Project description:BackgroundWe compiled a database of firefly species records from the Atlantic Forest hotspot in Brazil and made it available at GBIF. Data were gathered from literature and from several key entomological collections, including: Coleção entomológica Prof. José Alfredo Pinheiro Dutra (DZRJ/UFRJ) and Coleção do Laboratório de Ecologia de Insetos from Universidade Federal do Rio de Janeiro (CLEI/UFRJ); Coleção Entomológica do Instituto Oswaldo Cruz (CEIOC); Museu de Zoologia da Universidade de São Paulo (MZSP); Coleção Entomológica Pe. Jesus Santiago Moure from Universidade Federal do Paraná (DZUP/UFPR); and Coleção Entomológica from Universidade Federal Rural de Pernambuco (UFRPE). This database represents the largest contribution to a public repository of recorded occurrences from Neotropical fireflies.New informationThis dataset shows the occurrence and abundance of firefly species in the Atlantic Forest hotspot. Firefly species endemic to this biome are also present and considered in the study. These data can assist scientific and societal needs, by supporting future research projects and conservation decision-making.

Molecular Systematics of the Firefly Genus Luciola (Coleoptera: Lampyridae: Luciolinae) with the Description of a New Species from Singapore.

Project description:The firefly genus Luciola sensu McDermott contains 282 species that are distributed across major parts of Asia, Europe, Africa, Australia, and the Pacific islands. Due to phenotypic similarities, species identification using external morphological characters can be unreliable for this group. Consequently, decades of piecemeal taxonomic treatments have resulted in numerous erroneous and contentious classifications. Furthermore, our understanding of the group's evolutionary history is limited due to the lack of a robust phylogenetic framework that has also impeded efforts to stabilize its taxonomy. Here, we constructed molecular phylogenies of Luciola and its allies based on combined mitogenomes and Cytochrome c oxidase subunit 1 (COX1) sequences including a newly sequenced mitogenome of an unidentified taxon from Singapore. Our results showed that this taxon represents a distinct and hitherto undescribed evolutionary lineage that forms a clade with L. filiformis from Japan and L. curtithorax from China. Additionally, the Singaporean lineage can be differentiated from other congeners through several external and internal diagnostic morphological characters, and is thus described herein as a new species. Our phylogeny also strongly supported the paraphyly of Luciola with regard to L. cruciata and L. owadai, which were inferred to be more closely related to the genus Aquatica as opposed to other members of Luciola sensu stricto. The genus Hotaria was inferred as a derived clade within Luciola (sister to L. italica), supporting its status as a subgenus of Luciola instead of a distinct genus. This is the first time since 1909 that a new species of luminous firefly has been discovered in Singapore, highlighting the need for continued biodiversity research, even in small, well-studied and highly developed countries, such as Singapore.

Ten protein-coding genes stopped with TAA or TAG codon and the other three genes have an incomplete termination codon, a single T--. We sequenced the mitochondrial genome of fireflies to analyze phylogenetic relationships and deduce the evolution of their flashing signals.

Project description:Pyrocoelia analis (Fabricius, 1801) (Coleoptera, Lampyridae, Pyrocoelia) is a beautiful ornamental insect widely distributed in East and Southeast Asia. The complete mitogenome of P. analis has been sequenced. The mitogenome, total length of 14,785 bp, includes 13 protein-coding genes, 22 transfer RNAs, two ribosomal RNAs, and a noncoding D-loop region. The overall base composition of Pyrocoelia analis mitogenome is 34.63% for A, 13.69% for C, 42.79% for T, and 8.89% for G, with a high A + T bias of 77.42%. These mitogenome data might be useful for further phylogeography analyses and other related studies in Hymenoptera.

Complete mitochondrial genome of the Korean endemic firefly, Luciola unmunsana (Coleoptera: Lampyridae).

Project description:In this study, we announce the complete mitochondrial genome (mitogenome) of the Korean endemic firefly, Luciola unmunsana Doi, 1931. The full-length circular genome was 15,858 bp, with 77.94% A/T content. It contained the typical set of 37 metazoan genes: 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, and 2 ribosomal RNA (rRNA) genes, as well as an A + T-rich region. The gene arrangement of the species is identical to that of the ancestral arrangement found in the majority of insects. The maximum-likelihood tree, built using all PCGs and two rRNAs via randomized accelerated maximum likelihood (RAxML) showed that L. unmunsana was grouped as a sister to L. curtithorax with the highest nodal support. However, another Luciola species clustered with the Aquatica species such that the genus Luciola was a non-monophyletic group. Therefore, more sampling is required to clarify the phylogeny of Luciola.

Long-Read Genome Sequencing of Abscondita cerata (Coleoptera: Lampyridae), the Endemic Firefly of Taiwan.

Project description:Abscondita cerata is the most abundant and widely distributed endemic firefly species in Taiwan and is considered a key environmental and ecological indicator organism. In this study, we report the first long-read genome sequencing of Abs. cerata sequenced by Nanopore technology. The draft genome size, 967 Mb, was measured through a hybrid approach that consisted of assembling using 11.25-Gb Nanopore long reads and polishing using 9.47-Gb BGI PE100 short reads. The drafted genome was assembled into 4,855 contigs, with the N50 reaching 325.269 kb length. The assembled genome was predicted to possess 55,206 protein-coding genes, of which 20,862 (37.78%) were functionally annotated with public databases. 47.11% of the genome sequences consisted of repeat elements; among them DNA transposons accounted for the largest proportion (26.79%). A BUSCO (Benchmarking Universal Single Copy Orthologs) evaluation demonstrated that the genome and gene completeness were 84.8% and 79%, respectively. The phylogeny constructed using 1,792 single copy genes was consistent with previous studies. The comparative transcriptome between adult male head and lantern tissues revealed (1) the vision of Abs. cerata is primarily UV-sensitive to environmental twilight, which determines when it begins its nocturnal activity, (2) the major expressed OR56d receptor may be correlated to suitable humidity sensing, and (3) Luc1-type luciferase is responsible for Abs. cerata's luminescent spectrum.
