Project description:Saline environments are extreme habitats with a high diversity of microorganisms source of a myriad of biomolecules. These microorganisms are assigned as extremophiles recognized to be producers of new natural compounds, which can be synthesized by helping to survive under harshness and extreme conditions. In Brazil, in the saline and semi-arid region of Areia Branca (Caatinga biome), halotolerant bacteria (able to growth at high NaCl concentrations) were isolated from rhizosphere of native plants Blutaparon portulacoides and Spergularia sp. and their biopolymer production was studied. A total of 25 bacterial isolates were identified at genus level based on 16S rRNA gene sequence analysis. Isolates were mainly Gram-positive bacteria from Bacillaceae, Staphylococcaceae, Microbacteriaceae, and Bacillales XII incertae sedis families, affiliates to Bacillus, Staphylococcus, Curtobacterium, and Exiguobacterium genera, respectively. One of the Gram-negative isolates was identified as member of the Pseudomonadaceae family, genus Pseudomonas. All the identified strains were halotolerant bacteria with optimum growth at 0.6-2.0 M salt concentrations. Assays for biopolymer production showed that the halotolerant strains are a rich source of compounds as polyhydroxyalkanoates (PHA), biodegradable biopolymer, such as poly(3-hydroxybutyrate) (PHB) produced from low-cost substrates, and exopolysaccharides (EPS), such as hyaluronic acid (HA), metabolite of great interest to the cosmetic and pharmaceutical industry. Also, eight bacterial EPS extracts showed immunostimulatory activity, promising results that can be used in biomedical applications. Overall, our findings demonstrate that these biomolecules can be produced in culture medium with 0.6-2.0 M NaCl concentrations, relevant feature to avoid costly production processes. This is the first report of biopolymer-producing bacteria from a saline region of Caatinga biome that showed important biological activities.

Project description:The zebrafish embryo has repeatedly proved to be a useful model for the analysis of effects by environmental toxicants. This study was performed to investigate if an approach combining mechanism-specific bioassays with microarray techniques can obtain more in-depth insights into the ecotoxicity of complex pollutant mixtures as present, e.g., in freeze-dried whole sediment samples and their corresponding organic extracts in parallel. To this end, altered gene expression was compared to data from established bioassays as well as to results from chemical analysis. Microarray analysis revealed several classes of significantly regulated genes which could to a considerable extent be related to the hazard potential. Results indicate that potential classes of contaminants can be assigned to sediment extracts by both classical biomarker genes and corresponding expression profile analyses of known substances. However, it is difficult to distinguish between specific responses and more universal detoxification of the organism. Additionally, different gene expression was shown to be less influenced by the sampling site than by the method of exposure, which could be attributed to differential bioavailability of contaminants.

Microarray analyses were performed with early life stages of zebrafish exposed to sediment extracts or freeze-dried sediment from three sampling sites (Ehingen, Lauchert, Sigmaringen) along the Upper part of the Danube River, Germany. The expression profiles were compared within the sampling sites, between the exposure scheme and to the expression pattern of model toxicants, such as 4-chloroaniline, Cadmium, DDT, TCDD, and Valproic acid (Gene Expression Omnibus Series GSE9357). Additionally, mechanism-specific bioassays and chemical analysis of the sediments have been combined and compared to the present gene expression data.

Project description:RNA from Lake Michigan sediment was hybridized with a DNA probe for archaeal 16S rRNA. There was a peak of archaeal rRNA abundance in the oxic zone and another immediately below it. Six contributing species were identified by PCR amplification of extracted DNA with primers specific for archaeal rDNA: two related to Methanosarcina acetivorans and four related to marine crenarchaeotal sequences. rRNA quantification using a DNA probe specific for this crenarchaeotal assemblage showed it is most abundant in the oxic zone, where it accounts for about 10% of total archaeal rRNA.

Project description:The Caatinga is a semi-arid biome in northeast Brazil. The Paraguaçú River is located in the Caatinga biome, and part of its course is protected by the National Park of Chapada Diamantina (PNCD). In this study we evaluated the effect of PNCD protection on the water quality and microbial community diversity of this river by analyzing water samples obtained from points located inside and outside the PNCD in both wet and dry seasons. Results of water quality analysis showed higher levels of silicate, ammonia, particulate organic carbon, and nitrite in samples from the unprotected area compared with those from protected areas. Pyrosequencing of the 16S rRNA genes revealed that Burkholderiales was abundant in samples from all three sites during both seasons and was represented primarily by the genus Polynucleobacter and members of the Comamonadaceae family (e.g., genus Limnohabitans). During the dry season, the unprotected area showed a higher abundance of Flavobacterium sp. and Arthrobacter sp., which are frequently associated with the presence and/or degradation of arsenic and pesticide compounds. In addition, genes that appear to be related to agricultural impacts on the environment, as well as those involved in arsenic and cadmium resistance, copper homeostasis, and propanediol utilization, were detected in the unprotected areas by metagenomic sequencing. Although PNCD protection improves water quality, agricultural activities around the park may affect water quality within the park and may account for the presence of bacteria capable of pesticide degradation and assimilation, evidencing possible anthropogenic impacts on the Caatinga.

Project description:Roots of plants characteristic of the Brazilian caatinga showing necrosis symptoms were observed in Iguatu, CE, Brazil. To identify the species, morphological characterization was performed, through the morphometry of females, and molecular analysis of the ITS and 28S rDNA regions. The nematodes Pratylenchus zeae and P. brachyurus were identified as causal agents, confirming pathogenicity by Koch postulates. This is the first report of P. zeae and P. brachyurus in caaatinga plants in the state of Ceará, Brazil.

Project description:The jaguar, Panthera onca, is a top predator with the extant population found within the Brazilian Caatinga biome now known to be on the brink of extinction. Designing new conservation units and potential corridors are therefore crucial for the long-term survival of the species within the Caatinga biome. Thus, our aims were: 1) to recognize suitable areas for jaguar occurrence, 2) to delineate areas for jaguar conservation (PJCUs), 3) to design corridors among priority areas, and 4) to prioritize PJCUs. A total of 62 points records of jaguar occurrence and 10 potential predictors were analyzed in a GIS environment. A predictive distributional map was obtained using Species Distribution Modeling (SDM) as performed by the Maximum Entropy (Maxent) algorithm. Areas equal to or higher than the median suitability value of 0.595 were selected as of high suitability for jaguar occurrence and named as Priority Jaguar Conservation Units (PJCU). Ten PJCUs with sizes varying from 23.6 km2 to 4,311.0 km2 were identified. Afterwards, we combined the response curve, as generated by SDM, and expert opinions to create a permeability matrix and to identify least cost corridors and buffer zones between each PJCU pair. Connectivity corridors and buffer zone for jaguar movement included an area of 8.884,26 km2 and the total corridor length is about 160.94 km. Prioritizing criteria indicated the PJCU representing c.a. 68.61% of the total PJCU area (PJCU # 1) as of high priority for conservation and connectivity with others PJCUs (PJCUs # 4, 5 and 7) desirable for the long term survival of the species. In conclusion, by using the jaguar as a focal species and combining SDM and expert opinion we were able to create a valid framework for practical conservation actions at the Caatinga biome. The same approach could be used for the conservation of other carnivores.

Project description:Marine sediments harbor complex microbial communities that remain poorly studied relative to other biomes such as seawater. Moreover, bacteria in these communities produce antibiotics and other bioactive secondary metabolites, yet little is known about how these compounds affect microbial community structure. In this study, we used next-generation amplicon sequencing to assess native microbial community composition in shallow tropical marine sediments. The results revealed complex communities comprised of largely uncultured taxa, with considerable spatial heterogeneity and known antibiotic producers comprising only a small fraction of the total diversity. Organic extracts from cultured strains of the sediment-dwelling actinomycete genus Salinispora were then used in mesocosm studies to address how secondary metabolites shape sediment community composition. We identified predatory bacteria and other taxa that were consistently reduced in the extract-treated mesocosms, suggesting that they may be the targets of allelopathic interactions. We tested related taxa for extract sensitivity and found general agreement with the culture-independent results. Conversely, several taxa were enriched in the extract-treated mesocosms, suggesting that some bacteria benefited from the interactions. The results provide evidence that bacterial secondary metabolites can have complex and significant effects on sediment microbial communities. Ocean sediments represent one of Earth's largest and most poorly studied biomes. These habitats are characterized by complex microbial communities where competition for space and nutrients can be intense. This study addressed the hypothesis that secondary metabolites produced by the sediment-inhabiting actinomycete Salinispora arenicola affect community composition and thus mediate interactions among competing microbes. Next-generation amplicon sequencing of mesocosm experiments revealed complex communities that shifted following exposure to S. arenicola extracts. The results reveal that certain predatory bacteria were consistently less abundant following exposure to extracts, suggesting that microbial metabolites mediate competitive interactions. Other taxa increased in relative abundance, suggesting a benefit from the extracts themselves or the resulting changes in the community. This study takes a first step toward assessing the impacts of bacterial metabolites on sediment microbial communities. The results provide insight into how low-abundance organisms may help structure microbial communities in ocean sediments.

Project description:Mosquito fauna in the northeast semiarid region of Brazil, Caatinga biome, are poorly known. Studies on the diversity are scarce and the few surveys available focus on local fauna. In order to understand the ecological pattern of mosquito's distribution, information available from studies from 2008 to 2015 were gathered. A partitioning framework of the beta diversity, the turnover (βJTU) and nestedness (βJNE) components were used to determine dissimilarity among communities. Eighty-two morphospecies were recorded and 47 of the species were not shared between the areas. The most representative genera were Aedes, Anopheles, Psorophora, Haemagogus, Coquillettidia, and Mansonia, which all include species of medical interest. The communities had high rates of variation, and the mechanism of turnover accounted for the observed diversity pattern. Despite differences in collection methods, the observed dissimilarity may be related to the broad environmental heterogeneity of the biome, the intrinsic relationships of the species with their habitats, and the environmental degradation caused by different types of anthropogenic interference. Considering the mosquito species richness and endemicity, the hypothesis that the Caatinga harbor poor biodiversity is rejected. The spatial variation observed is of particular importance and should be taken into account for the knowledge of Caatinga biodiversity.
