Polydiacetylene-coated polyvinylidene fluoride strip aptasensor for colorimetric detection of zinc(II).

ABSTRACT: We report a new polydiacetylene (PDA) sensor strip for simple visual detection of zinc ions in aqueous solution. The specificity of this sensor comes from Zn²⁺ DNA aptamer probes conjugated onto PDA. Effects of aptamer length and structure on the sensitivity of PDA's color transition were first investigated. PDA conjugated with the optimal aptamer sequence was then coated onto a strip of polyvinylidene fluoride membrane and photopolymerized by UV exposure. The newly developed sensor successfully exhibited a blue-to-red chromatic change in a semi-quantitative manner in response to zinc ions. No discernable change was observed in solutions containing other common ions. Advantages of this sensor include its ease of fabrication, high specificity, and equipment-free detection, all of which are desirable for in-field applications and use in resource-limited settings.

Label-Free Colorimetric Detection of Influenza Antigen Based on an Antibody-Polydiacetylene Conjugate and Its Coated Polyvinylidene Difluoride Membrane.

Project description:This study presents an antibody-conjugated polydiacetylene (PDA) and its coated polyvinylidene difluoride (PVDF) membrane. The M149 antibody was hybridized to nano-vesicles consisting of pentacosa-10,12-diynoic acid (PCDA) and dimyristoylphosphatidylcholine (DMPC). After photo-polymerization at 254 nm, the effects on the PDA by antigenic injection were investigated with UV-vis spectroscopy, fluorescence spectroscopy, dynamic light scattering and transmission electron microscopy. Because PDA, an alternating ene-yne molecule, induces a blue-to-red color transition and an interesting fluorescent response by the distortion of its backbone, the biomolecular recognition of an antibody⁻antigen can be converted into an optical and fluorescent signal. Thus, an influenza antigen was successfully detected with the proposed label-free method. Furthermore, the vesicular system was improved by coating it onto a membrane type sensing platform for its stability and portability. The proposed antibody-PDA composite PVDF membrane has potential for rapid, easy and selective visualization of the influenza virus.

Electrically Stimulated Tunable Drug Delivery From Polypyrrole-Coated Polyvinylidene Fluoride.

Project description:Electrical stimulus-responsive drug delivery from conducting polymers such as polypyrrole (PPy) has been limited by lack of versatile polymerization techniques and limitations in drug-loading strategies. In the present study, we report an in-situ chemical polymerization technique for incorporation of biotin, as the doping agent, to establish electrosensitive drug release from PPy-coated substrates. Aligned electrospun polyvinylidene fluoride (PVDF) fibers were used as a substrate for the PPy-coating and basic fibroblast growth factor and nerve growth factor were the model growth factors demonstrated for potential applications in musculoskeletal tissue regeneration. It was observed that 18-h of continuous polymerization produced an optimal coating of PPy on the surface of the PVDF electrospun fibers with significantly increased hydrophilicity and no substantial changes observed in fiber orientation or individual fiber thickness. This PPy-PVDF system was used as the platform for loading the aforementioned growth factors, using streptavidin as the drug-complex carrier. The release profile of incorporated biotinylated growth factors exhibited electrosensitive release behavior while the PPy-PVDF complex proved stable for a period of 14 days and suitable as a stimulus responsive drug delivery depot. Critically, the growth factors retained bioactivity after release. In conclusion, the present study established a systematic methodology to prepare PPy coated systems with electrosensitive drug release capabilities which can potentially be used to encourage targeted tissue regeneration and other biomedical applications.

Improved Electrical Signal of Non-Poled 3D Printed Zinc Oxide-Polyvinylidene Fluoride Nanocomposites.

Project description:Polyvinylidene fluoride (PVDF) presents highly useful piezo and pyro electric properties but they are predicated upon the processing methods and the ensuing volume fraction of the β-phase. Production of PVDF with higher β-phase content for additive manufacturing (AM) is particularly desirable because it can enable the creation of custom parts with enhanced properties. Necessary steps from compounding to the testing of a 3D printed piezo sensitive sensor are presented in this paper. AM process variables and the influence of zinc oxide (ZnO) nanofiller on crystallinity, viscosity, and electromechanical properties of PVDF, have been explored. Fourier-transform infrared spectroscopy (FTIR) measurements confirm that a high cooling rate (HCR) of 30 °C min-1 promotes the conversion of the α-into the β-phase, reaching a maximum of 80% conversion with 7.5-12.5% ZnO content. These processing conditions increase the elastic modulus up to 40%, while maintaining the ultimate strength, ≈46 MPa. Furthermore, HCR 10% ZnO-PVDF produces four times higher volts per Newton when compared to low cooling rate, 5 °C min-1, pristine PVDF. A piezoelectric biomedical sensor application has been presented using HCR and ZnO nanofiller. This technique also reduces the need for post-poling which can reduce manufacturing time and cost.

Aptamer-Conjugated Polydiacetylene Colorimetric Paper Chip for the Detection of Bacillus thuringiensis Spores.

Project description:A colorimetric polydiacetylene (PDA) paper strip sensor that can specifically recognize Bacillus thuringiensis (BT) HD-73 spores is described in this work. The target-specific aptamer was combined with PDA, and the aptamer-conjugated PDA vesicles were then coated on polyvinylidene fluoride (PVDF) paper strips by a simple solvent evaporation method. The PDA-aptamer paper strips can be used to detect the target without any pre-treatment. Using the paper strip, the presence of BT spores is directly observable by the naked eye based on the unique blue-to-red color transition of the PDA. Quantitative studies using the paper strip were also carried out by analyzing the color transitions of the PDA. The specificity of this PDA sensor was verified with a high concentration of Escherichia coli, and no discernable change was observed. The observable color change in the paper strip occurs in less than 1 h, and the limit of detection is 3 × 107 CFU/mL, much below the level harmful to humans. The PDA-based paper sensor, developed in this work, does not require a separate power or detection device, making the sensor strip highly transportable and suitable for spore analysis anytime and anywhere. Moreover, this paper sensor platform is easily fabricated, can be adapted to other targets, is highly portable, and is highly specific for the detection of BT spores.

Quantitative Colorimetric Detection of Dissolved Ammonia Using Polydiacetylene Sensors Enabled by Machine Learning Classifiers.

Project description:Easy-to-use and on-site detection of dissolved ammonia are essential for managing aquatic ecosystems and aquaculture products since low levels of ammonia can cause serious health risks and harm aquatic life. This work demonstrates quantitative naked eye detection of dissolved ammonia based on polydiacetylene (PDA) sensors with machine learning classifiers. PDA vesicles were assembled from diacetylene monomers through a facile green chemical synthesis which exhibited a blue-to-red color transition upon exposure to dissolved ammonia and was detectable by the naked eye. The quantitative color change was studied by UV-vis spectroscopy, and it was found that the absorption peak at 640 nm gradually decreased, and the absorption peak at 540 nm increased with increasing ammonia concentration. The fabricated PDA sensor exhibited a detection limit of ammonia below 10 ppm with a response time of 20 min. Also, the PDA sensor could be stably operated for up to 60 days by storing in a refrigerator. Furthermore, the quantitative on-site monitoring of dissolved ammonia was investigated using colorimetric images with machine learning classifiers. Using a support vector machine for the machine learning model, the classification of ammonia concentration was possible with a high accuracy of 100 and 95.1% using color RGB images captured by a scanner and a smartphone, respectively. These results indicate that using the developed PDA sensor, a simple naked eye detection for dissolved ammonia is possible with higher accuracy and on-site detection enabled by the smartphone and machine learning processes.

Single-Layer and Double-Layer Filtration Materials Based on Polyvinylidene Fluoride-Co-hexafluoropropylene Nanofibers Coated on Melamine Microfibers.

Project description:In this work, we demonstrate selected optimization changes in the simple design of filtration masks to increase particle removal efficiency (PRE) and filter quality factor by combining experiments and numerical modeling. In particular, we focus on single-layer filters fabricated from uniform thickness fibers and double-layer filters consisting of a layer of highly permeable thick fibers as a support and a thin layer of filtering electrospun nanofibers. For single-layer filters, we demonstrate performance improvement in terms of the quality factor by optimizing the geometry of the composition. We show significantly better PRE performance for filters composed of micrometer-sized fibers covered by a thin layer of electrospun nanofibers. This work is motivated and carried out in collaboration with a targeted industrial development of selected melamine-based filter nano- and micromaterials.

Colorimetric Detection of Some Highly Hydrophobic Flavonoids Using Polydiacetylene Liposomes Containing Pentacosa-10,12-diynoyl Succinoglycan Monomers.

Project description:Flavonoids are a group of plant secondary metabolites including polyphenolic molecules, and they are well known for antioxidant, anti-allergic, anti-inflammatory and anti-viral propertied. In general, flavonoids are detected with various non-colorimetric detection methods such as column liquid chromatography, thin-layer chromatography, and electrochemical analysis. For the first time, we developed a straightforward colorimetric detection system allowing recognition of some highly hydrophobic flavonoids such as alpha-naphthoflavone and beta-naphthoflavone, visually using 10,12-pentacosadiynoic acid (PCDA) derivatized with succinoglycan monomers isolated from Sinorhizobium meliloti. Besides changes in visible spectrum, we also demonstrate fluorescence changes using our detection system in the presence of those flavonoids. The succinoglycan monomers attached to PCDA molecules may function as an unstructured molecular capturer for some highly hydrophobic flavonoids by hydrophobic interactions, and transmit their molecular interactions as a color change throughout the PCDA liposome.

Polyvinylidene fluoride/sulfonated graphene oxide blend membrane coated with polypyrrole/platinum electrode for ionic polymer metal composite actuator applications.

Project description:A polyvinylidene fluoride, sulfonated graphene oxide composite membrane coated with polypyrrole (Ppy) and platinum metal (Pt) was fabricated. The Fourier-transform infrared (FTIR) spectroscopic analysis was done to analyze the functional groups present in the composite material. Deposition of PPy/Pt electrode and surface morphology of PVDF/SGO/Pt/PPy was confirmed by scanning electron microscopic (SEM) images. The capacity of ion exchange and proton conductivity (PC) of PVDF/SGO/Pt/PPy were 1.4 meq g-1 of dry ion exchanger and 4.251 × 10-2 S cm-1, respectively. A two-link flexible manipulator based on the fabricated ionic polymer metal composite (IPMC) membranes was also developed where the electromechanical behaviour of a polymer-based actuator provides an important step in robotics applications.

Rapid and Efficient Removal of Anionic Dye in Water Using a Chitosan-Coated Iron Oxide-Immobilized Polyvinylidene Fluoride Membrane.

Project description:Anionic dyes are one of the most serious contaminants in water as these molecules are known to be toxic to many living organisms. Herein, we report the development of functionalized polyvinylidene fluoride membranes modified with chitosan-coated iron oxide nanomaterials (Fe-PVDF) for the efficient treatment of anionic dye-contaminated water. Aqueous solutions of anionic dyes could be captured rapidly by passing through the functionalized membrane under reduced pressure. Under neutral conditions, Fe-PVDF showed a maximum removal capacity of 74.6 mg/g for Evans blue (EB) through the adsorption process. In addition, the adsorption capacity was significantly enhanced up to 434.78 mg/g under acidic conditions. The adsorption process for EB matched well with the Langmuir model, indicating monolayer adsorption of the dye to the membrane surface. Moreover, Fe-PVDF can be reusable by a simple washing step in an alkaline solution, and thus, the composite membrane was applied several times without a significant decrease in its adsorption performance. The same composite membrane was further applied to the removal of five other different anionic dyes with high efficiencies. The adsorption mechanism can be explained by the electrostatic interaction between the positively charged chitosan and the negatively charged dye as well as the affinity of the sulfate groups in dye molecules for the surface of the iron oxide nanoparticles. The easy preparation and rapid decolorization procedures make this composite membrane suitable for efficient water treatment.

Colorimetric Detection of the SARS-CoV-2 Virus (COVID-19) in Artificial Saliva Using Polydiacetylene Paper Strips.

Project description:The spread and resurgence of the SARS-CoV-2 virus (COVID-19 disease) threatens human health and social relations. Prevention of COVID-19 disease partly relies on fabricating low-cost, point-of-care (POC) sensing technology that can rapidly and selectively detect the SARS-CoV-2 virus. We report a colorimetric, paper-based polydiacetylene (PDA) biosensor, designed to detect SARS-CoV-2 spike protein in artificial saliva. Analytical characterizations of the PDA sensor using NMR and FT-IR spectroscopy showed the correct structural elucidation of PCDA-NHS conjugation. The PDA sensor platform containing the N-Hydroxysuccinimide ester of 10, 12-pentacosadiynoic acid (PCDA-NHS) was divided into three experimental PCDA-NHS concentration groups of 10%, 20%, and 30% to optimize the performance of the sensor. The optimal PCDA-NHS molar concentration was determined to be 10%. The PDA sensor works by a color change from blue to red as its colorimetric output when the immobilized antibody binds to the SARS-CoV-2 spike protein in saliva samples. Our results showed that the PDA sensing platform was able to rapidly and qualitatively detect the SARS-CoV-2 spike protein within the concentration range of 1 to 100 ng/mL after four hours of incubation. Further investigation of pH and temperature showed minimal influence on the PDA sensor for the detection of COVID-19 disease. After exposure to the SARS-CoV-2 spike protein, smartphone images of the PDA sensor were used to assess the sensor output by using the red chromatic shift (RCS) of the signal response. These results indicate the potential and practical use of this PDA sensor design for the rapid, colorimetric detection of COVID-19 disease in developing countries with limited access to medical testing.
