Signal detection theory (SDT) shows that optimal choice bias depends not only on the prior but also the sensory uncertainty; however, it is unclear how animals integrate sensory inputs with various uncertainties and reward expectations to optimize choices. We developed a tone-frequency discrimination task for head-fixed mice in which we randomly presented either a long or short sound stimulus and biased the choice outcomes. The choice was less accurate and more biased toward the large-reward side in short- than in long-stimulus trials. Analysis with SDT found that mice did not use a separate, optimal choice threshold in different sound durations. Instead, mice updated one threshold for short and long stimuli with a simple reinforcement-learning rule. Our task in head-fixed mice helps understanding how the brain integrates sensory inputs and prior.
Project description:Early life stress (ELS) is strongly associated with negative outcomes in adulthood, including reduced motivation and increased negative mood. The mechanisms mediating these relations, however, are poorly understood. We examined the relation between exposure to ELS and reward-related brain activity, which is known to predict motivation and mood, at age 26, in a sample followed since kindergarten with annual assessments. Using functional neuroimaging, we assayed individual differences in the activity of the ventral striatum (VS) during the processing of monetary rewards associated with a simple card-guessing task, in a sample of 72 male participants. We examined associations between a cumulative measure of ELS exposure and VS activity in adulthood. We found that greater levels of cumulative stress during childhood and adolescence predicted lower reward-related VS activity in adulthood. Extending this general developmental pattern, we found that exposure to stress early in development (between kindergarten and grade 3) was significantly associated with variability in adult VS activity. Our results provide an important demonstration that cumulative life stress, especially during this childhood period, is associated with blunted reward-related VS activity in adulthood. These differences suggest neurobiological pathways through which a history of ELS may contribute to reduced motivation and increased negative mood.
Project description:BackgroundChildhood adversity increases the risk of psychopathology, but the neurobiological mechanisms underlying this vulnerability are not well-understood. In animal models, early adversity is associated with dysfunction in basal ganglia regions involved in reward processing, but this relationship has not been established in humans.MethodsFunctional magnetic resonance imaging was used to examine basal ganglia responses to: 1) cues signaling possible monetary rewards and losses; and 2) delivery of monetary gains and penalties, in 13 young adults who experienced maltreatment before age 14 years and 31 nonmaltreated control subjects.ResultsRelative to control subjects, individuals exposed to childhood adversity reported elevated symptoms of anhedonia and depression, rated reward cues less positively, and displayed a weaker response to reward cues in the left globus pallidus. There were no group differences in right hemisphere basal ganglia response to reward cues or in basal ganglia response to loss cues, no-incentive cues, gains, or penalties.ConclusionsResults indicate that childhood adversity in humans is associated with blunted subjective responses to reward-predicting cues as well as dysfunction in left basal ganglia regions implicated in reward-related learning and motivation. This dysfunction might serve as a diathesis that contributes to the multiple negative outcomes and psychopathologies associated with childhood adversity. The findings suggest that interventions that target motivation and goal-directed action might be useful for reducing the negative consequences of childhood adversity.
Project description:What kind of strategies subjects follow in various behavioral circumstances has been a central issue in decision making. In particular, which behavioral strategy, maximizing or matching, is more fundamental to animal's decision behavior has been a matter of debate. Here, we prove that any algorithm to achieve the stationary condition for maximizing the average reward should lead to matching when it ignores the dependence of the expected outcome on subject's past choices. We may term this strategy of partial reward maximization "matching strategy". Then, this strategy is applied to the case where the subject's decision system updates the information for making a decision. Such information includes subject's past actions or sensory stimuli, and the internal storage of this information is often called "state variables". We demonstrate that the matching strategy provides an easy way to maximize reward when combined with the exploration of the state variables that correctly represent the crucial information for reward maximization. Our results reveal for the first time how a strategy to achieve matching behavior is beneficial to reward maximization, achieving a novel insight into the relationship between maximizing and matching.
Project description:Recently, progress has been observed in the knowledge about Duchenne Muscular Dystrophy (DMD), which is a severe and commonly diagnosed genetic myopathy in childhood, historically resulting in early death. Currently, there are a lot of methods available to improve the clinical course of DMD and extend patients' life expectancy to more than 30 years of age. The key issue for DMD patients is the period between 16-18 years of age, which is described as a transition from pediatric- to adult-oriented healthcare. Adolescents and adults with DMD have highly complex healthcare needs associated with long-term steroid usage, orthopedic, ventilation, cardiac, and gastrointestinal problems. The current paper provides a comprehensive overview of special healthcare needs related to the transfer of a patient with DMD from child-oriented to adult-oriented care. Additionally, the need to organize effective care for adults with DMD is presented.
Project description:BackgroundBipolar disorder (BD) is a severe mental illness that can have high costs for youths (<18 years old) and adults. Relative to healthy controls (HC), individuals with BD often show impaired attention, working memory, executive function, and cognitive flexibility (the ability to adapt to changing reward/punishment contingencies). In our study of youths and young adults with BD, we investigated 1) how cognitive flexibility varies developmentally in BD, and 2) whether it is independent of other executive function deficits associated with BD.MethodsWe measured errors on a reversal-learning task, as well as spatial working memory and other executive function, among participants with BD (N=75) and HC (N=130), 7-27 years old. Regression analyses focused on the effects of diagnosis on reversal-learning errors, controlling for age, gender, IQ, spatial span, and executive function. Similar analyses examined non-reversal errors to rule out general task impairment.ResultsParticipants with BD, regardless of age, gender, or cognitive ability, showed more errors than HC on the response reversal stages of the cognitive flexibility task. However, participants with BD did not show more errors on non-reversal stages, even when controlling for other variables.LimitationsStudy limitations include the cross-sectional, rather than longitudinal, design; inability to measure non-linear age effects; and inclusion of medicated participants and those with psychiatric comorbidity.ConclusionsIndividuals with BD show a specific impairment in reversing a previously rewarded response, which persists across the transition from childhood to young adulthood. Tailored interventions targeting this deficit may be effective throughout this developmentally turbulent time.
Project description:The structure and stability of families have long stood as key predictors of juvenile delinquency. Boys from "broken homes" experience a higher prevalence of juvenile delinquency than those from intact families (Rebellon 2002, Wells and Rankin 1991). Unresolved is whether the consequences of frequently disrupted family contexts endure to shape criminal trajectories into adulthood. Long-term influence may also be indirect. Life course criminologists credit family formation during the transition to adulthood, and particularly marriage, for redirecting men's criminal trajectories, but children who experience repeated changes in family structure are more likely to experience precarious starts to their own eventual family formation. Using data from the Panel Study of Income Dynamics and its two child-centered supplemental studies (N=1,127), we find that the experience of repeated family structure change is associated with higher rates of arrest and incarceration during early adulthood for white men but not for black men. This association is partially mediated by a slower transition to marriage among men who experienced three or more changes in family structure during childhood.
Project description:Given the importance of gamma oscillations in normal and disturbed cognition, there has been growing interest in their developmental trajectory. In the current study, age-related changes in sensory cortical gamma were studied using the auditory steady-state response (ASSR), indexing cortical activity entrained to a periodic auditory stimulus. A large sample (n = 188) aged 8-22 years had electroencephalography recording of ASSR during 20-, 30-, and 40-Hz click trains, analyzed for evoked amplitude, phase-locking factor (PLF) and cross-frequency coupling (CFC) with lower frequency oscillations. Both 40-Hz evoked power and PLF increased monotonically from 8 through 16 years, and subsequently decreased toward ages 20-22 years. CFC followed a similar pattern, with strongest age-related modulation of 40-Hz amplitude by the phase of delta oscillations. In contrast, the evoked power, PLF and CFC for the 20- and 30-Hz stimulation were distinct from the 40-Hz condition, with flat or decreasing profiles from childhood to early adulthood. The inverted U-shaped developmental trajectory of gamma oscillations may be consistent with interacting maturational processes-such as increasing fast GABA inhibition that enhances gamma activity and synaptic pruning that decreases gamma activity-that may continue from childhood through to adulthood.
Project description:This article focuses on neighborhood and geographic change arising with the first "selection" of an independent residential setting: the transition out of the family home. Data from two sources-the Project on Human Development in Chicago Neighborhoods, and the Panel Study of Income Dynamics-are used to provide complementary analyses of trajectories of change in geographic location and neighborhood racial and economic composition during young adulthood. Findings indicate that for young adults who originate in segregated urban areas and remain in such areas, the period of young adulthood is characterized by continuity in neighborhood conditions and persistent racial inequality from childhood to adulthood. For young adults who exit highly segregated urban areas, this period is characterized by a substantial leveling of racial inequality, with African Americans moving into less-poor, less-segregated neighborhoods. However, the trend toward racial equality in young adulthood is temporary, as the gaps between whites and blacks grow as the young adults move further into adulthood. Crucial to the reemergence of racial inequality in neighborhood environments is the process of "unselected" change, or change in neighborhood conditions that occurs around young adults after they move to a new neighborhood environment.
Project description:Nativity differences in youths' health in the United States are striking, with the children of foreign-born parents showing more favorable outcomes than those of native-born parents. Very little is known about how inequalities evolve within the same individuals over time, or more generally about life cycle aspects of the health integration of youth with migration backgrounds. Using data from the National Longitudinal Study of Adolescent Health, I examine nativity differences in trajectories of weight gain during adolescence and early adulthood, as well as the degree to which trajectories are stratified by race/ethnicity and socioeconomic status. Do nativity differences converge, diverge or remain stable over time, and how are patterns socially stratified within and across nativity groups? I find that first-generation adolescents begin at a lower weight than their third generation peers and gain weight at a significantly slower pace, producing meaningful differences by early adulthood. More complex examination of the relationship between nativity and weight gain reveals additional differences by ethnicity: the foreign-born advantage over time does not extend as strongly to Hispanic adolescents. The findings demonstrate how the health-related integration of foreign-born youth is tied to race/ethnicity and socioeconomic circumstances, and suggest the need to examine the ways in which social circumstances and health change together.
