Hierarchical assembly of a dual-responsive macroscopic insulated molecular wire bundle in a gradient system.

ABSTRACT: Here, we report the hierarchical self-assembly of a cationic gemini amphiphile, Azo 1, in a composition gradient solution generated using solvent evaporation. As the gradient solution is formed, Azo 1 forms nanorods in the lower region of the solution. Depending on solvent composition, these nanorods can further develop into nanofibres, which can then intertwine to form double helices and other types of nanohelices in the upper region of the solution. Finally, a macroscopic wire bundle is formed via the fusion of nanohelices; this ribbon-like bundle exhibits elasticity and linear ohmic resistance properties. More intriguingly, this bundle exhibits photoresponsive properties that affect its deformation and conductivity, as well as a rapid electroresponse that affects its conductivity, indicating that it is feasible to control the charge pathway.

The Crystal Structure of a Biological Insulated Transmembrane Molecular Wire.

Project description:A growing number of bacteria are recognized to conduct electrons across their cell envelope, and yet molecular details of the mechanisms supporting this process remain unknown. Here, we report the atomic structure of an outer membrane spanning protein complex, MtrAB, that is representative of a protein family known to transport electrons between the interior and exterior environments of phylogenetically and metabolically diverse microorganisms. The structure is revealed as a naturally insulated biomolecular wire possessing a 10-heme cytochrome, MtrA, insulated from the membrane lipidic environment by embedding within a 26 strand ?-barrel formed by MtrB. MtrAB forms an intimate connection with an extracellular 10-heme cytochrome, MtrC, which presents its hemes across a large surface area for electrical contact with extracellular redox partners, including transition metals and electrodes.

Microfibres and macroscopic films from the coordination-driven hierarchical self-assembly of cylindrical micelles.

Project description:Anisotropic nanoparticles prepared from block copolymers are of growing importance as building blocks for the creation of synthetic hierarchical materials. However, the assembly of these structural units is generally limited to the use of amphiphilic interactions. Here we report a simple, reversible coordination-driven hierarchical self-assembly strategy for the preparation of micron-scale fibres and macroscopic films based on monodisperse cylindrical block copolymer micelles. Coordination of Pd(0) metal centres to phosphine ligands immobilized within the soluble coronas of block copolymer micelles is found to induce intermicelle crosslinking, affording stable linear fibres comprised of micelle subunits in a staggered arrangement. The mean length of the fibres can be varied by altering the micelle concentration, reaction stoichiometry or aspect ratio of the micelle building blocks. Furthermore, the fibres aggregate on drying to form robust, self-supporting macroscopic micelle-based thin films with useful mechanical properties that are analogous to crosslinked polymer networks, but on a longer length scale.

Insulated conjugated bimetallopolymer with sigmoidal response by dual self-controlling system as a biomimetic material.

Project description:Biological systems are known to spontaneously adjust the functioning of neurotransmitters, ion channels, and the immune system, being promoted or regulated through allosteric effects or inhibitors, affording non-linear responses to external stimuli. Here we report that an insulated conjugated bimetallopolymer, in which Ru(II) and Pt(II) complexes are mutually connected with insulated conjugations, exhibits phosphorescence in response to CO gas. The net profile corresponds to a sigmoidal response with a dual self-controlling system, where drastic changes were exhibited at two threshold concentrations. The first threshold for activation of the system is triggered by the depolymerization of the non-radiative conjugated polymer to luminescent monomers, while the second one for regulation is triggered by the switch in the rate-determining step of the Ru complex. Such a molecular design with cooperative multiple transition metals would provide routes for the development of higher-ordered artificial molecular systems bearing bioinspired responses with autonomous modulation.

Microfluidic Droplet-Facilitated Hierarchical Assembly for Dual Cargo Loading and Synergistic Delivery.

Project description:Bottom-up hierarchical assembly has emerged as an elaborate and energy-efficient strategy for the fabrication of smart materials. Herein, we present a hierarchical assembly process, whereby linear amphiphilic block copolymers are self-assembled into micelles, which in turn are accommodated at the interface of microfluidic droplets via cucurbit[8]uril-mediated host-guest chemistry to form supramolecular microcapsules. The monodisperse microcapsules can be used for simultaneous carriage of both organic (Nile Red) and aqueous-soluble (fluorescein isothiocyanate-dextran) cargo. Furthermore, the well-defined compartmentalized structure benefits from the dynamic nature of the supramolecular interaction and offers synergistic delivery of cargos with triggered release or through photocontrolled porosity. This demonstration of premeditated hierarchical assembly, where interactions from the molecular to microscale are designed, illustrates the power of this route toward accessing the next generation of functional materials and encapsulation strategies.

Liquid flow reversibly creates a macroscopic surface charge gradient.

Project description:The charging and dissolution of mineral surfaces in contact with flowing liquids are ubiquitous in nature, as most minerals in water spontaneously acquire charge and dissolve. Mineral dissolution has been studied extensively under equilibrium conditions, even though non-equilibrium phenomena are pervasive and substantially affect the mineral-water interface. Here we demonstrate using interface-specific spectroscopy that liquid flow along a calcium fluoride surface creates a reversible spatial charge gradient, with decreasing surface charge downstream of the flow. The surface charge gradient can be quantitatively accounted for by a reaction-diffusion-advection model, which reveals that the charge gradient results from a delicate interplay between diffusion, advection, dissolution, and desorption/adsorption. The underlying mechanism is expected to be valid for a wide variety of systems, including groundwater flows in nature and microfluidic systems.

A dual-responsive nanocapsule via disulfide-induced self-assembly for therapeutic agent delivery.

Project description:One-step synthesis of fluorescent molecules (SNBDP) containing one disulfide bond and two o-nitrobenzyl groups was demonstrated via multi-component Passerini reaction. This hydrophobic SNBDP could self-assemble into nanocapsules (SNBDP NCs) in aqueous solution via disulfide-induced assembly. The obtained nanocapsules were stable in aqueous solution for several weeks and exhibited enhanced fluorescence when nanocapsules were destroyed due to disaggregation-induced emission. The nanocapsules not only were reduction-sensitive and light-responsive, but also could be endocytosed by HeLa cells for cellular imaging. The enhanced fluorescence in the glutathione (GSH) pretreated HeLa cells showed that the compound was reduction-sensitive in living cells. In vitro WST-8 assays showed the nanocapsules were biocompatible and could further be used as drug delivery carriers. Indocyanine green (ICG), a clinically approved NIR dye, was loaded into the nanocapsules (ICG@SNBDP NCs). ICG@SNBDP NCs showed enhanced photothermal efficacy compared with same concentration of free ICG under 808-nm laser irradiation. Consequently, ICG@SNBDP NCs upon NIR irradiation can effectively kill cancer cells through local hyperthermia. These results highlight the potential of disulfide-induced nanocapsules as smart nanoparticles for cellular imaging and therapeutic agent delivery.

Hierarchical dynamics as a macroscopic organizing principle of the human brain.

Project description:Multimodal evidence suggests that brain regions accumulate information over timescales that vary according to anatomical hierarchy. Thus, these experimentally defined "temporal receptive windows" are longest in cortical regions that are distant from sensory input. Interestingly, spontaneous activity in these regions also plays out over relatively slow timescales (i.e., exhibits slower temporal autocorrelation decay). These findings raise the possibility that hierarchical timescales represent an intrinsic organizing principle of brain function. Here, using resting-state functional MRI, we show that the timescale of ongoing dynamics follows hierarchical spatial gradients throughout human cerebral cortex. These intrinsic timescale gradients give rise to systematic frequency differences among large-scale cortical networks and predict individual-specific features of functional connectivity. Whole-brain coverage permitted us to further investigate the large-scale organization of subcortical dynamics. We show that cortical timescale gradients are topographically mirrored in striatum, thalamus, and cerebellum. Finally, timescales in the hippocampus followed a posterior-to-anterior gradient, corresponding to the longitudinal axis of increasing representational scale. Thus, hierarchical dynamics emerge as a global organizing principle of mammalian brains.

Peptide-Controlled Assembly of Macroscopic Calcium Oxalate Nanosheets.

Project description:The fabrication of two-dimensional (2D) biomineral nanosheets is of high interest owing to their promise for applications in electronics, filtration, catalysis, and chemical sensing. Using a facile approach inspired by biomineralization in nature, we fabricate laterally macroscopic calcium oxalate nanosheets using β-folded peptides. The template peptides are composed of repetitive glutamic acid and leucine amino acids, self-organized at the air-water interface. Surface-specific sum frequency generation spectroscopy and molecular dynamics simulations reveal that the formation of oxalate nanosheets relies on the peptide-Ca2+ ion interaction at the interface, which not only restructures the peptides but also templates Ca2+ ions into a calcium oxalate dihydrate lattice. Combined, this enables the formation of a critical structural intermediate in the assembly pathway toward the oxalate sheet formation. These insights into peptide-ion interfacial interaction are important for designing novel inorganic 2D materials.

Dual responsive PMEEECL-PAE block copolymers: a computational self-assembly and doxorubicin uptake study.

Project description:The self-assembly behaviour of dual-responsive block copolymers and their ability to solubilize the anticancer drug doxorubicin (DOX) has been investigated using all-atom molecular dynamics (MD) simulations, MARTINI coarse-grained (CG) force field simulation and Scheutjens-Fleer self-consistent field (SCF) computations. These diblock copolymers, composed of poly{γ-2-[2-(2-methoxyethoxy)ethoxy]ethoxy-ε-caprolactone} (PMEEECL) and poly(β-amino ester) (PAE) are dual-responsive: the PMEEECL block is thermoresponsive (becomes insoluble above a certain temperature), while the PAE block is pH-responsive (becomes soluble below a certain pH). Three MEEECL20-AE M compositions with M = 5, 10, and 15, have been studied. All-atom MD simulations have been performed to calculate the coil-to-globule transition temperature (T cg) of these copolymers and finding appropriate CG mapping for both PMEEECL-PAE and DOX. The output of the MARTINI CG simulations is in agreement with SCF predictions. The results show that DOX is solubilized with high efficiency (75-80%) at different concentrations inside the PMEEECL-PAE micelles, although, interestingly, the loading efficiency is reduced by increasing the drug concentration. The non-bonded interaction energy and the RDF between DOX and water beads confirm this result. Finally, MD simulations and SCF computations reveal that the responsive behaviour of PMEEECL-PAE self-assembled structures take place at temperature and pH ranges appropriate for drug delivery.
