain Zn accumulation of maize with different N supply under field conditions. Results showed that only 1.2% to 39.4% of grain Zn accumulation derived from pre-silking Zn uptake, with Zn remobilization being negatively affected by increasing N supply. However, post-silking Zn uptake (0.8-2.3 mg plant-1) and its substantial contribution to grain Zn accumulation (60.6%-98.8%) were progressively enhanced with the increasing N supply. Furthermore, grain Zn concentration was positively associated with grain N concentration (r = 0.752***), post-silking N uptake (r = 0.695***), and post-silking Zn uptake (r = 738***). A significant positive relationship was also found between post-silking uptake of N and Zn (r = 0.775***). These results suggest that N nutrition is a critical factor for shoot Zn uptake and its allocation to maize grain. Dry weight, and N and Zn concentration of grain and straw were significantly enhanced with the increasing N from "no N" to "optimal N" supply (150 kg N ha-1 in 2009 and 105 kg N ha-1 in 2010), but further increasing N supply (250 kg N ha-1) generally resulted in a non-significant increase in both cropping seasons. During the grain development, N supply also generally tended to improve grain N and Zn concentrations, but decrease phosphorus (P) concentration and the molar ratio of P to Zn compared with null N application. These results suggest that grain Zn accumulation mainly originates from post-silking Zn uptake. Applying N at optimal rates ensures better shoot Zn nutrition and contributes to post-silking Zn uptake, maintaining higher grain Zn availability by decreasing the molar ratio of P to Zn, and resulting in benefits to human nutrition.

Project description:Understanding the genetic and molecular basis of grain yield is important for maize improvement. Here, we identified 49 consensus quantitative trait loci (cQTL) controlling maize yield-related traits using QTL meta-analysis. Then, we collected yield-related traits associated SNPs detected by association mapping and identified 17 consensus significant loci. Comparing the physical positions of cQTL with those of significant SNPs revealed that 47 significant SNPs were located within 20 cQTL regions. Furthermore, intensive reviews of 31 genes regulating maize yield-related traits found that the functions of many genes were conservative in maize and other plant species. The functional conservation indicated that some of the 575 maize genes (orthologous to 247 genes controlling yield or seed traits in other plant species) might be functionally related to maize yield-related traits, especially the 49 maize orthologous genes in cQTL regions, and 41 orthologous genes close to the physical positions of significant SNPs. In the end, we prospected on the integration of the public sources for exploring the genetic and molecular mechanisms of maize yield-related traits, and on the utilization of genetic and molecular mechanisms for maize improvement.Supplementary informationThe online version contains supplementary material available at 10.1007/s11032-021-01214-3.

Project description:In the North China Plain, the excessive application of nitrogen (N) fertilizer for ensuring high yield and a single application at sowing for simplifying management in farmer practice lead to low N use efficiency and environmental risk in maize (Zea mays L.) production. However, it is unclear whether and how late split application with a lower level of N fertilizer influences maize yield. To address this question, a two-year field experiment was conducted with two commercial maize cultivars (Zhengdan 958 and Denghai 605) using a lower level of N input (180 kg ha-1) by setting up single application at sowing and split application at sowing and later stages (V12, R1, and R2) with four different ratios, respectively. The maize yield with split-applied 180 kg ha-1 N did not decrease compared to the average yield with 240 kg ha-1 N input in farmer practice, while it increased by 6.7% to 11.5% in the four N split-application treatments compared with that of the single-application control. Morphological and physiological analyses demonstrated that late split application of N (i) increased the net photosynthetic rate and chlorophyll content and thus promoted the photosynthetic efficiency during the reproductive stages; (ii) promoted the sink capacity via improved kernel number, endosperm cells division, and grain-filling rate; and (iii) increased the final N content and N efficiency in the plant. Therefore, we propose that late split application of N could reduce N fertilizer input and coordinately improve N efficiency and grain yield in summer maize production, which are likely achieved by optimizing the source-sink relations during the grain-filling stage.

Project description:Increasing maize grain yield has been a major focus of both plant breeding and genetic engineering to meet the global demand for food, feed, and industrial uses. We report that increasing and extending expression of a maize MADS-box transcription factor gene, zmm28, under the control of a moderate-constitutive maize promoter, results in maize plants with increased plant growth, photosynthesis capacity, and nitrogen utilization. Molecular and biochemical characterization of zmm28 transgenic plants demonstrated that their enhanced agronomic traits are associated with elevated plant carbon assimilation, nitrogen utilization, and plant growth. Overall, these positive attributes are associated with a significant increase in grain yield relative to wild-type controls that is consistent across years, environments, and elite germplasm backgrounds.

Project description:All over the world, a common problem in the soil is the low content of available zinc (Zn), which is unevenly distributed and difficult to move. However, information on the foraging strategies of roots in response to heterogeneous Zn supply is still very limited. Few studies have analyzed the adaptability of maize inbred lines with different Zn efficiencies to different low Zn stress time lengths in maize. This study analyzed the effects of different time lengths of low Zn stress on various related traits in different inbred lines. In addition, morphological plasticity of roots and the response of Zn-related important gene iron-regulated transporter-like proteins (ZIPs) were studied via simulating the heterogeneity of Zn nutrition in the soil. In this report, when Zn deficiency stress duration was extended (from 14 to 21 days), under Zn-deficient supply (0.5 μM), Zn efficiency (ZE) based on shoot dry weight of Wu312 displayed no significant difference, and ZE for Ye478 was increased by 92.9%. Under longer-term Zn deficiency, shoot, and root dry weights of Ye478 were 6.5 and 2.1-fold higher than those of Wu312, respectively. Uneven Zn supply strongly inhibited the development of some root traits in the -Zn region. Difference in shoot dry weights between Wu312 and Ye478 was larger in T1 (1.97 times) than in T2 (1.53 times). Under heterogeneous condition of Zn supply, both the -Zn region and the +Zn region upregulated the expressions of ZmZIP3, ZmZIP4, ZmZIP5, ZmZIP7, and ZmZIP8 in the roots of two inbred lines. These results indicate that extended time length of low-Zn stress will enlarge the difference of multiple physiological traits, especially biomass, between Zn-sensitive and Zn-tolerant inbred lines. There were significant genotypic differences of root morphology in response to heterogeneous Zn supply. Compared with split-supply with +Zn/+Zn, the difference of above-ground biomass between Zn-sensitive and Zn-tolerant inbred lines under split-supply with -Zn/+Zn was higher. Under the condition of heterogeneous Zn supply, several ZmZIP genes may play important roles in tolerance to low Zn stress, which can provide a basis for further functional characterization.

Project description:The combined application of nitrogen (N) and zinc (Zn) fertilizers is a promising agronomic strategy for the biofortification of wheat grain with Zn for human nutrition. A glasshouse experiment was carried out to assess the effects of supplying N on the uptake, translocation and accumulation of Zn in tissues of two wheat genotypes (Quartzo and BRS Parrudo) with contrasting potential for grain Zn biofortification. Winter wheat genotypes were grown to maturity in 5 cm diameter, 100 cm length tubes filled with a mixture of sand, grit and gravel (40:40:20 v/v/v) over a layer of 0.1 m3 of gravel, and supplied a full nutrient solution with low Zn (0.15 μM) or high Zn (2.25 μM) and low N (0.4 mM) or high N (4.0 mM) concentrations. High N supply increased biomass production, Zn concentration and Zn content of straw and grain in both Quartzo and BRS Parrudo. Grain Zn content more than doubled when the supplies of Zn and N were both increased from low to high in both genotypes. Quartzo had a greater grain yield than BRS Parrudo. BRS Parrudo had greater grain Zn concentration and Zn content than Quartzo. A greater N supply promoted better uptake, translocation to the shoot and accumulation of Zn within the grain. Quartzo and BRS Parrudo differed in their partitioning of biomass and Zn between tissues. It might be possible to combine the greater grain yield of Quartzo with the greater grain Zn accumulation of BRS Parrudo to deliver a greatly improved genotype for human food security.

Project description:Maize is the staple food crop for millions of people in sub-Saharan Africa. Iron (Fe) and zinc (Zn) deficiency is a significant health risk that mainly affects low-income populations who rely solely on maize-based diets. This problem can be alleviated by developing micronutrient-rich maize grain. The aim of this study was to determine the adaptation and performance of hybrids for Fe and Zn concentration and grain yield under low soil nitrogen (N) and optimal conditions. Eighteen hybrids derived from lines and testers with low, medium, and high Fe and Zn concentration were grown during the summer rainy seasons of 2017 and 2018 at three locations under low and optimal N conditions. There were significant genotype and environment effects for grain yield, and Fe and Zn concentration, but the genotype by environment interaction effects were the largest, accounting for between 36% and 56% of variation under low N conditions. Low N levels significantly reduced grain yield, and Fe and Zn concentration. Hybrids G1, G2, G4, G7, G10, G11, and G16 were relatively stable, with relatively high mean Fe and Zn concentrations, and low additive main effects and multiplicative interaction (AMMI) stability values and iron stability index (FSI) and zinc stability index (ZSI) under low N conditions. These genotypes can be considered for production under low N stress conditions. Two environments (E4 and E3) were identified for good discriminatory power for genotype performance in terms of Fe and Zn content, respectively. Stable and high-yielding genotypes with high Fe and Zn concentration can be used as biofortified hybrids, which can contribute to a sustainable solution to malnutrition in the region, especially under low N conditions.
