Project description:Porous magnesium-aluminium layered double hydroxides (LDH) were prepared through intercalation and decomposition of hydrogen peroxide (H2O2). This process generates oxygen gas nano-bubbles that pierce holes in the layered structure of the material by local pressure build-up. The decomposition of the peroxide can be triggered by microwave radiation or chemically by reaction with iodide (I-) ions. The carbonate LDH version [Mg0.80Al0.20(OH)2](CO3)0.1∙mH2O was synthesized by microwave-assisted urea coprecipitation and further modified by iodide or H2O2 intercalation. High resolution Scanning Electron Microscopy (HR-SEM) and Brunauer-Emmet-Teller (BET) analysis were used to assess the morphology and surface area of the new porous materials. The presence of H2O2 in the interlayer region and later decomposition triggered by microwave radiation generated more pores on the surface of the LDH platelets, increasing their specific surface area from initially 9 m2/g to a maximum of 67 m2/g. X-Ray Diffraction showed that the formation of the pores did not affect the remaining crystal structure, allowing possible further functionalization of the material.

Project description:This article describes free radical scavenging activity of TEMPO (2, 2, 6, 6-tetramethylpiperidin-1-yloxyl) radical functionalized dendrimers of generation G0 - G4 against nitric oxide (NO), 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and hydrogen peroxide (H2O2) radicals. The total antioxidant activity of the dendrimers was also reported and compared with pure 4-hydoxy TEMPO (constituent of dendrimers) and ascorbic acid (vitamin C) standard. The activity of TEMPO functionalized G4 dendrimer against scavenging of nitric oxide and hydrogen peroxide radicals was found comparable to vitamin C. The data presented in this article supports the good antioxidant activity of the dendrimers newly reported in our previous publication (Radical dendrimers: Synthesis, anti-tumor activity and enhanced cytoprotective performance of TEMPO free radical functionalized polyurethane dendrimers).

Project description:Marinocine is a broad-spectrum antibacterial protein synthesized by the melanogenic marine bacterium Marinomonas mediterranea. This work describes the basis for the antibacterial activity of marinocine and the identification of the gene coding for this protein. The antibacterial activity is inhibited under anaerobic conditions and by the presence of catalase under aerobic conditions. Marinocine is active only in culture media containing l-lysine. In the presence of this amino acid, marinocine generates hydrogen peroxide, which causes cell death as confirmed by the increased sensitivity to marinocine of Escherichia coli strains mutated in catalase activity. The gene coding for this novel enzyme was cloned using degenerate PCR with primers designed based on conserved regions in the antimicrobial protein AlpP, synthesized by Pseudoalteromonas tunicata, and some hypothetical proteins. The gene coding for marinocine has been named lodA, standing for lysine oxidase, and it seems to form part of an operon with a second gene, lodB, that codes for a putative dehydrogenase flavoprotein. The identity of marinocine as LodA has been demonstrated by N-terminal sequencing of purified marinocine and generation of lodA mutants that lose their antimicrobial activity. This is the first report on a bacterial lysine oxidase activity and the first time that a gene encoding this activity has been cloned.

Project description:Hydrogen peroxide (H(2)O(2)) promotes seed germination of cereal plants and ascorbic acid which acts as antioxidant suppresses the germination of wheat seeds, but the role of H(2)O(2) scavenging on germination during seed maturation has not been demonstrated. We investigated relationship of germination, ascorbate, H(2)O(2) scavenging enzymes and sensitivity to ascorbic acid (AsA) maturing seeds of two typical wheat (Triticum aestivum L.) cultivars, cvs. Shirogane-Komugi and Norin61. Shirogane-Komugi had marked high germination ability than Norin61 during seed maturation. Although the H(2)O(2) content had no difference in the two culti-vars, sensitivity to AsA of Norin61 seeds was higher than that of Shirogane-Komugi seeds during seed maturation. The sensitivity to AsA closely correlated with germination characteristic in the two cultivars. Especially, at 28 days after pollination (DAP), sensitivity to AsA in Norin61 seeds was remarkably high. At that stage, no significant differences were observed in endogenous AsA level, ascorbate peroxidase (APX, EC 1.11.1.11) and dehydroascorbate reductase (DHAR, EC 1.8.5.1) activities in the two cultivars. However, catalase (CAT, EC 1.11.1.6) activity and CAT mRNA in Norin61 were remarkably higher than in Shirogane-Komugi. Sensitivity to AsA at 35 and 42 DAPs kept high levels in Norin61, and endogenous AsA and CAT activity in the seeds were significantly higher than in Shirogane-Komugi. These results revealed a direct correlation between germination and antioxidant sensitivity during the developmental stages of wheat seeds.

Project description:Controlled generation and removal of hydrogen peroxide play important roles in cellular redox homeostasis and signaling. We used a hydrogen peroxide biosensor HyPer, targeted to different compartments, to examine these processes in mammalian cells.Reversible responses were observed to various redox perturbations and signaling events. HyPer expressed in HEK 293 cells was found to sense low micromolar levels of hydrogen peroxide. When targeted to various cellular compartments, HyPer occurred in the reduced state in the nucleus, cytosol, peroxisomes, mitochondrial intermembrane space and mitochondrial matrix, but low levels of the oxidized form of the biosensor were also observed in each of these compartments, consistent with a low peroxide tone in mammalian cells. In contrast, HyPer was mostly oxidized in the endoplasmic reticulum. Using this system, we characterized control of hydrogen peroxide in various cell systems, such as cells deficient in thioredoxin reductase, sulfhydryl oxidases or subjected to selenium deficiency. Generation of hydrogen peroxide could also be monitored in various compartments following signaling events.We found that HyPer can be used as a valuable tool to monitor hydrogen peroxide generated in different cellular compartments. The data also show that hydrogen peroxide generated in one compartment could translocate to other compartments. Our data provide information on compartmentalization, dynamics and homeostatic control of hydrogen peroxide in mammalian cells.

Project description:Inflammation and oxidative stress are two major factors that are involved in the pathogenesis of atherosclerosis. A smart drug delivery system that responds to the oxidative microenvironment of atherosclerotic plaques was constructed in the present study. Simvastatin (SIM)-loaded biodegradable polymeric micelles were constructed from hyaluronic acid (HA)-coated poly(ethylene glycol)-poly(tyrosine-ethyl oxalyl) (PEG-Ptyr-EO) for the purpose of simultaneously inhibiting macrophages and decreasing the level of reactive oxygen species (ROS) to treat atherosclerosis. HA coating endows the micelle system the ability of targeting CD44-positive inflammatory macrophages. Owing to the ROS-responsive nature of PEG-Ptyr-EO, the micelles can not only be degraded by enzymes, but also consumes ROS by itself at the pathologic sites, upon which the accumulation of pro-inflammatory macrophages is effectively suppressed and oxidative stress is alleviated. Consequently, the cellular uptake experiment demonstrated that SIM-loaded HA-coated micelles can be effectively internalized by LPS-induced RAW264.7 cells and showed high cytotoxicity against the cells, but low cytotoxicity against LO2 cells. In mouse models of atherosclerosis, intravenously SIM-loaded HA-coated micelles can effectively reduce plaque content of cholesterol, resulting in remarkable therapeutic effects. In conclusion, the SIM-loaded micelle system provides a promising and innovative option against atherosclerosis.

Project description:The phytoplankton community in the oligotrophic open ocean is numerically dominated by the cyanobacterium Prochlorococcus, accounting for approximately half of all photosynthesis. In the illuminated euphotic zone where Prochlorococcus grows, reactive oxygen species are continuously generated via photochemical reactions with dissolved organic matter. However, Prochlorococcus genomes lack catalase and additional protective mechanisms common in other aerobes, and this genus is highly susceptible to oxidative damage from hydrogen peroxide (HOOH). In this study we showed that the extant microbial community plays a vital, previously unrecognized role in cross-protecting Prochlorococcus from oxidative damage in the surface mixed layer of the oligotrophic ocean. Microbes are the primary HOOH sink in marine systems, and in the absence of the microbial community, surface waters in the Atlantic and Pacific Ocean accumulated HOOH to concentrations that were lethal for Prochlorococcus cultures. In laboratory experiments with the marine heterotroph Alteromonas sp., serving as a proxy for the natural community of HOOH-degrading microbes, bacterial depletion of HOOH from the extracellular milieu prevented oxidative damage to the cell envelope and photosystems of co-cultured Prochlorococcus, and facilitated the growth of Prochlorococcus at ecologically-relevant cell concentrations. Curiously, the more recently evolved lineages of Prochlorococcus that exploit the surface mixed layer niche were also the most sensitive to HOOH. The genomic streamlining of these evolved lineages during adaptation to the high-light exposed upper euphotic zone thus appears to be coincident with an acquired dependency on the extant HOOH-consuming community. These results underscore the importance of (indirect) biotic interactions in establishing niche boundaries, and highlight the impacts that community-level responses to stress may have in the ecological and evolutionary outcomes for co-existing species.

Project description:Adaptation to hydrogen peroxide in Saccharomyces cerevisiae is profiled with expression arrays. Adaptation describes the process in which a mild dose of toxin (in this case, hydrogen peroxide) is able to protect against a later acute dose. Here, we study two adaptive protocols (0.1 mM H2O2 and 0.1 + 0.4 mM H2O2) and one acute protocol (0.4 mM H2O2) to identify processes uniquely involved in adaptation. Predictions from these studies are validated in expression profiling of deletion mutants of the transcription factors Yap1, Mga2, and Rox1.

Project description:MYB transcription factors are widely present in plants and play significant roles in abiotic stresses. However, most MYB genes have not been identified in plants and their functions in abiotic stresses are still unknown. In this study, one MYB gene, designated as PtrMYB3, was cloned from trifoliate orange (Poncirus trifoliata (L.) Raf.), and its function in salt tolerance was investigated. PtrMYB3 contains a conserved R2R3-MYB domain, which is the typical property of R2R3-MYB subfamily proteins. Expression profiling under abiotic stresses indicated that PtrMYB3 could be induced by salt, dehydration and cold stresses. PtrMYB3 was found to be localized to the nucleus and possessed transactivation activity. Overexpression of PtrMYB3 by genetic transformation in tobacco impaired its salt tolerance, whereas silencing of PtrMYB3 by VIGS (virus-induced gene silencing) in trifoliate orange conferred significantly enhanced salt tolerance, indicating that PtrMYB3 negatively regulates salt tolerance. Furthermore, a peroxidase gene (PtrPOD) was found to be greatly upregulated in PtrMYB3-silenced trifoliate orange, and a dual LUC (luciferase) assay confirmed that PtrMYB3 could suppress the expression of PtrPOD. The hydrogen peroxide (H2O2) accumulation in PtrMYB3 transgenic tobacco plants after salt stress was higher than the wild type (WT), further confirming that overexpression of PtrMYB3 inhibited PtrPOD-mediated H2O2 scavenging. Taken together, these results demonstrate that PtrMYB3 negatively regulates salt tolerance, at least in part, due to the excess accumulation of H2O2.

Project description:For the first time, the fate of radicals generated in heterogeneous chemical oxidation treatment systems has been accounted for and used to assess treatment performance in three reaction compartments; reaction with the target compound, rhodamine B (RhB), the aqueous phase scavengers, and the solid phase scavengers. Radicals formed during the ultra-violet (UV) activation of hydrogen peroxide (H2O2) (UV-AHP) and persulfate (S2O82-) (UV-APS) include hydroxyl (•OH) and sulfate radicals (SO4•-), respectively. •OH and SO4•-, used in oxidation treatment systems to degrade a broad spectrum of environmental contaminants, may also react with non-target chemical species (scavengers) that limit treatment efficiency. UV-AHP and UV-APS treatment systems were amended with solid phase alumina to assess scavenging by solid surfaces. The overall rate of reaction and rate of radical scavenging was greater for •OH than SO4•-. Scavenging by dissolved constituents was dominated by the oxidant used (H2O2, S2O82-); and the rate of radical scavenging by alumina was greater than the rate of RhB oxidation in all cases. Treatment efficiency was lower in the UV-AHP than in the UV-APS treatment system and was attributed to greater aqueous and solid phase scavenging rates. The cost of commercially available H2O2 ($0.031 mol-1) and PS ($0.24 mol-1) was used in conjunction with the overall treatment efficiency to assess specific cost of treatment. The specific cost to treat the probe compound with UV-AHP was greater than UV-APS and was attributed to the much lower treatment efficiency with UV-AHP. The much-desired high reaction rate constants between •OH and environmental contaminants, relative to SO4•-, may come at the cost of greater combined scavenging rates, and consequently lower treatment efficiency.
