Biodegradation of diesel fuel hydrocarbons by mangrove fungi from Red Sea Coast of Saudi Arabia.

ABSTRACT: Mangrove sediments were collected from major mangrove stands on the Red Sea Coast of Saudi Arabia. Forty five isolates belonging to 12 genera were purified and five isolates as well as their consortium were found to be able to grow in association with petroleum oil as sole carbon source under in vitro conditions. The isolated strains were identified based on internal transcribed spacer (ITS) rDNA sequence analysis. The fungal strains with the greatest potentiality to degrade diesel oil, without developing antagonistic activity, were identified as Alternaria alternata, Aspergillus terreus, Cladosporium sphaerospermum, Eupenicillium hirayamae and Paecilomyces variotii. As compared to the controls, these fungi accumulated significantly higher biomass, produced extracellular enzymes and liberated larger volumes of CO2. These observations with GC-MS data confirm that these isolates displayed rapid diesel oil bioremoval and when used together as a consortium, there was no antagonistic activity.

Project description:BackgroundMicrobial biodegradation of oil-hydrocarbons is one of the sustainable and cost-effective methods to remove petroleum spills from contaminated environments. The current study aimed to investigate the biodegradation abilities of three Fusarium isolates from oil reservoirs in Saudi Arabia. The novelty of the current work is that the biodegradation ability of these isolates was never tested against some natural hydrocarbons of variable compositions, such as Crude oil, and those of known components such as kerosene and diesel oils.MethodsThe isolates were treated with five selected hydrocarbons. The hydrocarbon tolerance test in solid and liquid media was performed. The scanning electron microscope (SEM) investigated the morphological changes of treated fungi. 2, 6-Dichlorophenol Indophenol (DCPIP), drop collapse, emulsification activity, and oil Spreading assays investigated the biodegradation ability. The amount of produced biosurfactants was measured, and their safety profile was estimated by the germination assay of tomato seeds.ResultsThe tolerance test showed enhanced fungal growth of all isolates, whereas the highest dose inhibition response (DIR) was 77% for Fusarium proliferatum treated with the used oil (p < 0.05). SEM showed morphological changes in all isolates. DCPIP results showed that used oil had the highest biodegradation by Fusarium verticillioides and Fusarium oxysporum. Mixed oil induced the highest effect in oil spreading, drop collapse, and emulsification assay caused by F. proliferatum. The highest recovery of biosurfactants was obtained by the solvent extraction method for F. verticillioides (4.6 g/L), F. proliferatum (4.22 g/L), and F. oxysporum (3.73 g/L). The biosurfactants produced by the three isolates stimulated tomato seeds' germination more than in control experiments.ConclusionThe current study suggested the possible oil-biodegradation activities induced by three Fusarium isolates from Riyadh, Saudi Arabia. The produced biosurfactants are not toxic against tomato seed germination, emphasizing their environmental sustainability. Further studies are required to investigate the mechanism of biodegradation activities and the chemical composition of the biosurfactants produced by these species.

Project description:Oil spills in the subarctic marine environment off the coast of Labrador, Canada, are increasingly likely due to potential oil production and increases in ship traffic in the region. To understand the microbiome response and how nutrient biostimulation promotes biodegradation of oil spills in this cold marine setting, marine sediment microcosms amended with diesel or crude oil were incubated at in situ temperature (4°C) for several weeks. Sequencing of 16S rRNA genes following these spill simulations revealed decreased microbial diversity and enrichment of putative hydrocarbonoclastic bacteria that differed depending on the petroleum product. Metagenomic sequencing revealed that the genus Paraperlucidibaca harbors previously unrecognized capabilities for alkane biodegradation, which were also observed in Cycloclasticus. Genomic and amplicon sequencing together suggest that Oleispira and Thalassolituus degraded alkanes from diesel, while Zhongshania and the novel PGZG01 lineage contributed to crude oil alkane biodegradation. Greater losses in PAHs from crude oil than from diesel were consistent with Marinobacter, Pseudomonas_D, and Amphritea genomes exhibiting aromatic hydrocarbon biodegradation potential. Biostimulation with nitrogen and phosphorus (4.67 mM NH4Cl and 1.47 mM KH2PO4) was effective at enhancing n-alkane and PAH degradation following low-concentration (0.1% [vol/vol]) diesel and crude oil amendments, while at higher concentrations (1% [vol/vol]) only n-alkanes in diesel were consumed, suggesting toxicity induced by compounds in unrefined crude oil. Biostimulation allowed for a more rapid shift in the microbial community in response to petroleum amendments, more than doubling the rates of CO2 increase during the first few weeks of incubation. IMPORTANCE Increases in transportation of diesel and crude oil in the Labrador Sea will pose a significant threat to remote benthic and shoreline environments, where coastal communities and wildlife are particularly vulnerable to oil spill contaminants. Whereas marine microbiology has not been incorporated into environmental assessments in the Labrador Sea, there is a growing demand for microbial biodiversity evaluations given the pronounced impact of climate change in this region. Benthic microbial communities are important to consider given that a fraction of spilled oil typically sinks such that its biodegradation occurs at the seafloor, where novel taxa with previously unrecognized potential to degrade hydrocarbons were discovered in this work. Understanding how cold-adapted microbiomes catalyze hydrocarbon degradation at low in situ temperature is crucial in the Labrador Sea, which remains relatively cold throughout the year.
