ABSTRACT: This is the first study to report a high-throughput approach integrating gene expression data from Spodoptera frugiperda guts and their associated metatranscriptomes. Our datasets provide information on the potential effects of environmental conditions on the expression profile of S. frugiperda larval guts, their associated metatranscriptome, and putative interactions between them.

Project description:BackgroundSpodoptera frugiperda (J. E. Smith), commonly known as fall armyworm (FAW), is one of the most destructive agricultural pests in the world and has posed a great threat to crops. The improper use of insecticides has led to rapid development of resistance. However, the genetic data available for uncovering the insecticide resistance mechanisms are scarce.MethodsIn this study, we used PacBio single-molecule real-time (SMRT) sequencing aimed at revealing the full-length transcriptome profiling of the FAW larval brain to obtain detoxification genes.ResultsA total of 18,642 high-quality transcripts were obtained with an average length of 2,371 bp, and 11,230 of which were successfully annotated in six public databases. Among these, 5,692 alternative splicing events were identified.

Spodoptera frugiperda kinetochore co-immunoprecipitations

Project description:Accurate chromosome segregation requires assembly of the multiprotein kinetochore complex at centromeres. In most eukaryotes, kinetochore assembly is primed by the histone H3 variant CenH3, which physically interacts with components of the inner kinetochore constitutive-centromere-associated-network (CCAN). Unexpected to its critical function, previous work identified that select eukaryotic lineages, including several insects, have lost CenH3, while having retained homologs of the CCAN. These findings imply alternative CCAN assembly pathways in these organisms that function in CenH3-independent manners. Here, we study the composition and assembly of CenH3-independent kinetochores of Lepidoptera. We show that lepidopteran kinetochores consist of previously identified CCAN homologs as well as additional components including a divergent CENP-T homolog, which are required for accurate mitotic progression. Our study focuses on CENP-T that we find both necessary and sufficient to recruit the Mis12 outer kinetochore complex. In addition, CRISPR-mediated gene editing in Bombyx mori establishes an essential function of CENP-T in vivo. Finally, the retention of CENP-T homologs in other independently-derived CenH3-deficient insects indicates a conserved mechanism of kinetochore assembly between these lineages. Our study provides the first functional insights into CCAN-based kinetochore assembly pathways that function independently of CenH3, thus contributing to the emerging picture of an unexpected plasticity to build a kinetochore.

Do NIR spectra collected from laboratory-reared mosquitoes differ from those collected from wild mosquitoes?

Project description:BACKGROUND:Near infrared spectroscopy (NIRS) is a high throughput technique that measures absorbance of specific wavelengths of light by biological samples and uses this information to classify the age of lab-reared mosquitoes as younger or older than seven days with an average accuracy greater than 80%. For NIRS to estimate ages of wild mosquitoes, a sample of wild mosquitoes with known age in days would be required to train and test the model. Mark-release-recapture is the most reliable method to produce wild-caught mosquitoes of known age in days. However, it is logistically demanding, time inefficient, subject to low recapture rates, and raises ethical issues due to the release of mosquitoes. Using labels from Detinova dissection results in a mathematical model with poor accuracy. Alternatively, a model trained on spectra from laboratory-reared mosquitoes where age in days is known can be applied to estimate the age of wild mosquitoes, but this would be appropriate only if spectra collected from laboratory-reared and wild mosquitoes are similar. METHODS AND FINDINGS:We performed k-means (k = 2) cluster analysis on a mixture of spectra collected from lab-reared and wild Anopheles arabiensis to determine if there is any significant difference between these two groups. While controlling the numbers of mosquitoes included in the model at each age, we found two clusters with no significant difference in distribution of spectra collected from lab-reared and wild mosquitoes (p = 0.25). We repeated the analysis using hierarchical clustering, and similarly, no significant difference was observed (p = 0.13). CONCLUSION:We find no difference between spectra collected from laboratory-reared and wild mosquitoes of the same age and species. The results strengthen and support the on-going practice of applying the model trained on spectra collected from laboratory-reared mosquitoes, especially first-generation laboratory-reared mosquitoes.

Baseline Susceptibility of Spodoptera frugiperda Populations Collected in India towards Different Chemical Classes of Insecticides.

Project description:Fall armyworm (FAW), Spodoptera frugiperda, is a major pest of maize in the Americas and recently invaded the Eastern hemisphere. It was first detected in India in 2018 and is considered a major threat to maize production. FAW control largely relies on the application of chemical insecticides and transgenic crops expressing Bacillus thuringiensis insecticidal proteins. Assessing FAW resistance and insecticide susceptibility is a cornerstone to develop sustainable resistance management strategies. In this study, we conducted more than 400 bioassays to assess the efficacy of nine insecticides from seven mode-of-action classes against 47 FAW populations collected in 2019 and 2020 across various geographical areas in India. The resistance status of the field-collected populations was compared to an Indian population sampled in 2018, and an insecticide susceptible reference population collected in 2005 in Brazil. Low to moderate resistance levels were observed for thiodicarb, chlorpyriphos, deltamethrin, chlorantraniliprole and flubendiamide in several populations (including the reference population collected in 2018). The highest resistance ratios were observed for deltamethrin which likely compromises recommended label rates for pyrethroid insecticides in general. Our data provide a useful baseline for future FAW resistance monitoring initiatives and highlight the need to implement insecticide resistance management strategies.

Virulence of entomopathogenic fungi against fall armyworm, <i>Spodoptera frugiperda</i> (Lepidoptera: Noctuidae) under laboratory conditions.

Project description:Maize is an essential crop of China. The recent invasion of Spodoptera frugiperda, also known as fall armyworm (FAW), poses a danger to the country's ability to maintain a sustainable level of productivity from this core crop. Entomopathogenic fungi (EPF) Metarhizium anisopliae MA, Penicillium citrinum CTD-28 and CTD-2, Cladosporium sp. BM-8, Aspergillus sp. SE-25 and SE-5, Metarhizium sp. CA-7, and Syncephalastrum racemosum SR-23 were tested to determine their effectiveness in causing mortality in second instars, eggs, and neonate larvae. Metarhizium anisopliae MA, P. citrinum CTD-28, and Cladosporium sp. BM-8 caused the highest levels of egg mortality, with 86.0, 75.3, and 70.0%, respectively, followed by Penicillium sp. CTD-2 (60.0%). Additionally, M. anisopliae MA caused the highest neonatal mortality of 57.1%, followed by P. citrinum CTD-28 (40.7%). In addition, M. anisopliae MA, P. citrinum CTD-28, and Penicillium sp. CTD-2 decreased the feeding efficacy of second instar larvae of FAW by 77.8, 75.0, and 68.1%, respectively, followed by Cladosporium sp. BM-8 (59.7%). It is possible that EPF will play an important role as microbial agents against FAW after further research is conducted on the effectiveness of these EPF in the field.

Differential protection of Cry1Fa toxin against Spodoptera frugiperda larval gut proteases by cadherin orthologs correlates with increased synergism.

Project description:The Cry proteins produced by Bacillus thuringiensis (Bt) are the most widely used biopesticides effective against a range of crop pests and disease vectors. Like chemical pesticides, development of resistance is the primary threat to the long-term efficacy of Bt toxins. Recently discovered cadherin-based Bt Cry synergists showed the potential to augment resistance management by improving efficacy of Cry toxins. However, the mode of action of Bt Cry synergists is thus far unclear. Here we elucidate the mechanism of cadherin-based Cry toxin synergism utilizing two cadherin peptides, Spodoptera frugiperda Cad (SfCad) and Manduca sexta Cad (MsCad), which differentially enhance Cry1Fa toxicity to Spodoptera frugiperda neonates. We show that differential SfCad- and MsCad-mediated protection of Cry1Fa toxin in the Spodoptera frugiperda midgut correlates with differential Cry1Fa toxicity enhancement. Both peptides exhibited high affinity for Cry1Fa toxin and an increased rate of Cry1Fa-induced pore formation in S. frugiperda. However, only SfCad bound the S. frugiperda brush border membrane vesicle and more effectively prolonged the stability of Cry1Fa toxin in the gut, explaining higher Cry1Fa enhancement by this peptide. This study shows that cadherin fragments may enhance B. thuringiensis toxicity by at least two different mechanisms or a combination thereof: (i) protection of Cry toxin from protease degradation in the insect midgut and (ii) enhancement of pore-forming ability of Cry toxin.

Expression of ricin B chain in Spodoptera frugiperda.

Project description:DNA encoding ricin B chain was derived from preproricin genomic DNA and ligated into the baculovirus transfer vector, pAcGP67A. Co-transfection of Spodoptera frugiperda Sf9 cells with BaculoGold DNA was followed by limiting dilution purification of recombinant baculovirus. Infection of SF9 cells at a multiplicity of infection of 1 in the presence of 25 mM lactose produced 3 mg/l of soluble, glycosylated, 34 kDa protein immunoreactive with monoclonal and polyclonal antibodies to ricin B chain. The recombinant ricin B chain had similar lectin-binding activity to plant ricin B chain. The recombinant protein reassociated with ricin toxin A chain similarly to ricin toxin B chain and the recombinant heterodimers had similar cell cytotoxicity to ricin.

Project description:We report the entire genome sequence of an isolate of Spodoptera frugiperda multiple nucleopolyhedrovirus from Nigeria, West Africa. The genome is 132,710 bp long and contains 144 open reading frames. The GC content is 40.3% and, based on baculovirus species demarcation criteria, the isolate belongs to the species Spodoptera frugiperda multiple nucleopolyhedrovirus.

Functional expression of a peritrophin A-like SfPER protein is required for larval development in Spodoptera frugiperda (Lepidoptera: Noctuidae).

Project description:Peritrophins are associated with structural and functional integrity of peritrophic membranes (PM), structures composed of chitin and proteins. PM lines the insect midgut and has roles in digestion and protection from toxins. We report the full-length cDNA cloning, molecular characterization and functional analysis of SfPER, a novel PM peritrophin A protein, in Spodoptera frugiperda. The predicted amino acid sequence indicated SfPER's domain structure as a CMCMC-type, consisting of a signal peptide and three chitin-binding (C) domains with two intervening mucin-like (M) domains. Phylogenetic analysis determined a close relationship between SfPER and another S. frugiperda PM peritrophin partial sequence. SfPER transcripts were found in larvae and adults but were absent from eggs and pupae. Chitin affinity studies with a recombinant SfPER-C1 peritrophin A-type domain fused to SUMO/His-tag confirmed that SfPER binds to chitin. Western blots of S. frugiperda larval proteins detected different sized variants of SfPER along the PM, with larger variants found towards the posterior PM. In vivo suppression of SfPER expression did not affect susceptibility of larvae to Bacillus thuringiensis toxin, but significantly decreased pupal weight and adult emergence, possibly due to PM structural alterations impairing digestion. Our results suggest SfPER could be a novel target for insect control.

What Defines a Host? Oviposition Behavior and Larval Performance of Spodoptera frugiperda (Lepidoptera: Noctuidae) on Five Putative Host Plants.

Project description:When an invasive species first breaches quarantine and establishes in yet another country, it invariably causes consternation for growers, in part because of incomplete understanding of the plants that are at risk. The Fall Armyworm, Spodoptera frugiperda (J.E. Smith) is the most recent example in Australia. The number of plants that this polyphagous noctuid is reported to attack is vast, including many crop species. Consequently, initial reactions from grower industry groups that perceived themselves at risk were to demand emergency use of insecticides. Yet the field evidence suggests that many crops might not be at risk and since S. frugiperda arrived in Australia, maize crops have suffered most damage, followed by sorghum. We question the accuracy of some of the claims of reported host plants of S. frugiperda and report experiments that compared oviposition behavior, neonate silking behavior, and larval performance on five crops: the known hosts maize and sorghum, and the putative hosts cotton, peanut, and pigeon pea. Maize ranked highest in all preference and performance measures, followed by sorghum and peanut, with pigeon pea and cotton ranking lowest. Although S. frugiperda can survive, develop, and pupate on the crop species we examined, cotton and pigeon pea are not preferred by the pest in either the larval or adult stages. We suggest that before a plant is listed as a host for a given insect that the evidence should be fully reported and carefully evaluated. Collecting an immature insect from a plant does not make that plant a host!
