Anther wall formation in Solanaceae species.

ABSTRACT: Anther wall formation was studied in 32 species belonging to 27 genera of Solanaceae. Dicotyledonous and basic types of wall formation were observed, as well as several deviations due to subsequent periclinal divisions in the layers formed (middle layers and sometimes the endothecium). One type of wall formation was observed in each species. Some genera are uniform in their type of wall formation, while others are heterogeneous; a similar situation was observed at the tribal level. Summarizing all reported information on anther wall formation in the Solanaceae, 64 % of species show the basic type, while the remaining 36 % show the dicotyledonous type. Thus, neither type predominates, and no single type characterizes genera, tribes or the entire family.

Ostkpr1 functions in anther cuticle development and pollen wall formation in rice.

Project description:BackgroundDuring pollen wall formation in flowering plants, a conserved metabolon consisting of acyl-CoA synthetase (ACOS), polyketide synthase (PKS) and tetraketide α-pyrone reductase (TKPR), is required for sporopollenin synthesis. Despite this, the precise function of each of these components in different species remains unclear.ResultsIn this study, we characterized the function of OsTKPR1, a rice orthologue of Arabidopsis TKPR1. Loss of function of OsTKPR1 delayed tapetum degradation, reduced the levels of anther cuticular lipids, and impaired Ubisch body and pollen exine formation, resulting in complete male sterility. In addition, the phenylpropanoid pathway in mutant anthers was remarkably altered. Localization studies suggest that OsTKPR1 accumulates in the endoplasmic reticulum, while specific accumulation of OsTKPR1 mRNA in the anther tapetum and microspores is consistent with its function in anther and pollen wall development.ConclusionsOur results show that OsTKPR1 is indispensable for anther cuticle development and pollen wall formation in rice, providing new insights into the biochemical mechanisms of the conserved sporopollenin metabolon in flowering plants.

An atypical strictosidine synthase, OsSTRL2, plays key roles in anther development and pollen wall formation in rice.

Project description:Strictosidine synthase (STR) plays an important role in the biosynthesis of terpenoid indole alkaloids (TIAs) and is expressed in a range of active meristematic tissues of higher plants. STR proteins are involved in different physiological and biochemical pathways. However, the function of STR proteins in rice development remains poorly understood. In this study, we identified 21 possible STR-like (OsSTRL) family members in rice genome and found that only one gene, OsSTRL2, exhibited a pre-emergency specific florescence expression pattern. Tissue-specific expression profile analysis, β-glucuronidase histochemical (GUS) staining and RNA in situ hybridization confirmed that OsSTRL2 was highly expressed in tapetal cells and microspores. Comparative protein sequence analysis indicated that OsSTRL2 lacked the key catalytic residue found in a typical STR (STR1), although it possessed conserved β-propellers and α-helices formed the basic structure of STR1. OsSTRL2 knockout mutant resulted to male sterility because of the defects in anther development and pollen wall formation. Subcellular localization of OsSTRL2-YFP revealed that the OsSTRL2 protein was primarily localized in the endoplasmic reticulum (ER). Therefore, OsSTRL2 is an atypical strictosidine synthase that plays crucial roles in regulating anther development and pollen wall formation in rice.

TCP24 modulates secondary cell wall thickening and anther endothecium development.

Project description:miR319-targeted TCP genes are believed to regulate cell division in leaves and floral organs. However, it remains unknown whether these genes are involved in cell wall development. Here, we report that TCP24 negatively regulates secondary wall thickening in floral organs and roots. The overexpression of the miR319a-resistant version of TCP24 in Arabidopsis disrupted the thickening of secondary cell walls in the anther endothecium, leading to male sterility because of arrested anther dehiscence and pollen release. Several genes linked to secondary cell wall biogenesis and thickening were down-regulated in these transgenic plants. By contrast, the inhibition of TCP24 using the ectopic expression of a TCP24-SRDX repressor fusion protein, or the silencing of TCP genes by miR319a overexpression, increased cell wall lignification and the enhanced secondary cell wall thickening. Our results suggest that TCP24 acts as an important regulator of secondary cell wall thickening and modulates anther endothecium development.

miR2118-dependent U-rich phasiRNA production in rice anther wall development.

Project description:Reproduction-specific small RNAs are vital regulators of germline development in animals and plants. MicroRNA2118 (miR2118) is conserved in plants and induces the production of phased small interfering RNAs (phasiRNAs). To reveal the biological functions of miR2118, we describe here rice mutants with large deletions of the miR2118 cluster. Our results demonstrate that the loss of miR2118 causes severe male and female sterility in rice, associated with marked morphological and developmental abnormalities in somatic anther wall cells. Small RNA profiling reveals that miR2118-dependent 21-nucleotide (nt) phasiRNAs in the anther wall are U-rich, distinct from the phasiRNAs in germ cells. Furthermore, the miR2118-dependent biogenesis of 21-nt phasiRNAs may involve the Argonaute proteins OsAGO1b/OsAGO1d, which are abundant in anther wall cell layers. Our study highlights the site-specific differences of phasiRNAs between somatic anther wall and germ cells, and demonstrates the significance of miR2118/U-phasiRNA functions in anther wall development and rice reproduction.

A new species of spiny Solanum (Solanaceae) from Peru.

Project description:A new species of Solanum is described from Peru. Solanum junctum S. Stern & M. Nee, sp nov. is a member of the Torva clade of the spiny solanums (Leptostemonum clade). The narrow corolla lobes and recurved prickles of Solanum junctum are similar to species in the Micracantha clade, but Solanum junctum differs in its branched inflorescences and upright green fruits. These characteristics are shared with other members the Torva clade; within this section Solanum junctum is morphologically most similar to Solanum subinerme and Solanum poinsettiifolium. Solanum subinerme has larger flowers, longer cauline prickles, and often has long straight prickles on the adaxial leaf surface that are lacking in Solanum junctum. Solanum poinsettiifolium has fewer spines, dense white tomentum on the abaxial leaf surfaces, stout unbranched inflorescences, and more extensive interpetalar corolla tissue than Solanum junctum.

The stress applied was 150mM NaCl and control plants were watered without the additional salt. Samples were collected at 0, 6, 12, 24, 48 and 96 hours after the first application of the salt.
