Easy access to heterobimetallic complexes for medical imaging applications via microwave-enhanced cycloaddition.

ABSTRACT: The Cu(I)-catalysed Huisgen cycloaddition, known as "click" reaction, has been applied to the synthesis of a range of triazole-linked porphyrin/corrole to DOTA/NOTA derivatives. Microwave irradiation significantly accelerates the reaction. The synthesis of heterobimetallic complexes was easily achieved in up to 60% isolated yield. Heterobimetallic complexes were easily prepared as potential MRI/PET (SPECT) bimodal contrast agents incorporating one metal (Mn, Gd) for the enhancement of contrast for MRI applications and one "cold" metal (Cu, Ga, In) for future radionuclear imaging applications. Preliminary relaxivity measurements showed that the reported complexes are promising contrast agents (CA) in MRI.

Enhanced Fe-Centered Redox Flexibility in Fe-Ti Heterobimetallic Complexes.

Project description:Previously, we reported the synthesis of Ti[N( o-(NCH2P( iPr)2)C6H4)3] and the Fe-Ti complex, FeTi[N( o-(NCH2P( iPr)2)C6H4)3], abbreviated as TiL (1), and FeTiL (2), respectively. Herein, we describe the synthesis and characterization of the complete redox families of the monometallic Ti and Fe-Ti compounds. Cyclic voltammetry studies on FeTiL reveal both reduction and oxidation processes at -2.16 and -1.36 V (versus Fc/Fc+), respectively. Two isostructural redox members, [FeTiL]+ and [FeTiL]- (2ox and 2red, respectively) were synthesized and characterized, along with BrFeTiL (2-Br) and the monometallic [TiL]+ complex (1ox). The solid-state structures of the [FeTiL]+/0/- series feature short metal-metal bonds, ranging from 1.94-2.38 Å, which are all shorter than the sum of the Ti and Fe single-bond metallic radii (cf. 2.49 Å). To elucidate the bonding and electronic structures, the complexes were characterized with a host of spectroscopic methods, including NMR, EPR, and 57Fe Mössbauer, as well as Ti and Fe K-edge X-ray absorption spectroscopy (XAS). These studies, along with hybrid density functional theory (DFT) and time-dependent DFT calculations, suggest that the redox processes in the isostructural [FeTiL]+,0,- series are primarily Fe-based and that the polarized Fe-Ti π-bonds play a role in delocalizing some of the additional electron density from Fe to Ti (net 13%).

An Easy Method of Synthesis CoxOy@C Composite with Enhanced Microwave Absorption Performance.

Project description:Design of interface-controllable magnetic composite towards the wideband microwave absorber is greatly significance, however, it still remains challenging. Herein, we designed a spherical-like hybrids, using the Co3O4 and amorphous carbon as the core and shell, respectively. Then, the existed Co3O4 core could be totally reduced by the carbon shell, thus in CoxOy core (composed by Co and Co3O4). Of particular note, the ratios of Co and Co3O4 can be linearly tuned, suggesting the controlled interfaces, which greatly influences the interface loss behavior and electromagnetic absorption performance. The results revealed that the minimum reflection loss value (RLmin) of -39.4 dB could be achieved for the optimal CoxOy@C sample under a thin thickness of 1.4 mm. More importantly, the frequency region with RL < -10 dB was estimated to be 4.3 GHz, ranging from 13.7 to 18.0 GHz. The superior wideband microwave absorption performance was primarily attributed to the multiple interfacial polarization and matched impedance matching ability.

Realization of Bidirectional, Bandwidth-Enhanced Metamaterial Absorber for Microwave Applications.

Project description:The ever-increasing interest towards metamaterial absorbers owes to its remarkable features such as ultra-thin nature and design flexibility. Subduing the inherent narrow bandwidth of such absorbers is the prime goal in metamaterial absorber research, as this can widen the applications areas. A greater challenge is to construct bidirectional absorber, which provides direction-insensitive absorption, as most of the existing designs exhibit single sided absorption due to the complete metal film used in the design. This work presents the realization of a bidirectional, bandwidth-enhanced metamaterial absorber with basic elements such as strips and squares optimized to have adjacent resonances leading to a bandwidth-enhanced absorption. The structural evolution of the constituent metallic components towards the formation of bandwidth-enhanced absorption is described. The bidirectional absorber exhibits more than 90% absorption between 13.40 GHz and 14.25 GHz from the two incident directions. The mechanism of absorption is studied with the surface current analysis and the effective parameters of the structure. The choice of the metallic components with four-fold rotation symmetry renders the proposed design to be polarization independent and wide-angle receptive. The numerical studies are verified experimentally at microwave frequencies, which shows a good agreement between them.

MetalDock: An Open Access Docking Tool for Easy and Reproducible Docking of Metal Complexes.

Project description:Despite the proven potential of metal complexes as therapeutics, the lack of computational tools available for the high-throughput screening of their interactions with proteins is a limiting factor toward clinical developments. To address this challenge, we introduce MetalDock, an easy-to-use, open access docking software for docking metal complexes to proteins. Our tool integrates the AutoDock docking engine with three well-known quantum software packages to automate the docking of metal-organic complexes to proteins. We used a Monte Carlo sampling scheme to obtain the missing Lennard-Jones parameters for 12 metal atom types and demonstrated that these parameters generalize exceptionally well. Our results show that the poses obtained by MetalDock are highly accurate, as they predict the binding geometries experimentally determined by crystal structures with high spatial reproducibility. Three different case studies are presented that demonstrate the versatility of MetalDock for the docking of diverse metal-organic compounds to different biomacromolecules, including nucleic acids.

Access control based on attribute certificates for medical intranet applications.

Project description:BackgroundClinical information systems frequently use intranet and Internet technologies. However these technologies have emphasized sharing and not security, despite the sensitive and private nature of much health information. Digital certificates (electronic documents which recognize an entity or its attributes) can be used to control access in clinical intranet applications.ObjectivesTo outline the need for access control in distributed clinical database systems, to describe the use of digital certificates and security policies, and to propose the architecture for a system using digital certificates, cryptography and security policy to control access to clinical intranet applications.MethodsWe have previously developed a security policy, DIMEDAC (Distributed Medical Database Access Control), which is compatible with emerging public key and privilege management infrastructure. In our implementation approach we propose the use of digital certificates, to be used in conjunction with DIMEDAC.ResultsOur proposed access control system consists of two phases: the ways users gain their security credentials; and how these credentials are used to access medical data. Three types of digital certificates are used: identity certificates for authentication; attribute certificates for authorization; and access-rule certificates for propagation of access control policy. Once a user is identified and authenticated, subsequent access decisions are based on a combination of identity and attribute certificates, with access-rule certificates providing the policy framework.ConclusionsAccess control in clinical intranet applications can be successfully and securely managed through the use of digital certificates and the DIMEDAC security policy.

A General Pathway to Heterobimetallic Triple-Decker Complexes.

Project description:A systematic study on the reactivity of the triple-decker complex [(Cp'''Co)2 (μ,η4 :η4 -C7 H8 )] (A) (Cp'''=1,2,4-tritertbutyl-cyclopentadienyl) towards sandwich complexes containing cyclo-P3 , cyclo-P4 , and cyclo-P5 ligands under mild conditions is presented. The heterobimetallic triple-decker sandwich complexes [(Cp*Fe)(Cp'''Co)(μ,η5 :η4 -P5 )] (1) and [(Cp'''Co)(Cp'''Ni)(μ,η3 :η3 -P3 )] (3) (Cp*=1,2,3,4,5-pentamethylcyclopentadienyl) were synthesized and fully characterized. In solution, these complexes exhibit a unique fluxional behavior, which was investigated by variable temperature NMR spectroscopy. The dynamic processes can be blocked by coordination to {W(CO)5 } fragments, leading to the complexes [(Cp*Fe)(Cp'''Co)(μ3 ,η5 :η4 :η1 -P5 ){W(CO)5 }] (2 a), [(Cp*Fe)(Cp'''Co)(μ4 ,η5 :η4 :η1 :η1 -P5 ){(W(CO)5 )2 }] (2 b), and [(Cp'''Co)(Cp'''Ni)(μ3 ,η3 :η2 :η1 -P3 ){W(CO)5 }] (4), respectively. The thermolysis of 3 leads to the tetrahedrane complex [(Cp'''Ni)2 (μ,η2 :η2 -P2 )] (5). All compounds were fully characterized using single-crystal X-ray structure analysis, NMR spectroscopy, mass spectrometry, and elemental analysis.

Project description:[2 + 3] cycloaddition reactions of fluorinated alkynes with 2-formylphenylboronic acids under the influence of Co(acac)2·2H2O in two-component solvents of acetonitrile/2-propanol at reflux temperature for 18 h took place smoothly, affording the corresponding fluoroalkylated indenol derivatives in good yields. This reaction shows excellent regioselectivity, giving 2-fluoroalkylated indenols, together with a very small amount of 3-fluoroalkylated indanones as side products.

Easy Access to Polyfunctionalized Chiral Decalins.

Project description:An unexpected ring expansion that converts hydrindanes into decalins via an unprecedented dyotropic reaction involving a mesylate group has been observed, and this paved the way for easy access to polyfunctionalized chiral decalins. These polyfunctionalized chiral decalins can be very useful building blocks for the synthesis of the thia analogues of many natural compounds. They can also be used in asymmetric catalysis and also in the synthesis of the new analogues of vitamin D with a modified D ring and side chain. The use of chiral sulfoxide ligands for asymmetric catalysis or asymmetric sulfur ylide-mediated epoxidation of carbonyl compounds is a very important topic in the field of organic chemistry, hence our results could be useful to the scientific community.

Nanogroove-Enhanced Hydrogel Scaffolds for 3D Neuronal Cell Culture: An Easy Access Brain-on-Chip Model.

Project description:In order to better understand the brain and brain diseases, in vitro human brain models need to include not only a chemically and physically relevant microenvironment, but also structural network complexity. This complexity reflects the hierarchical architecture in brain tissue. Here, a method has been developed that adds complexity to a 3D cell culture by means of nanogrooved substrates. SH-SY5Y cells were grown on these nanogrooved substrates and covered with Matrigel, a hydrogel. To quantitatively analyze network behavior in 2D neuronal cell cultures, we previously developed an automated image-based screening method. We first investigated if this method was applicable to 3D primary rat brain cortical (CTX) cell cultures. Since the method was successfully applied to these pilot data, a proof of principle in a reductionist human brain cell model was attempted, using the SH-SY5Y cell line. The results showed that these cells also create an aligned network in the 3D microenvironment by maintaining a certain degree of guidance by the nanogrooved topography in the z-direction. These results indicate that nanogrooves enhance the structural complexity of 3D neuronal cell cultures for both CTX and human SH-SY5Y cultures, providing a basis for further development of an easy access brain-on-chip model.

A [CoSiH<sub>2</sub>] Silylene Synthon Provides Modular Access to Homo- and Heterobimetallic [Co═Si═M] (M = Co, Fe) Silicide Complexes.

Project description:Base-stabilized [BP3iPr](H)2CoSiH2(DMAP) (1, [BP3iPr] = PhB(CH2PiPr2)3-; DMAP = 4-dimethylaminopyridine) is a rare instance of a synthon for the simplest "parent" silylene complex (LM═SiH2). Complex 1 was accessed in high yields via double Si-H bond activation in SiH4 by [BP3iPr]Co(DMAP), and in solution, it undergoes rapid exchange between bound and free DMAP by an associative mechanism (as determined by variable-temperature 1H NMR dynamic studies). The DMAP ligand of 1 is readily displaced by metal-based fragments that bind silicon and cleave the Si-H bonds of the SiH2 moiety to produce bimetallic [Co═Si═M] (M = Co, Fe) molecular silicides. Thus, treatment of 1 with 0.5 equiv of (LCoI)2(μ-N2) (L = a tripodal ligand) resulted in the spontaneous formation of [BP3iPr](H)2Co═Si═Co(H)2L (L = [BP2tBuPz], PhB(CH2PtBu2)2(pyrazolyl)- (3); Tp″, HB(3,5-diisopropylpyrazolyl)3- (4)) with the concomitant release of DMAP. The symmetrical silicide [BP3iPr](H)2Co═Si═Co(H)2[BP3iPr] (5) was prepared by treatment of a mixture of 1 and [BP3iPr]Co(DMAP) with 2 equiv of Ph3B, which in this case is required to sequester DMAP as the elimination product Ph3B-DMAP. A heterobimetallic silicide, [BP3iPr](H)2Co═Si═Fe(H)2[SiP3iPr] (7; [SiP3iPr] = PhSi(CH2PiPr2)3), was obtained via in situ KC8 reduction of [SiP3iPr]FeCl and subsequent addition of 1 and Ph3B. These transformations involving a metal-SiH2 derivative demonstrate a fundamentally new type of reactivity for silylene complexes and provide a unique synthetic method for construction of molecular silicide complexes.
