Data on comparison between FLEC and CLIMPAQ methods used for fast sorption measurements of VOCs on building materials.

ABSTRACT: A test emission chamber called CLIMPAQ has been coupled to a chromatography analyzer GC to measure volatile organic compounds (VOC) concentration during a sorption experiments (Fast sorption measurements of VOCs on building materials: Part 2 - Comparison between FLEC and CLIMPAQ methods, (Rizk et al., In press) [1]). The equations used to calculate the mass transfer coefficient and the thickness of the boundary layer developed on the surface of a material are presented. In addition, the experimental profiles obtained using the CLIMPAQ chamber is also presented in the presence and the absence of a building material. Finally, the impact of chamber size on the obtained concentration profile using different chambers is shown using 3 types of chambers having different volumes, 1 m(3), 30 m(3) and a micro chamber of 40 mL.

Building gene co-expression networks using transcriptomics data for systems biology investigations: Comparison of methods using microarray data.

Project description:Gene co-expression networks (GCN), built using high-throughput gene expression data are fundamental aspects of systems biology. The main aims of this study were to compare two popular approaches to building and analysing GCN. We use real ovine microarray transcriptomics datasets representing four different treatments with Metyrapone, an inhibitor of cortisol biosynthesis. We conducted several microarray quality control checks before applying GCN methods to filtered datasets. Then we compared the outputs of two methods using connectivity as a criterion, as it measures how well a node (gene) is connected within a network. The two GCN construction methods used were, Weighted Gene Co-expression Network Analysis (WGCNA) and Partial Correlation and Information Theory (PCIT) methods. Nodes were ranked based on their connectivity measures in each of the four different networks created by WGCNA and PCIT and node ranks in two methods were compared to identify those nodes which are highly differentially ranked (HDR). A total of 1,017 HDR nodes were identified across one or more of four networks. We investigated HDR nodes by gene enrichment analyses in relation to their biological relevance to phenotypes. We observed that, in contrast to WGCNA method, PCIT algorithm removes many of the edges of the most highly interconnected nodes. Removal of edges of most highly connected nodes or hub genes will have consequences for downstream analyses and biological interpretations. In general, for large GCN construction (with > 20000 genes) access to large computer clusters, particularly those with larger amounts of shared memory is recommended.

The effects of temperature and relative humidity on trichloroethylene sorption capacities of building materials under conditions relevant to vapor intrusion.

Project description:In this research, the sorption capacities of trichloroethylene (TCE) vapors were investigated at ppbv concentrations on building materials in the temperature range from 283.15 K to 303.15 K and at relative humidity levels of 50% and 85%. These are conditions that are relevant to vapor intrusion investigations. Such interactions of TCE with different materials at different temperatures/ relative humidity have been studied to a very limited extent, and not yet at all at the extremely low concentration ranges in vapor intrusion scenarios. The sorption capacities of the building materials decrease as temperature increases. The isotherms are for the most part linear, indicating that the adsorption process takes place in the Henry's Law regime, except for cinderblock. The isosteric heats of adsorption have been calculated. The sorption capacities of glass wool and nylon carpet increased slightly when the relative humidity increased from 0% to 85% whereas the sorption capacities of printer paper, drywall, and cinderblock decreased significantly at elevated humidity levels. The influence of humidity is complicated since under certain conditions it may enhance sorption or inhibit sorption. The sorption capacities of the studied materials are in a range indicating the possibility of these processes should not be overlooked during vapor intrusion investigations.

Dynamic Triple-Mode Sorption and Outgassing in Materials.

Project description:Moisture uptake and outgassing can be detrimental to a system by altering the chemical and mechanical properties of materials within the system over time. In this work, we conducted isotherm experiments to investigate dynamic moisture sorption and desorption in markedly different materials, i.e., a polymeric material, Sylgard-184 and a ceramic aluminosilicate material, Zircar RS-1200, at different temperatures (30?°C-70?°C) by varying the water activity (0.0-0.90). Sylgard-184 showed a linear sorption and outgassing behavior with no-hysteresis over the entire temperature and water activity range considered here. Whereas, the sorption and outgassing of Zircar RS-1200 was highly non-linear with significant hysteresis, especially at higher water activities, at all temperatures considered here. The type of hysteresis suggested the presence of mesopores in Zircar RS-1200, whereas the lack of hysteresis in Sylgard-184 indicates that it has a nonporous structure. A diffusion model coupled with a dynamic, triple-mode sorption (Langmuir, Henry, and pooling modes) model employed in this study matched our experimental data very well and provides mechanistic insight into the processes. Our triple-mode sorption model was adaptive enough to (1) model these distinctly different materials and (2) predict sorption and outgassing under conditions that are distinctly different from the parameterization experiments.

Fast methods for spatially correlated multilevel functional data.

Project description:We propose a new methodological framework for the analysis of hierarchical functional data when the functions at the lowest level of the hierarchy are correlated. For small data sets, our methodology leads to a computational algorithm that is orders of magnitude more efficient than its closest competitor (seconds versus hours). For large data sets, our algorithm remains fast and has no current competitors. Thus, in contrast to published methods, we can now conduct routine simulations, leave-one-out analyses, and nonparametric bootstrap sampling. Our methods are inspired by and applied to data obtained from a state-of-the-art colon carcinogenesis scientific experiment. However, our models are general and will be relevant to many new data sets where the object of inference are functions or images that remain dependent even after conditioning on the subject on which they are measured. Fracture toughness of porous materials - Experimental methods and data.

Project description:We provide numerical experimental data and detailed information about the sample preparation and the experimental methods, used by different research groups for measuring the fracture toughness of porous materials. These data are supplemental information to the publication "A Geometric Model for the Fracture Toughness of Porous Materials," [1], which is based on experimental data of ceramic and polymer materials. For the sake of completeness, we provide here also data from fracturing metallic foams. The corresponding theoretical curves, which are based on the model described in the reference, are given additionally in the diagrams. The utilized publications are not a comprehensive compilation of all corresponding measurements concerning porous materials, but should be seen as a typical set of respective experiments with the focus on the fracture toughness of porous materials. The discussion and interpretation are provided in the above-mentioned reference.

Comparison of soil sorption parameters of pesticides measured by batch and centrifugation methods using an andosol.

Project description:We compared the soil sorption coefficient (K d) measured by batch and centrifugation methods using a Japanese andosol and ten pesticides. Although the K d values measured by both methods increased with time, those obtained via the batch method tended to be higher during the test period. The difference in K d values between the two methods affected pesticide concentrations estimated in the soil solution, and the results estimated using K d values obtained via the batch method underestimated the observed trends.

A comparison of sample preparation methods for extracting volatile organic compounds (VOCs) from equine faeces using HS-SPME.

Project description:Introduction:Disturbance to the hindgut microbiota can be detrimental to equine health. Metabolomics provides a robust approach to studying the functional aspect of hindgut microorganisms. Sample preparation is an important step towards achieving optimal results in the later stages of analysis. The preparation of samples is unique depending on the technique employed and the sample matrix to be analysed. Gas chromatography mass spectrometry (GCMS) is one of the most widely used platforms for the study of metabolomics and until now an optimised method has not been developed for equine faeces. Objectives:To compare a sample preparation method for extracting volatile organic compounds (VOCs) from equine faeces. Methods:Volatile organic compounds were determined by headspace solid phase microextraction gas chromatography mass spectrometry (HS-SPME-GCMS). Factors investigated were the mass of equine faeces, type of SPME fibre coating, vial volume and storage conditions. Results:The resultant method was unique to those developed for other species. Aliquots of 1000 or 2000 mg in 10 ml or 20 ml SPME headspace were optimal. From those tested, the extraction of VOCs should ideally be performed using a divinylbenzene-carboxen-polydimethysiloxane (DVB-CAR-PDMS) SPME fibre. Storage of faeces for up to 12 months at - 80 °C shared a greater percentage of VOCs with a fresh sample than the equivalent stored at - 20 °C. Conclusions:An optimised method for extracting VOCs from equine faeces using HS-SPME-GCMS has been developed and will act as a standard to enable comparisons between studies. This work has also highlighted storage conditions as an important factor to consider in experimental design for faecal metabolomics studies.

A comparison of microfluidic methods for high-throughput cell deformability measurements.

Project description:The mechanical phenotype of a cell is an inherent biophysical marker of its state and function, with many applications in basic and applied biological research. Microfluidics-based methods have enabled single-cell mechanophenotyping at throughputs comparable to those of flow cytometry. Here, we present a standardized cross-laboratory study comparing three microfluidics-based approaches for measuring cell mechanical phenotype: constriction-based deformability cytometry (cDC), shear flow deformability cytometry (sDC) and extensional flow deformability cytometry (xDC). All three methods detect cell deformability changes induced by exposure to altered osmolarity. However, a dose-dependent deformability increase upon latrunculin B-induced actin disassembly was detected only with cDC and sDC, which suggests that when exposing cells to the higher strain rate imposed by xDC, cellular components other than the actin cytoskeleton dominate the response. The direct comparison presented here furthers our understanding of the applicability of the different deformability cytometry methods and provides context for the interpretation of deformability measurements performed using different platforms.
