On the synonymy of two Acantholycosa species (Araneae, Lycosidae) from the Altai.

ABSTRACT: Two species previously known from East Kazakhstan, Acantholycosa katunensis Marusik, Azarkina & Koponen, 2004, known from the holotype male, and Acantholycosa kurchumensis Marusik, Azarkina & Koponen, 2004, syn. n. known from females, are synonymized, and priority is given to Acantholycosa katunensis. Acantholycosa katunensis is reported for the first time in the Russian Altai. Both sexes of this species are illustrated, and a distribution map is provided.

The complete mitochondrial genome of <i>Halocosa hatanensis</i> (Araneae: Lycosidae).

Project description:The complete mitochondrial genome sequence of a desert wolf spider, Halocosa hatanensis (Lycosidae) was reported. The circular molecule is 14,257 base pairs (bp) in length and consisted of 77.8% AT nucleotides, containing two ribosomal RNA genes, 22 transfer RNAs, 13 protein-coding genes (PCGs), 1 control region, and 1 AT-rich region. The length of 13 PCGs is 10,782 bp in total, encoding 3638 amino acids. The phylogenetic analyses upon mitogenomes proves its relatively basal position within Lycosidae. It would provide further evolutionary research data for the wolf spider family Lycosidae.

Relevance of ddRADseq method for species and population delimitation of closely related and widely distributed wolf spiders (Araneae, Lycosidae).

Project description:Although species delimitation is often controversial, emerging DNA-based and classical morphology-based methods are rarely compared using large-scale samplings, even less in the case of widely distributed species that have distant, allopatric populations. In the current study, we examined species boundaries within two wolf spider species of the genus Pardosa (Araneae, Lycosidae), P. riparia and P. palustris. Wolf spiders constitute an excellent model for testing the relevance of traditional vs. modern methods in species and population delimitation because several closely related species are distributed over cross-continental geographic ranges. Allopatric populations of the two Pardosa species were sampled across Europe to Far East Russia (latitudinal range > 150°) and several dozen individuals were studied using morphological characters (morphometry of three measures for both sexes, plus five in males only and two in females only), DNA barcoding (COI sequencing) and double-digest restriction site associated DNA sequencing (ddRADseq). The results obtained allow for changing the taxonomic status of two Far East Russian populations to subspecies and ddRADseq proved to be a powerful tool for taxonomic research despite scarce sampling and inherent subjectivity of species delimitation in allopatry. Overall, this study pleads for both multi-criteria and more population-based studies in taxonomy.

Acantholycosa norvegica (Thorell, 1872) is reported from the Maritime Province for the first time. A key and illustrations to all six species that occur in Far East Asia are provided.

Transcriptome Profiling Analysis of Wolf Spider Pardosa pseudoannulata (Araneae: Lycosidae) after Cadmium Exposure.

Project description:Pardosa pseudoannulata is one of the most common wandering spiders in agricultural fields and a potentially good bioindicator for heavy metal contamination. However, little is known about the mechanisms by which spiders respond to heavy metals at the molecular level. In the present study, high-throughput transcriptome sequencing was employed to characterize the de novo transcriptome of the spiders and to identify differentially expressed genes (DEGs) after cadmium exposure. We obtained 60,489 assembled unigenes, 18,773 of which were annotated in the public databases. A total of 2939 and 2491 DEGs were detected between the libraries of two Cd-treated groups and the control. Functional enrichment analysis revealed that metabolism processes and digestive system function were predominately enriched in response to Cd stress. At the cellular and molecular levels, significantly enriched pathways in lysosomes and phagosomes as well as replication, recombination and repair demonstrated that oxidative damage resulted from Cd exposure. Based on the selected DEGs, certain critical genes involved in defence and detoxification were analysed. These results may elucidate the molecular mechanisms underlying spiders' responses to heavy metal stress.

A survey of the East Palaearctic Lycosidae (Araneae). 9. Genus Xerolycosa Dahl, 1908 (Evippinae).

Project description:Three species of Xerolycosa: Xerolycosa nemoralis (Westring, 1861), Xerolycosa miniata (C.L. Koch, 1834) and Xerolycosa mongolica (Schenkel, 1963), occurring in the Palaearctic Region are surveyed, illustrated and redescribed. Arctosa mongolica Schenkel, 1963 is removed from synonymy with Xerolycosa nemoralis and transferred to Xerolycosa, and the new combination Xerolycosa mongolica (Schenkel, 1963) comb. n. is established. One new synonymy, Xerolycosa undulata Chen, Song et Kim, 1998 syn.n. from Heilongjiang = Xerolycosa mongolica (Schenkel, 1963), is proposed. In addition, one more new combination is established, Trochosa pelengena (Roewer, 1960) comb. n., ex Xerolycosa.

Transcriptome response to temperature stress in the wolf spider <i>Pardosa pseudoannulata</i> (Araneae: Lycosidae).

Project description:The wolf spider Pardosa pseudoannulata is a dominant predator in paddy ecosystem and an important biological control agent of rice pests. Temperature represents a primary factor influencing its biology and behavior, although the underlying molecular mechanisms remain unknown. To understand the response of P. pseudoannulata to temperature stress, we performed comparative transcriptome analyses of spider adults exposed to 10°C and 40°C for 12 h. We obtained 67,725 assembled unigenes, 21,765 of which were annotated in P. pseudoannulata transcriptome libraries, and identified 905 and 834 genes significantly up- or down-regulated by temperature stress. Functional categorization revealed the differential regulation of transcription, signal transduction, and metabolism processes. Calcium signaling pathway and metabolic pathway involving respiratory chain components played important roles in adapting to low temperature, whereas at high temperature, oxidative phosphorylation and amino acid metabolism were critical. Differentially expressed ribosomal protein genes contributed to temperature stress adaptation, and heat shock genes were significantly up-regulated. This study represents the first report of transcriptome identification related to the Araneae species in response to temperature stress. These results will greatly facilitate our understanding of the physiological and biochemical mechanisms of spiders in response to temperature stress.

Two new species of Harpactea (Araneae, Dysderidae) from Turkey.

Project description:Two new species, Harpactea arnedoisp. n. and Harpactea kenceisp. n., are described on the basis of both sexes from the eastern Mediterranean and Aegean regions of Turkey. Harpactea kenceisp. n. can be easily distinguished from all other Turkish and European representatives of the genus by the structure of the flattened, massive embolus on the male copulatory organ. Although resembling Stalagtia in palpal morphology, we describe one of the new species as Harpactea arnedoisp. n. For both Turkish species, detailed morphological descriptions and diagnoses are presented together with figures of the copulatory organs.

Bridging the distributional gap of Tyloridastriata (Thorell, 1877) and new synonymy (Araneae: Tetragnathidae).

Project description:BackgroundAlthough Tyloridastriata has not been reported from India, observations on India Biodiversity Portal (IBP 2015), an open access repository for biodiversity information of Indian subcontinent, showed images resembling this species. The respective locality in Gujarat, India was explored and specimens were studied to confirm record of T.striata in India. Literature study showed some taxonomic lacunae which needed to be resolved.New informationThe tetragnathid spider Tyloridastriata (Thorell, 1877) is redescribed on examination of its long unknown syntypes from Kendari, Southeast Sulawesi deposited in the Natural History Museum of Giacomo Doria, Genoa, Italy and additional material from Kendari and other Indonesian regions. Description of Tyloridastellimicans (Simon, 1885) mentions its identity with T.striata, which however characterizes sub-adult of T.striata and is accordingly synonymized. Tyloridastriata is newly recorded from India which links the distributional gap between China to Australia and Comoros Islands. Distributional and taxonomic records of Tylorida species with T.striata- like globose abdomen infer probable synonymy, which is subject to genus revision.

Seasonal variations in body melanism and size of the wolf spider Pardosa astrigera (Araneae: Lycosidae).

Project description:Variations in species morphology and life-history traits strongly correlate with geographic and climatic characteristics. Most studies on morphological variations in animals focus on ectotherms distributed on a large geographic scale across latitudinal and/or altitudinal gradient. However, the morphological variations of spiders living in the same habitats across different seasons have not been reported. In this study, we used the wolf spider, Pardosa astrigera, as a model to determine seasonal differences in adult body size, melanism, fecundity, and egg diameter both in the overwintering and the first generation for 2010 and 2016. The results showed that in 2010, both females and males of the overwintering generation were significantly darker than the first generation. Moreover, the overwintering females were markedly larger and produced more and bigger eggs than the first generation in both 2010 and 2016. Considering the overwintering P. astrigera experiencing low temperature and/or desiccation stress, these results suggest that substantially darker and larger body of the overwintering generation is adaptive to adverse conditions.
