3D Micropatterned Surface Inspired by Salvinia molesta via Direct Laser Lithography.

ABSTRACT: Biomimetic functional surfaces are attracting increasing attention for their relevant technological applications. Despite these efforts, inherent limitations of microfabrication techniques prevent the replication of complex hierarchical microstructures. Using a 3D laser lithography technique, we fabricated a 3D patterned surface bioinspired to Salvinia molesta leaves. The artificial hairs, with crownlike heads, were reproduced by scaling down (ca. 100 times smaller) the dimensions of natural features, so that microscale hairs with submicrometric resolution were attained. The micropatterned surface, in analogy with the natural model, shows interesting properties in terms of hydrophobicity and air retention when submerged by water, even if realized with a hydrophilic material. Furthermore, we successfully demonstrated the capability to promote localized condensation of water droplets from moisture in the atmosphere.

Air Trapping Mechanism in Artificial Salvinia-Like Micro-Hairs Fabricated via Direct Laser Lithography.

Project description:Salvinia leaves represent an extraordinary example of how nature found a strategy for the long term retainment of air, and thus oxygen, on a surface, the so-called 'Salvinia effect', thanks to the peculiar three-dimensional and hierarchical shape of the hairs covering the leaves. Here, starting from the natural model, we have microfabricated hairs inspired by those present on the Salvinia molesta leaves, by means of direct laser lithography. Artificial hairs, like their natural counterpart, are composed of a stalk and a crown-like head, and have been reproduced in the microscale since this ensures, if using a proper design, an air-retaining behavior even if the bulk structural material is hydrophilic. We have investigated the capability of air retainment inside the heads of the hairs that can last up to 100 h, demonstrating the stability of the phenomenon. For a given dimension of the head, the greater the number of filaments, the greater the amount of air that can be trapped inside the heads since the increase in the number of solid⁻air interfaces able to pin the liquid phase. For this reason, such type of pattern could be used for the fabrication of surfaces for controlled gas retainment and gas release in liquid phases. The range of applications would be quite large, including industrial, medical, and biological fields.

Endocide-Induced Abnormal Growth Forms of Invasive Giant Salvinia (Salvinia molesta).

Project description:Giant salvinia (Salvinia molesta) is one of the most noxious invasive species in the world. The fern is known to have primary, secondary, and tertiary growth forms, which are also commonly hypothesized as growth stages. The identification of these forms is primarily based on the size and folding status of the floating leaves. However, we identified 12 forms in the greenhouse and the field. Our experiments showed that the folding of floating leaves is a reversible trait dependent on water access. The floating leaves quickly fold in response to water shortage, reducing water loss and needs, decreasing growth, and avoiding trichome damage. The leaves re-open to allow trichomes repel water and enhance growth when having adequate water supply. Larger secondary or tertiary forms do not produce small-leaf primary forms without high intensity stress. These results do not support the hypothesis that three growth forms represent sequential growth stages. The abnormal small-leaf forms are the result of endocide-induced autotoxicity and some of them never grow into other forms. The development of abnormal forms and reversible leaf folding strategy in response to high stress along with rapid asexual reproduction are major adaptive traits contributing to the invasiveness of S. molesta.

Engineering bacterial vortex lattice via direct laser lithography.

Project description:A suspension of swimming bacteria is possibly the simplest realization of active matter, i.e. a class of systems transducing stored energy into mechanical motion. Collective swimming of hydrodynamically interacting bacteria resembles turbulent flow. This seemingly chaotic motion can be rectified by a geometrical confinement. Here we report on self-organization of a concentrated suspension of motile bacteria Bacillus subtilis constrained by two-dimensional (2D) periodic arrays of microscopic vertical pillars. We show that bacteria self-organize into a lattice of hydrodynamically bound vortices with a long-range antiferromagnetic order controlled by the pillars' spacing. The patterns attain their highest stability and nearly perfect order for the pillar spacing comparable with an intrinsic vortex size of an unconstrained bacterial turbulence. We demonstrate that the emergent antiferromagnetic order can be further manipulated and turned into a ferromagnetic state by introducing chiral pillars. This strategy can be used to control a wide class of active 2D systems.

Fabrication of Multifaceted Micropatterned Surfaces with Laser Scanning Lithography.

Project description:The implementation of engineered surfaces presenting micrometer-sized patterns of cell adhesive ligands against a biologically inert background has led to numerous discoveries in fundamental cell biology. While existing surface patterning strategies allow for pattering of a single ligand it is still challenging to fabricate surfaces displaying multiple patterned ligands. To address this issue we implemented Laser Scanning Lithography (LSL), a laser-based thermal desorption technique, to fabricate multifaceted, micropatterned surfaces that display independent arrays of subcellular-sized patterns of multiple adhesive ligands with each ligand confined to its own array. We demonstrate that LSL is a highly versatile "maskless" surface patterning strategy that provides the ability to create patterns with features ranging from 450 nm to 100 μm, topography ranging from -1 to 17 nm, and to fabricate both stepwise and smooth ligand surface density gradients. As validation for their use in cell studies, surfaces presenting orthogonally interwoven arrays of 1×8 μm elliptical patterns of Gly-Arg-Gly-Asp-terminated alkanethiol self-assembled monolayers and human plasma fibronectin are produced. Human umbilical vein endothelial cells cultured on these multifaceted surfaces form adhesion sites to both ligands simultaneously and utilize both ligands for lamella formation during migration. The ability to create multifaceted, patterned surfaces with tight control over pattern size, spacing, and topography provides a platform to simultaneously investigate the complex interactions of extracellular matrix geometry, biochemistry, and topography on cell adhesion and downstream cell behavior.

A Reflectivity Enhanced 3D Optical Storage Nanostructure Application Based on Direct Laser Writing Lithography.

Project description:To enable high-density optical storage, better storage media structures, diversified recording methods, and improved accuracy of readout schemes should be considered. In this study, we propose a novel three-dimensional (3D) sloppy nanostructure as the optical storage device, and this nanostructure can be fabricated using the 3D laser direct writing technology. It is a 900 nm high, 1 × 2 µm wide Si slope on a 200 nm SiO2 layer with 200 nm Si3N4 deposited on top to enhance reflectivity. In this study, we propose a reflected spectrum-based method as the readout recording strategy to stabilize information readout more stable. The corresponding reflected spectrum varied when the side wall angle of the slope and the azimuth angle of the nanostructure were tuned. In addition, an artificial neural network was applied to readout the stored information from the reflected spectrum. To simulate the realistic fabrication error and measurement error, a 20% noise level was added to the study. Our findings showed that the readout accuracy was 99.86% for all 120 data sequences when the slope and azimuth angle were varied. We investigated the possibility of a higher storage density to fully demonstrate the storage superiority of this designed structure. Our findings also showed that the readout accuracy can reach its highest level at 97.25% when the storage step of the encoded structure becomes 7.5 times smaller. The study provides the possibility to further explore different nanostructures to achieve high-density optical storage.

Osteochondral Repair and Electromechanical Evaluation of Custom 3D Scaffold Microstructured by Direct Laser Writing Lithography.

Project description:ObjectiveThe objective of this study was to assess a novel 3D microstructured scaffold seeded with allogeneic chondrocytes (cells) in a rabbit osteochondral defect model.DesignDirect laser writing lithography in pre-polymers was employed to fabricate custom silicon-zirconium containing hybrid organic-inorganic (HOI) polymer SZ2080 scaffolds of a predefined morphology. Hexagon-pored HOI scaffolds were seeded with chondrocytes (cells), and tissue-engineered cartilage biocompatibility, potency, efficacy, and shelf-life in vitro was assessed by morphological, ELISA (enzyme-linked immunosorbent assay) and PCR (polymerase chain reaction) analysis. Osteochondral defect was created in the weight-bearing area of medial femoral condyle for in vivo study. Polymerized fibrin was added to every defect of 5 experimental groups. Cartilage repair was analyzed after 6 months using macroscopical (Oswestry Arthroscopy Score [OAS]), histological, and electromechanical quantitative potential (QP) scores. Collagen scaffold (CS) was used as a positive comparator for in vitro and in vivo studies.ResultsType II collagen gene upregulation and protein secretion was maintained up to 8 days in seeded HOI. In vivo analysis revealed improvement in all scaffold treatment groups. For the first time, electromechanical properties of a cellular-based scaffold were analyzed in a preclinical study. Cell addition did not enhance OAS but improved histological and QP scores in HOI groups.ConclusionsHOI material is biocompatible for up to 8 days in vitro and is supportive of cartilage formation at 6 months in vivo. Electromechanical measurement offers a reliable quality assessment of repaired cartilage.

3D printing of nanowrinkled architectures via laser direct assembly.

Project description:Structural wrinkles in nature have been widely imitated to enhance the surface functionalities of objects, especially three-dimensional (3D) architectured wrinkles, holding promise for emerging applications in mechanical, electrical, and biological processes. However, the fabrication of user-defined 3D nanowrinkled architectures is a long-pending challenge. Here, we propose a bottom-up laser direct assembly strategy to fabricate multidimensional nanowrinkled architectures in a single-material one-step process. Through the introduction of laser-induced thermal transition into a 3D nanoprinting process for leading the point-by-point nanoscale wrinkling and the self-organization of wrinkle structures, we have demonstrated the program-controlled and on-demand fabrication of multidimensional nanowrinkled structures. Moreover, the precise control of wrinkle morphology with an optimal wavelength of 40 nanometers and the regulation of the dynamic transformation of wrinkled cellular microstructures via interfacial stress mismatch engineering have been achieved. This study provides a universal protocol for constructing nearly arbitrary nanowrinkled architectures and facilitates a new paradigm in nanostructure manufacturing.

Bio-Inspired Micropatterned Platforms Recapitulate 3D Physiological Morphologies of Bone and Dentinal Cells.

Project description:Cells exhibit distinct 3D morphologies in vivo, and recapitulation of physiological cell morphologies in vitro is pivotal not only to elucidate many fundamental biological questions, but also to develop new approaches for tissue regeneration and drug screening. However, conventional cell culture methods in either a 2D petri dish or a 3D scaffold often lead to the loss of the physiological morphologies for many cells, such as bone cells (osteocytes) and dentinal cells (odontoblasts). Herein, a unique approach in developing a 3D extracellular matrix (ECM)-like micropatterned synthetic matrix as a physiologically relevant 3D platform is reported to recapitulate the morphologies of osteocytes and odontoblasts in vitro. The bio-inspired micropatterned matrix precisely mimics the hierarchic 3D nanofibrous tubular/canaliculi architecture as well as the compositions of the ECM of mineralized tissues, and is capable of controlling one single cell in a microisland of the matrix. Using this bio-inspired 3D platform, individual bone and dental stem cells are successfully manipulated to recapitulate the physiological morphologies of osteocytes and odontoblasts in vitro, respectively. This work provides an excellent platform for an in-depth understanding of cell-matrix interactions in 3D environments, paving the way for designing next-generation biomaterials for tissue regeneration.

Optically Clear and Resilient Free-Form µ-Optics 3D-Printed via Ultrafast Laser Lithography.

Project description:We introduce optically clear and resilient free-form micro-optical components of pure (non-photosensitized) organic-inorganic SZ2080 material made by femtosecond 3D laser lithography (3DLL). This is advantageous for rapid printing of 3D micro-/nano-optics, including their integration directly onto optical fibers. A systematic study of the fabrication peculiarities and quality of resultant structures is performed. Comparison of microlens resiliency to continuous wave (CW) and femtosecond pulsed exposure is determined. Experimental results prove that pure SZ2080 is ∼20 fold more resistant to high irradiance as compared with standard lithographic material (SU8) and can sustain up to 1.91 GW/cm² intensity. 3DLL is a promising manufacturing approach for high-intensity micro-optics for emerging ﬁelds in astro-photonics and atto-second pulse generation. Additionally, pyrolysis is employed to homogeneously shrink structures up to 40% by removing organic SZ2080 constituents. This opens a promising route towards downscaling photonic lattices and the creation of mechanically robust glass-ceramic microstructures.

STED-Inspired Cationic Photoinhibition Lithography.

Project description:Direct laser writing by two-photon lithography has been enhanced substantially during the past two decades by techniques borrowed from stimulated emission depletion (STED) microscopy. However, STED-inspired lithography was so far limited to radical polymerizations, mostly to acrylates and methacrylates. Cationic polymers did not derive benefits from this technique. Specifically, epoxide polymerization, which plays a paramount role in semiconductor clean-room technology, has not yet been reported with a second, depleting laser focus in the outer rim of the point spread function. We now found that using a thioxanthone as a sensitizer and sulfonium or iodonium salts as photoinitiators enables at least partial optical on/off switching of two-photon polymerization and, in the case of the sulfonium salt, allows for writing epoxy lines with widths shrunk by approx. two-thirds compared to lines written with two-photon polymerization alone.
