The synthesis and characterization of luminogenic, bioorthogonal iridium probes is described. These probes exhibit long photoluminescence lifetimes amenable to time-resolved applications. Project description:Oncosis is a non-apoptotic form of programmed cell death (PCD), which differs from apoptosis in both morphological changes and inner pathways, and might hold the key to defeating a major obstacle in cancer therapy - drug-resistance, which is often a result of the intrinsic apoptosis resistance of tumours. However, despite the fact that the term "oncosis" was coined and used much earlier than apoptosis, little effort has been made to discover new drugs which can initiate this form of cell death, in comparison to drugs inducing apoptosis or any other type of PCD. So herein, we present the synthesis of a series of mitochondria-targeting cyclometalated Ir(iii) complexes, which activated the oncosis-specific protein porimin and calpain in cisplatin-resistant cell line A549R, and determined their cytotoxicity against a wide range of drug-resistant cancer types. To the best of our knowledge, these complexes are the very first metallo-components to induce oncosis in drug-resistant cancer cells.

Project description:Heterogenized photoredox catalysts provide a path for sustainable chemical synthesis using highly tunable, reusable constructs. Here, heterogenized iridium complexes as photoredox catalysts were assembled via covalent attachment to metal oxide surfaces (ITO, ZrO2 , Al2 O3 ) in thin film or nanopowder constructs. The goal was to understand which materials provided the most promising constructs for catalysis. To do this, reductive dehalogenation of bromoacetophenone to acetophenone was studied as a test reaction for system optimization. All catalyst constructs produced acetophenone with high conversions and yields with the fastest reactions complete in fifteen minutes using Al2 O3 supports. The nanopowder catalysts resulted in faster and more efficient catalysis, while the thin film catalysts were more robust and easily reused. Importantly, the thin film constructs show promise for future photoelectrochemical and electrochemical photoredox setups. Finally, all catalysts were reusable 2-3 times, performing at least 1000 turnovers (Al2 O3 ), demonstrating that heterogenized catalysts are a sustainable catalyst alternative.

Project description:The first structure report of trichlorido[4'-(p-tolyl)-2,2':6',2''-terpyridine]iridium(III) dimethyl sulfoxide solvate, [IrCl(3)(C(22)H(17)N(3))] x C(2)H(6)OS, (I), is presented, along with a higher-symmetry setting of previously reported bis[4'-(p-tolyl)-2,2':6',2''-terpyridine]iridium(III) tris(hexafluoridophosphate) acetonitrile disolvate, [Ir(C(22)H(17)N(3))(2)](PF(6))(3) x 2 C(2)H(3)N, (II) [Yoshikawa, Yamabe, Kanehisa, Kai, Takashima & Tsukahara (2007). Eur. J. Inorg. Chem. pp. 1911-1919]. For (I), the data were collected with synchrotron radiation and the dimethyl sulfoxide solvent molecule is disordered over three positions, one of which is an inversion center. The previously reported structure of (II) is presented in the more appropriate C2/c space group. The iridium complex and one PF(6)(-) anion lie on twofold axes in this structure, making half of the molecule unique.

Project description:A unique chain-rupturing transformation that converts an ether functionality into two hydrocarbyl units and carbon monoxide is reported, mediated by iridium(i) complexes supported by aminophenylphosphinite (NCOP) pincer ligands. The decarbonylation, which involves the cleavage of one C-C bond, one C-O bond, and two C-H bonds, along with formation of two new C-H bonds, was serendipitously discovered upon dehydrochlorination of an iridium(iii) complex containing an aza-18-crown-6 ether macrocycle. Intramolecular cleavage of macrocyclic and acyclic ethers was also found in analogous complexes featuring aza-15-crown-5 ether or bis(2-methoxyethyl)amino groups. Intermolecular decarbonylation of cyclic and linear ethers was observed when diethylaminophenylphosphinite iridium(i) dinitrogen or norbornene complexes were employed. Mechanistic studies reveal the nature of key intermediates along a pathway involving initial iridium(i)-mediated double C-H bond activation.

Project description:Mitochondria generate energy but malfunction in many cancer cells, hence targeting mitochondrial metabolism is a promising approach for cancer therapy. Here we have designed cyclometallated iridium(iii) complexes, containing one TEMPO (2,2,6,6-tetramethylpiperidine-1-oxyl) spin label [C43H43N6O2Ir1·PF6]˙ (Ir-TEMPO1) and two TEMPO spin labels [C52H58N8O4Ir1·PF6]˙ (Ir-TEMPO2). Electron paramagnetic resonance (EPR) spectroscopy revealed spin-spin interactions between the TEMPO units in Ir-TEMPO2. Both Ir-TEMPO1 and Ir-TEMPO2 showed bright luminescence with long lifetimes (ca. 35-160 ns); while Ir-TEMPO1 displayed monoexponential decay kinetics, the biexponential decays measured for Ir-TEMPO2 indicated the presence of more than one energetically-accessible conformation. This observation was further supported by density functional theory (DFT) calculations. The antiproliferative activity of Ir-TEMPO2 towards a range of cancer cells was much greater than that of Ir-TEMPO1, and also the antioxidant activity of Ir-TEMPO2 is much higher against A2780 ovarian cancer cells when compared with Ir-TEMPO1. Most notably Ir-TEMPO2 was particularly potent towards PC3 human prostate cancer cells (IC50 = 0.53 μM), being ca. 8× more active than the clinical drug cisplatin, and ca. 15× more selective towards cancer cells versus normal cells. Confocal microscopy showed that both Ir-TEMPO1 and Ir-TEMPO2 localise in the mitochondria of cancer cells.

Project description:Transition metal complexes containing dimethyl sulfoxide (DMSO) are important precursors in catalysis and metallodrugs. Understanding the solid-state supramolecular structure is crucial for predicting the properties and biological activity of the material. Several crystalline phases of DMSO-coordinated iridium anions with different cations, potassium (1) and -butylammonium (1), were obtained and their structures determined by X-ray crystallography. Compound (1) is present in two solvatomorphic forms: ? and ?; the ? form contains disordered solvent water. In addition, the structures exhibit different rotamers of the -[IrCl(DMSO)] anion with the -DMSO ligands being oriented in and conformations. In consideration of these various conformers, the effects of the crystallized solvent and intermolecular interactions on the conformational preferences of the anion are discussed. In addition, density functional theory calculations were used to investigate the energies of the anions in the different conformations. It was found that hydrogen bonds between water and the DMSO complex stabilize the conformation which is the least stable form of the -DMSO complex. Consequently, by controlling the number of hydrogen-bond donors and acceptors and the amount of water, it may be possible to obtain different solvatomorphs of clinically significant metallodrugs.

Project description:Nearly all the neutral iridium complexes widely used as dopants in PhOLEDs are racemic mixtures; however, this study observed that these complexes can be separated into stable optically active ? and ? isomers and that their chirality is an intrinsic property. The circularly polarised phosphorescent photoluminescence (CPPPL) signals of ?/? isomers are perfect mirror images with opposite polarisation and equal intensity exhibiting a "handedness" for the polarisation. For the first time, we applied the ?/? iridium isomers as emitters in OLEDs, and the circularly polarised phosphorescent electroluminescence (CPPEL) spectra reveal completely positive or negative broad peaks consistent with the CPPPL spectra. The results demonstrate that the ?/? isomers have potential application for 3D OLEDs because they can exhibit high efficiency and luminance, and 3D display technology based on circularly polarised light is the most comfortable for the eyes.

Project description:New functionalized tris(2',6'-difluoro-2,3'-bipyridinato-N,C4')iridium(III) ((dfpypy)3Irs) complexes, including small molecules and their dendrimer embedded analogoues, were synthesized and characterized. It is demonstrated that both the fac-(dfpypy)3Ir-based polyphenylene dendrimers and (triisopropylsilyl)ethynyl (TIPSE)-substituted (dfpypy)3Ir complexes induce large bathochromic shifts (∼50 nm) of emission bands compared with fac-(dfpypy)3Ir. This is due to the pronounced 3π-π* character of emissive excited states and the extended conjugation. A further remarkable feature is the small bathochromic shift of the emissions of fac-tris(2-phenylpyridine)iridium (fac-(ppy)3Ir)-based polyphenylene dendrimers when compared to those of the iridium (Ir) complex core. Obviously, the triplet metal-to-ligand charge transfer makes emission less sensitive to extended conjugation than the 3π-π* transition. This finding suggests new concepts for designing blue phosphorescent dendrimer emitters. Both the dendrimers and the TIPSE-substituted (dfpypy)3Ir complexes represent new green and the trimethylsilyl-functionalized (dfpypy)3Ir new blue phosphorescent emitters. Incorporation of TIPSE moieties into the ligands of iridium complex gives rise to enhanced phosphorescence.

Project description:Cyclometalated iridium complexes have emerged as top-performing emitters in organic light-emitting diodes (OLEDs) and other optoelectronic devices. A persistent challenge has been the development of cyclometalated iridium complexes with deep blue luminescence that have the requisite color purity, efficiency, and stability to function in color displays. In this work we report a new class of cyclometalated iridium complexes with saturated blue luminescence. These complexes have the general structure Ir(C^C:NHC)2(C^C:ADC), where C^C:NHC is an N-heterocyclic carbene (NHC) derived cyclometalating ligand and C^C:ADC is a different type of cyclometalating ligand featuring an acyclic diaminocarbene (ADC). The complexes are prepared by a cascade reaction that involves nucleophilic addition of propylamine to an isocyanide precursor followed by base-assisted cyclometalation of the ADC intermediate. All three emit deep blue light with good quantum efficiencies (Φ PL = 0.13-0.48) and color profiles very close to the ideal primary blue standards for color displays.
