Correlation of NGX6 expression with clinicopathologic features and prognosis in colon cancer.

ABSTRACT: OBJECTIVE:The aim was to explore the correlation of NGX6 expression with clinicopathological features and prognosis in colon cancer. METHODS:Clinicopathological feature of 145 patients with colon cancer were analyzed.NGX6 expression was measured using immunohistochemistry methods. The correlation of NGX6 expression with clinicopathological features and prognosis were assessed. RESULTS:Among 145 cases of colon cancer, NGX6 positive expression were found in 76 (52.4%) cases and NGX6 negative expression were found in 69 (47.6%) cases. The expression of NGX6 was closely associated with size tumor, lymph node metastasis and TNM stage (P=0.002, 0.012, and 0.039, respectively). Kaplan-Meier analysis showed that NGX6 negative expression was associated with shorter disease-free survival (DFS) (P=0.029) and overall survival (OS) (P=0.015). Multivariate survival analysis demonstrated that NGX6 expression was the important independent prognostic factor for colon cancer (P=0.022). CONCLUSION:NGX6 is involved in the invasion and metastasis activity of colon cancer. NGX6 could may be applied as a novel and promising prognostic marker for colon cancer.

Project description:CD155, an immunoglobulin-like adhesion molecule, plays an important role in carcinoma such as cells migration, proliferation, metastasis, and tumor immune. The upregulation of CD155 has been found in several human malignancies, but its expression in cholangiocarcinoma (CCA) still remains unclear. The aim of this study is to investigate CD155 expression and its correlations with clinicopathologic data, angiogenesis, and prognosis in the patients with CCA.CD155 expression was investigated in 20 paired CCA tissues and corresponding paracancerous tissues by Western blotting and quantitative real-time polymerase chain reaction assays at protein and mRNA levels. Besides, this study evaluated the correlation between the tumor CD155 expression and the level of both vascular endothelial growth factor and intratumoral microvessel density by immunohistochemistry in 90 cases of CCA. Moreover, the clinical and prognostic significance of CD155 in CCA was assessed by immunohistochemistry.The protein and mRNA levels of CD155 were higher in CCA tumor tissues compared with corresponding paracancerous tissues (P<0.05). Immunohistochemical staining showed that CD155 was located in the cytoplasm of carcinoma cells and overexpressed in 61.2% (55/90) CCA tissues. Obviously, CD155 expression level was significantly correlated with tumor histological grade (P=0.002), lymph node metastasis (P<0.001), and tumor-node-metastasis (P=0.03). Additionally, Spearman rank correlation test demonstrated that CD155 expression was positively associated with vascular endothelial growth factor (r=0.481, P<0.001) and microvessel density (r=0.442, P<0.001) in CCA tissues. More importantly, CCA patients with high CD155 expression had a markedly shorter overall survival (P<0.001) and disease-free survival (P<0.001) after surgical resection, and multivariate analysis showed that high CD155 expression was an independent poor prognostic predictor of overall survival and disease-free survival (P<0.001).Our results revealed that upregulated CD155 correlated with aggressive clinicopathologic characteristics, angiogenesis, and poor prognosis in CCA and may be a promising prognostic biomarker for the CCA patients.

Project description:Colorectal cancer (CRC) is the third most commonly diagnosed malignancy and the second leading cause of cancer-related deaths in the world. It has been the most prevalent malignancy in Taiwan for consecutive thirteen years. Despite the diversity of its etiologic and pathophysiologic factors, a biological process named as epithelial-mesenchymal transition (EMT) is indispensable in the progression of epithelial cancer. Our aim is to investigate the correlation between the expression of 8 EMT-related proteins (E-cadherin, β-catenin, claudin-1, CD44, N-cadherin, fibronectin, vimentin, S100A4) and the clinicopathologic features of CRC in Taiwan, along with the DNA CpG epigenetic status of CD44 gene. In immunohistochemical assessment, decreased expression of E-cadherin is statistically associated with the progression of cancer stage, while decreased expression of claudin-1 as well as increased β-catenin nuclear translocation and N-cadherin expression is statistically associated with the progression of histopathologic grade. E-cadherin, nuclear β-catenin and claudin-1 are also associated with other important prognostic factors, including nodal metastasis, tumor deposits, and elevated serum CA 19-9 levels. In addition, the left-sided colon and rectal cancers show increased nuclear translocation of β-catenin compared to the right-sided colon cancers, while the rectal cancers show increased fibronectin expression compared to the right-sided and left-sided colon cancers. Moreover, vimentin is aberrantly expressed in one case of signet-ring cell carcinoma. The DNA methylation levels of CD44 gene promoter between the tumoral and non-tumorous tissues by NGS comparison showed statistical difference on six CpG sites. However, such difference may not be sufficient because these DNA methylation proportions are too low to inactivate CD44 gene. Our results demonstrate the expression of E-cadherin, claudin-1, and nuclear β-catenin is closely related to the clinicopathologic prognostic determinants of CRC in Taiwan. The DNA methylation level of CD44 gene and its protein expression, however, show no correlation with the clinicopathologic features in CRC.
