Project description:Segmentation of the acoustic environment into discrete percepts is an important facet of auditory scene analysis (ASA). Segmentation of auditory stimuli into perceptually meaningful and localizable groups is central to ASA in everyday situations; for example, separation of discrete words from continuous sentences when processing language. This is particularly relevant to schizophrenia, where deficits in perceptual organization have been linked to symptoms and cognitive dysfunction. Here we examined event-related potentials in response to grouped tones to elucidate schizophrenia-related differences in acoustic segmentation. We report for the first time in healthy subjects a sustained potential that begins with group initiation and ends with the last tone of the group. These potentials were reduced in schizophrenia, with the greatest differences in responses to first and final tones. Importantly, reductions in sustained potentials in schizophrenia patients were associated with greater negative symptoms and deficits in IQ, working memory, learning, and social cognition. These results suggest deficits in auditory pattern segmentation in schizophrenia may compound deficits in many higher-order facets of the disorder.

Project description:BackgroundThe P50 event-related potential sensory gating deficit, a failure to inhibit responses to repeated stimuli, is a leading endophenotype for schizophrenia (SZ). Both gamma and beta event-related oscillations (EROs) are major contributors to the auditory P50 response. However, the topographic distribution of gamma and beta ERO responses to initial (S1) and repeat (S2) stimuli and the association of these oscillations with P50 sensory gating are not clear.MethodsA total of 51 schizophrenic patients, 25 unaffected first-degree relatives, and 34 healthy comparison subjects were tested using a paired-click paradigm. Evoked power of gamma- and beta-band responses using wavelet analyses to S1 and S2 stimuli and gating of EROs and P50 were the main outcome measures.ResultsA P50 gating deficit was found in patients (P < .001) and at a trend level in relatives (P = .087). Patients showed widely distributed reductions in gamma and beta EROs to S1 stimuli and S2 stimuli, respectively, and impaired gating in both frequencies. Reduced gamma and beta ERO activity in patients was associated primarily with age of onset. Relatives did not differ significantly from control subjects in either EROs power or gating. Gating of P50, gamma, and beta were not significantly correlated (r = .18-.19, P > .05).ConclusionsThese results suggest that ERO deficits in gamma to S1 and beta to S2 stimuli and impaired ERO gating are associated with SZ, but are not related to genetic liability for the illness. The components of information processing assessed by gamma- and beta gating appear to be independent from those mediated by P50 suppression.

Project description:To better characterize neurophysiologic processes underlying olfactory dysfunction in schizophrenia, nose-referenced 30-channel electroencephalogram was recorded from 32 patients and 35 healthy adults (18 and 18 male) during detection of hydrogen sulfide (constant-flow olfactometer, 200 ms unirhinal exposure). Event-related potentials (ERPs) were transformed to reference-free current source density (CSD) waveforms and analyzed by unrestricted Varimax-PCA. Participants indicated when they perceived a high (10 ppm) or low (50% dilution) odor concentration. Patients and controls did not differ in detection of high (23% misses) and low (43%) intensities and also had similar olfactory ERP waveforms. CSDs showed a greater bilateral frontotemporal N1 sink (305 ms) and mid-parietal P2 source (630 ms) for high than low intensities. N1 sink and P2 source were markedly reduced in patients for high intensity stimuli, providing further neurophysiological evidence of olfactory dysfunction in schizophrenia.

Project description:The clinical high risk (CHR) period is a phase denoting a risk for overt psychosis during which subacute symptoms often appear, and cognitive functions may deteriorate. To compare biological indices during this phase with those during first episode schizophrenia, we cross-sectionally examined sex- and age-matched clinical high risk (CHR, n=21), first episode schizophrenia patients (FESZ, n=20) and matched healthy controls (HC, n=25) on oddball and novelty paradigms and assessed the N100, P200, P3a and P3b as indices of perceptual, attentional and working memory processes. To our knowledge, this is the only such comparison using all of these event-related potentials (ERPs) in two paradigms. We hypothesized that the ERPs would differentiate between the three groups and allow prediction of a diagnostic group. The majority of ERPs were significantly affected in CHR and FESZ compared with controls, with similar effect sizes. Nonetheless, in logistic regression, only the P3a and N100 distinguished CHR and FESZ from healthy controls, suggesting that ERPs not associated with an overt task might be more sensitive to prediction of group membership.

Project description:Recent behavioral and neuroimaging studies have suggested multisensory processing deficits in patients with schizophrenia (SCZ). Thus far, the neural mechanisms underlying these deficits are not well understood. Previous studies with unisensory stimulation have shown altered neural oscillations in SCZ. As such, altered oscillations could contribute to aberrant multisensory processing in this patient group. To test this assumption, we conducted an electroencephalography (EEG) study in 15 SCZ and 15 control participants in whom we examined neural oscillations and event-related potentials (ERPs) in the sound-induced flash illusion (SIFI). In the SIFI multiple auditory stimuli that are presented alongside a single visual stimulus can induce the illusory percept of multiple visual stimuli. In SCZ and control participants we compared ERPs and neural oscillations between trials that induced an illusion and trials that did not induce an illusion. On the behavioral level, SCZ (55.7%) and control participants (55.4%) did not significantly differ in illusion rates. The analysis of ERPs revealed diminished amplitudes and altered multisensory processing in SCZ compared to controls around 135 ms after stimulus onset. Moreover, the analysis of neural oscillations revealed altered 25-35 Hz power after 100 to 150 ms over occipital scalp for SCZ compared to controls. Our findings extend previous observations of aberrant neural oscillations in unisensory perception paradigms. They suggest that altered ERPs and altered occipital beta/gamma band power reflect aberrant multisensory processing in SCZ.

Project description:AntecedentThe event-related potential (ERP) components P300 and mismatch negativity (MMN) have been linked to cognitive deficits in patients with schizophrenia. The diagnosis of schizophrenia could be improved by applying machine learning procedures to these objective neurophysiological biomarkers. Several studies have attempted to achieve this goal, but no study has examined Multiple Kernel Learning (MKL) classifiers. This algorithm finds optimally a combination of kernel functions, integrating them in a meaningful manner, and thus could improve diagnosis.ObjectiveThis study aimed to examine the efficacy of the MKL classifier and the Boruta feature selection method for schizophrenia patients (SZ) and healthy controls (HC) single-subject classification.MethodsA cohort of 54 SZ and 54 HC participants were studied. Three sets of features related to ERP signals were calculated as follows: peak related features, peak to peak related features, and signal related features. The Boruta algorithm was used to evaluate the impact of feature selection on classification performance. An MKL algorithm was applied to address schizophrenia detection.ResultsA classification accuracy of 83% using the whole dataset, and 86% after applying Boruta feature selection was obtained. The variables that contributed most to the classification were mainly related to the latency and amplitude of the auditory P300 paradigm.ConclusionThis study showed that MKL can be useful in distinguishing between schizophrenic patients and controls when using ERP measures. Moreover, the use of the Boruta algorithm provides an improvement in classification accuracy and computational cost.

Project description:Using neuropsychological investigation and visual event-related potentials (ERPs), we aimed to compare the ERPs and cognitive function of nondemented Parkinson's disease (PD) patients with and without visual hallucinations (VHs) and of control subjects. We recruited 12 PD patients with VHs (PD-H), 23 PD patients without VHs (PD-NH), and 18 age-matched controls. All subjects underwent comprehensive neuropsychological assessment and visual ERPs measurement. A visual odd-ball paradigm with two different fixed interstimulus intervals (ISI) (1600?ms and 5000?ms) elicited visual ERPs. The frontal test battery was used to assess attention, visual-spatial function, verbal fluency, memory, higher executive function, and motor programming. The PD-H patients had significant cognitive dysfunction in several domains, compared to the PD-NH patients and controls. The mean P3 latency with ISI of 1600?ms in PD-H patients was significantly longer than that in controls. Logistic regression disclosed UPDRS-on score and P3 latency as significant predictors of VH. Our findings suggest that nondemented PD-H patients have worse cognitive function and P3 measurements. The development of VHs in nondemented PD patients might be implicated in executive dysfunction with altered visual information processing.

Project description:The detection of event-related potentials (ERPs) through electroencephalogram (EEG) analysis is a well-established method for understanding brain functions during a cognitive process. To increase the signal-to-noise ratio (SNR) and stationarity of the data, ERPs are often filtered to a wideband frequency range, such as 0.05-30 Hz. Alternatively, a natural-filtering procedure can be performed through empirical mode decomposition (EMD), which yields intrinsic mode functions (IMFs) for each trial of the EEG data, followed by averaging over trials to generate the event-related modes. However, although the EMD-based filtering procedure has advantages such as a high SNR, suitable waveform shape, and high statistical power, one fundamental drawback of the procedure is that it requires the selection of an IMF (or a partial sum of a range of IMFs) to determine an ERP component effectively. Therefore, in this study, we propose an intrinsic ERP (iERP) method to overcome the drawbacks and retain the advantages of event-related mode analysis for investigating ERP components. The iERP method can reveal multiple ERP components at their characteristic time scales and suitably cluster statistical effects among modes by using a tailored definition of each mode's neighbors. We validated the iERP method by using realistic EEG data sets acquired from a face perception task and visual working memory task. By using these two data sets, we demonstrated how to apply the iERP method to a cognitive task and incorporate existing cluster-based tests into iERP analysis. Moreover, iERP analysis revealed the statistical effects between (or among) experimental conditions more effectively than the conventional ERP method did.

Project description:Social comparison, a widespread phenomenon in human society, has been found to affect outcome evaluation. The need to belong to a social group may result in distinct neural responses to diverse social comparison outcomes. To extend previous studies by examining how social comparison with hierarchical characteristics is temporally processed, electroencephalography responses were recorded in the current study. Participants played a lottery game with two pseudo-players simultaneously and received both their own and the other two players' outcomes. Results of three event-related potential components, including the P2, the feedback-related negativity (FRN), and the late positive component (LPC), indicate that social comparison manifests in three stages. First, outcomes indicating a different performance from others elicited a larger P2 than evenness. Second, the FRN showed hierarchical sensitivity to social comparison outcomes. This effect manifested asymmetrically. Finally, large difference between the participant's outcome and the other two players' evoked a larger LPC than the medium difference and the even condition. We suggest that during social comparison, people detect if there is any difference between self and others, and then evaluate the information of this difference hierarchically, and finally interpret the situations in which oneself deviates from the group as most motivationally salient.

Project description:Recent research has shown that neurophysiological activation during action planning depends on the orientation to initial or final action goals for precision grips. However, the neural signature for a distinct class of grasping, power grips, is still unknown. The aim of the present study was to differentiate between cerebral activity, by means of event-related potentials (ERPs), and its temporal organization during power grips executed with an emphasis on either the initial or final parts of movement sequences. In a grasp and transportation task, visual cues emphasized either the grip (the immediate goal) or the target location (the final goal). ERPs differed between immediate and final goal-cued conditions, suggesting different means of operation dependent on goal-relatedness. Differences in mean amplitude occurred earlier for power grips than for recently reported precision grips time-locked to grasping over parieto-occipital areas. Time-locked to final object placement, differences occurred within a similar time window for power and precision grips over frontal areas. These results suggest that a parieto-frontal network of activation is of crucial importance for grasp planning and execution. Our results indicate that power grip preparation and execution for goal-related actions are controlled by similar neural mechanisms as have been observed during precision grips, but with a distinct temporal pattern.
