Project description:Quiver flag zero loci are subvarieties of quiver flag varieties cut out by sections of representation theoretic vector bundles. We prove the Abelian/non-Abelian correspondence in this context: this allows us to compute genus zero Gromov-Witten invariants of quiver flag zero loci. We determine the ample cone of a quiver flag variety, and disprove a conjecture of Craw. In the appendices (which can be found in the electronic supplementary material), which are joint work with Tom Coates and Alexander Kasprzyk, we use these results to find four-dimensional Fano manifolds that occur as quiver flag zero loci in ambient spaces of dimension up to 8, and compute their quantum periods. In this way, we find at least 141 new four-dimensional Fano manifolds.

Project description:Plasmonic nanoclusters, an ordered assembly of coupled metallic nanoparticles, support unique spectral features known as Fano resonances due to the coupling between their subradiant and superradiant plasmon modes. Within the Fano resonance, absorption is significantly enhanced, giving rise to highly localized, intense near fields with the potential to enhance nonlinear optical processes. Here, we report a structure supporting the coherent oscillation of two distinct Fano resonances within an individual plasmonic nanocluster. We show how this coherence enhances the optical four-wave mixing process in comparison with other double-resonant plasmonic clusters that lack this property. A model that explains the observed four-wave mixing features is proposed, which is generally applicable to any third-order process in plasmonic nanostructures. With a larger effective susceptibility ?(3) relative to existing nonlinear optical materials, this coherent double-resonant nanocluster offers a strategy for designing high-performance third-order nonlinear optical media.

Project description:PurposeTo report clinical outcomes of a four-flanged intrascleral fixation technique using toric and multifocal intraocular lens.ObservationsWe describe two cases of premium intraocular lens (IOL) implantation after which the patients fully recovered their visual function following a four-point sutureless scleral fixation technique via a 2.8-mm corneal incision. In the first case, a monofocal toric hydrophobic lens consisting of two haptic plates with four holes for suturing was fixated with 5-0 polypropylene monofilament. In the second case, a bifocal hydrophobic lens with the same haptic design was fixated. No conjunctival or scleral sutures, glue, or flap formation was required during the surgery. There were no complications related to the surgical process.Conclusions and importanceA four-flanged intrascleral fixation technique may benefit patients with poor zonular support who have high expectations for postoperative visual quality.

Project description:Observation of Fano resonances in various physical phenomena is usually ascribed to the coupling of discrete states with background continuum, as it has already been reported for various physical phenomena. Here, we report on Fano lineshapes of nonthermal GHz phonons generated and observed with pumped Brillouin light scattering in gold-silicon thin membranes, overlapping the broad zero-shift background of yet questionable origin. The system's broken mid-plane symmetry enabled the generation of coherent quasi-symmetric and quasi-antisymmetric Lamb acoustic waves/phonons, leading to the four orders-of-magnitude enhancement of Brillouin light scattering. Notably, the membrane asymmetry resulted also in the mode-dependent Stokes and anti-Stokes Fano lineshapes asymmetry.

Project description:Four-dimensional (4D) printing relies on multimaterial printing, reinforcement patterns, or micro/nanofibrous additives as programmable tools to achieve desired shape reconfigurations. However, existing programming approaches still follow the so-called origami design principle to generate reconfigurable structures by self-folding stacked 2D materials, particularly at small scales. Here, we propose a programmable modular design that directly constructs 3D reconfigurable microstructures capable of sophisticated 3D-to-3D shape transformations by assembling 4D micro-building blocks. 4D direct laser writing is used to print two-photon polymerizable, stimuli-responsive hydrogels to construct building blocks at micrometer scales. Denavit-Hartenberg (DH) parameters, used to define robotic arm kinematics, are introduced as guidelines for how to assemble the micro-building blocks and plan the 3D motion of assembled chain blocks. Last, a 3D-printed microscaled transformer capable of changing its shape from a race car to a humanoid robot is devised and fabricated using the DH parameters to guide the motion of various assembled compartments.

Project description:Chromatin conformation plays an important role in a variety of genomic processes, including genome replication, gene expression, and gene methylation. Hi-C data is frequently used to analyze structural features of chromatin, such as AB compartments, topologically associated domains, and 3D structural models. Recently, the genomics community has displayed growing interest in chromatin dynamics. Here, we present 4DMax, a novel method, which uses time-series Hi-C data to predict dynamic chromosome conformation. Using both synthetic data and real time-series Hi-C data from processes, such as induced pluripotent stem cell reprogramming and cardiomyocyte differentiation, we construct smooth four-dimensional models of individual chromosomes. These predicted 4D models effectively interpolate chromatin position across time, permitting prediction of unknown Hi-C contact maps at intermittent time points. Furthermore, 4DMax correctly recovers higher order features of chromatin, such as AB compartments and topologically associated domains, even at time points where Hi-C data is not made available to the algorithm. Contact map predictions made using 4DMax outperform naïve numerical interpolation in 87.7% of predictions on the induced pluripotent stem cell dataset. A/B compartment profiles derived from 4DMax interpolation showed higher similarity to ground truth than at least one profile generated from a neighboring time point in 100% of induced pluripotent stem cell experiments. Use of 4DMax may alleviate the cost of expensive Hi-C experiments by interpolating intermediary time points while also providing valuable visualization of dynamic chromatin changes.

Project description:Four-dimensional imaging, which indicates imaging in three spatial dimensions as a function of time, provides useful evidence to investigate the interactions of rising bubbles. However, this has been largely unexplored for microbubbles, mostly due to problems associated with strong light scattering and shallow depth of field in optical imaging. Here, tracking x-ray microtomography is used to visualize rising microbubbles in four dimensions. Bubbles are tracked by moving the cell to account for their rise velocity. The sizes, shapes, time-dependent positions, and velocities of individual rising microbubbles are clearly identified, despite substantial overlaps between bubbles in the field of view. Our tracking x-ray microtomography affords opportunities for understanding bubble-bubble (or particle) interactions at microscales - important in various fields such as microfluidics, biomechanics, and floatation.

Project description:Four-dimensional computed tomography (4DCT), or dynamic CT, is an emerging modality with a wealth of orthopedic applications for both clinical practice and research. This technology creates CT volumes of a moving structure at multiple time points to depict real-time motion. Recent advances in acquisition technology and reduction in radiation dosage have allowed for increased adoption of the modality and have made imaging of joint motion feasible and safe. Musculoskeletal 4DCT has been used primarily to investigate wrist motion; however, the utility of 4DCT has been shown in other areas, including the shoulder, elbow, hip, knee and ankle. Imaging these joints through a full range of motion provides new insight into dynamic phenomena such as instability, impingement and joint kinematics. Although 4DCT has not yet been widely adopted in orthopedic practice and research, future use has the potential to enable a deeper understanding of musculoskeletal conditions and to improve patient care.

Project description:PurposeSpherical orthokeratology and soft multifocal contact lenses are commonly used for myopia control and correction, but have been shown to increase spherical aberration, coma and total higher order root mean square (HORMS) aberrations. There are limited myopia control contact lens options for patients with moderate to high astigmatism. The purpose of this study was to quantify changes in higher order aberrations (HOA) in myopic astigmatic eyes fitted with toric orthokeratology (TOK) and soft toric multifocal (STM) contact lenses.MethodsOcular wavefront aberrations were measured in both eyes of 30 adult subjects and are reported through the 6th radial order over a 5 mm, dilated pupil. All eyes met refractive criteria of myopia (-5.00 D to plano) and cylinder (-3.50 to -1.25 D). Three measurements were taken at baseline and after 10 ± 2 days of lens wear (TOK, STM). Sixteen subjects achieved logMAR high contrast visual acuity of 0.30 or better in both eyes and were included in this analysis. Repeated measures analysis of variance and post-hoc paired t-tests were used, as appropriate, with Benjamini-Hochberg correction.ResultsHigher order root mean square, spherical aberration (C12), and coma RMS (C7, C8) increased with TOK (0.641 [0.222], 0.409 [0.157], 0.426 [0.187] µm, respectively) and STM (0.481 [0.107], 0.223 [0.139], 0.320 [0.130] µm, respectively) from baseline (all p < 0.001). TOK was elevated compared to STM for HORMS (p = 0.03), spherical aberration (p = 0.001) and coma RMS (p = 0.04).ConclusionsToric orthokeratology induced more HORMS, spherical aberration and coma RMS than STM in myopic astigmats; however, both lens types showed an increase in HOA compared to baseline, which placed patients outside the age and pupil size matched normative ranges. While the optical changes that accompany these modalities are helpful for myopia management, the induction of HOAs may have unintended consequences on visual performance.

Project description:PurposeThere has been little research on myopia management options for patients with astigmatism. This study quantified changes in peripheral refraction induced by toric orthokeratology (TOK) and soft toric multifocal (STM) contact lenses.MethodsThirty adults with refractive error of plano to -5.00 D (sphere) and -1.25 to -3.50 D (cylinder) were enrolled. Cycloplegic autorefraction was measured centrally, ±20 degrees, and ±30 degrees from the line of sight nasally (N) and temporally (T) on the retina. Measurements were made at baseline, after 10 ± 2 days of TOK wear (without lenses on eye), and after 10 ± 2 days of STM wear (with lenses on the eyes) and compared with repeated-measures analysis of variance.ResultsCompared to baseline, TOK induced a myopic shift in defocus (M) at all locations (all P < 0.01), but STM only induced a myopic shift at 20 T in both eyes and 30 N/T in the left eye (all P < 0.01). TOK resulted in more myopic defocus than STM at all locations (all P < 0.05) except 20 T in the left eye. TOK induced more J0 astigmatism at all locations (all P < 0.02), except 20 N in the right eye; J0 with STM was different than baseline at 20 N in both eyes and 30 N in the right eye (all P < 0.02). TOK induced more J0 astigmatism than STM at all locations (all P < 0.01), except 20 T in the left eye. Differences in J45 astigmatism, when significant, were clinically small.ConclusionsGreater amounts of peripheral myopic defocus and J0 astigmatism were induced by TOK compared to STM, which may influence efficacy for myopia management.
