ABSTRACT: The crystal structure of the title compound, C6H10O2, an ?,?-unsaturated carb-oxy-lic acid, displays carb-oxy-lic acid inversion dimers linked by pairs of O-H?O hydrogen bonds. The packing is characterized by layers of acid dimers. All the non-H atoms of the (E)-hex-2-enoic acid mol-ecule lie almost in the same plane (r.m.s. deviation for the non-H atoms = 0.018?Å).

Project description:The crystal structure of (E)-dodec-2-enoic acid, C12H22O2, an α,β-unsaturated carb-oxy-lic acid with a melting point (295 K) near room temperature, is characterized by carb-oxy-lic acid inversion dimers linked by pairs of O-H⋯O hydrogen bonds. The carb-oxy-lic acid group and the following three carbon atoms of the chain of the (E)-dodec-2-enoic acid mol-ecule lie almost in one plane (r.m.s. deviation for the four C atoms and two O atoms = 0.012 Å), whereas the remaining carbon atoms of the hydro-carbon chain adopt a nearly fully staggered conformation [moduli of torsion angles vary from 174.01 (13) to 179.97 (13)°].

In the crystal, centrosymmetrically related mol-ecules are linked by pairs of O-H⋯O hydrogen bonds into dimers, forming layers parallel to the (041) plane.

Project description:The title compound, C21H33N3O3, is a tri-substituted cyclo-hex-yloxy triazine. In the crystal, the triazine rings form (C3i-PU) Piedfort units. The inter-centroid distance of the π-π inter-action involving the triazine rings is 3.3914 (10) Å. In the crystal, mol-ecules are linked by C-H⋯O hydrogen bonds, forming ribbons propagating along [1-10]. There are also weak C-H⋯N and C-H⋯O contacts present, linking inversion-related ribbons, forming a three-dimensional structure.

Crystal structure of 11-[4-(hex-yloxy)phen-yl]-1,2,4-triazolo[4,3-a][1,10]phenanthroline.

Project description:The title compound, C25H24N4O, was prepared from 2-chloro-phenanthroline and hexyl-oxyphenyl-tetra-zole. The main difference between the two independent mol-ecules (A and B) in the asymmetric unit is the orientation of the all-anti-configured hex-yloxy chain: in A the C-O-C-C torsion angle is 175.9 (2)° whereas it is -88.3 (3)° in B. The benzene substitution in the bay of the triazolophenanthroline results in a helical distorsion of the heterocyclic core, the dihedral angles between the mean planes formed by quinoline and benzotriazole ring systems are 13.73 (9) for mol-ecule A and 14.87 (8)° for B. The dihedral angles between the triazole ring and the attached benzene ring are 45.87 (15) in A and 53.93 (14)° in B. The angular annulation of four rings and the benzene substituent results in a helical distortion of the aromatic framework. The crystal is formed from layers composed of centrosymmetric pairs of A 2, B 2 mol-ecules with inter-digitating alkyl chains.

Project description:The title compound, C(23)H(20)O(2), is a precursor of a pyrene-based supra-molecular element for non-covalent attachment to a carbon nanotube. The asymmetric unit contains three independent mol-ecules. The carb-oxy-lic acid group in each of these mol-ecules serves as an inter-molecular hydrogen-bond donor and acceptor, generating the commonly observed double O-H⋯O hydrogen-bond motif in an eight-membered ring. Weaker C-H⋯O, π-π [centroid-centroid distance = 3.968 (4) Å] and C-H⋯π inter-actions are also found in the crystal structure.

Crystal structure of mammalian acid sphingomyelinase.

Project description:Acid sphingomyelinase (ASMase, ASM, SMPD1) converts sphingomyelin into ceramide, modulating membrane properties and signal transduction. Inactivating mutations in ASMase cause Niemann-Pick disease, and its inhibition is also beneficial in models of depression and cancer. To gain a better understanding of this critical therapeutic target, we determined crystal structures of mammalian ASMase in various conformations. The catalytic domain adopts a calcineurin-like fold with two zinc ions and a hydrophobic track leading to the active site. Strikingly, the membrane interacting saposin domain assumes either a closed globular conformation independent from the catalytic domain, or an open conformation, which establishes an interface with the catalytic domain essential for activity. Structural mapping of Niemann-Pick mutations reveals that most of them likely destabilize the protein's fold. This study sheds light on the molecular mechanism of ASMase function, and provides a platform for the rational development of ASMase inhibitors and therapeutic use of recombinant ASMase.

In the crystal, inter-molecular N-H⋯O bonds lead to the formation of polymer chains propagating along [011].
