Permeability Barrier of Gram-Negative Cell Envelopes and Approaches To Bypass It.

ABSTRACT: Gram-negative bacteria are intrinsically resistant to many antibiotics. Species that have acquired multidrug resistance and cause infections that are effectively untreatable present a serious threat to public health. The problem is broadly recognized and tackled at both the fundamental and applied levels. This paper summarizes current advances in understanding the molecular bases of the low permeability barrier of Gram-negative pathogens, which is the major obstacle in discovery and development of antibiotics effective against such pathogens. Gaps in knowledge and specific strategies to break this barrier and to achieve potent activities against difficult Gram-negative bacteria are also discussed.

Isotope Fractionation Pinpoints Membrane Permeability as a Barrier to Atrazine Biodegradation in Gram-negative Polaromonas sp. Nea-C.

Project description:Biodegradation of persistent pesticides like atrazine often stalls at low concentrations in the environment. While mass transfer does not limit atrazine degradation by the Gram-positive Arthrobacter aurescens TC1 at high concentrations (>1 mg/L), evidence of bioavailability limitations is emerging at trace concentrations (<0.1 mg/L). To assess the bioavailability constraints on biodegradation, the roles of cell wall physiology and transporters remain imperfectly understood. Here, compound-specific isotope analysis (CSIA) demonstrates that cell wall physiology (i.e., the difference between Gram-negative and Gram-positive bacteria) imposes mass transfer limitations in atrazine biodegradation even at high concentrations. Atrazine biodegradation by Gram-negative Polaromonas sp. Nea-C caused significantly less isotope fractionation (?(C) = -3.5 ‰) than expected for hydrolysis by the enzyme TrzN (?(C) = -5.0 ‰) and observed in Gram-positive Arthrobacter aurescens TC1 (?(C) = -5.4 ‰). Isotope fractionation was recovered in cell-free extracts (?(C) = -5.3 ‰) where no cell envelope restricted pollutant uptake. When active transport was inhibited with cyanide, atrazine degradation rates remained constant demonstrating that atrazine mass transfer across the cell envelope does not depend on active transport but is a consequence of passive cell wall permeation. Taken together, our results identify the cell envelope of the Gram-negative bacterium Polaromonas sp. Nea-C as a relevant barrier for atrazine biodegradation.

Low-Avidity Autoantibodies against Bactericidal/Permeability-Increasing Protein Occur in Gram-Negative and Gram-Positive Bacteremia.

Project description:Antibody autoreactivity against bactericidal/permeability-increasing protein (BPI) is strongly associated with Pseudomonas aeruginosa infection in cystic fibrosis (CF), non-CF bronchiectasis (BE), and chronic obstructive pulmonary disease (COPD). We examined the pathogen-specific nature of this autoreactivity by examining antibodies to BPI in bacteremia patients. Antibodies to BPI and bacterial antigens were measured in sera by ELISA from five patient cohorts (n = 214). Antibody avidity was investigated. Bacteremic patient sera (n = 32) exhibited IgG antibody autoreactivity against BPI in 64.7% and 46.7% of patients with positive blood cultures for P. aeruginosa and Escherichia coli, respectively. Autoantibody titers correlated with IgG responses to bacterial extracts and lipopolysaccharide (LPS). A prospective cohort of bacteremic patient sera exhibited anti-BPI IgG responses in 23/154 (14.9%) patients with autoreactivity present at the time of positive blood cultures in patients with Gram-negative and Gram-positive bacteria, including 8/60 (13.3%) patients with Staphylococcus aureus Chronic tissue infection with S. aureus was associated with BPI antibody autoreactivity in 2/15 patients (13.3%). Previously, we demonstrated that BPI autoreactivity in CF patient sera exhibits high avidity. Here, a similar pattern was seen in BE patient sera. In contrast, sera from patients with bacteremia exhibited low avidity. These data indicate that low-avidity IgG responses to BPI can arise acutely in response to bacteremia and that this association is not limited to P. aeruginosa This is to be contrasted with chronic respiratory infection with P. aeruginosa, suggesting that either the chronicity or the site of infection selects for the generation of high-avidity responses, with biologic consequences for airway immunity.

Damaging effects of ethylenediaminetetra-acetate and penicillins on permeability barriers in Gram-negative bacteria.

Project description:1. The permeability barrier against benzylpenicillin has been found to be passive in four strains of penicillinase-producing Gram-negative bacteria (three of Klebsiella aerogenes and one of Escherichia coli). 2. If the three K. aerogenes strains are grown in the presence of sub-inhibitory concentrations of benzylpenicillin, ampicillin or phenethicillin the resultant bacterial cells have deficient permeability barriers. Concentrations of ampicillin or benzylpenicillin less than one-tenth of those required to inhibit growth cause destruction of more than half the permeability barrier in these strains. 3. Benzylpenicillin, ampicillin and phenethicillin have no effect upon the permeability barriers of resting cells from the three K. aerogenes strains. 4. Treatment of resting cells with trisodium EDTA, although failing to sensitize K. aerogenes to lysozyme, severely damages permeability barriers in this species. 5. The magnesium and calcium salts of EDTA do not have the same capacity as the sodium salt for causing damage to permeability barriers in K. aerogenes and E. coli. Damage caused by trisodium EDTA can be at least partially reversed by treatment with Ca(2+) or Mg(2+) ions. It is suggested that EDTA damage is caused by removal of either Ca(2+) or Mg(2+) ions, or both, from the bacterial cell envelope. 6. Bacterial cells with deficient permeability barriers as a result of either growth in the presence of a penicillin or treatment with EDTA remain viable, and revert to their usual permeability after growth in nutrient broth.
