Ecology and Genetic Structure of the Parasitoid Phobocampe confusa (Hymenoptera: Ichneumonidae) in Relation to Its Hosts, Aglais Species (Lepidoptera: Nymphalidae).

Project description:The biology of parasitoids in natural ecosystems remains very poorly studied, though they are key species for their functioning. Here we focused on Phobocampe confusa, a Nymphalini specialist, responsible for high mortality rates in charismatic butterfly species in Europe (genus Aglais). We studied its ecology and genetic structure in connection with those of its host butterflies in Sweden. To this aim, we gathered data from 428 P. confusa individuals reared from 6094 butterfly larvae (of A. urticae, A. io, and in two occasions of Araschnia levana) collected over two years (2017 and 2018) and across 19 sites distributed along a 500 km latitudinal gradient. We found that P. confusa is widely distributed along the latitudinal gradient. Its distribution seems constrained over time by the phenology of its hosts. The large variation in climatic conditions between sampling years explains the decrease in phenological overlap between P. confusa and its hosts in 2018 and the 33.5% decrease in the number of butterfly larvae infected. At least in this study, P. confusa seems to favour A. urticae as host. While it parasitized nests of A. urticae and A. io equally, the proportion of larvae parasitized is significantly higher for A. urticae. At the landscape scale, P. confusa is almost exclusively found in vegetated open land and near deciduous forests, whereas artificial habitats are negatively correlated with the likelihood of a nest to be parasitized. The genetic analyses on 89 adult P. confusa and 87 adult A. urticae using CO1 and AFLP markers reveal a low genetic diversity in P. confusa and a lack of genetic structure in both species, at the scale of our sampling. Further genetic studies using high-resolution genomics tools will be required to better understand the population genetic structure of P. confusa, its biotic interactions with its hosts, and ultimately the stability and the functioning of natural ecosystems.

Sequence analysis of a reovirus isolated from the winter moth Operophtera brumata (Lepidoptera: Geometridae) and its parasitoid wasp Phobocampe tempestiva (Hymenoptera: Ichneumonidae).

Project description:A reovirus was isolated from Operophtera brumata (ObRV) and its parasitoid wasp Phobocampe tempestiva. Each of the 10 dsRNA genome segments of ObRV was sequenced and shown to contain a single open reading frame (ORF). Conserved motifs ([+ve] 5'-AAATAAA ...(G)/(T)AGGTT-3') were found at the termini of each segment, with the exception of Seg-6 and Seg-8, where the 5' termini were 5'-AACAAA...-3'. The putative proteins encoded by each segment were compared with those of other members of the family Reoviridae. Phylogenetic comparisons to published sequences for the RNA-dependent RNA polymerase genes from other reoviruses indicated that ObRV is most closely related to members of the genus Cypovirus. However, unlike the cypoviruses, ObRV has a double-layered capsid structure. When the protein encoded by ObRV Seg-10 was expressed (by inserting the open reading frame into a baculovirus expression vector) no 'occlusion bodies' were observed in the recombinant baculovirus infected insect cell cultures. This suggests that unlike the cypoviruses, Seg-10 of ObRV does not contain a polyhedrin gene. Further phylogenetic comparisons also identified relationships between Seg-2 and Seg-10 of ObRV, and genes of Diadromus pulchellus Idnoreovirus 1 (DpIRV1), suggesting that ObRV represents a new species from the genus Idnoreovirus.

The mitochondrial genome of <i>Vulgichneumon</i> sp. (Hymenoptera: Ichneumonidae).

Project description:The Vulgichneumon sp. belongs to the subfamily Ichneumoninae of Ichneumonidae. The mitogenome (GenBank accession number: MN178162) of Vulgichneumon sp. was sequenced, the first representative of the mitogenome of the subfamily. The nearly complete mitogenome is 15,306 bp totally, consisting of 13 protein-coding genes, 2 rRNAs and 22 transfer RNAs. The nucleotide composition biases toward A and T, which together made up 83.6% of the entirety. Bayesian inference analysis supported the monophyly of Evanioidea, Chalcidoidea, Platygastroidea, Proctotrupoidea, Ceraphronoidea, and Ichneumonoidea. This result also suggested that the clade that contains other six superfamilies was the sister group to the clade that consist of Trigonaloidea, Megalyroidea and Ichneumonoidea.

Extensive sampling reveals the phenology and habitat use of Afrotropical parasitoid wasps (Hymenoptera: Ichneumonidae: Rhyssinae).

Project description:Tropical invertebrates, such as the ichneumonids of tropical forests, are poorly known. Here, we report the first results of extensive sampling at Kibale National Park, Uganda, by providing some of the first tropical ecological data for the ichneumonid subfamily Rhyssinae. We sampled ichneumonids with 34 Malaise traps for a year in 10 sites, in habitats ranging from primary forest to farmland. We also gathered weather and vegetation data. The total sampling effort was 373 trap months and we caught 444 rhyssines in six species. We caught the most rhyssines in dry weather, and towards the end of the sampling year. The rhyssines showed a clear preference for decaying logs and for primary forest. We fitted a model which can be used to predict future catches at the site, and draw conclusions on when rhyssines emerge and on their adult lifespan. Sampling extensively gave us a wealth of ecological data on a poorly known parasitoid wasp subfamily. We recommend that future tropical sampling collect ecological data, and that existing data from previous large-scale surveys be used for ecological analyses.

A new genus and species of Metopiinae (Hymenoptera, Ichneumonidae) from Mexico.

Project description:A new genus of Metopiinae, Ojuelos Khalaim, gen. n., with a single species, Ojuelos juachicus Khalaim & Ruíz-Cancino, sp. n., is described from Jalisco Province in central Mexico. Ojuelos belongs to the group of three genera (Cubus Townes & Townes, Colpotrochia Holmgren and Triclistus Förster) having a high lamella separating the antennal sockets and reaching the median ocellus (this lamella possesses a dorsal groove in it, just below the median ocellus), but differs from these genera primarily by 1) face and clypeus not convex in lateral view, 2) face separated from clypeus by a shallow transverse impression, 3) mandible with lower tooth very strongly reduced, 4) flagellomeres of antenna strongly transverse, and 5) dorsal carinae of propodeum reduced. A portion of the key to the genera of Metopiinae is provided to distinguish the new genus.
