ABSTRACT: South East Asia pest thrips species, Thrips parvispinus (Karny), is a serious pest on a number of agricultural and horticultural crops in a number of plant families. Based on an integrated approach of morphology and DNA barcoding, invasion of this serious pest is reported first time from India on papaya plantations. Molecular data have corroborated with the morphological identification. Haplotyping data suggested that the Indonesia may be a probable source of invasion of this pest to India.

Microbiome profiling of the onion thrips, Thrips tabaci Lindeman (Thysanoptera: Thripidae).

Project description:The gut microbial community structure of adult Thrips tabaci collected from 10 different agro-climatically diverse locations of India was characterized by using the Illumina MiSeq platform to amplify the V3 region of the 16S rRNA gene of bacteria present in the sampled insects. Analyses were performed to study the bacterial communities associated with Thrips tabaci in India. The complete bacterial metagenome of T. tabaci was comprised of 1662 OTUs of which 62.25% belong to known and 37.7% of unidentified/unknown bacteria. These OTUs constituted 21 bacterial phyla of 276 identified genera. Phylum Proteobacteria was predominant, followed by Actinobacteria, Firmicutes, Bacteroidetes and Cyanobacteria. Additionally, the occurrence of the reproductive endosymbiont, Wolbachia was detected at two locations (0.56%) of the total known OTUs. There is high variation in diversity and species richness among the different locations. Alpha-diversity metrics indicated the higher gut bacterial diversity at Bangalore and lowest at Rahuri whereas higher bacterial species richness at T. tabaci samples from Imphal and lowest at Jhalawar. Beta diversity analyses comparing bacterial communities between the samples showed distinct differences in bacterial community composition of T. tabaci samples from different locations. This paper also constitutes the first record of detailed bacterial communities associated with T. tabaci. The location-wise variation in microbial metagenome profile of T. tabaci suggests that bacterial diversity might be governed by its population genetic structure, environment and habitat.

Cypermethrin resistance and reproductive types in onion thrips, Thrips tabaci (Thysanoptera: Thripidae).

Project description:Cypermethrin resistance and reproductive types were examined for T. tabaci strains. Some arrhenotokous and thelytokous strains encoded the sodium channel mutation (T929I) involved in cypermethrin resistance. However, the resistance levels varied to some degree among the strains. A cytochrome P450 inhibitor, piperonyl butoxide, showed different synergistic effects on the strains examined. These results suggest that fundamental and additional levels of cypermethrin resistance in T. tabaci are conferred respectively by reduced sensitivity of the sodium channel and by cytochrome P450-mediated detoxification.

Detection and Localization of Wolbachia in Thrips palmi Karny (Thysanoptera: Thripidae).

Project description:Thrips palmi Karny is a globally distributed polyphagous agricultural pest. It causes huge economic loss by its biological behaviors like feeding, reproduction and transmission of tospoviruses. Since T. palmi shows close morphological similarities with other thrips species, we employed mitochondrial cytochrome oxidase 1 (mtCO1) gene as a molecular marker. BLAST analysis of this sequence helped us to identify the collected specimen as T. palmi. We observed the female to male ratio of about 3:1 from collected samples and suspected the presence of Wolbachia. The presence of Wolbachia was detected by PCR using genus specific primers of 16S rRNA gene. Further confirmation of Wolbachia strain was achieved by conducting PCR amplification of three ubiquitous genes ftsZ, gatB and groEL. A phylogenetic tree was constructed with concatenated sequences of ftsZ and gatB gene to assign supergroup to Wolbachia. Finally, we localized Wolbachia in abdominal region of the insect using fluorescent in situ hybridization with the help of confocal microscope. Our result confirmed the presence of Wolbachia supergroup B strain for the first time in T. palmi.

DNA Barcoding: A Reliable Method for the Identification of Thrips Species (Thysanoptera, Thripidae) Collected on Sticky Traps in Onion Fields.

Project description:Several thrips species (Insecta, Thysanoptera) are globally known as important crop pests and vectors of viral diseases, but their identification is difficult because of their small body size and inconspicuous morphological differences. Sequencing variation in the mitochondrial cytochrome c oxidase I (COI) region has been proven to be useful for the identification of species of many groups of insect pests. Here, DNA barcoding has been used to identify thrips species collected with the use of sticky traps placed in an open onion field. A total of 238 thrips specimens were analyzed, 151 of which could be identified to species and 27 to genera belonging to the family Thripidae. Fifty-one specimens could not be assigned to any genus, with the closest BLAST match in the GenBank queries being below 98%, whilst six specimens were not recognized as Thysanoptera. The results indicate that, although there are a few pest thrips species not yet barcoded, most of the species that may cause damage to crops in Europe are represented in GenBank and other databases, enabling correct identification. Additionally, DNA barcoding can be considered a valuable alternative to the classic morphology method for identification of major thrips species.

Onion Thrips (Thysanoptera: Thripidae) Feeding Promotes Infection By Pantoea ananatis in Onion.

Project description:Onion thrips, Thrips tabaci Lindeman, is a primary insect pest of onions (Allium cepa) worldwide. Onion thrips cause feeding damage by destroying epidermal tissue. They are also vectors of Pantoea ananatis (Serrano) Mergaert, the bacteria that causes center rot. Onions with center rot develop white streaks with water-soaked margins along the onion leaves, which turn necrotic and lead to bulb rot during storage. The role of thrips feeding on the establishment and progression of bacterial infection in onions has not been investigated. Onions infested with thrips and inoculated with P. ananatis had more necrotic tissue and symptoms were more severe with increasing thrips density. We conducted a fluorescence microscopy study that examined how P. ananatis (expressing a fluorescence protein gene) colonized a control group of onions without thrips in comparison to a test group of onions with thrips. We found that P. ananatis colonized some onions in the control group because of naturally existing wounds in the epidermal tissue but more colonization was found in the thrips infested group because of the increased presence of entry points caused by thrips feeding. Overall, our results demonstrate that wounds caused by thrips feeding facilitate center rot development by providing entry sites for the bacteria into leaf tissue.

Complete mitochondrial genome sequence for the <i>Thrips hawaiiensis</i> (Thysanoptera: Thripidae).

Project description:Thrips hawaiiensis (Morgan) (Thysanoptera: Thripidae) is a common Thysanoptera insect widely distributed in Asia and the Pacific, it damages various plants. In this study the complete mitochondrial genome of T. hawaiiensis was sequenced and characterized by using next-generation sequencing technique. The total length of the complete genome is 15,357 bp and A + T content of 77.8% (GeneBank accession No. MW582621). The T. hawaiiensis mitochondrial genome consists of 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA genes (tRNAs) and 2 non-coding control regions (Dloop region). According to previous studies, only a few complete mitochondrial genomes from Order Thysanoptera have been reported. Thus, T. hawaiiensis complete mitochondrial genome sequence reported will provide molecular information for mitochondrial genome research on Thysanoptera.

Repellency of Plant Extracts against the Legume Flower Thrips Megalurothrips sjostedti (Thysanoptera: Thripidae).

Project description:Megalurothrips sjostedti Trybom is an important pest of cowpea (Vigna unguiculata L.) in Africa. To propose an alternative to chemical control, the repellency of 24 plant extracts was evaluated against adult female thrips of M. sjostedti in the laboratory. Plant extracts in ethanol were separately applied on a filter paper disk in a still air visual cue olfactometer. The results showed highly significant differences in repellency among extract type, concentration and their interactions. We classified the level of repellency into four categories as strong, good, moderate and weak or non- repellent based on hierarchical ascendant classification. We identified Piper nigrum, Cinnamomum zeylanicum, Cinnamomum cassia as strong repellents. Five extracts were classified as good, eight as moderate and the remaining eight extracts were weak or non-repellent. Repellency of the extracts increased with the concentration suggesting that the behavioral response of M. sjostedti was dose-dependent. Mono- and sesquiterpene hydrocarbon compounds from seven highly repellent extracts were identified by gas chromatography-mass spectrometry (GC/MS). The use of repellent extracts could be useful in developing integrated pest management strategies for thrips on legume crops. In this regard, the specific modes of action of the identified compounds need to be investigated to incorporate them into the existing crop protection strategies.

Iris Yellow Spot Virus Prolongs the Adult Lifespan of Its Primary Vector, Onion Thrips (Thrips tabaci) (Thysanoptera: Thripidae).

Project description:Iris yellow spot virus (IYSV) from the genus Tospovirus, family Peribunyaviridae, reduces yield in several crops, especially Allium spp. IYSV is primarily transmitted by onion thrips (Thrips tabaci), but little is known about how IYSV impacts the biology of its principal vector. In a controlled experiment, the effect of IYSV on the lifespan and fecundity of onion thrips was examined. Larvae were reared on IYSV-infected onions until pupation. Individual pupae were confined until adults eclosed, and the lifespan and total progeny produced per adult were monitored daily. Thrips were tested for the virus in reverse-transcriptase polymerase chain reaction using specific primers to confirm the presence of IYSV. Results indicated that 114 and 35 out of 149 eclosing adults tested positive (viruliferous) and negative (nonviruliferous) for IYSV, respectively. The viruliferous adults lived 1.1-6.1 d longer (average of 3.6 d) than nonviruliferous adults. Fecundity of viruliferous and nonviruliferous onion thrips was similar with 2.0 ± 0.1 and 2.3 ± 0.3 offspring produced per female per day, respectively. Fecundity for both viruliferous and nonviruliferous thrips also was significantly positively correlated with lifespan. These findings suggest that the longer lifespan of viruliferous onion thrips adults may allow this primary vector of IYSV to infect more plants, thereby exacerbating IYSV epidemics.

Detection of Gene Flow from Sexual to Asexual Lineages in Thrips tabaci (Thysanoptera: Thripidae).

Project description:Populations of Thrips tabaci are known to have two sympatric but genetically isolated reproductive modes, arrhenotoky (sexual reproduction) and thelytoky (asexual reproduction). Herein, we report behavioral, ecological and genetic studies to determine whether there is gene flow between arrhenotokous and thelytokous T. tabaci. We did not detect significant preference by arrhenotokous males to mate with females of a particular reproductive mode, nor did we detect significant behavioral differences between arrhenotokous males mated with arrhenotokous or thelytokous females in their pre-copulation, copulation duration and mating frequency. Productive gene transfer resulting from the mating between the two modes was experimentally confirmed. Gene transfer from arrhenotokous T. tabaci to thelytokous T. tabaci was further validated by confirmation of the passage of the arrhenotokous male-originated nuclear gene (histone H3 gene) allele to the F2 generation. These behavioral, ecological and genetic studies confirmed gene transfer from the sexual arrhenotokous mode to the asexual thelytokous mode of T. tabaci in the laboratory. These results demonstrate that asexual T. tabaci populations may acquire genetic variability from sexual populations, which could offset the long-term disadvantage of asexual reproduction.

Effects of flower thrips (Thysanoptera: Thripidae) on nutritional quality of banana (Zingiberales: Musaceae) buds.

Project description:The abundance of banana flower thrips (Thrips hawaiiensis Morgan) in a banana (Musa acuminata Colla "Williams" cultivar) plantation was investigated using yellow sticky traps (29.70 cm × 21.00 cm) in 2015. Banana flower thrips occurred throughout the year with monthly variation, and the maximum occurrence was observed in October and November during the bud burst (73.80 ± 6.32 adults/trap) and young fruit (70.06 ± 5.69 adults/trap) periods. The damage rates were as follows: interior flowers >3rd-layer flowers > 2nd-layer flowers > 1st-layer flowers > young fruits. This result indicates that thrips migrated to lower bracts, young fruits, and other flower buds as bracts gradually opened. Results also showed that the reducing sugar, vitamin C, protein and ash contents in thrips-damaged flowers were all significantly lower than those in undamaged flowers, while there was no significant difference between damaged and undamaged young banana fruit. Our results indicated that the abundances of banana flower thrips were closely associated with the growing stage of banana. Thrips mainly infested flower buds and caused a reduction in nutrients for the host plant, especially the reducing sugar and vitamin C contents, which reduced the nutritional quality of banana fruits and the quality of flower bud by-products of banana.
