Modes of Antiviral Action of Chemical Portions and Constituents from Woad Root Extract against Influenza Virus A FM1.

ABSTRACT: Woad root has been used for the prevention of influenza for hundreds of years in many Asian countries. In this study, the antiviral modes of clemastanin B (CB), epigoitrin, phenylpropanoid portion (PEP), and the mixture of phenylpropanoids, alkaloids, and organic acid portions (PEP + ALK + OA) from wood root extract against influenza virus A FM1 were investigated. The results revealed that CB, epigoitrin, PEP, and PEP + ALK + OA exert their anti-influenza activity via inhibiting the virus multiplication, prophylaxis, and blocking the virus attachment. The primary mode of action of PEP and PEP + ALK + OA is the inhibition of virus replication. The inhibitory effect on virus attachment and multiplication is the main modes for epigoitrin. All the compounds or chemical portions from woad root extract tested in this study do not have direct virucidal activity. Our results provided the comprehensive analysis of the antiviral mechanism of wood root extract.

Project description:Zanthoxylum paracanthum Kokwaro (Rutaceae) is an endemic Kenyan and Tanzanian plant used in folk medicine by local populations. Although other Zanthoxylum species have been studied, only Z. paracantum stem extracts have been profiled, even though the roots are also used as herbal remedies. As root extracts may be another source of pharmaceutical compounds, the CH2Cl2/MeOH (1:1) root bark extract was studied in this report. Eight root bark compounds were isolated and their structural identities were confirmed by mass spectrometry (MS) and nuclear magnetic resonance (NMR) (using COSY, HSQC, NOESY and HMBC) analyses. The structural identities were determined as follows: the fatty acid-myristic acid (1); the sterol-stigmasterol (2); the lignan-sesamin (3); two β-carboline alkaloids-10-methoxycanthin-6-one (6) and canthin-6-one (7); and three phenanthridine alkaloids-8-acetonyldihydrochelerythrine (4), arnottianamide (5) and 8-oxochelerythrine (8). Some of these compounds were identified in the species for the first time. These compounds and the extract were then tested in vitro against methicillin-resistant Staphylococcus aureus (MRSA), Escherichia coli (ATCC 25922), Staphylococcus aureus (ATCC 29213) and Candida albicans (ATCC 10231) before tests for antiproliferative activity against the human breast cancer (HCC 1395), human prostate cancer (DU 145) and normal (Vero E6) cell lines were conducted. Minimum inhibition concentration values of 3.91, 1.95, 0.98 and 7.81 µg/mL against MRSA, S. aureus, E. coli and C. albicans, respectively, were recorded. Among the isolates, canthin-6-one was the most active, followed by 10-methoxycanthin-6-one. The root extract and some of the compounds also had antiproliferative activity against the HCC 1395 cell line. Stigmasterol and canthin-6-one had IC50 values of 7.2 and 0.42. The root bark extract also showed activity, at 8.12 µg/mL, against the HCC 1395 cells. Out of the chemical isolates, 10-methoxycanthin-6-one and canthin-6-one showed the strongest inhibition of the DU 145 cells. The root extract had significant antimicrobial and antiproliferative activities, supporting the traditional use of this plant in treating microbial infections and cancer-related ailments.

Project description:The genus Gnidia, with species close to 152, is traditionally used to treat wide ranges of ailments in humans and animals. Gnidia involucrata is one of the species found in Ethiopia and traditionally used as a laxative, antirheumatic agent, insecticide, antibacterial agent, and antimalarial agent. In view of its traditional use, the root bark was sequentially extracted with n-hexane, EtOAc, and MeOH to afford 0.78%, 4%, and 6% crude extracts, respectively. The chromatographic separation of the EtOAc extract using silica gel column chromatography yielded three pure compounds: tetratriacontanyl caffeate (1), 12-O-dodeca-2,4-dienoylphorbol-13-acetate (2), and naringenin (3). This is the first report of the isolation of 1 and its kind from the genus and most probably from the Thymelaeaceae family. The structures of these compounds were characterized and identified by NMR and mass spectrometric analyses and comparison with literature data. The EtOAc extract and isolated compounds were assessed for their in vitro antibacterial and antioxidant activities. The EtOAc extract (1.5 mg/mL) showed significant inhibitory activity against S. aureus, E. coli, P. mirabilis, and K. pneumonia bacterial strains with the highest inhibition zone observed against S. aureus (23 mm), which is even greater than that of the reference drug ciprofloxacin (22 mm). However, the inhibition displayed on these bacterial strains for the three pure compounds was marginal with variable degrees of potency between the compounds. The better activity of the crude extract could be due to the synergistic interactions of several phytochemicals present in the extract, which cannot be the case when pure compounds are evaluated alone. The antioxidant activities of the extracts and isolated compounds were evaluated using DPPH and ferric thiocyanate methods. The EtOAc and MeOH extracts and compounds 1 and 2 were found to inhibit the DPPH radical by 70.7, 66.9, 85.8, and 52.8%, respectively. The EtOAc extract and compound 1 inhibited peroxidation of lipids by 84 and 86%, respectively. The radical scavenging displayed by compound 1 was significant compared with that displayed by ascorbic acid, indicating the strong antilipid peroxidation potential of the extract of root barks of G. involucrata. Therefore, the extracts of the root bark of G. involucrata can be used as a remedy in combating diseases caused by bacteria and free radicals provided that further comprehensive evaluation could be recommended for the conclusive decision on potential candidacy of this plant.
