Floral Volatiles in Parasitic Plants of the Orobanchaceae. Ecological and Taxonomic Implications.

ABSTRACT: The holoparasitic broomrapes, Orobanche spp. and Phelipanche spp. (Orobanchaceae), are root parasites that completely depend on a host plant for survival and reproduction. There is considerable controversy on the taxonomy of this biologically and agronomically important family. Flowers of over 25 parasitic Orobanchaceae and a number of close, parasitic and non-parasitic, relatives emitted a complex blend of volatile organic compounds (VOCs), consisting of over 130 VOCs per species. Floral VOC blend-based phylogeny supported the known taxonomy in internal taxonomic grouping of genus and eliminated the uncertainty in some taxonomical groups. Moreover, phylogenetic analysis suggested separation of the broomrapes into two main groups parasitizing annual and perennial hosts, and for the annual hosts, into weedy and non-weedy broomrapes. We conclude that floral VOCs are a significant tool in species identification and possibly even in defining new species and can help to improve controversial taxonomy in the Orobanchaceae.

Project description:Background and aimsFlowers emit a wide range of volatile compounds which can be critically important to interactions with pollinators or herbivores. Yet most studies of how the environment influences plant volatiles focus on leaf emissions, with little known about abiotic sources of variation in floral volatiles. Understanding phenotypic plasticity in floral volatile emissions has become increasingly important with globally increasing temperatures and changes in drought frequency and severity. Here quantitative relationships of floral volatile emissions to soil water content were analysed.MethodsPlants of the sub-alpine herb Ipomopsis aggregata and hybrids with its closest congener were subjected to a progressive dry down, mimicking the range of soil moistures experienced in the field. Floral volatiles and leaf gas exchange were measured at four time points during the drought.Key resultsAs the soil dried, floral volatile emissions increased overall and changed in composition, from more 1,3-octadiene and benzyl alcohol to higher representation of some terpenes. Emissions of individual compounds were not linearly related to volumetric water content in the soil. The dominant compound, the monoterpene α-pinene, made up the highest percentage of the scent mixture when soil moisture was intermediate. In contrast, emission of the sesquiterpene (E,E)-α-farnesene accelerated as the drought became more intense. Changes in floral volatiles did not track the time course of changes in photosynthetic rate or stomatal conductance.ConclusionsThis study shows responses of specific floral volatile organic compounds to soil moisture. The non-linear responses furthermore suggest that extreme droughts may have impacts that are not predictable from milder droughts. Floral volatiles are likely to change seasonally with early summer droughts in the Rocky Mountains, as well as over years as snowmelt becomes progressively earlier. Changes in water availability may have impacts on plant-animal interactions that are mediated through non-linear changes in floral volatiles.

Project description:Background and aimsAustralian sexually deceptive Chiloglottis orchids attract their specific male wasp pollinators by means of 2,5-dialkylcyclohexane-1,3-diones or 'chiloglottones', representing a newly discovered class of volatiles with unique structures. This study investigated the hypothesis that UV-B light at low intensities is directly required for chiloglottone biosynthesis in Chiloglottis trapeziformis.MethodsChiloglottone production occurs only in specific tissue (the callus) of the labellum. Cut buds and flowers, and whole plants with buds and flowers, sourced from the field, were kept in a growth chamber and interactions between growth stage of the flowers and duration and intensity of UV-B exposure on chiloglottone production were studied. The effects of the protein synthesis inhibitor cycloheximide were also examined.Key resultsChiloglottone was not present in buds, but was detected in buds that were manually opened and then exposed to sunlight, or artificial UV-B light for ?5?min. Spectrophotometry revealed that the sepals and petals blocked UV-B light from reaching the labellum inside the bud. Rates of chiloglottone production increased with developmental stage, increasing exposure time and increasing UV-B irradiance intensity. Cycloheximide did not inhibit the initial production of chiloglottone within 5?min of UV-B exposure. However, inhibition of chiloglottone production by cycloheximide occurred over 2?h of UV-B exposure, indicating a requirement for de novo protein synthesis to sustain chiloglottone production under UV-B.ConclusionsThe sepals and petals of Chiloglottis orchids strongly block UV-B wavelengths of light, preventing chiloglottone production inside the bud. While initiation of chiloglottone biosynthesis requires only UV-B light, sustained chiloglottone biosynthesis requires both UV-B and de novo protein biosynthesis. The internal amounts of chiloglottone in a flower reflect the interplay between developmental stage, duration and intensity of UV-B exposure, de novo protein synthesis, and feedback loops linked to the starting amount of chiloglottone. It is concluded that UV-B light contributes directly to chiloglottone biosynthesis. These findings suggest an entirely new and unexpected biochemical reaction that might also occur in taxa other than these orchids.
