Efficient one-pot synthesis of 1-arylcycloprop-2-ene-1-carboxamides.

ABSTRACT: An expeditious and cost-efficient method for synthesis of 1-arylcycloprop-2-ene-1-carboxamides was developed. This one-pot protocol involving coupling of amines with acyl chlorides, generated upon treatment of cyclopenylcarboxylic acids with oxalyl chloride, is applicable for the preparation of sensitive products with a reactive, unsubstituted strained double bond.

The protocol employs piperidine-iodine as a dual system catalyst and ethanol, a green solvent. The main advantages of this approach are that it is a metal-free and clean reaction, has low catalyst loading, and requires no tedious workup.

Pot economy and one-pot synthesis.

Project description:The one-pot synthesis of a target molecule in the same reaction vessel is widely considered to be an efficient approach in synthetic organic chemistry. In this review, the characteristics and limitations of various one-pot syntheses of biologically active molecules are explained, primarily involving organocatalytic methods as key tactics. Besides catalysis, the pot-economy concepts presented herein are also applicable to organometallic and organic reaction methods in general.

Cyanoacetamides (IV): versatile one-pot route to 2-quinoline-3-carboxamides.

Project description:Cyanoacetic acid derivatives are the starting materials for a plethora of multicomponent reaction (MCR) scaffolds. Herein, we describe scope of a valuable general protocol for the synthesis of arrays of 2-aminoquinoline-3-carboxamides from cyanoacetamides and 2-aminobenzaldehydes or heterocyclic derivatives via a Friedländer reaction variation. In many cases, the reactions involve a very convenient work up by simple precipitation and filtration. More than 40 new products are described. We foresee our protocol and the resulting derivatives becoming very valuable to greatly expanding the scaffold space of cyanoacetamide derivatives.

Efficient One-Pot, Two-Component Modular Synthesis of 3,5-Disubstituted Pyrazoles.

Project description:The pyrazole scaffold is one of the most prevalent and important tool in medicinal chemistry. Here, we report a method for preparing 3,5-diarylpyrazoles in good to excellent yield by reacting hydrazones of aryl aldehydes with substituted acetophenones in ethanol in the presence of dimethyl sulfoxide/cat. I2/cat. HCl. The reverse process, reacting hydrazones of substituted acetophenones with aryl aldehydes under the same conditions, also provides 3,5-diarylpyrazoles in good to excellent yields. Reaction of hydrazones of aldehydes with 2'-aryloxy ketones in the presence of cat. HCl in ethanol and the catalyst-free reaction of phenacyl bromides with hydrazones of aldehydes in ethanol also gave good to excellent yields of 3,5-diarylpyrazoles.

Silica: An efficient catalyst for one-pot regioselective synthesis of dithioethers.

Project description:The development of a silica-promoted highly selective synthesis of 1,2 or 1,3-dithioethers via solvent-free one-pot tandem reactions of an allyl bromide with excess thiol at room temperature is described. The choice of silica gel, either pre-calcined or moistened with water, exhibited notable regioselectivity in the formation of dithioethers. Plausible mechanistic routes were explored and postulated.

One Pot Synthesis of Copper Oxide Nanoparticles for Efficient Antibacterial Activity

Project description:The unique semiconductor and optical properties of copper oxides have attracted researchers for decades. However, using fruit waste materials such as peels to synthesize the nanoparticles of copper oxide (CuO NPs) has been rarely described in literature reviews. The main purpose of this part of the research was to report on the CuO NPs with the help of apple peel extract under microwave irradiation. Metal salts and extracts were irradiated at 540 W for 5 min in a microwave in a 1:2 ratio. The crystallinity of the NPs was confirmed by the XRD patterns and the crystallite size of the NPs was found to be 41.6 nm. Elemental mapping of NPs showed homogeneous distributions of Cu and O. The NPs were found to contain Cu and O by EDX and XPS analysis. In a test involving two human pathogenic microbes, NPs showed antibacterial activity and the results revealed that the zone of inhibition grew significantly with respect to the concentration of CuO NPs. In a biofilm, more specifically, NPs at 25.0 µg/mL reduced mean thickness and biomass values of S. aureus and E. coli biofilms by >85.0 and 65.0%, respectively, with respect to untreated controls. In addition, environmentally benign materials offer a number of benefits for pharmaceuticals and other biomedical applications as they are eco-friendly and compatible.

Easy, efficient and versatile one-pot synthesis of Janus-type-substituted fullerenols.

Project description:An efficient one-pot synthesis for Janus-type fullerenol derivatives and how to characterize them is reported. This synthesis provides access to asymmetrically substituted fullerenol with five substituents on one pole of the fullerene and polyhydroxylation moieties, mostly ether and hydroxy groups, on the rest of the fullerene core. As substituents a broad variety of primary amines can be used to obtain Janus-type amphiphilic fullerenols in good to excellent yield. These fullerenol amphiphiles can serve as suitable precursors for further reactions resulting in new applications for fullerenols.

Efficient One-Pot Synthesis of Novel Caffeic Acid Derivatives as Potential Antimalarials.

Project description:New protocol for the preparation of the novel caffeic acid derivatives using the Wittig reaction has been applied to follow the principles of green chemistry. The compounds have been evaluated against chloroquine-sensitive and chloroquine-resistant P. falciparum strains. Their cytotoxicity to normal human dermal fibroblasts and their propensity to induce hemolysis have been also determined. Ethyl (2E)-3-(2,3,4-trihydroxyphenyl)-2-methylpropenoate has exhibited the highest antiplasmodial activity against P. falciparum strains without the cytotoxic and hemolytic effects. This derivative is significantly more potent than caffeic acid parent structure. The application of our one-step procedure has been shown to be rapid and efficient. It allows for an easy increase of input data to refine the structure-activity relationship model of caffeates as the antimalarials. The one-step approach meets the conditions of "atom economy" and eliminates hazardous materials. Water has been used as the effective medium for the Wittig reaction to avoid toxic organic solvents.

One-Pot Synthesis of Cellulose/MXene/PVA Foam for Efficient Methylene Blue Removal.

Project description:Ti3C2Tx MXene has attracted considerable interest as a new emerging two-dimensional material for environmental remediation due to its high adsorption capacity. However, its use is greatly limited by its poor mechanical properties, low processability and recyclability, and the low dispersity of such powder materials. In this work, a porous adsorbent (C-CMP) containing cellulose nanocrystals (CNC), Ti3C2Tx MXene and polyvinyl alcohol (PVA) was prepared by a simple and environmentally-friendly foaming method. Glutaraldehyde was used as crosslinker to improve the mechanical properties and boost the adsorption efficiency of methylene blue (MB) molecules. Fourier transform infrared (FT-IR), elemental analysis (EDX) and thermogravimetric analysis (TGA) further confirmed that the preparation of the C-CMP foam and cross-linking reaction were successful. Scanning electron microscope (SEM) indicated that the macropores were distributed homogeneously. The adsorption experiment showed that maximum adsorption capacity of MB can reach 239.92 mg·g-1 which was much higher than anionic dye (methyl orange, 45.25 mg·g-1). The adsorption behavior fitted well with the Langmuir isotherm and pseudo-second-order kinetic models. Thermodynamic analysis indicated that the adsorption process was spontaneous and endothermic. Based on FT-IR, EDX and X-ray photoelectron spectroscopy (XPS) analysis, the adsorption mechanism between C-CMP and MB molecules was attributed to electrostatic interaction.

Project description:(-)-Ambrox is recognised as the prototype of all ambergris odorants. Widely used in perfumery, (-)-Ambrox is an important ingredient due to its unique scent and excellent fixative function. An environmentally friendly and practical preparation of (-)-Ambrox is still unavailable at present although a lot of attention has been paid to this hot research topic for many years. A one-pot synthesis of (-)-Ambrox was studied starting from (-)-sclareol through oxidation with hydrogen peroxide in the presence of a quaternary ammonium phosphomolybdate catalyst {[C5H5NC16H33] [H2PMo12O40]}, which gave the product a 20% overall yield.
