A new species of Mollitrichosiphum Suenaga from Taiwan Island (Hemiptera, Aphididae), based on morphological characteristics and DNA sequences.

ABSTRACT: A new species of Mollitrichosiphum Suenaga, Mollitrichosiphum tumorisiphum Qiao & Jiang, sp. n., from Fagus longipetiolata in Taiwan island is described. Siphunculi of Mollitrichosiphum tumorisiphum in alatae are distinctly swollen on the distal part, unlike those of the other known species in the genus. Updated keys to apterous and alate viviparous females of all known Chinese species of Mollitrichosiphum are provided. The specimens studied are deposited in the National Zoological Museum of China, Institute of Zoology, Chinese Academy of Sciences, Beijing, China and the Natural History Museum, London, United Kingdom.

Microbiota associated with Mollitrichosiphum aphids (Hemiptera: Aphididae: Greenideinae): diversity, host species specificity and phylosymbiosis.

Project description:Symbiotic association is universal in nature, and an array of symbionts play a crucial part in host life history. Aphids and their diverse symbionts have become a good model system to study insect-symbiont interactions. Previous symbiotic diversity surveys have mainly focused on a few aphid clades, and the relative importance of different factors regulating microbial community structure is not well understood. In this study, we collected 65 colonies representing eight species of the aphid genus Mollitrichosiphum from different regions and plants in southern China and Nepal and characterized their microbial compositions using Illumina sequencing of the V3 - V4 hypervariable region of the 16S rRNA gene. We evaluated how microbiota varied across aphid species, geography and host plants and the correlation between microbial community structure and host aphid phylogeny. Heritable symbionts dominated the microbiota associated with Mollitrichosiphum, and multiple infections of secondary symbionts were prevalent. Ordination analyses and statistical tests highlighted the contribution of aphid species in shaping the structures of bacterial, symbiont and secondary symbiont communities. Moreover, we observed a significant correlation between Mollitrichosiphum aphid phylogeny and microbial community composition, providing evidence for a pattern of phylosymbiosis between natural aphid populations and their microbial associates.

Phylosymbiotic Structures of the Microbiota in Mollitrichosiphum tenuicorpus (Hemiptera: Aphididae: Greenideinae).

Project description:Aphids harbor an array of symbionts that provide hosts with ecological benefits. Microbial community assembly generally varies with respect to aphid species, geography, and host plants. However, the influence of host genetics and ecological factors on shaping intraspecific microbial community structures has not been fully understood. In the present study, using Illumina sequencing of the V3 - V4 hypervariable region of the 16S rRNA gene, we characterized the microbial compositions associated with Mollitrichosiphum tenuicorpus from different regions and plants in China. The primary symbiont Buchnera aphidicola and the secondary symbiont Arsenophonus dominated the microbial flora in M. tenuicorpus. Ordination analyses and statistical tests suggested that geography and aphid genetics primarily contributed to the variation in the microbiota of M. tenuicorpus. We further confirmed the combined effect of aphid genetics and geography on shaping the structures of symbiont and secondary symbiont communities. Moreover, the significant correlation between aphid genetic divergence and symbiont community dissimilarity provides evidence for intraspecific phylosymbiosis in natural systems. Our study helped to elucidate the eco-evolutionary relationship between symbiont communities and aphids within one given species.

Complete mitochondrial genome of <i>Mollitrichosiphum tenuicorpus</i> (Okajima, 1908) (Hemiptera: Aphididae: Greenideinae).

Project description:Here we report the complete mitochondrial genome of the aphid species Mollitrichosiphum tenuicorpus. The M. tenuicorpus mitogenome is 15,727 bp in length and comprising 37 genes typically present in insect mitogenomes, a control region, and a unique repeat region. All protein-coding genes (PCGs) terminate with TAA or TAG except for cox1, which is terminated with T-. The secondary structure of trnS (AGN) loses the dihydrouridine (DHU) arm, but all the other transfer RNAs show the typical clover-leaf secondary structure. The repeat region between trnE and trnF is 458 bp, with a 217-bp repeat unit repeating 2.11 times. Phylogenetic analysis of the M. tenuicorpus mitogenome using the maximum-likelihood optimality criterion places it in a strongly supported sister position to Eutrichosiphum pasaniae. These data show that mitogenome sequences could be useful in resolving phylogenetic relationships of the Greenideinae.

Project description:Ericaphisvoegtlini sp. n. living on Chamaebatiafoliolosa (Rosaceae) in California is described based on apterous and alate viviparous females. The new species differs from all other species of the genus Ericaphis Börner, 1939 in several important morphological characters including very long and rigid dorsal setae and distinctly swollen siphunculi with clearly visible polygonal reticulation.

Project description:We report and describe the first species of Atheroides Haliday presumed to be native to North America, collected at the Valles Caldera National Preserve, New Mexico, USA. We hypothesize its placement among the Siphini based on morphological, phylogenetic analysis and extend the distribution of the genus to the Holoarctic. We expand the key of the known Atheroides to include the new species and discuss the current hypotheses of the geographic distribution of the type species, Atheroidesserrulatus Haliday.

Species differentiation of Chinese mollitrichosiphum (Aphididae: greenideinae) driven by geographical isolation and host plant acquirement.

Project description:The impact of both the uplift of the Qinghai-Tibetan Plateau (QTP) and the separation of the Taiwan and Hainan Islands on the evolution of the fauna and flora in adjacent regions has been a topic of considerable interest. Mollitrichosiphum is a polyphagous insect group with a wide range of host plants (14 families) and distributions restricted to Southeast Asia. Based on the mitochondrial Cytochrome C Oxidase Subunit I (COI) and Cytochrome b (Cytb) genes, the nuclear elongation factor-1? (EF-1?) gene, and the detailed distribution and host plant data, we investigated the species differentiation modes of the Chinese Mollitrichosiphum species. Phylogenetic analyses supported the monophyly of Mollitrichosiphum. The divergence time of Mollitrichosiphum tenuicorpus (c. 11.0 mya (million years ago)), Mollitrichosiphum nandii and Mollitrichosiphum montanum (c. 10.6 mya) was within the time frame of the uplift of the QTP. Additionally, basal species mainly fed on Fagaceae, while species that fed on multiple plants diverged considerably later. Ancestral state reconstruction suggests that Fagaceae may be the first acquired host, and the acquisition of new hosts and the expansion of host range may have promoted species differentiation within this genus. Overall, it can be concluded that geographical isolation and the expansion of the host plant range may be the main factors driving species differentiation of Mollitrichosiphum.
