A gold nanohole array based surface-enhanced Raman scattering biosensor for detection of silver(I) and mercury(II) in human saliva.

ABSTRACT: A surface-enhanced Raman scattering (SERS) biosensor has been developed by incorporating a gold nanohole array with a SERS probe (a gold nanostar@Raman-reporter@silica sandwich structure) into a single detection platform via DNA hybridization, which circumvents the nanoparticle aggregation and the inefficient Raman scattering issues. Strong plasmonic coupling between the Au nanostar and the Au nanohole array results in a large enhancement of the electromagnetic field, leading to amplification of the SERS signal. The SERS sensor has been used to detect Ag(I) and Hg(II) ions in human saliva because both the metal ions could be released from dental amalgam fillings. The developed SERS sensor can be adapted as a general detection platform for non-invasive measurements of a wide range of analytes such as metal ions, small molecules, DNA and proteins in body fluids.

Tailoring plasmonic properties of gold nanohole arrays for surface-enhanced Raman scattering.

Project description:The wide plasmonic tuning range of nanotriangle and nanohole array patterns fabricated by nanosphere lithography makes them promising in surface-enhanced Raman scattering (SERS) sensors. Unfortunately, it is challenging to optimize these patterns for SERS sensing because their optical response is a complex mixture of localized surface plasmon resonance (SPR) and propagating surface plasmon polariton (SPP). In this paper, transmission and reflection measurements are combined with finite difference time domain simulations to identify and separate each plasmonic mode, discerning which resonance leads to the electromagnetic field enhancement. The SERS enhancement is found to be dominated by the absorption, which is shifted from the transmission and reflection dips usually used as tuning points, and by the 'gap' defects formed within the pattern. These effects have different spectral and geometric dependences, forming two optimization curves which can be used to predict the best performance for a given excitation wavelength. The developed model is verified with experimental SERS measurements for several nanohole sizes and periodicities, and then used to give optimal fabrication parameters for a range of measurement conditions. The results will promote the application of two-dimensional plasmonic nanoarrays in SERS sensors.

Development of Hybrid Silver-Coated Gold Nanostars for Nonaggregated Surface-Enhanced Raman Scattering.

Project description:In the ongoing search for ever-brighter surface-enhanced Raman scattering (SERS) nanoprobes, gold nanostars (AuNSs) have emerged as one of the best geometries for producing SERS in a nonaggregated state. Despite their high enhancement factor, optical extinction from plasmon-matched nanoparticles can greatly attenuate the overall SERS intensity. Herein, we report the development of a new hybrid bimetallic NS-based platform that exhibits superior resonant SERS (SERRS) properties. In this new nanoplatform, coating AuNSs with a subtotal layer of silver (AuNS@Ag) can further increase their SERRS brightness by an order of magnitude when being interrogated by an off-resonant excitation source. Silica-encapsulated AuNS@Ag nanoprobes were injected intradermally into a rat pelt, where SERRS was readily detected with higher signal-to-noise than nanoprobes prepared from AuNS. Moreover, these off-resonance AuNS@Ag nanoprobes did not cause any gross photothermal damage to tissue, which was observed with the plasmon-matched AuNSs. This novel SERRS-active hybrid nanoprobe exhibits high SERRS brightness and offers promising properties for future applications in sensing and molecular imaging.

Detection of nitrite with a surface-enhanced Raman scattering sensor based on silver nanopyramid array.

Project description:Nutrient pollution is of worldwide environmental and health concerns due to extensive use of nitrogen fertilizers and release of livestock waste, which induces nitrite compounds in aquatic systems. Herein a surface-enhanced Raman scattering (SERS) sensor is developed for nitrite detection based on coupling between the plasmonic gold nanostars and the silver nanopyramid array. When nitrite is present in the assay, an azo group is formed between the 1-naphthylamine-functionalized silver nanopyramids and the 4-aminothiophenol-functionalized gold nanostars. This not only generates the SERS spectral fingerprint for selective detection, but also creates "hot spots" at the gap between the Au nanostars and the Ag nanopyramids where the azo group is located, amplifying SERS signals remarkably. Finite-difference time-domain (FDTD) simulation shows a SERS enhancement factor of 4?×?1010?at the "hot spots". As a result, the SERS sensor achieves a limit of detection of 0.6?pg/mL toward nitrite in water, and enables nitrite detection in real-world river water samples. In addition, this sensor eliminates the use of any Raman reporter and any expensive molecular recognition probe such as antibody and aptamer. This highly sensitive, selective and inexpensive SERS sensor has unique advantages over colorimetric, electrochemical and fluorescent devices for small molecule detection.

Nanorough gold for enhanced Raman scattering.

Project description:Conventional Raman scattering is a workhorse technique for detecting and identifying complex molecular samples. In surface enhanced Raman scattering, a nanorough metallic surface close to the sample enhances the Raman signal enormously. In this work, the surface is on a clear epoxy substrate. The epoxy is cast on a silicon wafer, using 20 nm of gold as a mold release. This single step process already produces useful enhanced Raman signals. However, the Raman signal is further enhanced by (1) depositing additional gold on the epoxy substrate and (2) by using a combination of wet and dry etches to roughen the silicon substrate before casting the epoxy. The advantage of a clear substrate is that the Raman signal may be obtained by passing light through the substrate, with opaque samples simply placed against the surface. Results were obtained with solutions of Rhodamine 6G in deionized water over a range of concentrations from 1 nM to 1 mM. In all cases, the signal to noise ratio was greater than 10:1.

Gold Nanocone Array with Extensive Electromagnetic Fields for Highly Reproducible Surface-Enhanced Raman Scattering Measurements.

Project description:Surface-enhanced Raman scattering (SERS) is a technique used to distinguish the constitution of disease-related biomarkers in liquid biopsies, such as exosomes and circulating tumor cells, without any recognition elements. Previous studies using metal nanoparticle aggregates and angular nanostructures have achieved the detection of various biomarkers owing to strong hot spots and electromagnetic (EM) fields by localized surface plasmon resonance (LSPR). Although these SERS platforms enable significant enhancement of Raman signals, they still have some problems with the fabrication reproducibility of platforms in obtaining reproducible SERS signals. Therefore, highly reproducible fabrication of SERS platforms is required. Here, we propose the application of a polymer-based gold (Au) nanocone array (Au NCA), which extensively generates an enhanced EM field near the Au NCA surface by LSPR. This approach was experimentally demonstrated using a 785 nm laser, typically used for SERS measurements, and showed excellent substrate-to-substrate reproducibility (relative standard deviation (RSD) < 6%) using an extremely simple fabrication procedure and very low laser energy. These results proved that a Au NCA can be used as a highly reproducible SERS measurement to distinguish the constitution of biomarkers.

Graphene-Gold Nanoparticles Hybrid-Synthesis, Functionalization, and Application in a Electrochemical and Surface-Enhanced Raman Scattering Biosensor.

Project description:Graphene is a single-atom-thick two-dimensional carbon nanosheet with outstanding chemical, electrical, material, optical, and physical properties due to its large surface area, high electron mobility, thermal conductivity, and stability. These extraordinary features of graphene make it a key component for different applications in the biosensing and imaging arena. However, the use of graphene alone is correlated with certain limitations, such as irreversible self-agglomerations, less colloidal stability, poor reliability/repeatability, and non-specificity. The addition of gold nanostructures (AuNS) with graphene produces the graphene-AuNS hybrid nanocomposite which minimizes the limitations as well as providing additional synergistic properties, that is, higher effective surface area, catalytic activity, electrical conductivity, water solubility, and biocompatibility. This review focuses on the fundamental features of graphene, the multidimensional synthesis, and multipurpose applications of graphene-Au nanocomposites. The paper highlights the graphene-gold nanoparticle (AuNP) as the platform substrate for the fabrication of electrochemical and surface-enhanced Raman scattering (SERS)-based biosensors in diverse applications as well as SERS-directed bio-imaging, which is considered as an emerging sector for monitoring stem cell differentiation, and detection and treatment of cancer.

Carbon-Assistant Nanoporous Gold for Surface-Enhanced Raman Scattering.

Project description:Surface-enhanced Raman scattering (SERS) technology can amplify the Raman signal due to excited localized surface plasmon (LSP) from SERS substrates, and the properties of the substrate play a decisive role for SERS sensing. Several methods have been developed to improve the performance of the substrate by surface modification. Here, we reported a surface modification method to construct carbon-coated nanoporous gold (C@NPG) SERS substrate. With surface carbon-assistant, the SERS ability of nanoporous gold (NPG) seriously improved, and the detection limit of the dye molecule (crystal violet) can reach 10-13 M. Additionally, the existence of carbon can avoid the deformation of the adsorbed molecule caused by direct contact with the NPG. The method that was used to improve the SERS ability of the NPG can be expanded to other metal structures, which is a convenient way to approach a high-performance SERS substrate.

3D hierarchically porous magnetic molybdenum trioxide@gold nanospheres as a nanogap-enhanced Raman scattering biosensor for SARS-CoV-2.

Project description:The global pandemic of COVID-19 is an example of how quickly a disease-causing virus can take root and threaten our civilization. Nowadays, ultrasensitive and rapid detection of contagious pathogens is in high demand. Here, we present a novel hierarchically porous 3-dimensional magnetic molybdenum trioxide-polydopamine-gold functionalized nanosphere (3D mag-MoO3-PDA@Au NS) composed of plasmonic, semiconductor, and magnetic nanoparticles as a multifunctional nanosculptured hybrid. Based on the synthesized 3D mag-MoO3-PDA@Au NS, a universal "plug and play" biosensor for pathogens is proposed. Specifically, a magnetically-induced nanogap-enhanced Raman scattering (MINERS) detection platform was developed using the 3D nanostructure. Through a magnetic actuation process, the MINERS system overcomes Raman signal stability and reproducibility challenges for the ultrasensitive detection of SARS-CoV-2 spike protein over a wide dynamic range up to a detection limit of 10-15 g mL-1. The proposed MINERS platform will facilitate the broader use of Raman spectroscopy as a powerful analytical detection tool in diverse fields.

Arrayed nanopore silver thin films for surface-enhanced Raman scattering.

Project description:Active substrates are crucial for surface-enhanced Raman scattering (SERS). Among these substrates, large uniform area arrayed nanoporous silver thin films have been developed as active substrates. Arrayed nanoporous silver thin films with unique anisotropic morphologies and nanoporous structures can be fabricated onto the nanoporous anodic aluminum oxide (AAO) of controlled pore size and interspacing by precisely tuning the sputtering parameters. These thin films preserve locally enhanced electromagnetic fields by exciting the surface plasmon resonance, which is beneficial for SERS. In this study, nanoporous silver thin films were transferred into polymethylmethacrylate (PMMA) and polydimethylsiloxane (PDMS) substrates using our recently invented template-assisted sol-gel phase inverse-imprinting process to form two different nanopore thin films. The as-formed Ag nanoporous thin films on PMMA and PDMS exhibited intensively enhanced SERS signals using Rhodamine 6G (R6G) as the model molecule. The two nanopore thin films exhibited opposite pore size-dependent SERS tendencies, which were elucidated by the different enhancement tendencies of the electric field around pores of different diameters. In particular, the Ag nanoporous thin film on PMMA exhibited an R6G detection limit of as low as 10-6 mol L-1, and the SERS enhancement factor (EF) was more than 106. The low detection limit and large EF demonstrated the high sensitivity of the as-prepared SERS substrates for label-free detection of biomolecules. Compared with conventional smooth films, this nanopore structure can facilitate future application in biomolecular sensors, which allows the detection of single molecules via an electronic readout without requirement for amplification or labels.

Gap-enhanced gold nanodumbbells with single-particle surface-enhanced Raman scattering sensitivity.

Project description:Gap-enhanced Raman tags (GERTs) have been widely used for surface-enhanced Raman scattering (SERS) imaging due to their excellent SERS performances. Here, we reported a synthetic strategy for novel gap-enhanced dumbbell-like nanoparticles with anisotropic shell coatings. Controlled shell growth at the tips of gold nanorods was achieved by using cetyltrimethylammonium bromide (CTAB) as a capping agent. A mechanism related to the shape-directing effects of CTAB was proposed to explain the findings. Optimized gap-enhanced gold dumbbells exhibited highly enhanced SERS responses compared to rod cores, with an enhancement ratio of 101.5. We further demonstrated that gap-enhanced AuNDs exhibited single-particle SERS sensitivity with an acquisition time as fast as 0.1 s per spectrum, showing great potential for high-speed SERS imaging.
