Computational multifocus fluorescence microscopy for three-dimensional visualization of multicellular tumor spheroids.

Project description:SignificanceThree-dimensional (3D) visualization of multicellular tumor spheroids (MCTS) in fluorescence microscopy can rapidly provide qualitative morphological information about the architecture of these cellular aggregates, which can recapitulate key aspects of their in vivo counterpart.AimThe present work is aimed at overcoming the shallow depth-of-field (DoF) limitation in fluorescence microscopy while achieving 3D visualization of thick biological samples under study.ApproachA custom-built fluorescence microscope with an electrically focus-tunable lens was developed to optically sweep in-depth the structure of MCTS. Acquired multifocus stacks were combined by means of postprocessing algorithms performed in the Fourier domain.ResultsImages with relevant characteristics as extended DoF, stereoscopic pairs as well as reconstructed viewpoints of MCTS were obtained without segmentation of the focused regions or estimation of the depth map. The reconstructed images allowed us to observe the 3D morphology of cell aggregates.ConclusionsComputational multifocus fluorescence microscopy can provide 3D visualization in MCTS. This tool is a promising development in assessing the morphological structure of different cellular aggregates while preserving a robust yet simple optical setup.

Three-dimensional metal-intercalated covalent organic frameworks for near-ambient energy storage.

Project description:A new form of nanoporous material, metal intercalated covalent organic framework (MCOF) is proposed and its energy storage property revealed. Employing density functional and thermodynamical analysis, we find that stable, chemically active, porous materials could form by stacking covalent organic framework (COF) layers with metals as a gluing agent. Metal acts as active sites, while its aggregation is suppressed by a binding energy significantly larger than the corresponding cohesive energy of bulk metals. Two important parameters, metal binding and metal-metal separation, are tuned by selecting suitable building blocks and linkers when constructing COF layers. Systematic searches among a variety of elements and organic molecules identify Ca-intercalated COF with diphenylethyne units as optimal material for H2 storage, reaching a striking gravimetric density ~ 5 wt% at near-ambient conditions (300 K, 20 bar), in comparison to < 0.1 wt% for bare COF-1 under the same condition.

Probing Molecular Motions in Metal-Organic Frameworks by Three-Dimensional Electron Diffraction.

Project description:Flexible metal-organic frameworks (MOFs) are known for their vast functional diversities and variable pore architectures. Dynamic motions or perturbations are among the highly desired flexibilities, which are key to guest diffusion processes. Therefore, probing such motions, especially at an atomic level, is crucial for revealing the unique properties and identifying the applications of MOFs. Nuclear magnetic resonance (NMR) and single-crystal X-ray diffraction (SCXRD) are the most important techniques to characterize molecular motions but require pure samples or large single crystals (>5 × 5 × 5 μm3), which are often inaccessible for MOF synthesis. Recent developments of three-dimensional electron diffraction (3D ED) have pushed the limits of single-crystal structural analysis. Accurate atomic information can be obtained by 3D ED from nanometer- and submicrometer-sized crystals and samples containing multiple phases. Here, we report the study of molecular motions by using the 3D ED method in MIL-140C and UiO-67, which are obtained as nanosized crystals coexisting in a mixture. In addition to an ab initio determination of their framework structures, we discovered that motions of the linker molecules could be revealed by observing the thermal ellipsoid models and analyzing the atomic anisotropic displacement parameters (ADPs) at room temperature (298 K) and cryogenic temperature (98 K). Interestingly, despite the same type of linker molecule occupying two symmetry-independent positions in MIL-140C, we observed significantly larger motions for the isolated linkers in comparison to those reinforced by π-π stacking. With an accuracy comparable to that of SCXRD, we show for the first time that 3D ED can be a powerful tool to investigate dynamics at an atomic level, which is particularly beneficial for nanocrystalline materials and/or phase mixtures.

Spontaneous resolution of racemic camphorates in the formation of three-dimensional metal-organic frameworks.

Project description:Reported here is a rare example of enantioselective processes between organic racemic ligands (DL-camphorates) with in situ formed chiral metal complexes. Such enantioselectivity leads to a double spontaneous resolution of two pairs of distinctly different racemates [the LambdaDelta-Zn(Htea) units and the DL-camphorate ligands] into two chiral three-dimensional frameworks containing only one enantiommeric form from each racemate. This work demonstrates unique enantioselectivity in the self-assembly of metal-organic frameworks containing multiple chiral features, which points to a new avenue for the preparation of chiral framework materials and for the resolution of racemates.

Three-dimensional visualization of lithium metal anode via low-dose cryogenic electron microscopy tomography.

Project description:The structure of lithium (Li) metal anode, including the Li metal and the solid electrolyte interphase (SEI), is critical to the investigation of cycle stability or decay mechanisms. The three-dimensional (3D) visualization of Li metal and SEI, however, has not been demonstrated yet, owing to the lack of 3D characterization techniques and the susceptibility of Li metal anode toward oxygen, moisture, as well as electron beam. Herein, we introduce a successful 3D presentation of deposited Li metal and SEI established via low-dose cryogenic electron microscopy tomography. The Li metal anode is imaged in low-dose mode at different tilt angles and then aligned and reconstructed into a 3D image through an expectation-maximization algorithm. The spherical Li deposits and SEI are confirmed in the 3D tomography of Li metal anode. It is also discovered that the Li metal corrodes and SEI turns concave owing to possible self-discharge after long-time rest.

Dissolving uptake-hindering surface defects in metal-organic frameworks.

Project description:Metal-organic frameworks (MOFs) have unique properties which make them perfectly suited for various adsorption and separation applications; however, their uses and efficiencies are often hindered by their limited stability. When most MOFs are exposed to water or humid air, the MOF structure, in particular at the surface, is destroyed, creating surface defects. These surface defects are surface barriers which tremendously hinder the uptake and release of guest molecules and, thus, massively decrease the performance in any application of MOFs. Here, the destruction by exposure to water vapor is investigated by using well-defined MOF films of type HKUST-1 as a model system for uptake experiments with different-sized probe molecules as well as for spectroscopic investigations, complemented by density functional theory calculations of the defect structure. In addition to the characterization of the surface defects, it is found that the pristine MOF structure can be regenerated. We show that the surface defects can be dissolved by exposure to the synthesis solvent, here ethanol, enabling fast uptake and release of guest molecules. These findings show that the storage of MOF materials in a synthesis solvent results in healing of surface defects and enables ideal performance of MOF materials.

Visualization and Quantification of Geometric Diversity in Metal-Organic Frameworks.

Project description:With ever-growing numbers of metal-organic framework (MOF) materials being reported, new computational approaches are required for a quantitative understanding of structure-property correlations in MOFs. Here, we show how structural coarse-graining and embedding ("unsupervised learning") schemes can together give new insights into the geometric diversity of MOF structures. Based on a curated data set of 1262 reported experimental structures, we automatically generate coarse-grained and rescaled representations which we couple to a kernel-based similarity metric and to widely used embedding schemes. This approach allows us to visualize the breadth of geometric diversity within individual topologies and to quantify the distributions of local and global similarities across the structural space of MOFs. The methodology is implemented in an openly available Python package and is expected to be useful in future high-throughput studies.

Two-Dimensional Band Structure in Honeycomb Metal-Organic Frameworks.

Project description:Two-dimensional (2D) metal-organic frameworks (MOFs) have been recently proposed as a flexible material platform for realizing exotic quantum phases including topological and anomalous quantum Hall insulators. Experimentally, direct synthesis of 2D MOFs has been essentially confined to metal substrates, where the strong interaction with the substrate masks the intrinsic electronic properties of the MOF. In addition to electronic decoupling from the underlying metal support, synthesis on weakly interacting substrates (e.g., graphene) would enable direct realization of heterostructures of 2D MOFs with inorganic 2D materials. Here, we demonstrate synthesis of 2D honeycomb MOFs on epitaxial graphene substrate. Using low-temperature scanning tunneling microscopy (STM) and atomic force microscopy (AFM) complemented by density-functional theory (DFT) calculations, we show the formation of a 2D band structure in the MOF decoupled from the substrate. These results open the experimental path toward MOF-based designer electronic materials with complex, engineered electronic structures.

Two-dimensional, conductive niobium and molybdenum metal-organic frameworks.

Project description:The incorporation of second-row transition metals into metal-organic frameworks could greatly improve the performance of these materials across a wide variety of applications due to the enhanced covalency, redox activity, and spin-orbit coupling of late-row metals relative to their first-row analogues. Thus far, however, the synthesis of such materials has been limited to a small number of metals and structural motifs. Here, we report the syntheses of the two-dimensional metal-organic framework materials (H2NMe2)2Nb2(Cl2dhbq)3 and Mo2(Cl2dhbq)3 (H2Cl2dhbq = 3,6-dichloro-2,5-dihydroxybenzoquinone), which feature mononuclear niobium or molybdenum metal nodes and are formed through reactions driven by metal-to-ligand electron transfer. Characterization of these materials via X-ray absorption spectroscopy suggests a local trigonal prismatic coordination geometry for both niobium and molybdenum, consistent with their increased covalency relative to related first-row transition metal compounds. A combination of vibrational spectroscopy, magnetic susceptibility, and electronic conductivity measurements reveal that these two frameworks possess distinct electronic structures. In particular, while the niobium compound displays evidence for redox-trapping and strong magnetic interactions, the molybdenum phase is valence-delocalized with evidence of large polaron formation. Weak interlayer interactions in the neutral molybdenum phase enable solvent-assisted exfoliation to yield few-layer hexagonal nanosheets. Together, these results represent the first syntheses of metal-organic frameworks containing mononuclear niobium and molybdenum nodes, establishing a route to frameworks incorporating a more diverse range of second- and third-row transition metals with increased covalency and the potential for improved charge transport and stronger magnetic coupling.

Three-Dimensional Hierarchical Porous Nanotubes Derived from Metal-Organic Frameworks for Highly Efficient Overall Water Splitting.

Project description:Effective design of bifunctional catalysts for both hydrogen evolution reaction (HER) and oxygen evolution reaction (OER) is important but remains challenging. Herein, we report a three-dimensional (3D) hierarchical structure composed of homogeneously distributed Ni-Fe-P nanoparticles embedded in N-doped carbons on nickel foams (denoted as Ni-Fe-P@NC/NF) as an excellent bifunctional catalyst. This catalyst was fabricated by an anion exchange method and a low-temperature phosphidation of nanotubular Prussian blue analogue (PBA). The Ni-Fe-P@NC/NF displayed exceptional catalytic activity toward both HER and OER and delivered an ultralow cell voltage of 1.47 V to obtain 10 mA cm-2 with extremely excellent durability for 100 h when assembled as a practical electrolyser. The extraordinary performance of Ni-Fe-P@NC/NF is attributed to the abundance of unsaturated active sites, the well-defined hierarchical porous structure, and the synergistic effect between multiple components. Our work will inspire more rational designs of highly active non-noble electrocatalysts for industrial energy applications.
