How to Regenerate and Protect Desert Riparian Populus euphratica Forest in Arid Areas.

ABSTRACT: We found that the most suitable flooding disturbance model for regenerating Populus euphratica forest was two to three times per year with a duration of 15-20 days and an intensity of 25-30?m(3)/s. The flooding should take place during the seed emergence to young tree growth stages, and should be based on flooding experiments and data from vegetation quadrats and ecological water conveyance. Furthermore, we found that tree-ring width index for P. euphratica declined as the groundwater depth increased, and ascertained that the minimum groundwater depths for young trees, near-mature trees, mature trees and over-mature trees were 4.0?m, 5.0-5.4?m, 6.9?m and 7.8?m, respectively. These were derived from a quantitative relationship model between groundwater depth and tree-ring width index. The range for ecological water conveyance volume was 311-320 million m(3) in the lower reaches of the Tarim River. This study not only provides a technical basis for sustainable ecological water conveyance in the Tarim River Basin, but also offers a theoretical guide and scientific information that could be used in similar areas to regenerate and protect Populus euphratica around the world.

Project description:Populus euphratica Oliv. (Euphrates poplar), as the dominant tree species of desert riparian forests along the Central Asian inland rivers, plays a critical role in protecting arid land ecosystems. In recent decades, climate change and excessive water resources utilization activities have led to the environmental degradation of desert riparian forests along the Tarim River in northwest China. Understanding the forest stand structure and spatial distribution pattern provide important guidance for monitoring forest dynamics in support of sustainable management. However, few studies have examined how riparian forests stand attributes differ in response to environmental heterogeneity. In this study, terrestrial laser scanning (TLS) was applied to acquire a total of 1648 individual P. euphratica tree's 3D structure attributes within 18 plots along the upper, middle, and lower reaches of the Tarim River, which included tree height (TH), diameter at breast height (DBH), crown diameter (CD), crown projection area (CPA), stand density index (SDI), age structure ratios, and spatial pattern. The results showed that the average tree segmentation and structure determination accuracies of TLS were 93.2 and 94.6%. From the upper to the lower reaches, the average TH and CD decreased by 3.8 and 0.3 m, while the DBH increased by 4.2 cm. The SDI and CPA exhibited the following order: upper reaches (454 n ha-1, 82.3%) > middle reaches (382 n ha-1, 67.3%) > lower reaches (263 n ha-1, 39.1%), the differences were significant at 0.05 level. The population age structure changed from growing population in the upper reaches to stable population in the middle and a temporarily stable population in the lower reaches. The pair correlation g(r) function determined random distribution pattern in the upper reaches [g(r) = 1.2], an aggregated pattern in the middle [g(r) = 3.1], and lower reaches [g(r) = 9.7]. The decline in groundwater depth and soil moisture increased aggregated distribution pattern (R = 0.67 and 0.56, P < 0.05) of the P. euphratica along the mainstream of Tarim River. The results enrich our understanding of the current development stage of P. euphratica, which is important for optimizing management strategies and realizing the sustainability of floodplain ecosystems.

Project description:BACKGROUND: Compared with other Populus species, Populus euphratica Oliv. exhibits better tolerance to abiotic stress, especially those involving extreme temperatures. However, little is known about gene regulation and signaling pathways involved in low temperature stress responses in this species. Recent development of Illumina/Solexa-based deep-sequencing technologies has accelerated the study of global transcription profiling under specific conditions. To understand the gene network controlling low temperature perception in P. euphratica, we performed transcriptome sequencing using Solexa sequence analysis to generate a leaf transcriptome at a depth of 10 gigabases for each sample. RESULTS: Using the Trinity method, 52,081,238 high-quality trimmed reads were assembled into a non-redundant set and 108,502 unigenes with an average length of 1,047 bp were generated. After performing functional annotations by aligning all-unigenes with public protein databases, 85,584 unigenes were annotated. Differentially expressed genes were investigated using the FPKM method by applying the Benjamini and Hochberg corrections. Overall, 2,858 transcripts were identified as differentially expressed unigenes in at least two samples and 131 were assigned as unigenes expressed differently in all three samples. In 4 °C-treated sample and -4 °C-treated sample, 1,661 and 866 differently expressed unigenes were detected at an estimated absolute log2-fold change of > 1, respectively. Among them, the respective number of up-regulated unigenes in C4 and F4 sample was 1,113 and 630, while the respective number of down-regulated ungenes is 548 and 236. To increase our understanding of these differentially expressed genes, we performed gene ontology enrichment and metabolic pathway enrichment analyses. A large number of early cold (below or above freezing temperature)-responsive genes were identified, suggesting that a multitude of transcriptional cascades function in cold perception. Analyses of multiple cold-responsive genes, transcription factors, and some key transduction components involved in ABA and calcium signaling revealed their potential function in low temperature responses in P. euphratica. CONCLUSIONS: Our results provide a global transcriptome picture of P. euphratica under low temperature stress. The potential cold stress related transcripts identified in this study provide valuable information for further understanding the molecular mechanisms of low temperature perception in P. euphratica.

Project description:Heterophylly, or leaf morphological changes along plant shoot axes, is an important indicator of plant eco-adaptation to heterogeneous microenvironments. Despite extensive studies on the genetic control of leaf shape, the genetic architecture of heterophylly remains elusive. To identify genes related to heterophylly and their associations with plant saline tolerance, we conducted a leaf shape mapping experiment using leaves from a natural population of Populus euphratica. We included 106 genotypes grown under salt stress and salt-free (control) conditions using clonal seedling replicates. We developed a shape tracking method to monitor and analyze the leaf shape using principal component (PC) analysis. PC1 explained 42.18% of the shape variation, indicating that shape variation is mainly determined by the leaf length. Using leaf length along shoot axes as a dynamic trait, we implemented a functional mapping-assisted genome-wide association study (GWAS) for heterophylly. We identified 171 and 134 significant quantitative trait loci (QTLs) in control and stressed plants, respectively, which were annotated as candidate genes for stress resistance, auxin, shape, and disease resistance. Functions of the stress resistance genes ABSCISIC ACIS-INSENSITIVE 5-like (ABI5), WRKY72, and MAPK3 were found to be related to many tolerance responses. The detection of AUXIN RESPONSE FACTOR17-LIKE (ARF17) suggests a balance between auxin-regulated leaf growth and stress resistance within the genome, which led to the development of heterophylly via evolution. Differentially expressed genes between control and stressed plants included several factors with similar functions affecting stress-mediated heterophylly, such as the stress-related genes ABC transporter C family member 2 (ABCC2) and ABC transporter F family member (ABCF), and the stomata-regulating and reactive oxygen species (ROS) signaling gene RESPIRATORY BURST OXIDASE HOMOLOG (RBOH). A comparison of the genetic architecture of control and salt-stressed plants revealed a potential link between heterophylly and saline tolerance in P. euphratica, which will provide new avenues for research on saline resistance-related genetic mechanisms.

Project description:Grassland degradation has become a serious problem in some areas, making it necessary to quantitatively evaluate this process and its related factors. The study area was the arid windy sandy area in eastern Ningxia. The purpose of this study was to explore how soil properties and quality change during the process of grassland degradation in arid windy sandy areas. We looked at undegraded, lightly degraded, moderately degraded, and severely degraded desert steppe to study the physical, chemical, and biological changes at 0-5 cm, 5-15 cm, and 15-30 cm soil depths at different degradation degrees. We also analyzed the correlations across soil factors, established the minimum data set, and used the soil quality index (SQI) to evaluate the soil quality of grassland at different degradation degrees. The results showed that with grassland degradation, the soil bulk density increased; the soil clay, moisture, organic matter, total nitrogen, and available potassium content decreased; and the number of soil bacteria, actinomycetes, and fungi, as well as the activity of urease, polyphenol oxidase, protease, phosphatase, and sucrase, decreased. As soil depth increased, soil bulk density increased; the soil moisture, organic matter, available potassium, and available phosphorus content decreased; and soil microorganisms accumulated in the upper soil of undegraded, lightly, and moderately degraded grassland. There was also a positive correlation among the soil clay content, moisture content, organic matter content, total nitrogen content, available potassium content, microorganism quantity, and enzyme activity, while soil bulk density was negatively correlated with the above factors. The minimum data set for the soil quality evaluation of the degraded desert steppe was comprised of soil organic matter content, soil total nitrogen content, soil available phosphorus content, and phosphatase activity. Based on the minimum data set, we calculated the SQI of the grassland at different degradation degrees and found that the ranking based on overall soil quality was undegraded >lightly degraded >moderately degraded >severely degraded grassland. The results showed that the degradation of desert steppe in arid windy sandy areas had relatively consistent effects on the physical, chemical, and biological traits of the soil. The minimum data set can be used to replace the total data set when evaluating the soil quality of the desert steppe at different degrees of degradation.
