Isolation and characterization of nine microsatellite loci from Bemisia tabaci (Hemiptera: Aleyrodidae) biotype B.

ABSTRACT: Nine microsatellites were isolated from Bemisia tabaci (Gennadius) biotype B and screened across 60 individuals from two populations (biotype B) to examine polymorphism. Two to 12 alleles were observed per locus. Observed and expected heterozygosities ranged from 0.033 to 0.967 and 0.033 to 0.854, respectively. There was no significant deviation from Hardy-Weinberg equilibrium and no significant linkage disequilibrium between loci. One locus showed evidence for null alleles. These loci will be useful in future studies of the genetic structure of worldwide biotypes and gene flow analyses between and within biotypes of B. tabaci.

Characterization of a newly discovered symbiont of the whitefly Bemisia tabaci (Hemiptera: Aleyrodidae).

Project description:Bemisia tabaci (Hemiptera: Aleyrodidae) is a species complex containing >28 cryptic species, some of which are important crop pests worldwide. Like many other sap-sucking insects, whiteflies harbor an obligatory symbiont, "Candidatus Portiera aleyrodidarum," and a number of secondary symbionts. So far, six genera of secondary symbionts have been identified in B. tabaci. In this study, we report and describe the finding of an additional bacterium in the indigenous B. tabaci cryptic species China 1 (formerly known as B. tabaci biotype ZHJ3). Phylogenetic analysis based on the 16S rRNA and gltA genes showed that the bacterium belongs to the Alphaproteobacteria subdivision of the Proteobacteria and has a close relationship with human pathogens of the genus Orientia. Consequently, we temporarily named it Orientia-like organism (OLO). OLO was found in six of eight wild populations of B. tabaci China 1, with the infection rate ranging from 46.2% to 76.8%. Fluorescence in situ hybridization (FISH) of B. tabaci China 1 in nymphs and adults revealed that OLOs are confined to the bacteriome and co-occur with "Ca. Portiera aleyrodidarum." The vertical transmission of OLO was demonstrated by detection of OLO at the anterior pole end of the oocytes through FISH. Quantitative PCR analysis of population dynamics suggested a complex interaction between "Ca. Portiera aleyrodidarum" and OLO. Based on these results, we propose "Candidatus Hemipteriphilus asiaticus" for the classification of this symbiont from B. tabaci.

Isolation and identification of culturable bacteria from honeydew of whitefly, Bemisia tabaci (G.) (Hemiptera: Aleyrodidae).

Project description:Bemisia tabaci (G.) is an important pest and a vector of Gemini viruses infecting plants. During the process of feeding B. tabaci excretes honeydew which is rich in nutrients, and an excellent medium for microbial growth. Recent report proved that volatile emitted by the honeydew associated bacteria of aphid, Acyrthosiphon pisum Harris was involved in natural enemy calling. Thus understanding the honeydew associated bacteria is of paramount importance from the non-chemical method of insect pest management. In this perspective, very less information is available on bacteria associated with the honeydew excreted by B. tabaci. Therefore, in the present study we have isolated and characterized three culturable bacteria from the honeydew of B. tabaci viz. Bacillus endophyticus, Bacillus niacini and Roseomonas species by employing 16Sr DNA BLASTx analyses which revealed that both B. endophyticus and B. niacini had high similarity (> 99%) to the respective species, while Roseomonas sp. showed only 95% similarity to the existing Roseomonas sp. specificity of honeydew association of Roseomonas sp. was confirmed by developing specific primers as this genus is reported from immunocompromised persons and recently from ticks and mites. The present study also indicated the possible host-plant origin of these honeydew associated bacteria.

Host Plant Affects Symbiont Abundance in Bemisia tabaci (Hemiptera: Aleyrodidae).

Project description:Symbionts contribute nutrients that allow insects to feed on plants. The whitefly Bemisia tabaci Middle East-Asia Minor 1 (MEAM1) is a polyphagous pest that depends on symbionts to provide key nutrients that are deficient in the diet. Here, we established three whitefly populations on eggplants, cucumbers, and tomatoes and observed that they harbored the same symbiont taxa in different quantities. The amount of the primary symbiont, Portiera, decreased with increasing concentrations of host-plant essential amino acids (EAAs). Whitefly populations transferred to different plant species exhibited fluctuations in Portiera amounts in the first three or four generations; the amount of Portiera increased when whitefly populations were transferred to plant species with lower EAAs proportions. As for the secondary symbionts, the whitefly population of eggplants exhibited lower quantities of Hamiltonella and higher quantities of Rickettsia than the other two populations. The changes of both symbionts' abundance in whitefly populations after host-plant-shifting for one generation showed little correlation with the EAAs' proportions of host plants. These findings suggest that host-plant nitrogen nutrition, mainly in the form of EAAs, influences the abundance of symbionts, especially Portiera, to meet the nutritional demands of whiteflies. The results will inform efforts to control pests through manipulating symbionts in insect-symbiont associations.

Transcriptome analysis of host-associated differentiation in Bemisia tabaci (Hemiptera: Aleyrodidae).

Project description:Host-associated differentiation is one of the driving forces behind the diversification of phytophagous insects. In this study, host induced transcriptomic differences were investigated in the sweetpotato whitefly Bemisia tabaci, an invasive agricultural pest worldwide. Comparative transcriptomic analyses using coding sequence (CDS), 5' and 3' untranslated regions (UTR) showed that sequence divergences between the original host plant, cabbage, and the derived hosts, including cotton, cucumber and tomato, were 0.11-0.14%, 0.19-0.26%, and 0.15-0.21%, respectively. In comparison to the derived hosts, 418 female and 303 male transcripts, respectively, were up-regulated in the original cabbage strain. Among them, 17 transcripts were consistently up-regulated in both female and male whiteflies originated from the cabbage host. Specifically, two ESTs annotated as Cathepsin B or Cathepsin B-like genes were significantly up-regulated in the original cabbage strain, representing a transcriptomic response to the dietary challenges imposed by the host shifting. Results from our transcriptome analysis, in conjunction with previous reports documenting the minor changes in their reproductive capacity, insecticide susceptibility, symbiotic composition and feeding behavior, suggest that the impact of host-associated differentiation in whiteflies is limited. Furthermore, it is unlikely the major factor contributing to their rapid range expansion/invasiveness.

Population genetic structure of Bemisia tabaci MED (Hemiptera: Aleyrodidae) in Korea.

Project description:The sweet potato whitefly, Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) is a major agricultural pest that causes economic damages worldwide. In particular, B. tabaci MED (Mediterranean) has resulted in serious economic losses in tomato production of Korea. In this study, 1,145 B. tabaci MED females from 35 tomato greenhouses in different geographic regions were collected from 2016 to 2018 (17 populations in 2016, 13 in 2017, and five in 2018) and analyzed to investigate their population genetic structures using eight microsatellite markers. The average number of alleles per population (NA) ranged from 2.000 to 5.875, the expected heterozygosity (HE) ranged from 0.218 to 0.600, the observed heterozygosity (HO) ranged from 0.061 to 0.580, and the fixation index inbreeding coefficient (FIS) ranged from -0.391 to 0.872 over the three years of the study. Some significant correlation (p < 0.05) was present between genetic differentiations (FST) and geographical distance, and a comparatively high proportion of variation was found among the B. tabaci MED populations. The B. tabaci MED populations were divided into two well-differentiated genetic clusters within different geographic regions. Interestingly, its genetic structures converged into one genetic cluster during just one year. The reasons for this genetic change were speculated to arise from different fitness, insecticide resistance, and insect movement by human activities.

Bacterial Endosymbiont Diversity Among Bemisia tabaci (Hemiptera: Aleyrodidae) Populations in Florida.

Project description:The sweetpotato whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae), is a pest of many economically important agricultural crops and a vector of plant viruses. Bemisia tabaci harbors facultative endosymbiont species that have been implicated in pest status, including tolerance to insecticides, virus transmission efficiency and tolerance to high-temperatures. The facultative endosymbionts reported in B. tabaci include Arsenophonus, Hamiltonella, Wolbachia, Cardinium, Fritschea and Rickettsia. We collected whitefly populations from weed and crop hosts in south Florida and identified the whitefly species as well as the facultative endosymbionts present in these populations by molecular analysis. In addition, a phylogenetic analysis of whiteflies and their endosymbionts was performed. The only facultative endosymbionts found among the B. tabaci populations collected in Florida were Hamiltonella and Rickettsia. The phylogenetic analysis revealed the low genetic diversity of whiteflies and their endosymbionts. Additionally, the phylogenetic tree clustered Rickettsia from Florida in the R1 genetic group. The results will aid to understand the role of the bacterial endosymbionts in the whitefly host.

Aphid-induced Defences in Chilli Affect Preferences of the Whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae).

Project description:The sweetpotato whitefly (WF), Bemisia tabaci, is a major pest that damages a wide range of vegetable crops in Malaysia. WF infestation is influenced by a variety of factors, including previous infestation of the host plant by other insect pests. This study investigated the effects of previous infestation of host chilli plants by the green peach aphid (Myzus persicae) on the olfactory behavioural response of B. tabaci, using free-choice bioassay with a Y-tube olfactometer. We analysed volatile organic compounds (VOCs) emitted by non-infested and M. persicae-infested chilli plants using solid-phase microextraction and gas chromatography-mass spectrometry. Our results showed that female WFs preferred non-infested to pre-infested plants. Collection and analysis of volatile compounds emitted by infested plants confirmed that there were significant increases in the production of monoterpenes (cymene; 1,8-cineole), sesquiterpenes (β-cadinene, α-copaene), and methyl salicylate (MeSA) compared to non-infested plants. Our results suggest that host plant infestation by aphids may induce production of secondary metabolites that deter B. tabaci from settling on its host plants. These results provide important information for understanding WF host selection and dispersal among crops, and also for manipulating WF behaviour to improve IPM in chilli.

Methyl benzoate exhibits insecticidal and repellent activities against Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae).

Project description:Methyl benzoate (MB) is a plant-derived volatile organic compound with insecticidal properties, but such activity has not been evaluated against the sweetpotato whitefly Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae), a major crop pest. In this study, we tested methyl benzoate control efficacy on B. tabaci infecting tomato plants in a greenhouse, specifically measuring contact and fumigant toxicity, as well as repellent activity. For direct spray applications of 0% (control), 0.1%, 0.25%, 0.5%, 1%, 2% MB onto tomato leaves infested with adults of B. tabaci (< 5-d-old), 2% MB showed the highest corrected mortality (100%) at 24 h post-treatment. For residual toxicity in which the same MB solutions were sprayed onto tomato leaves and allowed to dry for 2 h before < 5-d-old adults were released, the 2% MB also showed the highest corrected mortality (100%) at 48 h post-treatment. The lethal median concentration (LC50) for eggs, fourth-instar nymphs, and adults were 0.3%, 0.2%, and 0.2%, respectively. In pot culture experiments, 1% MB concentration was found more effective at killing nymphs and preventing adult eclosion than all other concentrations, and gave 100 percent population reduction compared with the control. MB repelled adult whiteflies and caused 96.5% fumigant toxicity within 10 h post-treatment. Repellency and anti-oviposition rates against B. tabaci had median effective doses of 0.24% and 0.16%, respectively. Our results suggest that MB has strong potential as an environmentally friendly biopesticide for control of B. tabaci but field trials and further greenhouse studies are required to establish efficacy under more natural conditions.

Proteomic and transcriptomic responses to thiamethoxam in the Sweetpotato whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae)

Project description:We investigated the transcriptional response to thiamethoxam in the Bemisia tabaci using Illumina sequencing technology. A total of 1,338 genes were differently expressed in the thiamethoxam-resistant whiteflies. All the B biotype whiteflies were maintained on cabbage. The susceptible strain (TH-S) was cultured without exposure to any chemical insecticides. Before the sample collected, almost 10,000 thiamethoxam-resistant adult whiteflies were treated with 2,000 mg/L thiamethoxam (~LC80) to eliminate the heterozygous individuals. Then the survivors were collected after 48 hours and designated as the TH-2000 sample. Then approximately 1,500 adult TH-S and TH-2000 were collected, respectively, The RNA was extracted and sequenced using Illumina HiSeq 2000.
