European molecular epidemiology and strain diversity of feline calicivirus.

Project description:Feline calicivirus (FCV) causes a variable syndrome of upper respiratory tract disease, mouth ulcers and lameness. A convenience-based prospective sample of oropharyngeal swabs (n=426) was obtained from five countries (France, Germany, Greece, Portugal and the UK). The prevalence of FCV by virus isolation was 22.2 per cent. Multivariable analysis found that animals presenting with lymphoplasmacytic gingivitis stomatitis complex were more likely to test positive for FCV infection. Furthermore, vaccinated cats up to 48?months of age were significantly less likely to be infected with FCV than unvaccinated animals of similar ages. Phylogenetic analysis based on consensus sequences for the immunodominant region of the capsid gene from 72 FCV isolates identified 46 strains. Thirteen of the 14 strains with more than one sequence were restricted to individual regions or sites in individual countries; the exception was a strain present in two sites close to each other in France. Four strains were present in more than one household. Five colonies, four of which were rescue shelters, had multiple strains within them. Polymerase sequence suggested possible rare recombination events. These locally, nationally and internationally diverse FCV populations maintain a continuous challenge to the control of FCV infection and disease.

Project description:Caliciviruses that cause diarrhea have been reported in both industrial and developing countries, including China, in recent years. Here, we report the complete genome of a porcine calicivirus strain, Ah-1, which is prevalent in swine groups in Anhui Province. This viral genome is 7,342 nucleotides (nt) long, excluding the poly(A) of the 3' end, which is 202 nt shorter in the 3' untranslated region (UTR) than that of the other Chinese porcine calicivirus strain (Ch-sw-sav1; GenBank accession number FJ387164), previously isolated in the Shanghai area, China, though they shared 98.8% sequence identity over the whole genome excluding the 202-nt-shorter region.

Distribution and genetic characterizations of Cryptosporidium spp. in pre-weaned dairy calves in Northeastern China's Heilongjiang Province.

Project description:BackgroundCryptosporidium spp. are common parasites of humans and animals. Farm animals, especially pre-weaned calves, are considered to be one of main animal reservoir hosts of Cryptosporidium in the transmission of human cryptosporidiosis. The aim of this study was to determine the distribution and genotypes of Cryptosporidium spp. in pre-weaned calves using molecular tools and to assess zoonotic transmission and elucidate the public health significance in northeastern China.Methodology/principal findingsA total of 151 fecal specimens from pre-weaned calves were collected in Heilongjiang Province and were screened for Cryptosporidium by PCR. The average prevalence of Cryptosporidium was 47.68% (72/151). Cryptosporidium spp. were characterized by DNA sequencing of the small subunit (SSU) rRNA gene and the 60-kDa glycoprotein (gp60) gene. Based on the SSU rRNA gene, five Cryptosporidium spp. were identified, including C. bovis (n = 34), C. andersoni (n = 26), C. ryanae (n = 5), C. meleagridis (n = 5) and C. parvum (n = 2). The SSU rRNA nucleotide sequences were identical to each other, respectively, within C. ryanae, C. parvum, C. meleagridis and C. andersoni. Four types of C. bovis were found in the SSU rRNA gene, with two novel types. The gp60 gene was successfully sequenced in one C. parvum isolate and three C. meleagridis isolates, with IIdA19G1 for C. parvum and IIIeA22G2R1 for C. meleagridis.Conclusion/significanceMolecular analysis indicates that Cryptosporidium spp. are endemic in pre-weaned calves in Heilongjiang Province. The findings of C. parvum and C. meleagridis suggested the possibility of zoonotic transmission and public health significance. The transmission dynamics of C. parvum and C. meleagridis needed to be clarified by further molecular epidemiologic studies from humans and animals. Whether calves could act as the natural reservoirs of C. meleagridis needed to be confirmed by more systematic experimental infection studies.

Characterization of a strain of feline calicivirus isolated from a dog faecal sample.

Project description:To expand the epidemiological understanding of feline calicivirus (FCV) in dogs, genotypic and phenotypic characterizations of a FCV strain isolated from a puppy showing enteritis were performed. After isolation in cell culture, the novel isolate was analysed by RT-PCR and the amplicons obtained were sequenced. In order to characterize the growth properties of the isolate, the size of the plaques, the temperature of inactivation and the kinetics of growth were evaluated. Moreover, the novel strain was used to perform a serological study on 86 canine serum samples and 81 feline sera by virus-neutralization assay. The comparative analysis of nucleotide and amino acid sequences of the isolate, named FCV-Te/10/07, revealed the highest identity to strain FCV-F65. The growth kinetic revealed that strain Te/10/07 grew more rapidly than F9 strain. By virus-neutralization assay, dogs from the same region of the isolate showed antibodies against the FCV-F9 vaccinal strain in 63.9% (55/86) of sera, while antibodies against the Te/10/07 were found in seven sera (8.13%). In cats neutralizing antibodies against Te/10/07 strain were recovered in 50.62% (41/81) of samples tested, even if 38 sera were positives for F9 strain with similar titres or higher. In three cats neutralization to Te/10/07 alone was seen. Our results confirmed the interspecific circulation of FCV strains among different animal species, but new investigations are needed to establish whether FCV is pathogenic in the dog and the role of interspecies circulation in pathogen spreading.

MLST subtypes and population genetic structure of Cryptosporidium andersoni from dairy cattle and beef cattle in northeastern China's Heilongjiang Province.

Project description:Cattle are the main reservoir host of C. andersoni, which shows a predominance in yearlings and adults of cattle. To understand the subtypes of C. andersoni and the population genetic structure in Heilongjiang Province, fecal specimens were collected from 420 dairy cattle and 405 beef cattle at the age of 12-14 months in eight cattle farms in five areas within this province and were screened for the presence of Cryptosporidium oocysts by microscopy after Sheather's sugar flotation technique. The average prevalence of Cryptosporidium spp. was 19.15% (158/825) and all the Cryptosporidium isolates were identified as C. andersoni by the SSU rRNA gene nested PCR-RFLP using SspI, VspI and MboII restriction enzymes. A total of 50 C. andersoni isolates were randomly selected and sequenced to confirm the RFLP results before they were subtyped by multilocus sequence typing (MLST) at the four microsatellite/minisatellite loci (MS1, MS2, MS3 and MS16). Four, one, two and one haplotypes were obtained at the four loci, respectively. The MLST subtype A4,A4,A4,A1 showed an absolute predominance and a wide distribution among the six MLST subtypes obtained in the investigated areas. Linkage disequilibrium analysis showed the presence of a clonal population genetic structure of C. andersoni in cattle, suggesting the absence of recombination among lineages. The finding of a clonal population genetic structure indicated that the prevalence of C. andersoni in cattle in Heilongjiang Province is not attributed to the introduction of cattle. Thus, prevention and control strategies should be focused on making stricter measures to avoid the occurrence of cross-transmission and re-infection between cattle individuals. These molecular data will also be helpful to explore the source attribution of infection/contamination of C. andersoni and to elucidate its transmission dynamics in Heilongjiang Province, even in China.
