Janus particles for biological imaging and sensing.

ABSTRACT: Janus particles, named after the two-faced Roman god Janus, have different surface makeups, structures or compartments on two sides. This review highlights recent advances in employing Janus particles as novel analytical tools for live cell imaging and biosensing. Unlike conventional particles used in analytical science, two-faced Janus particles provide asymmetry and directionality, and can combine different or even incompatible properties within a single particle. The broken symmetry enables imaging and quantification of rotational dynamics, revealing information beyond what traditional measurements offer. The spatial segregation of molecules on the surface of a single particle also allows analytical functions that would otherwise interfere with each other to be decoupled, opening up opportunities for novel multimodal analytical methods. We summarize here the development of Janus particles, a few general methods for their fabrication and, more importantly, the emerging and novel applications of Janus particles as multi-functional imaging probes and sensors.

Amphiphilic Janus Particles Confined in Symmetrical and Janus-Like Slits.

Project description:We use Monte Carlo simulations to investigate the behavior of Janus spheres composed of attractive and repulsive parts confined between two parallel solid surfaces. The slits with identical and competing walls are studied. The adsorption isotherms of Janus particles are determined, and the impact of the density in the pore on the morphology is discussed in detail. So far, this issue has not been systematically investigated. New, unique structures are observed along the isotherms, including the bilayer and three-layer structures located at different distances from the walls. We analyze how selected parameters affect the positional and orientational ordering in these layers. In some cases, the particles form highly ordered hexagonal lattices.

An Optically Controlled Microscale Elevator Using Plasmonic Janus Particles.

Project description:In this article, we report how Janus particles, composed of a silica sphere with a gold half-shell, can be not only stably trapped by optical tweezers but also displaced controllably along the axis of the laser beam through a complex interplay between optical and thermal forces. Scattering forces orient the asymmetric particle, while strong absorption on the metal side induces a thermal gradient, resulting in particle motion. An increase in the laser power leads to an upward motion of the particle, while a decrease leads to a downward motion. We study this reversible axial displacement, including a hysteretic jump in the particle position that is a result of the complex pattern of a tightly focused laser beam structure above the focal plane. As a first application we simultaneously trap a spherical gold nanoparticle and show that we can control the distance between the two particles inside the trap. This photonic micron-scale "elevator" is a promising tool for thermal force studies, remote sensing, and optical and thermal micromanipulation experiments.

Substrate wettability guided oriented self assembly of Janus particles.

Project description:Self-assembly of Janus particles with spatial inhomogeneous properties is of fundamental importance in diverse areas of sciences and has been extensively observed as a favorably functionalized fluidic interface or in a dilute solution. Interestingly, the unique and non-trivial role of surface wettability on oriented self-assembly of Janus particles has remained largely unexplored. Here, the exclusive role of substrate wettability in directing the orientation of amphiphilic metal-polymer Bifacial spherical Janus particles, obtained by topo-selective metal deposition on colloidal Polymestyere (PS) particles, is explored by drop casting a dilute dispersion of the Janus colloids. While all particles orient with their polymeric (hydrophobic) and metallic (hydrophilic) sides facing upwards on hydrophilic and hydrophobic substrates respectively, they exhibit random orientation on a neutral substrate. The substrate wettability guided orientation of the Janus particles is captured using molecular dynamic simulation, which highlights that the arrangement of water molecules and their local densities near the substrate guide the specific orientation. Finally, it is shown that by spin coating it becomes possible to create a hexagonal close-packed array of the Janus colloids with specific orientation on differential wettability substrates. The results reported here open up new possibilities of substrate-wettability driven functional coatings of Janus particles, which has hitherto remained unexplored.

Preparation of Cylindrical Janus Particles Using a Stirring Method.

Project description:We previously discovered a novel method for the preparation of polymer particles that have a cylindrical shape. Polystyrene (PS) or poly methyl methacrylate (PMMA) spherical particles were deformed into a cylindrical shape by stirring with a magnetic stirrer in a polyvinylpyrrolidone (PVP) aqueous solution. In this study, cylindrical "Janus" particles consisting of PS and PMMA were prepared by this stirring method. In the case of spherical Janus particles, cylindrical particles were obtained after stirring; however, the direction of the interface between the PS and PMMA phases was random. However, in the case of snowman-like Janus particles, cylindrical Janus particles with the interface at the center of the long axis were successfully prepared. This indicated that the extension direction can be controlled owing to the anisotropic shape and supported the proposed deformation mechanism of the cylindrical particles. Moreover, amphiphilic cylindrical Janus particles were also successfully prepared by hydrolysis of only one phase to introduce carboxy groups.

Remote Control of T Cell Activation Using Magnetic Janus Particles.

Project description:We report a strategy for using magnetic Janus microparticles to control the stimulation of T cell signaling with single-cell precision. To achieve this, we designed Janus particles that are magnetically responsive on one hemisphere and stimulatory to T cells on the other side. By manipulating the rotation and locomotion of Janus particles under an external magnetic field, we could control the orientation of the particle-cell recognition and thereby the initiation of T cell activation. This study demonstrates a step towards employing anisotropic material properties of Janus particles to control single-cell activities without the need of complex magnetic manipulation devices.

Biofunctional Janus particles promote phagocytosis of tumor cells by macrophages.

Project description:Herein, a versatile strategy for the construction of biofunctional Janus particles (JPs) through the combination of Pickering emulsion and copper-free click chemistry is developed for the study of particle-mediated cell-cell interactions. A variety of biomolecules including bovine serum albumin (BSA), ferritin, transferrin (Tf), and anti-signal regulatory protein alpha antibodies (aSIRPα), etc., can be incorporated into the Janus platform in a spatially defined manner. JPs consisting of Tf and aSIRPα (Tf-SPA1-aSIRPα JPs) demonstrate a significantly improved binding affinity to either macrophages or tumor cells compared to their uniformly modified counterparts. More importantly, Tf-SPA1-aSIRPα JPs mediate more efficient phagocytosis of tumor cells by macrophages as revealed by real-time high-content confocal microscopy. This study demonstrates the potential advantages of JPs in mediating cell-cell interactions and may contribute to the emerging cancer immunotherapy.

Biological Deep Temperature Imaging with Fluorescence Lifetime of Rare-Earth-Doped Ceramics Particles in the Second NIR Biological Window.

Project description:Contactless thermal imaging generally relies on mid-infrared cameras and fluorescence imaging with temperature-sensitive phosphors. Fluorescent thermometry in the near-infrared (NIR) region is an emerging technique for analysing deep biological tissues but still requires observation depth calibration. We present an NIR fluorescence time-gated imaging (TGI) thermometry technology based on fluorescence lifetime, an intrinsic fluorophore time constant unrelated to observation depth. Fluorophore used is NaYF4 co-doped with Nd3+ and Yb3+ that emits fluorescence at 1000?nm. An agarose gel-based phantom with the fluorophore embedded at a 5-mm depth was covered by sheets of meat to vary the observation depth. The temperature was determined independently from depth by sequences of NIR fluorescence decay images, and the rate of change in the fluorescence lifetime per temperature was almost constant (-0.0092 ~ -0.010?°C-1) at depths ranging from 0 to 1.4?mm of meat, providing non-contact and absolute measurements of temperature in deep biological tissues.

Janus particles as artificial antigen-presenting cells for T cell activation.

Project description:Here we show that the multifunctionality of Janus particles can be exploited for in vitro T cell activation. We engineer bifunctional Janus particles on which the spatial distribution of two ligands, anti-CD3 and fibronectin, mimics the "bull's eye" protein pattern formed in the membrane junction between a T cell and an antigen-presenting cell. Different levels of T cell activation can be achieved by simply switching the spatial distribution of the two ligands on the surfaces of the "bull's eye" particles. We find that the ligand pattern also affects clustering of intracellular proteins. This study demonstrates that anisotropic particles, such as Janus particles, can be developed as artificial antigen-presenting cells for modulating T cell activation.

A general strategy to synthesize chemically and topologically anisotropic Janus particles.

Project description:Emulsion polymerization is the most widely used synthetic technique for fabricating polymeric particles. The interfacial tension generated with this technique limits the ability to tune the topology and chemistry of the resultant particles. We demonstrate a general emulsion interfacial polymerization approach that involves introduction of additional anchoring molecules surrounding the microdroplets to synthesize a large variety of Janus particles with controllable topological and chemical anisotropy. This strategy is based on interfacial polymerization mediated by an anchoring effect at the interface of microdroplets. Along the interface of the microdroplets, the diverse topology and surface chemistry features of the Janus particles can be precisely tuned by regulating the monomer type and concentration as well as polymerization time. This method is applicable to a wide variety of monomers, including positively charged, neutrally charged, and negatively charged monomers, thereby enriching the community of Janus particles.

Arrested-motility states in populations of shape-anisotropic active Janus particles.

Project description:The emergence of large-scale collective phenomena from simple interactions between individual units is a hallmark of active matter systems. Active colloids with alignment-dominated interparticle interactions tend to develop orientational order and form motile coherent states, such as flocks and swarms. Alternatively, a combination of self-propulsion and excluded-volume interactions results in self-trapping and active phase separation into dense clusters. Here, we reveal unconventional arrested-motility states in ensembles of active discoidal particles powered by induced-charge electrophoresis. Combining experiments and computational modeling, we demonstrate that the shape asymmetry of the particles promotes the hydrodynamically assisted formation of active particles' bound states in a certain range of excitation parameters, ultimately leading to a spontaneous collective state with arrested motility. Unlike the jammed clusters obtained through self-trapping, the arrested-motility phase remains sparse, dynamic, and reconfigurable. The demonstrated mechanism of phase separation seeded by bound state formation in ensembles of oblate active particles is generic and should be applicable to other active colloidal systems.
