Project description:Bacteriophage O1 is a Myoviridae A1 group member used historically for identifying Salmonella. Sequencing revealed a single, linear, 86,155-base-pair genome with 39% average G+C content, 131 open reading frames, and 22 tRNAs. Closest protein homologs occur in Erwinia amylovora phage ?Ea21-4 and Escherichia coli phage wV8. Proteomic analysis indentified structural proteins: Gp23, Gp36 (major tail protein), Gp49, Gp53, Gp54, Gp55, Gp57, Gp58 (major capsid protein), Gp59, Gp63, Gp64, Gp67, Gp68, Gp69, Gp73, Gp74 and Gp77 (tail fiber). Based on phage-host codon differences, 7 tRNAs could affect translation rate during infection. Introns, holin-lysin cassettes, bacterial toxin homologs and host RNA polymerase-modifying genes were absent.
Project description:IntroductionMany colleges and medical schools are incorporating boot camps and transition programs for students prior to their first year of medical school. In these prematriculation programs, students are given early introductions to teaching modalities and study strategies. This resource provides a realistic case scenario that introduces students to the basic case format while guiding them through the thought process needed to work through a case. Additionally, it provides students with small-group skills that will be beneficial when working through cases in the first and second years of medical school.MethodsThe case features Mary Beth Chrispy, a 25-year-old second-year medical student who visits her primary care physician with complaints of fatigue and difficulty with focusing on her studies. Presenting questions throughout the case help guide students in making necessary connections and allow the facilitator to lead the students through each section of the case. There is no prereading to be done before the first meeting of the case. Students need to be willing to participate in the motions of the case so that they are prepared for their actual casework in the first year of medical school.ResultsSix questions pertaining to the case were placed on a self-assessment, and the students' ability to answer the questions, based on their work throughout the week, was measured. On five out of six questions, 94.4% of students (N = 19) answered correctly. On the remaining question, 100% of the students were able to answer correctly. We concluded that the activity met our learning objectives.DiscussionInstitutions that would like to include this case in their own prematriculation programs may want to utilize a standardized patient during its introductory clinical encounter.
Project description:Post-excavation iron corrosion may be accelerated by the presence of Cl- , leading to conservation methods designed to remove Cl. This study exploits a unique opportunity to assess 35 years of conservation applied to cast-iron cannon shot excavated from the Mary Rose. A combination of synchrotron X-ray powder diffraction (SXPD), absorption spectroscopy (XAS), and fluorescence (XRF) mapping have been used to characterise the impact of conservation on the crystalline corrosion products, chlorine distribution, and speciation. The chlorinated phase akaganeite, β-FeO(OH,Cl), was found on shot washed in corrosion inhibitor Hostacor IT with or without an additional reduction stage. No chlorinated phases were observed on the surface of shot stored in sodium sesquicarbonate (Na2 CO3 /NaHCO3 ); however, hibbingite, β-Fe2 (OH)3 Cl, was present in metal pores. It is proposed that surface β-FeO(OH,Cl) formed in the early stages of active conservation owing to oxidation of β-Fe2 (OH)3 Cl at near-neutral pH.
Project description:Josef Jadassohn (1863-1936) and his assistant, Felix Lewandowsky (1879-1921), were eminent German dermatologists who had several dermatology conditions linked eponymously. One such condition is Jadassohn-Lewandowsky syndrome, which is a type of pachyonychia congenita - a disease that is characterized by severe thickening of the nail due to massive nail hyperkeratosis. This report describes Jadassohn-Lewandowsky syndrome and the men behind this disease.
Project description:Sister Mary Joseph Nodule (SMJN) is a metastatic umbilical lesion secondary to a primary malignancy of any viscera, stomach and colon being most common in men, and ovary in women.In this article, we present the case of SMJN in a 54-year old female patient. An urgent diagnostic workup was performed with a computerized tomography of abdominal cavity and pelvis showing an expansive tumorous formation covering uterus with a carcinomatosis of peritoneum. After biopsy, immunohistochemical profile suggested adenocarcinoma of the ovarian origin. The patient was then referred to the Oncology Consilier of Gynecology Department and further continued followed by the Oncology team.
Project description:Rickettsial diseases have long been diagnosed with serum antibodies cross-reactive against Proteus vulgaris (Weil-Felix reaction). Although Weil-Felix antibodies are associated with the development of immunity, their rickettsial target and contribution to disease pathogenesis are not established. Here, we developed a transposon for insertional mutagenesis of Rickettsia conorii, isolating variants defective for replication in cultured cells and in spotted fever pathogenesis. Mutations in the polysaccharide synthesis operon (pso) abolish lipopolysaccharide O-antigen synthesis and Weil-Felix serology and alter outer-membrane protein assembly. Unlike wild-type R. conorii, pso mutants cannot elicit bactericidal antibodies that bind O antigen. The pso operon is conserved among rickettsial pathogens, suggesting that bactericidal antibodies targeting O antigen may generate universal immunity that could be exploited to develop vaccines against rickettsial diseases.
